ABSTRACT Purpose: Spontaneous intracerebral hemorrhage (ICH) is a major cause of death and disability with a huge economic burden worldwide. Cerebrolysin (CBL) has been previously used as a nootropic drug. Necroptosis is a programmed cell death mechanism that plays a vital role in neuronal cell death after ICH. However, the precise role of necroptosis in CBL neuroprotection following ICH has not been confirmed. Methods: In the present study, we aimed to investigate the neuroprotective effects and potential molecular mechanisms of CBL in ICH-induced early brain injury (EBI) by regulating neural necroptosis in the C57BL/6 mice model. Mortality, neurological score, brain water content, and neuronal death were evaluated by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining, Evans blue extravasation, Western blotting, and quantitative real-time polymerase chain reaction (PCR). Results: The results show that CBL treatment markedly increased the survival rate, neurological score, and neuron survival, and downregulated the protein expression of RIP1 and RIP3, which indicated that CBL-mediated inhibition of necroptosis, and ameliorated neuronal death after ICH. The neuroprotective capacity of CBL is partly dependent on the Akt/GSK3β signaling pathway. Conclusions: CBL improves neurological outcomes in mice and reduces neuronal death by protecting against neural necroptosis.

@#[Abstract] Objective: To explore the role of miR-9-5p in the biological behaviors of breast cancer cells and its possible regulatory mechanism. Methods: online OncomiR database was used to analyze the differential expression of miR-9-5p in breast cancer tissues and normal breast tissues. qPCR was used to detect the miR-9-5p expression in breast cancer cell lines and normal breast cells. Based on target gene prediction software TargetScan, ONECUT2 (one cut homeobox 2) was predicted to be the target gene of miR-9-5p. Dual luciferase reporter system was used to validate the relationship between miR-9-5p and its promising target gene ONECUT2. MDA-231 cells were transfected with miR-9-5p mimic, ONECUT2 siRNAs as well as the corresponding control sequences. The protein and mRNA levels of stemness-associated gene NOTCH1, NANOG and SOX9 (SRY (sex-determing region of Y chromosome) -Box transcription Factor 9) were detected by WB and qPCR. The effect of transfection on proliferation, apoptosis and chemo-resistance of cells was detected by BrdU method, Annexin Ⅴ method and MTS Assay, respectively. The ALDEFLUOR experiment was used to detect the effects of miR-9-5p and its target gene ONECUT2 on tumor stemness. NSG mouse breast cancer chemotherapy model was established, and the in vivo experiments further verified the effect of ONECUT2 on tumor malignant biological behaviors, such as cell stemness and chemo-resistance. Results: miR-9-5p was highly expressed in breast cancer tissues (P=0.007) and breast cancer MDA-231 cell line (P=0.0005), and was positively correlated with the poor prognosis of breast cancer patients (P=0.0016). Compared to control group, miR-9-5p could target and negatively regulate ONECUT2 expression, further increase ALDH+ cell population (P=0.0006), as well as increase the expressions of stemness-associated genes NOTCH1, NANOG and SOX9. Besides, miR-9-5p increased the anti-apoptosis ability (P=0.0003) and chemo-resistance of MDA-231 cells; however, miR-9-5p/ONECUT2 exerted no significant effect on the proliferation ability of MDA-231 cells (P>0.05). Compared with the control group, the volume of xenografts in mice of MDA-231/ONECUT2 group after DTX chemotherapy was significantly lower than that in the control group (P<0.05), and the protein expressions of NOTCH1, SOX9 and the mRNA expression of ABC transporter in the transplanted tumor tissues were significantly reduced (P<0.05 or P<0.01). Conclusions: The highly expressed miR-9-5p in breast cancer induces tumor stemness and anti-apoptotic ability by targeting ONECUT2 and enhances its resistance to chemotherapy.

