RESUMO

Retinal vein occlusion(RVO), the second most prevalent retinal vascular disease, has complex pathophysiological mechanism. Except for mechanical pressure on blood vessel, inflammation and endothelin have been confirmed to be involved in the pathogenesis of RVO. However, its specific mechanism remains unclear. Hypertension, diabetes and dyslipidemia have been previously shown to be the most common risk factors in elder population, while recent studies found that coagulation and hemorheological abnormalities are more common in people under 50 years old. Ocular risk factors including glaucoma, high corrected intraocular pressure and retinal vessels abnormality, have gained more and more attention. These factors probably exert a synergistic effect when present simultaneously in the same patient. Therefore, early identification and intervention of those factors could lower the incidence of RVO. This article aims to review recent research and summarize existing mechanism and theories, giving some new research ideas for potential therapy targets and providing references for identification and management of risk factors.

RESUMO

Based on the systematic retrieval and the reported components of Sojae Semen Nigrum and Sojae Semen Praeparatum, this study conducted in-depth analysis of conversion of components in the fermentation process, and discussed types and possible mec-hanisms of conversion of chemical components, so as to provide the basis for studying technology, medicinal ingredients and quality standards. According to the analysis, there is a certain degree of conversion of nutrients(like protein, sugar, lipid), bioactive substances(like isoflavones, saponins, γ-aminobutyric acid) and other substances(like nucleosides, melanoids, biamines, etc) in the process of fermentation.

RESUMO

The historical evolution, fermentation technology and key links of Sojae Semen Praeparatum (SSP) were sorted out by consulting ancient books and modern literature, and the influencing factors and control methods of quality were analyzed and summarized in order to provide reference for the quality control of SSP. After analysis, it was found that in the fermentation process of SSP, fermentation strains, miscellaneous bacteria, temperature and humidity were all important factors affecting the quality of SSP. The condition control of "post fermentation" process has been paid more attention to in the past dynasties. In addition, the delicious SSP recognized in ancient times should be made from mold fermentation, and the breeding and application of fermented mold may be the key point to solve the quality problem of SSP. Therefore, based on the evaluation indexes of SSP in the past dynasties, it is of great significance to study and optimize the technological conditions such as strain, temperature and humidity in depth to improve the quality of SSP.

Objective:To explore<italic> </italic>the efficacy and mechanism of Danggui Shaoyaosan on rats of nonalcoholic fatty liver disease (NAFLD). Method:Sixrty SPF SD male rats were randomly divided into normal group, model group,essentiale (0.144 g·kg<sup>-1</sup>) and low, middle and high-dose of Danggui Shaoyaosan groups (2.44, 4.88, 9.76 g·kg<sup>-1</sup>). High fat diet were fed to bulid the NAFLD model, and each treatment group was given corresponding drugs at the same time. After 8 weeks, the serum and liver tissue were collected to detect the contents or activities of total cholesterol (TC), triglyceride (TG), alanine aminotransferase (ALT), aspartic acid aminotransferase (AST), tumor necrosis factor-<italic>α</italic> (TNF-<italic>α</italic>) and interleukin-10 (IL-10) in serum, the contents of TC, TG and free fatty acid (FFA) in liver tissue, Real-time fluorescence quantitative polymerase chain reaction(Real-time PCR) and Western blot were used to observe the gene and protein expressions of Toll-like receptor 4 (TLR4), myeloid different factory 88 (MyD88) and c-Jun n-terminal kinase (JNK) and the protein expression of phosphorylation JNK(p-JNK) in liver tissue. Hematoxylin-eosin (HE) staining and Oil red staining to observe the pathological morphological changes of liver. Result:Compared with control group, the contents or activities of TC, TG, ALT, AST and TNF-<italic>α</italic> in serum, the contents of TC, TG and FFA in liver and the gene and protein expressions of TLR4, MyD88, JNK, and the protein expression of p-JNK in liver tissue of model group were distinctly increased (<italic>P</italic><0.01), the content of IL-10 was significantly decreased (<italic>P</italic><0.01). Compared with model group, the contents or activities of TC, TG, ALT, AST and TNF-<italic>α</italic> in serum, the contents of TC, TG and FFA in liver and the mRNA and protein expressions of TLR4, MyD88 and JNK, and the protein expression of p-JNK in liver tissue of Danggui Shaoyaosan groups were significantly reduced (<italic>P</italic><0.05,<italic>P</italic><0.01), the content of IL-10 in serum of Danggui Shaoyaosan groups was distinctly increased(<italic>P</italic><0.05,<italic>P</italic><0.01), HE staining and Oil red staining show that the degree of liver steatosis was alleviated obviously by Danggui Shaoyaosan. Conclusion:Danggui Shaoyaosan has a better treatment on NAFLD by inhibiting TLR4/MyD88/JNK pathway and alleviating the inflammation response.

