RESUMO

Objective@#To investigate blood pressure and vascular remodeling of OSAS by establishing the chronic-intermittent hypoxia model in rat.@*Methods@#Experiments were performed on 35 adult male Sprague-Dawley rats. Animals were randomly divided into four groups: unhandled control group (with 5 rats in it), CIH group at 9/6/3 weeks (with 10 ratsin each group). Rats in CIH group went through 8-hour intermittent hypoxia everyday, and those in control group were raising normally. After 9-week experiment, blood pressure was measured. The changes of the following indexes were observed: pathological changes of aorta and the middle aorta thickness (HE staining), the collagen of aorta wall (Masson staining). The experimental data were analyzed by SPSS 24.0 statistical software. The variance was analyzed by one-way analysis of variance, and the irregularity was selected using the calibration t test.@*Results@#The systolic and diastolic blood pressures of the CIH9, 6, and 3 weeks groups and the control group were: (127±13) and (79±9), (124±11) and (81±7), (101±11) and (75±9), (91±10) and (65±9) mmHg (1 mmHg=0.133 kPa). The systolic blood pressure and diastolic blood pressure of the rats in the week of CIH 9 and 6 weeks were significantly higher than the control group (F=14.64, P=0.000; F=6.81, P=0.000). There was no significant difference in the mean blood pressure between the three groups of CIH and the control group. Membrane thickness in CIH9, 6 and 3 weeks and control group were: (20±2), (19±2), (14±2), (13±3) μm. Compared with the control group, the aortic pathology and thickness of the middle layer of the CIH9 and 6 weeks group were significantly thicker (F=20.24, P=0.000), but there was no significant difference between the CIH3 week group and the control group; the collagen deposition was unchanged compared with the control group.@*Conclusion@#Intermittent hypoxia for 6 weeks or more in rats resulted in the increasement of blood pressure, morphological changes of aorta and vascular remodeling in thickened media.

RESUMO

BACKGROUND: Electro-acupuncture therapy shows good central and peripheral analgesic effects. Several studies have shown that transforming growth factor β (TGF-β) plays an important role in the pathogenesis of knee osteoarthritis. OBJECTIVE: To investigate the mechanism underlying electro-acupuncture treatment of knee osteoarthritis. METHODS: A total of 80 healthy male 3-month-old Sprague Dawley rats were randomly and evenly divided into normal, model, electro-acupuncture and drug groups. Rat models of knee osteoarthris were estbalished by ligating the femoral veins and forcing rats to do activies. At 1 month after knee osteoarthris induction, the electro-acupuncture group rats received electro-acupuncture therapy at two acupoints Neixiyan (EX-LE4) and Dubi (ST 35) with a depth of 0.1 cun (pulse 2 Hz, 20 minutes, once a day). The drug group rats were intraarticularly administerd sodium hyaluronate (0.1 mL/administration, once a week). After 2-week treatment, synovial tissue of the knee joint was harvested to determine the exprssion of TGF-β1, TGF-β1 receptor Ⅰand TGF-β1 receptor Ⅱ. RESULTS AND CONCLUSION: TGF-β1 expression in synovial tissue of the knee joint was significantly increased after knee osteoarthris (P < 0.05), but after electro-accupuncture therepy or sodium hyaluronate treatment, TGF-β1 expression was significantly decreased (P < 0.05), moreover, TGF-β1 receptor Ⅰ, Ⅱ expression was signficantly decreased (P < 0.05). These findings suggest that electro-acupuncture for treatment of knee osteoartheis improves the symptoms of osteoarthris by downregulating TGF-β1 expression, and reduction in TGF-β1 receptor Ⅰ, Ⅱ expression promotes the recovery of knee osteoarthris.

RESUMO

This study was designed to prove simultaneously blocking both EGFR and COX-2-medi-ated pathways may be an efficient means of inhibiting cancer cell growth in NPC. Method: A combination of tarceva (EGFR-selectivetyrosine kinase inhibitors) with celecoxib(Cox-2 inhibitor) was studied on its effects on cell growth, cell cycle progression, apoptosis and protein expression in CNE-2 cell lines by cell growth assay, flow cy-tometric analysis assay and Western blotting. Result: ①The inhibition rate of cell growth was higher in the group treated with a combination of two agents than that the sum of rates of the two groups treated with only one agent (P0.05). ③The group of combination showed less expressions of p-EGFR and COX-2 than any other group. Conclusion Simultaneously blocking EGFR and COX-2 mediated pathways would significantly in-hibit the growth of CNE-2 cell line, increase G_1 arrest and reduce the expression levels of p-EGFR and COX-2.
