RESUMO

Purpose@#We aimed to develop a novel method for orthotopic colon cancer model, using tissue adhesive in place of conventional surgical method. @*Materials and Methods@#RFP HCT 116 cell line were used to establish the colon cancer model. Fresh tumor tissue harvested from a subcutaneous injection was grafted into twenty nude mice, divided into group A (suture method) and group B (tissue adhesive method). For the group A, we fixed the tissue on the serosa layer of proximal colon by 8-0 surgical suture. For the group B, tissue adhesive (10 μL) was used to fix the tumor. The mortality, tumor implantation success, tumor metastasis, primary tumor size, and operation time were compared between the two groups. Dissected tumor tissue was analyzed for the histology and immunohistochemistry. Also, we performed tumor marker analysis. @*Results@#We observed 30% increase in graft success and 20% decrease in mortality, by using tissue adhesive method, respectively. The median colon tumor size was significantly increased by 4 mm and operation time was shortened by 6.5 minutes. The H&E showed similar tumor structure between the two groups. The immunohistochemistry staining for cancer antigen 19-9, carcinoembryonic antigen, cytokeratin 20, and Ki-67 showed comparable intensities in both groups. Real-time quantitative reverse transcription analysis showed eight out of nine tumor markers are unchanged in the tissue adhesive group. Western blot indicated the tissue adhesive group expressed less p-JNK (apototic marker) and more p-MEK/p-p38 (proliferation marker) levels. @*Conclusion@#We concluded the tissue adhesive method is a quick and safe way to generate orthotopic, colon cancer model.

Objective To investigate the different analgesic effects of acupuncture by selecting points along and not along the meridian in treating migraine, analyze the necessity of selecting points along the meridian in the treatment of migraine with acupuncture, and to provide evidence for the acupuncture treatment for migraine.Method Sixty-nine migraine patients were selected as the study subjects and randomized into Shaoyang meridian group, Yangming meridian group and a non-meridian point group by adopting a complete randomized design, 23 cases in each group, to respectively receive acupuncture at the Yuan-primary point of Foot Shaoyang Meridian Qiuxu (GB40), the Yuan-primary point of Foot Yangming Meridian Chongyang (ST42), and a non-meridian point at the midpoint between Qiuxu and Chongyang. Of the three groups, only Shaoyang meridian group selected point along the meridian. The improvements of pain intensity in the three groups were compared, and the pain intensity was measured by using Visual Analogue Scale (VAS). Nonparametric test was used to analyze whether there were significant differences in comparing the improvements of pain in the three groups.Result According to the statistical analyses, the pain intensities were improved significantly in each group (P<0.001), while there were no significant between-group differences in comparing the improvement of pain intensity (P>0.05).Conclusion In the treatment of migraine with acupuncture, selecting points along the meridian and not along the meridian (including non-meridian point) produces equivalent analgesic effect.

RESUMO

<p><b>OBJECTIVE</b>To study the chemical constituents from the roots of Cudrania cochinchinensis.</p><p><b>METHOD</b>The chemical constituents of C. cochinchinensis were isolated and purified by silica gel, polyamide column chromatography, Their structures were identified on the basis of spectroscopic data.</p><p><b>RESULT</b>Six compounds were isolated and identified. Their structures were established as 3, 5, 7, 4"-tetrahydroxyflavanone-7-O-(6"-acetyl)-glucoside (1), 3, 5, 7, 4'-tetra hydroxyflavanone-7-O-glucoside (2), 2', 4', 5, 7-tetrahydroxy-6-prenyldihydroflavanone (3), 5, 7, 4'-trihysdroxy-6-prenylisoflavanone (4), 1, 3, 5, 6-tetrahydroxyxanthone (5), stilbene-2, 4, 3', 5'-tetraol (6).</p><p><b>CONCLUSION</b>Compound 1 was isolated from this genus, while compounds 4 and 5 were isolated from this plant for the first time.</p>

RESUMO

<p><b>AIM</b>To observe protective effects and involved mechanism of ginsenoside Rb3 on hypoxic/ischemic brain injury, using cultured hippocampal neurons, rat hippocampal slices and intact animals.</p><p><b>METHODS</b>(1) Mice were tightly closed in glasses of 150 ml, and then survival time of them was observed. (2) During simulated ischemia and after reoxygenation, changes of orthodromic population spikes (OPS) in the area CA1 of hippocampal slice were investigated. (3) By using histochemistry, the expressions of NOS in CA1 area of rat hippocampus after hypoxic exposure were observed. (4) Using LDH detection, tests of total NOS, iNOS and cNOS activity, the protective effects of ginsenoside Rb3 were investigated on cultured hippocampal neurons treated with hypoxia.</p><p><b>RESULTS</b>(1) Given ginsenoside Rb3 (10 mmol/L), mice survived significantly longer than that of control group. (2) The occurrence of HIP (hypoxic injury potentials) decreased after administration of ginsenosides Rb3 (60 micromol/L) in many slices, while the recovery rate and amplitude of OPS after reoxygenation were significantly higher than those of the control group. (3) In CA1 area of rat hippocampus, NOS-positive neurons increased at the end of 24 h hypoxia and further 24 h reoxygenation, while the number of NOS-positive neurons decreased after treatment with ginsenoside Rb3. (4) The LDH leakage rate of cultured rat hippocampal neurons increased at the end of hypoxia, while it decreased after treatment with Rb3. Moreover the total NOS, especially iNOS activity of these neurons also decreased.</p><p><b>CONCLUSION</b>Ginsenoside Rb3 has a significant protective effect on hypoxic-ischemic injury of neurons, and this involves the stabilization of the cell membrane, the inhibition of the expression and activity of NOS, especially iNOS activity.</p>

RESUMO

Objective To discuss the illness mechanism and the reason of defective energy of the stomach and intestines with acute panereatitis.Method 43 cases of acute pancreatitis was studied which had the stomach and intestinal insufficiency to determine lesion degree of the stomach and intestines acording to the change of abdomen signs.Results The data found that the patient of light acute pancreatitis had light abdominal distention,cirumscribed abdominal tenderness and rebounding pain,weaken gurgling sound;most advanced acute pancreatitis had severe abdominal distention,abdominal tenderness and rebounding pain,almost no gurgling sound.Conclusion Abdominal sign could be a indicator to determine patient's condition of acute pancreatitis and a standard to diagnose acute pancreatitis.