Objective: To explore related influence factors of the long-term curative effect of 3D conformal radiotherapy for patients with esophagus cancer. Methods: 472 patients with esophageal cancer were carried out simulative location by using CT, and they were implemented radical operative 3D conformal radiotherapy. And their basic documents and image documents were analyzed by using single factor analysis, and then these screened factors were token into Cox model to carry out multiple-factor analysis so as to seek the influence factor of long-term curative effect. Results: The 1 year, 3-year and 5-year survival rate of all of patients were 71.0%, 33.1% and 26.2%, respectively. The results of single factor analysis showed that series of factors, included parts of stratification, length of lesion, the longest diameter of the lesion, largest anteroposterior diameter of lesion, the largest transverse diameter of lesion, wall thickness of lesions, GTV-E, GTV-LN, GTV-T, whether out invasion, whether lymph node metastasis, lymph node metastasis model and the number of lymph nodes under X-ray, were significantly relative with long-term prognosis of patients. And then, the multiple-factor analysis of Cox model showed that the longest diameter of the lesion and GTV-T were the independent factor that can influence the long-term prognosis (OR=1.647, OR=1.379, P<0.05). Conclusion:GTV-T and the longest diameter of lesion on the CT image of patients with esophagus cancer can influence the long-term curative effect of 3D conformal radiotherapy for esophagus cancer. Therefore, they should be applied as the important evaluation index in formulating therapeutic schedule.

As an extension of proteomics, peptidomics has been widely used in medical and biological researches. However, the effect of reproducibility of identification method on peptidomics is not yet clear. In this work, the urine sample of a healthy people was analyzed for seven times in parallel by nano-liquid chromatography-high-resolution tandem mass spectrometry. To illustrate the variability and stability among these experiments, the number of spectra, the utilization of total spectra, the number of identified peptides, the number of proteins, and the ionic strength and retention time of peptides were counted and compared. The average number of peptides was 208 and the standard deviation was 38. 7. After all of data were combined, 426 peptides belonging to 114 proteins were obtained, while only 109 peptides coming from 35 proteins were identified in each experiment, indicating that there were both an randomness and a relative stability for LC-MS analysis. Increasing the number of parallel experiments would expand the data set of peptidomics, but the rate of increase would decrease over 3 or more measurements. Compared with peptides, the results of peptidomics were more stable at protein level, indicating that proteins were more robustly peptidomics biomarker than the peptides.

Objective: To optimize the purifying process of water-extraction solution of Xiaoyan Tuire Granules Oral Liquid by flocculation clarifying method, reserve more active components while less impurities, and improve the clarity of oral liquid. Methods: After clearing out the impurities composition of water-extraction solution of Xiaoyan Tuire Granules Oral Liquid, the flocculant of chitosan chosen by screening test was used to analyze the flocculation effect. The retention rate of aesculetin, the removal rate of impurity, and supernatant turbidity were set as mainly indexes to estimate the influence of the dosage of flocculant, temperature, fast mixing speed, and fast mixing time. Results: The optimal flocculating process conditions which were analyzed by experiments were as follows: the dosage of chitosan was 1.25 g/L, the flocculation temperature was 40 ℃, the fast mixing speed was 350 r/min, and the fast mixing time was 3 min. Under these conditions, the retention rate of aesculetin was 88.92%, the removal rates of protein and tanning were 62.96% and 67.19%, the supernatant turbidity was 5.0 NTU after the oral liquid was flocculated for 24 h. Conclusion: Using chitosan as flocculant in the water-extraction solution of Xiaoyan Tuire Granules Oral Liquid, it could improve the removal rate of impurities effeitively as well as keep higher active components and enhance the clarity of oral liquid.