The microorganism in human body is related to human health and disease,which plays a significant role in the exclusion of pathogen invading,participation of immune regulation and occurrence and development of the tumors.Studies have indicated that dysbacteriosis is often detected in cancer patients;especially,enteric dysbacteriosis can promote the occurrence and development of the tumors by influencing homeostatic balance of intestinal tract,gut metabolite and immunological function.Moreover,proportion detection of intestinal flora can be used as a non-invasive screening method and a diagnostic indicator for early detection of the malignant neoplasms.This review summarizes the recent studies on the associations of intestinal flora with human diseases and the possible mechanisms causing tumorigenesis,which may provide a novel research paradigm for cancer prevention and treatment.

<p><b>OBJECTIVE</b>To establish a method to determine lead and Isotope Ratios in whole blood by inductively coupled plasma mass spectrometry (ICP-MS).</p><p><b>METHODS</b>The whole blood samples was removed protein by 5% nitric acid , Online join thallium (Tl) as internal standard substance, used lyophilized bovine blood lead and cadmium standard material (GBW09139h and GBW09140h) for quality control of blood lead concentrations. Lead isotope standard substances (GBW04426) is used to determine the correction factor, lead isotope ratios will lead isotope standard reference material NIST SRM981 by icp-ms with samples for testing.</p><p><b>RESULTS</b>Optimize the detection method, detection of blood lead and lead isotope, and the method of linear range r >0.9999, GBW09139h and GBW09140h test results are within the scope of quality control. NIST SRM981 isotope determination precision RSD<1%, NIST SRM981 test results and the certificate of value close to.</p><p><b>CONCLUSION</b>The method is simple and convenient data is reliable, can meet the total lead (pb) in blood and former isotope simultaneous determination.</p>

Objective To investigate the serum lipids levels of Chinese men with chronic paraplegia.Methods A total of 650 male paraplegics admitted to the China Rehabilitation Research Center (CRRC) for rehabilitation therapy from Jan 2004-Dec 2008 were investigated. The inclusion criteria were a definite paraplegic history (T2-L5 spinal cord injury); complete data; age 18-60 years; and no history of stroke. TG, TC, LDL and HDL serum lipid profiles were studied. Results These male paraplegics showed significantly higher mean TG and TC levels and lower mean HDL levels than the nation-wide averages for similar males ( TG 1.72 mmol/L vs. 1.36 mmol/L; TC4.42 mmol/L vs. 4.30 mmoL/L; HDL 0.99 mmol/L vs. 1.18 mmol/L). No difference in average LDL levels was found. TG, TC and LDL levels were correlated positively with age, but HDL was not. Duration of paraplegia correlated positively with HDL levels, but negatively with LDL. The prevalence of dyslipidemia was 67% , and the prevalence of low HDL was 64%. Conclusions The serum lipid profiles of the young male paraplegics showed a high prevalence of dislipidemia, significantly low HDL levels with a high prevalence of low HDL, and no correlation with age. TG, TC and LDL levels increased with age. The duration of paraplegia was correlated positively with HDL but negatively with LDL, probably due to increased exercise of the upper limbs.

@#ObjectiveTo investigate the value of serum creatinine(SCr) to monitor the renal function of patients with spinal cord injury(SCI).Methods1298 SCI patients were investigated in Beijing Charity Hospital from January 2004 to May 2009. 249 healthy people were involved as control. The data of SCr were analysed.ResultsThe level of SCr for the SCI patients was (56.81±14.33) μmol/L with normal distribution, 95% CI was 28.73~84.89 μmol/L; The level of SCr for the healthy people was (75.98±11.34) μmol/L, 95% CI was 57.36~101.80 μmol/L. The level of SCr was significantly lower in the SCI patients comparing to the healthy people (P<0.01). Among the SCI patients, the level of SCr was (59.59±13.76) μmol/L for male while (46.41±11.31) μmol/L for female(P<0.05). There was no difference in level of SCr among different ages of the SCI patients(P>0.05). The characteristics were likely in the two groups. There was no difference in level of SCr between paraplegic (55.54±14.96) μmol/L and quadriplegic (57.67±13.83) μmol/L (P>0.05).ConclusionThe characteristics of level of SCr were likely in SCI patients and healthy people with normal distribution. The level of SCr was lower in SCI patients comparing to healthy people, which was higher for male SCI patients comparing to female SCI patients.

