RESUMO

Objective To investigate the safety and clinical significance of total thyroidectomy performed for benign thyroid disease. Methods Eighty-eight patients with benign thyroid disease were treated with total thyroidectomy. The postoperative complications were analyzed. Results With primary total thyroidectomy, the incidences of transient hypocalcemia and transient recurrent laryngeal nerve paralysis were 2.5% and 1.2% respectively. RESUMO

<p><b>OBJECTIVE</b>To establish a novel bioartificial liver (BAL) consisting of spheroids of porcine hepatocytes in a hollow-fiber bioreactor, and to perform an in vitro study on its metabolic effects on the serum from severe hepatitis B patients.</p><p><b>METHODS</b>Hepatocytes were isolated from pup pigs and cultured as aggregate spheroids through rotation and vibration. Phase-contrast microscopy, transmission electron microscopy, and scanning electron microscopy were used for morphological detection of hepatocyte spheroids. The hepatocyte spheroids were then transferred into the shell of a polysulfone hollow-fiber bioreactor, creating a novel BAL. Diluted serum samples of severe hepatitis B patients were circulated for 3 hours each into the bioreactor, by using an extracorporeal circulatory system. Every half hour, including both before and after perfusion, serum samples were collected to assay total bilirubin (TBIL), total protein (TP), albumin (ALB), and globulin (GLB) concentrations in order to judge the metabolic effects of this novel BAL.</p><p><b>RESULTS</b>Most hepatocytes had formed spheroids with high viability after 24 hours in culture. After 3 hours of perfusion, when compared with the control group, the serum concentration of TBIL in the treatment group decreased significantly (P < 0.01), but the serum concentrations of TP and ALB increased significantly (P < 0.05).</p><p><b>CONCLUSIONS</b>Hepatocytes can be conveniently cultured as aggregate spheroids through a rotation and vibration method. The novel BAL is efficient in removing bilirubin from the serum of severe hepatitis B patients, and in supplying the serum with ALB. Thus, the BAL might provide effective therapy for patients with severe hepatitis B.</p>

Objective To find a preferable cryopreservative solution and enhance the viability further.Methods Donor hepatocytes were isolated from hybrid pig weighting 5~8 kg by means of collagenase.Then the hepatocyte suspension was put into a liquid nitrogen container for one month with three different cryoprotectants (only use DMSO,combined use of DMSO+HES and combined use of DMSO+HES+1.3 mmol/L Ca 2+).The samples were thawed rapidly and detected in the viability and morphology after a month.Fractional hepatocytes were cultured five days in containing Ca 2+ and uncontaining Ca 2+ culture media to observe the expression of function.Results Porcine hepatocytes,which were preserved in different culture media,show the different viability and morphology manifestation.Among the total the method of only uses DMSO show worst effect.Then is the DMSO+HES group.Combined cryopreservative method with HES+DMSO+1.3 mmol/LCacl 2 has a best effect and functional expression.Hepatocytes were cultured in culture media containing Ca 2+is better than those were cultured in culture media uncontaining Ca 2+.Conclusion The combined cryopreservative system in which HES+DMSO+1.3 mmol/LCacl 2 as the cardinal component can improve the preservative quality significantly.

RESUMO

Objective To study the value of the neuron-specific enolase(NSE) in the serum of patients with cerebral stroke in diagnosis, seriousness and evaluation of prognosis. Methods The levels of NSE in the serum of 77 patients with cerebral stroke(36 cases with hemorrhage, 41 cases with infarction) were measured by radio-immunoassay (RIA)method.Results The serum leves of NSE both in cerebral infarction and cerebral hemorrhage patients were obviously higher than those of the control group( P 0.05). In the first week there was a positive correlation between NSE serum level and neurological deficit scores,the volume of haemorrhage, the volume of infarction( P 0.05) . Conclusion The NSE level in serum may be served as a objective index of seriousness and early diagnosis in stroke, but no helpful in differential diagnosis and evaluation of prognosis.
