RESUMO

@#Water content determination is an important aspect of quality research in the whole process of pharmaceutical development, production, storage and use.Therefore, it is significant to develop an accurate and robust method for the determination of water content in the quality control of pharmaceuticals.In this article, the selection and strategy for the study of water determination methods are explained on the basis of general water determination principles and application examples in Chinese and foreign pharmacopoeias, together with the analysis of sample structure, solubility and water content.This article aims to provide basis for the development of moisture testing methods and formulation of proper quality standards for pharmaceuticals.

RESUMO

OBJECTIVE:To investigate antitussive,antiasthmatic,phlegm-resolving and anti-inflammatory effects of Fritillaria cirrhosa and Fritillaria unibracteata. METHODS:Mice and guinea pig were randomly divided into blank control group (distilled water),ethanol extract groups,water extraction groups and powder groups of F. cirrhosa and F. unibracteata(1.13 g/kg for mice, 0.42 g/kg for guinea pig,calculated by crude drug),positive control group,with 10 mice(guinea pig)in each group. Each group was given relevant medicine intragastrically once a day for consecutive 5 d. Relative indicators were detected 30 min after last medication. The ammonia water induced cough method was used to investigate antitussive effect (dextromethorphan hydrobromide as positive drug,0.013 g/kg)by determining coughing latent period and coughing times within 3 min. After inducing asthma with 2% histamine phosphate for 15 s, antiasthmatic effect (aminophylline as positive drug, 0.033 g/kg) was investigated by determining coughing latent period and the number of guinea pig with convulsive fall. The phenol red injection method was used to investigate phlegm-resolving effect (ambroxol as positive drug,21.304 g/kg) by determining the content of phenol red in the trachea of mice in phenol red expectoration test. The anti-inflammatory effect(dexamethasone acetate as positive drug,9.225×10-4 g/kg)was investigated by determining ear swelling degree and inhibition rate of ear swelling mice in xylene-induced inflammation test. RESULTS:Compared with blank control group, coughing latent period and cough induction latent period prolonged significantly in ethanol extract of F. cirrhosa and F. unibracteata groups,F. cirrhosa and F. unibracteata powder groups,positive control group, and coughing time decreased significantly within 3 min. The number of mice with convulsive fall decreased significantly in F. cirrhosa powder group, F. unibracteata ethanol group,F. unibracteata powder group and positive control group. The content of phenol red in the trachea of mice increased significantly,while ear swelling and inhibitory rate of ear swelling decreased significantly,with statistical significance (P<0.05 or P<0.01). There was no statistical significance in each index of mice between powder group and positive control group (P>0.05). Compared with corresponding groups of F. cirrhosa,coughing latent period of mice prolonged significantly in F. unibracteata ethanol extract group (P<0.05), other indexes had no statistical significance(P>0.05). CONCLUSIONS:The antitussive,antiasthmatic,phlegm-resolving and anti-inflammatory effects of F. cirrhosa are similar to or worse than those of F. unibracteata.

RESUMO

Objective To develop a method for the determination of total anthraquinones in Xiaofan lotion by HPLC.Methods Rhubarb anthraquinones were simultaneously separated and assayed on a Kromasil C18 column(250 mm×4.6 mm,5 μm) with the mobile phase of 0.1%phosphoric acid-methanol (15∶85) at 30 ℃.The detection wavelength was 207 nm.Results The linear relationship is good for Aloe-emodin and other five standard ingredients.The average recovery was 99.45%, RSD 1.68%.Conclusion The method is simple, rapid and accurate.It is suitable for determination of total anthraquinones in Xiaofan lotion.

Objective To study the curative effect of edaravone in the treatment of patients with transient cerebral ischemia and its mechanism.Methods 92 patients with TIA treated in our hospital from January 2015 to December 2015 were selected,they were randomly divided into observation group and control group,with 46 cases in each group.Both two groups were given conventional treatment,while the observation group added intravenous infusion of edaravone injection,two groups were treated for 2 weeks.Then,the efficacy and adverse reactions during treatment in two groups were compared.The levels of serum tumor necrosis factor alpha(TNF -α),transforming growth factor beta 1(TGF -β1)and matrix metalloproteinase -9(MMP -9)were detected by enzyme -linked immunosorbent assay(ELISA)in two groups before treatment and 1d,4d,7d,14d after treatment.The levels of high/low shear viscos-ity(HSV /LSV),plasma viscosity(PV),as well as fibrinogen(Fib)were also compared before treatment and 14d after treatment.Results The total effective rate of the observation group was 89.13%,which was significantly higher than 71.74% in the control group(χ2 =4.420,P <0.05).The levels of HSV,LSV,PV and Fib in two groups 14d after treatment were significantly lower than before treatment,which of the observation group were significantly lower than the control group,the differences were statistically significant(P <0.05).The serum levels of TNF -α[(5.99 ± 2.36)pg/mL,(5.82 ±2.34)pg/mL],TGF -β1 [(13.94 ±3.18)pg/L,(12.33 ±3.57)pg/L]and MMP -9 [(42.94 ±11.65)ng/mL,(38.26 ±17.58)ng/mL]in the observation group at 7d and 14d after treatment were sig-nificantly lower than before treatment,the differences were statistically significant (t =4.703,6.031,5.667;5.074, 7.623,6.150;all P <0.05),and which in the observation group were lower than the control group in the same peri-od,the differences were statistically significant(t =2.025,5.344,5.894;2.616,6.120,2.228;all P <0.05).The levels of above indicators 14d after treatment in the control group were significantly lower than before treatment,the differences were statistically significant(P <0.05).There were no serious adverse reactions in the two groups during the treatment.Conclusion Edaravone has obvious curative effect in the treatment of patients with TIA,which works by regulating the hemodynamic and the levels of TNF -α,TGF -β1 and MMP -9,it is safe and worthy of promotion.

Objective:To determin the effect of PLGA microspheres loading with PTH(1 34)[PTH(1 34)/PLGA]on the differentiation of MC3T3E1 cells.Methods:MC3T3E1 cells were divided into control group,continuous or intermittent PTH(1 34)adminstration groups,PLGA microsphere group and PTH(1 34)/PLGA group.Osteogenesis differentiation was observed by alkaline phosphatase activity(ALP),alizarin red staining and RTPCR.Results:The PTH(1 34)/PLGA with 1 0 -9 mol/L final release concentration enhanced ALP activity and mineralization,increased the mRNA expression of RUNX2,ALP and VEGF.Conclusion:Controlledrelease of PTH(1 34)from PLGA microspheres can promote the osteogenesis differentiation of MC3T3E1 cells.
