RESUMO

The effective components of flavonoids in the "Pueraria lobata-Hovenia dulcis" drug pair have low bioavailability in vivo due to their unstable characteristics. This study used microemulsions with amphoteric carrier properties to solve this problem. The study drew pseudo-ternary phase diagrams through titration compatibility experiments of the oil phase with emulsifiers and co-emulsifiers and screened the prescription composition of blank microemulsions. The study used average particle size and PDI as evaluation indicators, and the central composite design-response surface method(CCD-RSM) was used to optimize the prescription; high-dosage drug-loaded microemulsions were obtained, and their physicochemical properties, appearance, and stability were evaluated. The results showed that when ethyl butyrate was used as the oil phase, polysorbate 80(tween 80) as the surfactant, and anhydrous ethanol as the cosurfactant, the maximum microemulsion area was obtained. When the difference in results was small, K_(m )of 1∶4 was chosen to ensure the safety of the prescription. The prescription composition optimized by the CCD-RSM was ethyl butyrate(16.28%), tween 80(9.59%), and anhydrous ethanol(38.34%). When the dosage reached 3% of the system mass, the total flavonoid microemulsion prepared had a clear and transparent appearance, with average particle size, PDI, and potential of(74.25±1.58)nm, 0.277±0.043, and(-0.08±0.07) mV, respectively. The microemulsion was spherical and evenly distributed under transmission electron microscopy. The centrifugal stability and temperature stability were good, and there was no layering or demulsification phenomenon, which significantly improved the in vitro dissolution of total flavonoids.

RESUMO

ObjectiveTo investigate the effects of hydroxycamptothecin liposomes (LHCPT) on myocardial fibrosis in rats with heart failure by regulating the sphingosine kinase 1 (SphK1)/sphingosine-1-phosphate (S1P) signaling pathway. MethodsSD rats were divided into control group, model group, hydroxycamptothecin (HCPT) group, LHCPT group, captopril group, and LHCPT+K6PC-5 (SphK1 activator) group, with 12 rats in each group. The heart failure rat models in all groups except the control group were established by intraperitoneal injection of doxorubicin and then the corresponding drugs were given once a day. After four weeks, we applied color Doppler ultrasound to detect left ventricular end systolic diameter (LVESD), left ventricular end diastolic diameter (LVEDD), and left ventricular ejection fraction (LVEF) in rats; HE and Masson staining for myocardial pathological damage and myocardial fibrosis in rats, respectively; ELISA method for the levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in rat myocardial tissues; qRT-PCR for the expression of transforming growth factor-β1 (TGF-β1), type I collagen (Collagen I), and type Ⅲ collagen (Collagen Ⅲ) in rat myocardial tissues; Western blot for the expression of SphK1 and S1P proteins in rat myocardial tissues. ResultsCompared with the control group, the model group showed severe myocardial pathological damage and myocardial fibrosis, increased LVESD, LVEDD, levels of TNF-α and IL-6, expression of TGF-β1, Collagen I, Collagen Ⅲ, SphK1, S1P and decreased LVEF (P<0.05). Compared with the model group, the HCPT group, LHCPT group and captopril group showed alleviated myocardial pathological damage and myocardial fibrosis, decreased LVESD, LVEDD, levels of TNF-α and IL-6, expression of TGF-β1, Collagen I, Collagen Ⅲ, SphK1, S1P and increased LVEF (P<0.05). Compared with the LHCPT group, the LHCPT+K6PC-5 group showed aggravated myocardial pathological damage and myocardial fibrosis, increased LVESD, LVEDD, levels of TNF-α and IL-6, expression of TGF-β1, Collagen I, Collagen Ⅲ, SphK1, S1P and decreased LVEF (P<0.05). RESUMO

