RESUMO

OBJECTIVE To investigate the effects and mechanism of luteolin on osteogenic repair of bone defects. METHODS The targets and potential pathways of luteolin in the treatment of bone defects were screened by network pharmacology method, and then the top 2 targets were selected by Hub gene screening for molecular docking verification, with binding energy as the evaluation standard. In vitro experiments were conducted on rat bone mesenchymal stem cells (BMSC) and rat umbilical vein endothelial cells (RUVEC). Phenotypic validation was performed using alkaline phosphatase staining, alizarin red S staining, and in vitro angiogenesis experiments. Western blot assay was used to detect the protein expressions of phosphatidylinositol 3 kinase (PI3K) and protein kinase 1 (Akt1), so as to validate the mechanism of luteolin on osteogenic differentiation of BMSC and angiogenesis of RUVEC in vitro. RESULTS The results of network pharmacology showed that the effects of luteolin on vascular formation and bone repair in bone defects were mainly related to Akt1, SRC, estrogen receptor 1, epidermal growth factor receptor, cyclooxygenase 2, matrix metalloproteinase 9 targets, and were closely related to PI3K-Akt signaling pathway. The results of molecular docking showed that luteolin binding to Akt1 and SRC proteins was stable. The results of in vitro experiments showed that luteolin could significantly improve the expressions and activities of alkaline phosphatase in BMSC, increased the number of calcium salt deposits and calcified nodules, and promoted calcification of BMSC. Compared with luteolin 0 μmol/L group, the angiogenesis ability of RUVEC was enhanced significantly in luteolin 1, 10 μmol/L groups, the length of blood vessels and the protein expressions of PI3K and Akt1 were significantly increased (P<0.05 or P<0.01); the higherthe concentration, the better the effect. CONCLUSIONS Luteolin may play a role in promoting angiogenesis and bone repair at the fracture site by activating PI3K/Akt signaling pathway and promoting the protein expressions of PI3K and Akt1.

RESUMO

Objective:To study the active components and their potential mechanism of Yanghe Decoction for the treatment of chronic osteomyelitis (CO) via the methods of network pharmacology and molecular docking. Methods:Active components and action targets of Yanghe Decoction were screened from TCMSP, BATMAN-TCM and relevant literature. GeneCards, OMIM, DisGeNET, and PharmGKB databases were used to predict the targets for the CO. Cytoscape 3.8.0 software and STRING database were used to build the networks of "Chinese materia medica-active components-potential targets" and "protein-protein interaction", and according to topological parameters in the network, the core active components as well as Hub genes were screened. MCODE plug was used to accomplish clustering analysis of protein modules in PPI network. Then, intersection targets were enriched and analyzed by GO and KEGG in KOBAS database. Finally, molecular docking was carried out with the help of Autodock tool platform to predict the binding ability between the main active components and key targets. Results:A total of 120 active components of Yanghe Decoction and 402 targets were obtained; 1 464 CO-related targets were screened, and there were 103 intersection target genes of Yanghe Decoction-CO, 110 active components related to intersection targets, which mainly contained some flavonoids and Phytosterols, such as quercetin, Kaempferol, and Beta-Sitosterol. There were 9 Hub genes, including TNF, IL6, AKT1, etc., and 4 protein modules which involved the regulation of immune inflammatory response, vascular microcirculation, bone development, and formation, material synthesis and metabolism and other physiological processes. 193 signaling pathways and 1 552 GO results were acquired in KOBAS database. Molecular docking results showed that the active compounds had good binding activity with key targets based on the minimum binding energy of less than - 5 kcal/mol.Conclusion:The mechanism in the treatment of CO with Yanghe Decoction is a complex process of multiple components, multiple targets, and multiple pathways. It mainly regulates targets such as TNF, IL-6, CXCL8, VEGFA, and AKT1 through pathways such as TNF signaling pathway, IL-17 signaling pathway, and Toll-like receptors, participating in local inflammatory reactions, microcirculation, and bone cell metabolism in chronic osteomyelitis, and interfering with the immune escape mechanism of pathogenic bacteria.

RESUMO

Objectives To study the knowledge, attitude and practice (KAP) status of hospice care among probationer nurses in Xi'an and analyze their influence factors with the purpose of providing reference for the education of hospice care in medical colleges and hospitals. Methods By a convenience sampling method, the probationer nurses from four medical colleges were investigated with the hospice care KAP questionnaire in January 2018.And the data were processed by SPSS 18.0 for descriptive analysis,t test, variance analysis and correlation analysis. Results A total of 1 534 questionnaires were issued,1 523 taken back, and 1 467 valid, with an effective return-ratio as 95.63%.The knowledge score, attitude score and practice score were (12.31 ± 3.44),(129.25 ± 9.97) and (23.15 ± 3.96) with the accuracy ratio as 61.5%,68.0%,and 77.2% respectively. The knowledge was correlated significantly with education, nation, religion, domicile and learning experience (P<0.05),the attitude correlated significantly with gender, education, nation, religion, domicile and learning experience (P<0.05),and the practice correlated significantly with gender, education, hospital level, religion, monthly family income per person, and learning experience (P<0.05).There were positive correlations between the knowledge and the attitude (r=0.39,P<0.01),the knowledge and the practice (r=0.25,P<0.01),and the attitude and the practice (r=0.49,P<0.01).Conclusion The probationer nurses lack the knowledge of hospice care and the awareness of it is weak.Consequently, the medical colleges and hospitals should strengthen the education to help the probationer nurses develop effective hospice care behaviors.

RESUMO

Sufficient embryos are needed for the preservation of Beagle dogs germplasm resources and the prepara-tion of gene?modified human disease animal models. Up to now, the induced ovulation technique has no effect on dogs,it is hard to obtain mature oocytes in vivo, although the scientists try a lot in many aspects, but still could not make a break?through. The in vitro maturation rate is too low to support the preservation of germplasm resources, application in gene?modified disease models and biomedical research. Aiming to provide useful information on breakthrough in dog oocytes mat?uration, this review will summarize the effect of different age and reproductive stage, different morphology and size of the oo?cytes and lipid droplet on the in vitro maturation of dog oocytes.

RESUMO

Objective To investigate the diagnostic value of determination of serum carcinoembryonic antigen (CEA) levels for primary liver carcinoma and metastatic liver cancer. Methods Serum CEA levels in 365 patients with liver cancer (273 with HCC and 92 with metastatic liver cancer accordingly) and 33 controls were determined with ELISA. Data were analysed with SPSS.Results Serum CEA levels were (7.52?18.04)?g/L in patients with liver cancer and (0.99?0.79)?g/L in controls. Taking 2.5 ?g/L as the cut-off level, the positive rate was 29.86% and 6.06% respectively. The values were significantly higher than those in controls (P