@#ObjectiveTo explore the arrhythmia happened in patients with cervical spinal cord injury (CSCI). Methods61 patients with CSCI were retrospectively analyzed. ResultsThe overall incidence of sinus bradycardia was 26.2%, which was higher in the cases within 3 months (52.4%) than over (12.5%); and was higher in cases injured the cord above C5(46.2%)than lower (C6-8,11.4%). ConclusionThe risk of arrhythmia is still high in chronic phase, which still need to monitor.

@#ObjectiveTo investigate the safety and clinical value of treadmill exercise testing for the patients with acute myocardial infarction in the early recovery. Methods36 patients with acute myocardial infarction performed treadmill exercise testing with Bruce protocol 15~57 d after infarction. ResultsThe outcome of 19 cases was positive, and the outcome of other 16 cases was negative. 1 case couldn't finish the testing, whose outcome could not be analyzed because his exercise time was too short. The average cardic fuction was (5.42±3.12) METs. ConclusionIt is safe that the patients with acute myocardial infarction performed treadmill exercise testing early.

@#ObjectiveTo investigate the relationship between high-sensitivity C reactive protein (hs-CRP) and oxygen uptake efficiency slope (OUES) in patents with cardiovascular disease. MethodsExercise tests, following a symptom-limited standard Bruce protocol with simultaneous respiratory gas-exchange measurements, were performed on a treadmill in 17 post-PCI patients, 21 stable angina pectoris and 24 patients with essential hypertension (EH). The oxygen uptake (VO2), minute ventilation (VE) were determined. The OUES was derived from the relation between VO2 (ml/min) and VE (L/min) during incremental exercise and was determined by VO2=a×lgVE+b, where a = OUES. The OUES was calculated from data of the first 75% (OUES75) and 100% (OUES100) of exercise duration. The serum hs-CRP was measured by immunoassays, and the left ventricular ejection fraction (LVEF) was measured with echocardiography. ResultsOUES75 (r=-0.506,P<0.001), OUES100 (r=-0.567,P<0.001) and LVEF (r=-0.286,P<0.01) were correlated with lg(hs-CRP). In stepwise multivariable linear regression models, lg(hs-CRP) (β=-0.374,P=0.006), body height (β=-1.854,P=0.036), body weight (β=-5.033,P=0.034)were independent risk factors of lower OUES75. lg(hs-CRP) (β=-0.396,P=0.003), body height (β=-2.157,P=0.013), body weight (β=-5.912,P=0.011) were independent risk factors of lower OUES100. ConclusionThe serum leveles of hs-CRP may be the useful marker that reflecting cardiopulmonary functional reserve and left ventricular function.

Objective To investigate the cognitive impairment of middle-aged patients with type 2 diabetes mellitus(T2DM). Methods Wechsler adult intelligence scale-Chinese revised edition (WATS-RC) was used as the tool to examine the cognitive function of middle-aged patients with T2DM and the healthy controls in 51 cases. The t-test was used to compare the differences in score between patients and the control group. The patients were divided into two groups with or without cognitive dysfunction. The logistic regression analysis was made for all the variables to confirm risk factors for the cognitive dysfunction in diabetes. Results The WAITS-RC scores of digit-symbol coding test and block design test were much lower(P<0.05) in diabetes versus control group. The other tests showed no significant difference. The cognitive function was negatively correlated with HbA_1>c and HOMA-IR by logistic regression analysis. Conclusions For middle-aged patients with type 2 diabetes, their cognitive functions such as learning ability, analysis and comprehensive capabilities are impaired. The cognitive function is closely related with HbA_1c and HOMA-IR.

