RESUMO

An anisakid nematode larva found in cod sold in the state of Minas Gerais, Brazil was studied by light and scanning electron microscopy and by a molecular approach. Mitochondrial cytochrome c-oxidase subunit 2 (mtDNA cox-2), 28S rRNA and ITS1, 5.8S and ITS2 regions were amplified using the polymerase chain reaction and sequenced to evaluate the phylogenetic relationships of the larva. The genetic profile confirmed that this larva belongs to the species Pseudoterranova decipiens (sensu stricto). This is the first molecular and ultrastructural study of Pseudoterranova decipiens (sensu stricto) in imported cod sold in Brazil. The health implications of these findings are discussed.


Uma larva de Nematoda anisakídeo encontrada em bacalhau comercializado no Estado de Minas Gerais, Brasil foi estudada por microscopias de luz e eletrônica de varredura e por uma abordagem molecular. As regiões da subunidade 2 da citocromo c-oxidase mitocondrial (mtDNA cox-2), 28S rRNA e ITS1, 5.8S e ITS2 foram amplificadas usando a reação em cadeia da polimerase e sequenciadas para avaliar as relações filogenéticas da larva. O perfil genético confirmou que esta larva pertence à espécie Pseudoterranova decipiens (sensu stricto). Esse é o primeiro estudo molecular e ultraestrutural de Pseudoterranova decipiens (sensu stricto) de bacalhau importado vendido no Brasil. As implicações destes resultados para a saúde são discutidas.

Three new sequences of Mitochondrial cytochrome c-oxidase subunit 2 (mtDNA cox-2) from C. pelagicum parasite of Spheniscus magellanicus, the Magelanicus penguin, were determined from Brazilian waters. The sequences presented 99 and 98% of similarity with C. pelagicum sequences from Argentina, deposited on GenBank for the same genetic region and with a strong statistical support inferred from the phylogenetic tree. The morphological and ultrastructural studies that were carried out confirmed the genetic analysis.


Foram determinadas três novas sequências da região do Citocromo c-oxidase da subunidade II do DNA mitocondrial (cox-2 mtDNA) de Contracaecum pelagicum, parasito de Spheniscus magellanicus, pinguim Magalhães, de águas brasileiras. As sequências apresentaram 99 e 98% de similaridade com sequências de C. pelagicum da Argentina depositadas no GenBank da mesma região genética com forte suporte estatístico inferido pela arvore filogenética. Estudos morfológicos e ultraestruturais realizados confirmaram a identidade genética.

RESUMO

This article presents an improvement to the Kato-Katz (KK) method, making it faster and more efficient for the visualisation of fertile eggs in stool samples. This modified KK method uses sodium acetate formalin as a fixative and reveals the intensity of infection in less than 1 h, reducing the diagnostic time without increasing the cost. This modified method may contribute to future epidemiological studies in both hospitals and the field due to its rapid and precise diagnostic, which allow for immediate treatment.

Raphidascaris (Sprentascaris) lanfrediae sp. nov. is described from the intestine of the freshwater fish Satanoperca jurupari (Heckel) (Cichlidae) from the Guamá River, state of Pará, Brazil. The prevalence in fish (n = 59) was 27 percent with intensity of one-124 (mean 16) nematodes per fish. The new species is characterized mainly by the markedly larger size of ventricular appendix in relation to the oesophagus, presence of short male caudal alae, 14-16 subventral pairs of preanal papillae and six pairs of postanal papillae.

Absonifibula estuarina sp. n. (Diclidophoridae, Absonifibulinae), is described from the gills of juvenile striped weakfish, Cynoscion guatucupa (Cuvier), from the southwestern Atlantic, Argentinean coast. This marine fish migrates to estuarine areas to spawn where exclusively juveniles are found parasitized; adult fish in marine water were never found to be parasitized by this monogenean. A. estuarina sp. n. is characterized mainly by the pedunculate clamps dissimilar in size, the shape of anterior jaw with sclerite 'a' attached to a sub-trapezoidal lamellate extension and fused to sclerites 'c' and 'd'. It differs from Absonifibula bychowskyi Lawler & Overstreet, 1976, the only known species of the genus, in the shape and arrangement of the genital corona, which is armed with six similar hooks disposed in circle and the sub-trapezoidal shape of lamellate extension ('b'). The restriction to juvenile sciaenids is a shared feature among the Absonifibulinae indicating an estuary-dependent life cycle.

Camallanus tridentatus is redescribed on the basis of the examination of specimens obtained from the stomach, caeca and intestine of the naturally infected arapaima Arapaima gigas (Schinz) from the Mexiana Island, Amazon River Delta, Brazil. Data on the surface morphology of adults inferred from confocal laser scanning and scanning electron microscopical observations are also provided. The study revealed some taxonomically important, previously unreported morphological features in this species, such as the presence of the poorly sclerotized left spicule and deirids. C. tridentatus distinctly differs from other congeneric species parasitizing freshwater fishes in South America mainly in the structure of the buccal capsule and the female caudal end. C. maculatus Martins, Garcia, Piazza and Ghiraldelli is considered a junior synonymm of Camallanus cotti Fujita.

