Construction and identification of CCDC67 gene lentiviral vector / 中华内分泌外科杂志

Objective To construct a]entiviral expression vector carrying CCDC67 and to obtain CCDC67 efficient and stable expression in mammalian cells.Methods The DNA fragment of CCDC67 coding sequence was amplified by PCR with designed primer from the cDNA library including CCDC67 gene,and then subcloned into GV208 vector with in-fusion technique to generate the lentiviral expression vector,GV208-CCDC67.The positive clones were screened by PCR and the correct CCDC67 was confirmed by sequencing.Recombinant lentiviruses were produced in 293T cells after the cotransfection of GV208-CCDC67,and packaging plasmids of pHelper 1.0 and pHelper 2.0 Green fluorescent protein (GFP)expression in 293T cells was observed to evaluate gene delivery efficiency.CCDC67 protein expression in 293T cells was detected by Western blot.Resuits The lentiviral expression vector carrying CCDC67 was successfully constructed and CCDC67 protein expression was detected in 293T cells.Conclusion The recombinant lentivirus GV208-GFP-CCDC67 is successfully constructed and it lays a foundation for further molecular function study of CCDC67.

Clinical analysis of thyroid and breast multiple primary carcinomas / 中华内分泌外科杂志

Objective To investigate the relationship between thyroid and breast multiple primary carci-nomas.Methods A total of 24 cases of multiple primary tumors of thyroid and breast admitted from Jan .2011 to Jun.2013 in First Affiliated Hospital of Zhengzhou University were retrospectively analyzed .Results The 24 samples were confirmed as the tumor of thyroid and breast by ultrasonography .The patients underwent operation and adjuvant therapy after fine-needle aspiration biopsy , mammography and other relevant examinations .All the malignancies of thyroid and breast were confirmed by pathology .Among the 24 cases, 19 patients were invasive ductal carcinomas , 2 patients were diagnosed as intraductal carcinomas , 1 patient was confirmed as intraductal papillary carcinoma , 1 case was invasive lobular carcinoma with invasive ductal carcinoma , and 1 case was inva-sive ductal carcinoma with intraductal carcinoma .All the thyroid malignancies were papillary thyroid carcinomas . The results of immunohistochemistry indicated that , among the 24 patients of thyroid and breast multiple primary carcinomas , 11 cases were ER (+) in both thyroid cancers and breast cancers , moreover , 8 cases were ER (-) in both thyroid cancers and breast cancers .The difference between the expression of ER in the tumors of thyroid and breast had no statistical significance (P>0.05).Conclusions Ultrasonography is a simple and effective im-aging method that can discover the multiple primary tumors of thyroid and breast .For patients of thyroid cancer , breast cancer should be excluded routinely .The thyroid should also be examined simultaneously in breast cancer patients.Once the possibility of one gland tumor is discovered , we should pay more attention to the other gland . Particularly, the other gland should be examined carefully for patients with ER (+).

Loss of fragile histidine triad gene expression in papillary thyroid carcinoma / 中华内分泌外科杂志

Objective To study fragile histidine triad gene(FHIT) gene mRNA expression in 60 samples of papillary thyroid carcinoma(PTC) and matched adjacent non-concerous tissues (NCE),and to explore the role of FHIT gene in the development in PTC.Methods Reverse transcription polymerase chain reaction(RT-PCR) was used to detect FHIT gene mRNA expression in 60 samples of PTC and matched NCE.Results In PTC tissues,the positive rate of FHIT gene mRNA expression was 45.0％ (27/60),lower than that in NCE tissues (100.0％,60/60).FHIT gene mRNA expression was correlated with pathology stage,TNM stage and lymph node metastasis (P ＜ 0.05).Conclusion FHIT loss and transcript abnormality might play important roles in proliferation and metastasis of PTC,and might be a useful marker for evaluating the biological behavior of PTC.

Inductive effect of Prunella vulgaris Injection on K562 cells apoptosis / 中草药

Objective To investigate the antileukemia effect of Prunella vulgaris Injection(PVI) in order to offer experimental data for the treatment of leukemias with PVI in clinic.Methods Inhibition of PVI on K562 cell proliferation and IC_(50) value was assayed by MTT assay.The growth curve of K562 cells was drawn also.The cellular morphology was observed by invert microscope,Giemsa staining,and MTT assay.The apoptosis induced the effect of PVI to K562 cells was observed by trypan-blue exclusion test and flow cytometer(FCM).The expression of apoptosis related protein bcl-2 and bax was measured by immunocytochemistry,and the quantitative analysis was made with figure analysis system.Results PVI obviously inhibited the proliferation of K562 cells in a dose-dependent manner(r=0.985 0).The IC_(50) was 0.113 mg/mL.(After) K562 cells were treated with PVI(50 mg/mL),the morphological characters of apoptosis were observed.The results of FCM showed that the apoptosis rats [(37.15?1.46)%] in the treatment group were significant higher than that in control group [(5.56?0.68)%](P

Establishment of multidrug resistance to adriamycin in Ehrlich ascites carcinoma in vivo / 中国药理学通报

Resistance to adriamycin was developed in an orignally adriamycin- sensitive strain of Ehrlich ascites carcinoma (EAC) in vivo by treament with the adriamycin during 50 weekly passages of the tumor. The results showed that the growth of the resistant line was slower than that of the EAC, total ascites cell volume,total cell number and mean single-cell volume were significantly smaller of the resistant line than those of original sensitive line. Also, itwas found the homogerously staining region and double minute chromosome in the cells of resistant line. The adriamycin-resistant tumor produced cross resistance to vincristine.mitomycin C,and actinomycin D,while section resistance to VP-16. There was no change in the sensitivity of tumor to oridonin.

AUTORADIOGRAPHIC STUDIES OF THE DISTRIBUTION OF〔~(14)C〕PROBIMANE IN MICE / 中国药理学通报

Probimane was first synthesized and developed in Shanghai Institute of Materia Medica, Chinese Academy of Science as a novel antit umor agent. Male mice were injected iv 〔14C〕 probimane 3.7?105Bq/20g( 5.62? 107Bq/g ) . At 0.5h,higher radioactivities were found in S87 tumor, kidney, bladder, and vertebra, moderate in skim, lung, liver, blood vessel and gut. The radioactivities tended to decre-aseafter 3h except in the tumor, kidney, and gut in which radioactivities persisted. After 12h, only a trace of radioactivity was detected n intestine. The distribution of probimane correlated with its therepeutic efficacy and toxicity.

DISTRIBUTION OF 〔~(14)C〕 BIMOLONE IN MICE BY WHOLE BODY AUTORADIOGRAPHY / 中国药理学通报

Distribution of 〔14C〕 bimolane in mice was studied by means of whole body autoradiography. At 30min, and lh, 3h, 9 h, 24h after iv, the mice were briefly anesthetized with ether and frozen by immersing indry ice/acetone. The mice were processed for whole body auto-radiography by continual sections with LKB-2250 PMV big whole body automatic-manual frozen cutter. The exeriments showed that 〔24C〕 bimolane distributed rapidly into the various tissues after iv administrtion. The highest radioactivity was found in lung, kidney, gastrointestinal tract, then the tumor, skin etc, the lowest radioactivity was found in brain.