@#Objective To investigate the efficacy of the oxygen uptake efficiency slope (OUES), obtained from submaximal exercise, as an index of cardiopulmonary functional reserve in adults. Methods Exercise tests, following a symptom-limited standard Bruce protocol with simultaneous respiratory gas measurement, were performed on a treadmill in 33 healthy adults free of clinically recognized cardiovascular disease and 31 patients with heart disease. The OUES was derived from the relation between oxygen uptake (VO2) and minute ventilation (VE) during incremental exercise and was determined by VO2=alog10VE+b, where a=OUES. The OUES was calculated from data of the first 75%, 90% and 100% of exercise duration (as 75%OUES,90%OUES and 100%OUES). Results The 100%OUES,90%OUES and 75%OUES were not significantly different from each other(F=0.212,P=0.809). The 75%OUES was slightly lower (1.9%) than the 100%OUES. The 100%OUES, the 90%OUES and the 75%OUES correlated with the peak VO2 (r=0.836,r=0.824 and r=0.777, P<0.001, respectively). The VAT correlated with the peak VO2 (r=0.878,P<0.001),and the VE/VCO2 at VAT correlated with the peak VO2 (r=-0.584,P<0.001). The 100%OUES correlated with the 90%OUES and the 75%OUES (r=0.991 and r =0.945, P<0.001, respectively). Conclusion The OUES is an index of exercise performance and cardiopulmonary functional reserve on healthy adults and patients with cardiac disease. The 75%OUES from submaximal exercise can be an index of cardiopulmonary functional reserve for patients with cardiac disease.

@#: Objective To investigate the effects of exercise therapy on activity of daily living (ADL), perception of dyspnea and quality of life (QOL) in stable patients with chronic obstructive pulmonary disease (COPD). Methods 40 COPD stable patients accepted treadmill exercise for 8 weeks. 6-min walk test (6MWT), the parameters of cardiopulmonary exercise test, ADL, perception of dyspnea (Borg Scale), and St. George's respiratory questionnaire (SGRQ) were measured before and after exercise. Results ADLs in different levels (P<0.05), especially in higher level (P<0.01), were significantly improved in all patients after exercise. Perception of dyspnea (Borg Scale) was also improved significantly (P<0.01). There was positive correlation between ADL and QOL (r=0.455, P<0.01), but negative between dyspnea and QOL (r=-0.412,P<0.05). Conclusion Exercise therapy can improve the ADL and perception of dyspnea in daily activities in patients with COPD, as well as QOL.@*@#

@#Chronic obstructive pulmonary disease (COPD) influenced the activities of daily living (ADL) of COPD patients. The six-minute walking test is considered as a good marker of functional capacity to perform ADLs. However, it does not identify the activities roundly. A good ADL scale can be used to assess functional incapacity of ADL in patients with COPD, and present sensitivity to changes following pulmonary rehabilitation.

@#Objective To investigate the relationship between objectively determined daily physical activities and blood pressure levels in Chinese adults. Methods 125 individuals who participated in routine physical examinations of health were investigated, include systolic pressure, diastolic pressure, pulse pressure, and daily walking steps determined by pedometer. Results The daily walking steps correlated negatively with systolic pressure(Male r=-0.613, P<0.01; Female r=-0.430, P<0.01) and diastolic pressure (male r=-0.686, P<0.01; female r=-0.388,P<0.01). Daily walking steps of normotensive, pre-hypertensive, and hypertensive groups were (18249±6066) step/d, (10395±3788) step/d, (6232±3967) step/d for males, and (12564±6927) step/d, (11458±5084) step/d, (6153±3569) step/d for females respectively (P<0.05). Conclusion The daily physical activity determined by walking steps correlated negatively with blood pressure in Chinese adults. Hypertensives often live in low active or sedentary lifestyle. Active lifestyle (≥10000 step/d) may help keep normal blood pressure for Chinese adults.