Circadian rhythm is an internal regulatory mechanism formed in organisms in response to the circadian periodicity in the environment, which modulates the pathophysiological events, occurrence and development of diseases, and the response to treatment in mammals. It significantly influences the susceptibility, injury, and recovery of ischemic stroke, and the response to therapy. Accumulating evidence indicates that circadian rhythms not only regulate the important physiological factors of ischemic stroke events, such as blood pressure and coagulation-fibrinolysis system, but also participate in the immuno-inflammatory reaction mediated by glial cells and peripheral immune cells after ischemic injury and the regulation of neurovascular unit(NVU). This article aims to link molecular, cellular, and physiological pathways in circadian biology to the clinical consequences of ischemic stroke and to illustrate the impact of circadian rhythms on ischemic stroke pathogenesis, the regulation of NVU, and the immuno-inflammatory responses. The regulation of circadian rhythm by traditional Chinese medicine is reviewed, and the research progress of traditional Chinese medicine intervention in circadian rhythm is summarized to provide a reasonable and valuable reference for the follow-up traditional Chinese medicine research and molecular mechanism research of circadian rhythm.

RESUMO

Neuroblastoma (NB) is the most common extracranial solid tumor in children and has the features of high recurrence rate and low survival rate, and therefore, early diagnosis, treatment response evaluation, and recurrence monitoring are of great significance for NB patients. Liquid biopsy refers to the detection of cells and nucleic acids in fluid specimens, mainly blood. It is noninvasive and can overcome tumor heterogeneity, thus making it possible to achieve the early diagnosis and dynamic detection of NB. This review introduces the latest advances in clinical research on the application of liquid biopsy in NB.

RESUMO

OBJECTIVE@#To construct cytarabine-resistant acute myeloid leukemia (AML) cell lines, and explore the correlation between Sirt1, PGC-1α expression levels and drug resistance.@*METHODS@#Human acute promyelocytic leukemia Kasumi-1 cells were induced by the method of gradually increasing the concentration of Ara-C drug. The IC50 value of Kasumi-1 cells before and after drug addition was detected by CCK-8 method, so as to construct Ara-C resistant cell lines. The expression levels of Sirt1 and PGC-1α mRNA in Kasumi-1 drug-resistant cell lines and their parental cell lines were detected by real-time fluorescence quantitative PCR, and the expression levels of Sirt1 and PGC-1α protein in kasumi-1 drug-resistant cell lines and their parental cell lines were detected by Western blot.@*RESULTS@#The constructed Kasumi-1 cell line had common morphological characteristics of drug-resistant cell lines under microscope, and the drug resistance index was greater than 5, indicating that Kasumi-1 drug-resistant cells had good drug resistance after the construction. The RT-qPCR and Western blot assays showed that the expression levels of Sirt1 and PGC-1α mRNA and protein in the drug-resistant cell lines were higher than those of the parental cell lines (P<0.001).@*CONCLUSION@#AML cell lines resistant to Ara-C can be successfully induced by the method of gradually increasing the concentration, and the co-high expression of Sirt1 and PGC-1α may mediate the drug resistance of AML cells.

RESUMO

Coumarins are the main active components in Psoraleae Fructus. To study the multi-component pharmacokinetics of Psoraleae Fructus, this study established a sensitive and rapid ultra-pressure liquid chromatography coupled to tandem mass spectrometry(UPLC-MS/MS) method for simultaneous determination of psoralen, isopsoralen, psoralenoside, and isopsoralenoside in rat plasma. After validation, the method was applied to the investigation of pharmacokinetics of psoralen, isopsoralen, psoralenoside, and isopso-ralenoside in rats after single and multiple administration of Psoraleae Fructus extract. The results revealed that the exposure of psoralen and isopsoralen in rat plasma was high after a single intragastric administration of Psoraleae Fructus extract, with an AUC_(0-∞) of 443 619-582 680 and 167 314-276 903 ng·mL~(-1)·h~(-1), respectively. Compared with these two compounds, the exposure of psoralenoside and isopsoralenoside was lower with marked gender difference. After 7-day administration of Psoraleae Fructus extract to rats, the AUC_(0-∞) of psoralen and isopsoralen was 29 701-81 783 and 39 234-89 914 ng·mL~(-1)·h~(-1), respectively, which was significantly lower than that at the first day(P<0.05), and that of psoralenoside and isopsoralenoside was 7 360-19 342 and 8 823-45 501 ng·mL~(-1)·h~(-1), respectively. There was no significant gender difference in exposure of psoralenoside and isopsoralenoside in male and female rats. However, the exposure of psoralenoside and isopsoralenoside in male rats was reduced(P<0.05), and the t_(1/2) and mean residence time(MRT) were shortened, suggesting that the removal of these two compounds from the body was accelerated.