<p><b>BACKGROUND</b>Acute lung injury (ALI) is a common syndrome associated with high morbidity and mortality in emergency medicine. Cell apoptosis plays a key role in the pathogenesis of ALI. Hydrogen sulfide (H(2)S) plays a protective role during acute lung injury. We designed this study to examine the role of H(2)S in the lung alveolar epithelial cell apoptosis in rats with ALI.</p><p><b>METHODS</b>Sixty-nine male Sprague Dawley rats were used. ALI was induced by intra-tail vein injection of oleic acid (OA). NaHS solution was injected intraperitonally 30 minutes before OA injection as the NaHS pretreatment group. Single sodium hydrosulfide pretreatment group and control group were designed. Index of quantitative assessment (IQA), wet/dry weight (W/D) ratio and the percentage of polymorphonuclear leukocyte (PMN) cells in the bronchoalveolar lavage fluid (BALF) were determined. H(2)S level in lung tissue was measured by a sensitive sulphur electrode. Apoptosis was evaluated by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining and Fas protein was measured by immunohistochemical staining.</p><p><b>RESULTS</b>The level of endogenous H(2)S in lung tissue decreased with the development of ALI induced by OA injection. Apoptosis and Fas protein in alveolar epithelial cells increased in the ALI of rats but NaHS lessened apoptosis and Fas protein expression in alveolar epithelial cells of rats with ALI.</p><p><b>CONCLUSION</b>Endogenous H(2)S protects rats from oleic acid-induced ALI, probably by inhibiting cell apoptosis.</p>

<p><b>OBJECTIVE</b>To immunopurify human endometrial endothelial cells (HEEC) from fresh surgical specimens of endometrial cancers and normal endometrial tissues, and investigate their biological characteristics.</p><p><b>METHODS</b>Endothelial cells of endometrial cancers and normal endometrial tissues were isolated using anti-CD31 conjugated magnetic microbeads. The isolated endothelial cells were cultured in vitro and their origins were identified. Their angiogenic characteristics were observed by MTT, wound healing, Transwell cell invasion and tube formation assays.</p><p><b>RESULTS</b>Flow cytometry revealed that the immunopurification technique yielded endothelial cell purity of > 95% in all samples. All purified HEEC were characterized as endothelial cells on the basis of expression of the classical endothelial markers vWF and CD31 as shown by immunofluorescence examination. Although the tumor-associated HEEC didn't show more rapid proliferation than normal HEEC, they exhibited enhanced migration ability (P = 0.006), potent invasiveness (P = 0.033), and elevated tube formation in vitro (P = 0.029).</p><p><b>CONCLUSIONS</b>Human endometrial endothelial cells can be efficiently isolated from endometrial cancer and normal endometrial tissues by immunomagnetic methods. Tumor-associated HEEC exhibit enhanced migratory ability, potent invasiveness, and elevated tube formation in vitro.</p>

Objective To know the application effects of two different hand prepping by avagard before the operation.Methods Divided operation-actors into the experimental group (121 cases) and the control group (116 cases)randomly.Kneading hand prepping by 9200 and 9230 avagard was used in the experimental group, while the brushing by 9241- D and 9250 was used in the control group.Compared the effects between the two groups.Results There was no significant difference about disinfection effects between the two groups, while the compliance of kneading hand prepping was more better than the other hand prepping method.Condusions Kneading hand prepping by avagard 9200 should be used before surgical operation.

Objective To investigate the present status of Kaschin-Beck disease(KBD) in Qingzhou at present, and provide a scientific basis for decision-making in controlling the disease. Methods According to The National Technical Scheme for Endemic Disease Control in 2007, children aged 7 to 12 were chosen to receive clinical and X-ray examination in 3 villages of 2 towns in Qingzhou. Local adults aged 16 years and older were clinically diagnosed in 10 villages of 4 towns and divided into different groups according to the clinical degrees and their ages. The Diagnostic Criteria of Kashin Beck Disease(GB 16003-1995) was carded out by clinical and X-ray diagnosis. Results Two hundred and sixty school children aged 7 to 12 were clinically diagnosed, and the patients of I degree upwards of KBD were not detected. One hundred and ninety-eight school children aged 7 to 12 were examined by X-ray, and the detectable rate was 0(0/198). In 7099 adults aged 16 years old upwards clinically tested, 502 patients of degree I and higher of KBD were detected and the detectable rate was 7.07%(502/7099). The patients distributed mainly in the population aged 36 years old and older, accounting for 99.00% (497/502). Conclusions Although KBD in children have been controlled in Qingzhou, it is still serious in adults. So the monitoring of KBD should be carried on and prevention and control for KBD in adults should be strengthened.