@#Objective To investigate the relationship between daily walking-steps and serum lipids.Methods 120 individuals who participated in routine physical examinations of health were investigated, objective measurements included triacylglycerol (TG), total cholesterol (TC), low density lipoprotein (LDL) and high density lipoprotein (HDL), and daily walking-steps were determined by pedometer.Results The daily walking-steps were correlated negatively to TG ( r=-0.351, P<0.01), and positively to HDL ( r=0.284, P<0.05) for male after adjustment for age.Conclusion Daily walking-steps determined by pedometer are correlated negatively to TG levels, and positively to HDL levels for male. No correlations are found between daily walking-steps and levels of TC, LDL for male. No correlations are found between daily walking-steps and serum lipids for female.

@# Objective To investigate the relationship between daily physical activities determined by walking-steps and the obesity indices including body weight,Body Mass Index (BMI),waist and hip circumference and their ratio in adults. Methods 125 individuals who participated in routine physical examinations of health were measured with body weight, BMI, waist and hip circumference/ratio, daily physical activities determined by walking steps. Results For males, daily physical activities correlated negatively with body weight (r=-0.397,P<0.01), BMI (r=-0.499,P<0.01), waist circumference (r=-0.490, P<0.01) and the ratio of waist vs. hip circumference (r=-0.478,P<0.01), whereas body weight(r=-0.342,P<0.01), BMI (r=-0.249,P<0.05), and hip circumference (r=-0.303,P<0.05) for females. Daily walking-steps were significantly different among the groups of normal weight (11491±6100) steps/d, overweight (9314±5722) steps/d, and obesity (6141±2985) steps/d (P<0.01). Conclusion Daily physical activities determined by walking steps correlated significantly with obesity indices.

@# Objective To compare the effect of breathing oxygen or air on respiratory function training for chronic obstructive pulmonary disease (COPD). Methods 32 patients with stable COPD, pulmonary function Ⅱ~Ⅳ, were divided into 2 groups: oxygen (10males and 6 females and air (12 males and 4 females). All the patients participated in a standardized pulmonary rehabilitation program. They were assessed with St. George respiratory questionnaire, pulmonary function test, 6-minute walking distance, cardiopulmonary exercise test and Borg scale, maximal oxygen consumption (VO2max) before and after training. Results After 8 weeks of pulmonary rehabilitation, the VO2max in oxygen group increased compared with that of the air. Conclusion Supplying oxygen during training can increase the tolerance of COPD patients.

<p><b>OBJECTIVE</b>To analyze the capacity of personal protection on poison emergency items in Chinese disease control and prevention institutes.</p><p><b>METHODS</b>Evaluation analysis based on data obtained from investigating personal protection equipments and professional knowledge quizzing about personal protection in poisoning control of 57 different level centers for disease control and prevention selected from China by a multi-stage stratified sampling.</p><p><b>RESULTS</b>All 80.70% of the institutes possessed the protection equipments, provincial, municipal, county level institutes were 100.00%, 95.24%, 66.67%, respectively, with significant statistical difference (H = 7.94, P < 0.05). The average level of the type of individual protective equipments in disease control and prevention institutes was (5.42 +/- 4.00) kinds, the average points in category of provincial, municipal, county level institutes were (11.33 +/- 3.67), (6.52 +/- 3.16), (3.47 +/- 3.10) kinds, being statistically significant (F = 17.30, P < 0.05); type difference counts of disease control and prevention institutes in economic development, secondary, less-developed regions were (6.41 +/- 4.03), (3.55 +/- 3.35), (6.45 +/- 4.07) kinds, with statistical significance (F = 3.70, P < 0.05). Protection equipments chiefly possessed were latex gloves, gauze masks and C-protective clothing. Protective clothing and respiratory protective equipments were insufficient evidently. The average points in testing personal protection basic knowledge were (71.39 +/- 12.52) points; there were no differences between different institutes with different economic regions, levels, technical posts and title degrees.</p><p><b>CONCLUSION</b>Certain advances have been achieved in recent years in personal protection capacity of institutes for disease control and prevention, but far from the actual demands, and maybe no enough effective response on emergency occurred.</p>
