RESUMO

Objective To exploxe the analgesic effect of general anesthesia combined with different concentrations of ropivacaine and transverse abdominis plane block on laparoscopic appendectomy(LA).Methods LA patients from Xiamen University Affiliated Zhongshan Hospital from January 2022 to July 2023 were included.All patients underwent general anesthesia combined ropivacaine plane block of the transverse abdominis muscle.According to the different concentrations of ropivacaine,patients were divided into high concentration group(0.50%,20 mL),medium concentration group(0.375%,20 mL)and low concentration group(0.25%,20 mL).The dosage of fentanyl,postoperative bleeding,time of first press of the analgesic pump,and number of presses among three groups of patients were recorded and compared.The postoperative vital signs(mean arterial pressure and heart rate),pain level(NRS),comfort level(BCS)(1 h,6 h,12 h,and 24 h),and incidence of postoperative adverse reactions were observed and compared.Results This study included a total of 90 LA patients,with 30 cases in each group.The first press of the analgesic pump in the low concentration group was significantly shorter than that in the medium and high concentration groups(P<0.05),while the number of presses was significantly higher than the other two groups(P<0.05).The mean arterial pressure and heart rate significantly decreased with the increase of ropivacaine concentration at 1 hour after surgery(P<0.05);compared with 1 hour after surgery,the average arterial pressure and heart rate of patients in the medium concentration group and low concentration group significantly increased at 6 hours after surgery(P<0.05),and no significant changes occurred thereafter.The NRS scores of patients in the high concentration group were significantly lower than those in the medium and low concentration groups at all four postoperative time points(P<0.05);the NRS scores of patients in the medium concentration group were significantly lower than those in the low concentration group at four postoperative time points(P<0.05).The BCS scores of the low concentration higher than those of the medium concentration group and low concentration group an 1 h,6 h and 24 h.The incidence of adverse reactions in the high concentration group was significantly higher than that in the low concentration group and the medium concentration group(P<0.05);there was no significant difference in the incidence of adverse reactions between the medium concentration group and the low concentration group(P>0.05).Conclusion General anesthesia combined with 0.50%ropivacaine abdominal transverse muscle plane block has a significant analgesic effect and high comfort for patients undergoing LA,but the risk of postoperative adverse reactions is high,and the plan needs to be adjusted according to the actual situation.

Aim To research the effect of gender on immune function and keratinocyte damage in imiquimod(IMQ)induced psoriasis mice.Methods IMQ-induced psoriasis mice were freely divided into female and male model groups, and female and male normal groups were set up smeared with an equal amount of petroleum.PASI score and HE staining were used to evaluate skin lesion and pathology; Western blot and immunity fluorescence were used to detect the expression of proliferating cell nuclear antigen(PCNA), Ki67, keratin 1(K1), keratin 10(K10), and involucrin in skin lesions; spleen index of mice was calculated; immunohistochemistry was used to detect the expression of CD4, IFN-γ, IL-4, and IL-17 in skin and spleen; flow cytometry was used to detect the changes of Th1 and Th17 cell subsets in spleen.Results Compared with female and male normal groups, PASI score of female and male model groups increased, skin lesions were abnormally thickened and differentiated, the level of PCNA and Ki67, the spleen index, the Th1 and Th17 cell subsets in the spleen both increased, K1, K10, and involucrin decreased, the levels of CD4, IFN-γ, IL-17 in skin lesions and spleen were elevated, but the level of IL-4 showed the opposite trend.There was no statistical difference in the above indicators between the female and male model groups.Conclusion Gender has no effect on the abnormal activation of T cell immune function and the proliferation and differentiation of keratinocytes in psoriasis-like mice induced by IMQ.