<p><b>OBJECTIVE</b>To investigate prognostic factors related to early and late intrahepatic recurrences after curative hepatectomy for patients with hepatocellular carcinoma (HCC).</p><p><b>METHODS</b>A retrospective review was conducted on medical records of patients with HCC treated by curative hepatectomy from January 2002 to January 2009. Clinicopathologic data were evaluated for their possible association with postoperative intrahepatic recurrence in univariate and multivariate analysis using Cox proportional hazard model. Recurrence time calculated by Kaplan-Meier method was compared using Log-rank test. Receiver operator characteristic curve (ROC) analysis with calculation of the area under the curve (AUC), sensitivity, and specificity where appropriated and risk stratification were applied to assess predictive ability of prognostic factors.</p><p><b>RESULTS</b>All 101 patients underwent curative hepatectomy. During follow-up period, 75 patients developed postoperative intrahepatic recurrence, among whom, 63 experienced early recurrence (84.0%) and the remaining had late recurrence (16.0%). The 1-, 2-, 3-and 5-year cumulative recurrent rates were 48.5% (49/101), 62.4% (63/101), 70.3% (71/101) and 74.3% (75/101), respectively. Multivariate analysis identified that tumor residual resectional margin, increased BCLC staging and severity of concomitant liver cirrhosis as independent prognostic factors predicting early recurrence while age ≥ 60 years and presence of tumor capsule predicting late recurrence. Cutoff point values (PI ≥ 2.798) predicted early recurrence with AUC 0.897 (95%CI = 0.829 - 0.965), sensitivity 76.6%and specificity 88.9% calculated from ROC. Median recurrent time of early recurrence and late recurrence reached statistically difference after risk stratification, 20.2 months vs. 4.4 months (χ(2) = 29.198, P = 0.000), 46.6 months vs. 28.6 months (Log-rank test, χ(2) = 8.479, P = 0.004), respectively.</p><p><b>CONCLUSIONS</b>Postoperative recurrence for HCC after curative hepatectomy should be differentiated as early recurrence and late recurrence, since each is associated with different risk factors, indicating possible different mechanism responsible for postoperative recurrence. Risk stratification can be used for prediction of different type of recurrence.</p>

The hemagglutinin (HA) gene from H5N1 avian influenza virus and the chicken interleukin 2 (chiIL-2) gene were inserted into a expressing vector p12LS to construct a recombinant transferring vector p12LSH5AIL2, in which HA gene under the control of the promoter Ps was in inverse tandem connection with the chiIL-2 gene under the control of the promoter PE/L. The p12LSH5AIL2 was then used to transfect the chicken embryo fibroblasts (CEF) pre-infected with a wild-type fowlpox virus 282E4 strain, to generate a recombinant fowlpox virus coexpressing the inserted HA and chiIL2 genes (rFPV-H5AIL2). The rFPV-H5AIL2 was obtained and purified by blue plaque screening on the CEF. The in vitro expression of HA gene by rFPV-H5AIL2 was detected in the recombinant fowlpox virus-infected CEFs with an indirect immunofluorescence assay, and the expression of the chiIL2 gene by rFPV-H5AIL2 was confirmed by detection of the chiIL2 mRNA by RT-PCR and by detection of chiIL2 by the indirect immunofluorescence assay. Experiments on SPF and commercial chickens demonstrated that the titer for HI antibodies induced by the rFPV-H5AIL2 was significantly higher than that by the rFPV-HA. The group immunized with the rFPV-H5AIL2 exhibited the similar ratios of protective efficacy and virus shedding as the group immunized with the rFPV-HA in SPF chicken. However, in commercial chicken, the group immunized with the rFPV-H5AIL2 generated significantly higher protection against H5N1 avian influenza virus challenge and lower virus shedding than the group immunized with the rFPV-HA. This study paved the way for further development of a new AIV recombinant vaccine.

<p><b>OBJECTIVE</b>To observe the differentiation and distribution of epidermal stem cell (ESC) after skin soft tissue expansion, and to initially probe into the growth mechanism of expanded skin tissue.</p><p><b>METHODS</b>Samples of normal skin and expanded skin (mean effusion period 45 days) were harvested from head and cervical region in 15 patients who underwent II stage surgery after skin expansion. Samples were divided into scalp adjacent to the center of expander group (expanded scalp, 3 cm from the vertical axis of the expander), scalp from lateral part of the expander group (expanded scalp, 5 - 7 cm lateral to the vertical axis of the expander), cervical skin expansion group, un-expanded scalp control group, and un-expanded cervical skin control group, according to the position of skin harvested. The tissue structure of skin in each group was observed with HE staining, and the differentiation and distribution characteristics of cytokeratin 19 (CK19) positive cells were observed with immunohistochemical staining.</p><p><b>RESULTS</b>Compared with those in the un-expanded control groups, uneven, relatively thickened and obviously folded epidermis with more cell layers and cells with obvious aggregation close to the basal layer were observed in the expanded groups, but those cells were not well-arranged and the transition of polarity was not obvious. The continuity of CK19 positive cells in the basal layer of skin was observed in each of the expanded group with immunohistochemical staining, and positive cells increased obviously and arranged in multilayer in certain parts of basal layer. Clustered or dispersed CK19 positive cells were also observed outside the basal layer. No above-mentioned phenomenon was observed in the un-expanded control group.</p><p><b>CONCLUSIONS</b>The proliferation and differentiation of ESC with ectopic distribution may enhance the repair process after skin soft tissue expansion.</p>