The life history of the trematode Pygidiopsis macrostomum Travassos, 1928 is described for the first time. Rediae and cercariae were obtained from naturally infected snails Heleobia australis (d´Orbigny), a new first intermediate host. Metacercariae were found encysted in the mesenteries of three naturally infected guppies, Phalloptychus januarius (Hensel), Jenynsia multidentata (Jenyns) (new host records) and Poecilia vivipara Bloch and Schneider. Experimental infections were successfully completed in the intermediate hosts H. australis and Poe. vivipara reared in the laboratory and hamsters Mesocricetus auratus Waterhouse were utilised as a definitive host.

RESUMO

A new nematode species, Capillostrongyloides arapaimae sp. n., is described from the intestine and pyloric caeca of the arapaima, Arapaima gigas (Schinz), from the Mexiana Island, Amazon river delta, Brazil. It is characterized mainly by the length of the spicule (779-1,800 µm), the large size of the body (males and gravid females 9.39-21.25 and 13.54-27.70 mm long, respectively) and by the markedly broad caudal lateral lobes in the male. It is the third species of genus Capillostrongyloides reported to parasitize Neotropical freshwater fishes.

The tegument of the polyopisthocotylean monogenean Atriaster heterodus Lebedev & Parukhin, 1969 was studied using transmission electron microscopy. The outer syncytial layer of the tegument is connected to the internal cell bodies by cytoplasmic extensions which interweave between the muscular fibres. The free surface of the syncytium has projections of the external membrane which are similar to microvilli. The undulating basal membrane, with numerous narrow elongate projections, is associated with the basal lamina situated between the syncytial and muscular layers. The cell bodies and syncytial layer of the tegument exhibit two types of vesicles, one with fibrous contents and one with electron-dense contents; these were analysed using two cytochemical tests, the E-PTA and alcian blue methods, used for the first time on monogeneans

Larval stages and adults of Procamallanus (Spirocamallanus) pereirai Annereaux, 1946 are described from naturally infected Paralonchurus brasiliensis (Steindachner) (Sciaenidae) from the coast of the State of Rio de Janeiro, Brazil. The translucent first-stage larvae have a denticulate process at the anterior end, no buccal capsule or esophagus undifferentiated into anterior muscular and posterior glandular parts and an elongate tail; third-stage larvae have a tail with three terminal projections, a buccal capsule divided into an anterior portion with 12-20 ridges running to the left and a posterior smooth portion, and an esophagus with muscular and glandular regions. Fourth-stage larvae exhibit a buccal capsule lacking a distinct basal ring with ridges running to the right and a tail with two terminal processes, as in adults. New host records are reported and their role in its life-cycle are discussed.

Bicotylophora trachinoti (Mac Callum, 1921) from Trachinotus carolinus L.; Pseudanthocotyloides heterocotyle (Van Beneden, 1871) from Cetengraulis edentulus (Cuvier), and Decapterus punctatus (Cuvier), new host records; Pseudomazocraes selene Hargis, 1957 from Selene vomer (L.) and Caranx latus Agassiz, new host record, are reported for the first time in Brazil from the coast of Rio de Janeiro State. The marine fishes Diplectrum sp. and Pomatomus saltatrix (L.) are respectively new host records for Pseudotagia cupida (Hargis, 1956) and Macrovalvitrema sinaloense Caballero & Bravo Hollis, 1955. Measurements, original figures and photos in scanning electron microscopy of B. trachinoti are presented. The egg with filaments is reffered for the first time in the genus Pseudanthocotyloides

Cribomazocraes travassosi n. sp. is described from Harengula clupeola (Cuvier, 1829) from Rio de Janeiro, coast, Brazil. It differs from C. nagibinae, Mamaev, 1981 in the size and shape of opisthohaptor and lappet and in the smaller size of anchors. From C. bychowskyi it also differs in the extension of vittelaria. A key for the species of the genus is presented

From the gills of 100 Micropogonia furnieri (Desmarest, 1823) from Atlantic coast of Rio de Janeiro State, were recovered Macrovalvitrema sinaloense Caballero & Bravo-Hollis, 1955, Pterinotrematoides mexicanum Caballero & Bravo-Hollis, 1955, Rhamnocercus rhmnocercus Monaco, Wood & Mizelle, 1954 and Encotyllabe spari Yamaguti, 1934. M. furnieri represents a new host record for them and a new geographical dsitribution is referred for M. sinaloense, P. mexicanum and R. rhmnocercus.