<p><b>OBJECTIVE</b>To investigate the correlation between Young's modulus of the thyroid tissue measured by in shear wave elastography (SWE) and the clinical manifestations of Hashimoto's thyroiditis in different stages.</p><p><b>METHODS</b>A total of 104 patients with the clinical diagnosis of Hashimoto's thyroiditis were enrolled in this study, including 26 with hyperthyroidism, 29 with normal thyroid function, 27 with subclinical hypothyroidism, 22 with clinical hypothyroidism, with 50 healthy volunteers serving as the healthy control group. All the subjects underwent SWE to obtain the Young's modulus value of the thyroid tissue. Univariate analysis of variance was used to compare the Young's modulus among the groups, and Pearson correlation analysis was used to analyze the correlation between the Young's modulus of the thyroid tissue and serum levels of thyroid microsomal antibody (TMAb) and thyroid globulin antibody (TGAb).</p><p><b>RESULTS</b>In the 4 groups of patients, the Young's modulus increased significantly in the order of hyperthyroidism group, normal thyroid function group, subclinical hypothyroidism group and clinical hypothyroidism group (F=60.983, P<0.01). The Young's modulus was significantly lower in hyperthyroidism group than in the other 3 groups (P<0.05), and was significantly lower in normal thyroid function group than in subclinical hypothyroidism group and clinical hypothyroidism group (P<0.05).</p><p><b>CONCLUSION</b>The Young's modulus of the thyroid tissue measured by shear wave elastography is related with the clinical manifestations of Hashimoto's thyroiditis in different stages, but the relevance needs to be further confirmed by multi-center, randomized, controlled studies involving a larger sample size.</p>

BACKGROUND: The nervous system has been proved to play an important role in fracture healing, and different nervous components exert different effects on bony callus.OBJECTIVE: To observe the callus volume and morphological changes in the fracture healing progress of the rats with spinal cord hemisection and fracture.METHODS: Forty-eight male Sprague-Dawley rats were divided into two groups, and the models of simple fracture and spinal cord hemisection by cutting the right spinal cord combined with fracture were established. The tibia was removed at 14 and 28 days after surgery, and the wet weight was measured, the fracture healing was observed by X-ray examination, morphological changes of the callus were observed through hematoxylin-eosin staining, and the expression level of calcitonin gene-related peptide mRNA in the broken bones was detected by SYBR Green qPCR.RESULTS AND CONCLUSION: In the combination group, the wet weight of the contralateral tibia was more than that of the affected side (P < 0.05), which both were more than those in the simple group (P < 0.05). X-ray findings showed that the combination group had the largest lateral callus, followed by the affected side, and the smallest in the simple group.Hematoxylin-eosin staining showed that in the combination group, the contralateral cells arranged in disorder, while the fibrous callus and cells in regular arrangement were found in the affected side; there were bony callus and cells in regular arrangement in the simple group at 28 days after surgery. SYBR Green qPCR results revealed that the expression level of calcitonin gene-related peptide mRNA in the simple group was significantly higher than that in the combination group (P < 0.05). To conclude, complete innervation is a necessary factor for fracture healing.

<p><b>OBJECTIVE</b>To investigate the expression level of cytokeratin20 mRNA (CK20 mRNA) of cancer cells in peripheral blood from colorectal cancer patients, detected by fluorescence quantitative polymerase chain reaction (FQ-PCR), and to evaluate its clinical value.</p><p><b>METHODS</b>To systemically study the reproducibility, quantitative range and amplification efficiency of CK20 mRNA detection by FQ-PCR, analyze and compare the result consistency with conventional RT-PCR and immunohistochemistry, separately. The expression level of CK20 mRNA of cancer cells in peripheral blood was examined in 136 colorectal cancer cases with or without hepatic metastasis.</p><p><b>RESULTS</b>The within-run and between-run CV of FQ-PCR to assay CK20 mRNA were 3.6% and 5.3%, respectively, quantitative range was from 10(3) copies/ml to 10(8) copies/ml and amplification efficiency was 87.4%. Comparing with traditional RT-PCR and immunohistochemistry, the Kappa value was 0.87 and 0.83, respectively. The expression level of CK20 mRNA of cancer cells in peripheral blood from colorectal cancer patients was (3.52 +/- 1.47) x 10(4) copies/ml, and the expression positive rate was 48.5%. None was found among the 75 cases in the control group. The positive rate of CK20 mRNA of cancer cells in peripheral blood was 9.5%, 25.0%, 48.8% and 87.5% in the patients at Dukes stage A, B, C and D, respectively (P < 0.05). The positive rate of CK20 mRNA was 87.5% in patients with hepatic metastasis and 32.3% in patients without hepatic metastasis (P < 0.05). CK20 mRNA showed a tendency to decline in 35 cases of colorectal cancer within the 1st, 3rd and 5th week after operation. There was no difference among the data of pre-operation cases and on the 1st, 3rd week (P > 0.05), but a significant difference between pre-operation and the 5th week (P < 0.05).</p><p><b>CONCLUSION</b>FQ-PCR is a rapid and sensitive method for quantitating CK20 mRNA. The expression of CK20 mRNA of cancer cells in peripheral blood from colorectal cancer patients has a correlation with recurrence and metastasis of the tumors. Detection of CK20 mRNA is helpful to monitor hematogenous dissemination of colorectal cancers.</p>