<p><b>OBJECTIVE</b>To research the origin of Zibei Xiqi which included in Volume 3 of < Bencao Gangmu Shiyi > (A supplement to the compendium of materia medica).</p><p><b>METHOD</b>The origin of Zibei Xiqi was confirmed with textural, botanical characteristics research and drug name, therapeutic effects and so on.</p><p><b>RESULT</b>The origin of Zibei Xiqi was confirmed.</p><p><b>CONCLUSION</b>The Zibei Xiqi which is recorded in < Bencao Gangmu Shiyi > should be the immature herbs of Shuang Hudie [Tripterospermum chinense, Gentianaceae], and suggest to correct the original records of the drug name.</p>

RESUMO

To investigate the historical origins of folk herbal medicine 'Xuelihua', the origin of 'Xuelihua' was confirmed by textural, botanical characteristics research, investigation of therapeutic effects and field survey. The 'Xuelihua' which was originally recorded in 'Bencao Gangmu Shiyi' (A Supplement to the Compendium of Materia Medica) should be 'Riben Shegencao' (Ophirrhiza japonica) of Rubiaceae family.

RESUMO

<p><b>OBJECTIVE</b>To observe the distribution of epidermal stem cell (ESC) after soft tissue expansion, and to explore dynamic change in ESC under mechanical stress and kinetic mechanism of skin expansion.</p><p><b>METHODS</b>Skin samples were collected from patients after expansion of the scalp. They were divided into three groups: A group (scalp harvested 3 cm away from the center of dilator), B group (scalp tissues at the edge of dilator), and control group (scalp without dilatation). The tissue structures were observed with optical microscope with HE staining. The distribution and differentiation characteristics of cell keratin 19 (CK19) positive cells were observed with inverted phase contrast microscope after immunohistochemistry staining.</p><p><b>RESULTS</b>HE staining showed that the epidermis was thickened and distributed densely with uneven, rugged and increased layers in A, B groups. With immunohistochemistry staining, CK19 positive cells appeared in multilayers in basal membrane, a few of them were in cluster or dispersed , with" hollowing" structure formation. These phenomena were not seen in control group.</p><p><b>CONCLUSION</b>ESC can proliferate with abnormal distribution and "hollowing" structure formation after mechanical dilatation, which may be related to dynamic changes in basal layer cells.</p>

RESUMO

<p><b>OBJECTIVE</b>To isolate and determine the chemical constituents from Ranunculus japonicus.</p><p><b>METHOD</b>Compounds were isolated by column chromatography, and identified by MS, NMR data.</p><p><b>RESULT</b>Nine compounds were obtained andelucidated as scoparone (1), tricin (2), protocatechuic acid (3), luteolin (4), anemonin (5), scopoletin (6), 5-hydroxy-6, 7-dimethoxyflavone (7), 5-hydroxy-7, 8-dimethoxyflavone (8), ternatolide (9), respectively.</p><p><b>CONCLUSION</b>Compounds 1-9 were isolated from R. japonicus for the first time while compounds 7, 8 were separated from Ranunculus genus first time.</p>

RESUMO

<p><b>OBJECTIVE</b>To establish an porcine model of whole pancreaticoduodenal transplantation with portal venous drainage and enteric drainage for ensuring physiologically normal function without hyperinsulinemia and reducing postoperative complications.</p><p><b>METHODS</b>Twenty sichuan native outbreding white pigs weighing 25-30 kg were divided equally into two groups to serve as the donors and recipients. Cooling of the grafts was accomplished with in situ flush with 4 degrees C UW preservation solution via an aortic cannula. A whole pancreatoduodenal graft with the segment of abdominal aorta and the portal vein was harvested from the donor pigs. Type I diabetes model was established by complete removal of the recipient pancreas. The whole pancreatoduodenal graft was preserved and shaped in UW solution, and the subphrenic abdominal aorta of the recipient was joined with the donor abdominal aorta via a side-to-end anastomosis, and venous reflux was reconstructed between the donor portal vein and the recipient superior mesenteric vein. Side-to-side intestinal anastomosis was performed between the donor duodenum and the recipient jejunum.</p><p><b>RESULTS</b>Ten pancreaticoduodenal transplantations (PVE+ED style) were done, and pancreatic graft thrombosis and embolism occurred only in 1 pig 6 days after transplantation.</p><p><b>CONCLUSION</b>The model of whole pancreaticoduodenal transplantation with portal venous drainage and enteric drainage is stable and reliable.</p>