The type of basic media and the contents of plant growth substances were investigated by orthogonal design experiment,and also the effects of different culture conditions on the growth of suspension cells and the accumulation of total flavonoids in Eucommia ulmoides were studied.The results showed that B5 medium supplemented with 0.5mg/L NAA,0.6mg/L 6-BA and 30g/L sucrose,at initial pH 5.0~5.5,20g(FW)/L inoculation quantity and 110 r/min of rotation speed was a preferable culture conditions for E.ulmoides suspension cells growth and flavonoids synthesis.The results of metabolic kinetics analysis for E.ulmoides cell suspension culture showed that the logistic and Luedeking-Piret equations can be used for describing the kinetics of cell growth,sucrose consumption and flavonoids production during the process.The maximum specific growth rate(?m),the actual growth yield based on sucrose(YG) and maintenance coefficient(m) were 0.417/d,0.619g/g and 0.0206g/(g?d-1) respectively.All these outcomes could give a basis for establishing the suspension cell culture of E.ulmoides and production of the natural active components in large-scale.

<p><b>OBJECTIVE</b>To investigate the possible association between polymorphism of CC16 gene exon 1 and asthma, the genotype and allele frequencies of CC16 gene exon 1 in the asthmatic patients of Han population in southwest China were analyzed.</p><p><b>METHODS</b>The authors determined the genotypes of CC16 gene exon 1 with polymerase chain reaction technique and restricted enzyme analysis, and then compared the genotype and allele frequencies of the gene of the asthmatic group with those of the healthy control group.</p><p><b>RESULTS</b>There was no significant difference in genotype and allele frequencies of CC16 gene between the asthmatic group and control group. There was no association between the genotype and allele frequencies of gene and the severity of asthma.</p><p><b>CONCLUSION</b>CC16 gene may be not a susceptibility gene of asthmatic patients of Han population in southwest China.</p>

RESUMO

Aim The chronic toxicity of tea polyphenols(TP) in dogs was observed. Methods 18 dogs were randomly divided into high, middle and low dose TP groups and one control group . When TP was given(po) by 650 mg?kg-1?d-1,65 mg?kg-1?d-1,6.5 mg?kg-1?d-1 respectively for 90 d and after TP was stopped for two weeks, the general conditions, ECG, blood routine, urine routine, serum lipids, blood sugar, coagulation time, and hepatic and kidney function were detected respectively. Results These mdices were not different from each other among the 4 groups before, during and after administration of TP . The level of serum total cholesterol was progressively decreased in TP groups. Conclusion TP has no toxic effect on dogs and can decrease the level of serum total cholesterol in normal dogs.

In this study,nearly 200 endophytic bacteria were isolated from different part of Huperzia serrata, over 60 bacterium with clear antifungal activity were selected from those cultures.Among them,strain H-6 exhibited the highest antifungal activity which was strongly inhibits the growth of many plant pathogenic fungi such as Sclerotinia scleroliorum,Fusarium graminearumt,Sclerotinia libertiana,Phytophthora capsici Leonia and Sesame fusarium wilt.According to the characteristics of morphology,physiology and biochem- istry tests and the comparison of 16S rDNA sequence,the strain H-6 was similar to the Burkholderia.So strain H-6 was identified as Burkholderia sp.H-6.The results also showed that Burkholderia sp.H-6 was markedly different from Burkholderia cepacia that was applied widely in agriculture as antagonistic bacteria. The medium and culture conditions of the strain all were optimized by single factor and orthogonal experiments.In the investigation of the culture condition,growth was carried out in a basal medium(potato juice)and gradually supplemented with the various ingredients to be investigated.The major ingredients be- ing investigated included carbon sources and nitrogen sources.The optimal antifungal activity production condition is growth in a medium(potato juice with 2.5%mannitol and 0.1%NaNO3),initial pH 4.0 at 28℃.