RESUMO

Objective: To define the current status and analyze the medical quality of interventional therapy for patients with atrial fibrillation (AF) in China. Methods: This survey was performed in all seven large regions of China, one to three regional major medical centers were selected from each region. Medical records of patients underwent interventional therapy for AF in the year 2017 were randomly inspected. CHA2DS2-VASc score, prescribed anticoagulant after ablation, indication of left atrial appendage occlusion (LAAO), and complications in the medical records were analyzed. Results: A total of 10 800 AF catheter ablations and 447 LAAOs were performed in 17 regional medical centers in 2017. There were 10/17 centers performing AF catheter ablation<500 cases and 7/17 centers performing LAAO<20 cases. A total of 1 347 cases of catheter ablation and 160 cases of LAAO were selected for further analysis. Among all selected cases, 15.8% (238/1 505) non-valvar AF cases recorded CHA2DS2-VASc scores. The anticoagulation rate after AF catheter ablation was 98.6% (1 328/1 347), anticoagulation rate was higher than 90% in 16 out of 17 centers. The complication and severe complication rates of AF catheter ablation were 0.9% (12/1 347) and 0.4% (5/1 347), respectively. The differences of complication and severe complication rates in AF catheter ablation were similar between centers performing<500 cases and centers performing ≥500 cases (0.5% (2/413) vs. 1.1% (10/934), P>0.05; 0.5% (2/413) vs. 0.3% (3/934), P>0.05). The coincidence rate of LAAO indication was 81.3% (130/160), and the rate was higher in center performing ≥20 cases than in centers performing<20 cases (84.8% (106/125) vs. 68.6% (24/35), P<0.05). The complication and severe complication rates of LAAO were 3.1% (5/160) and 1.9% (3/160). The rate of complications in LAAO was higher in center performing<20 cases than in centers performing ≥20 cases (8.6% (3/35) vs. 1.6% (2/125), P<0.05), and there was no significant difference in severe complication rate (5.7% (2/35) vs. 0.8% (1/125), P>0.05). Conclusions: Interventional therapy for AF in China is generally standardized and safe. The overall incidence of complications post AF interventional ablation is low, the anticoagulation rate after AF catheter ablation is high, and the adherence rate of LAAO indication is fair. The indicators mentioned above vary widely among centers.

RESUMO

Objective::To investigate the effects of plant-soil feedback on secondary metabolites in roots, stems and leaves of Acanthopanax senticosus seedlings. Method::One-year-old seedlings of A. senticosus were planted in the soil where no A. senticosus had been planted before (group 1), soil where A. senticosus had been planted for 3 years (group 2), and soil where A. senticosus had been planted for many years in the greenhouse pot experiment, and the secondary metabolites of its roots, stems and leaves were then analyzed. Result::L-Phenylalanine, 3, 4-dihydroxybenzoic acid, syringin, chlorogenic acid, caffeic acid, eleutheroside E, isofraxidin, rutin, hyperoside, and quercetin had significant differences in leaves and roots of A. senticosus seedlings in the soil of group 3, but there was no significant difference in chlorogenic acid and eleutheroside E in stem. Eleutheroside E, isofraxidin, rutin and hyperoside were not detected in the leaves of seedlings planted in group 3.Most of the secondary metabolites in the roots of A. senticosus seedlings showed positive feedback, while in the stems of Acanthopanax senticosus seedlings, caffeine, A. senticosus glycosides, hypericin and quercetin showed negative feedback, and most of the secondary metabolites in the leaves of A. senticosus seedlings showed positive feedback. Conclusion::The plant and soil showed different feedback in different parts of the growth process of A. senticosus seedlings, and the soil where A. senticosus had not been planted was more advantageous to the secondary metabolites of A. senticosus seedlings. The results of the study provide a basis for the study of the effect of plant-soil feedback on the A. senticosus, and provide the theoretical basis and technical support for the artificial cultivation of A. senticosus.

Method::Through the greenhouse pot experiment, plant height, leaf color value (SPAD), antioxidant enzymes [protein, superoxide dismutase(SOD), catalase(CAT), peroxidase(POD), aseorbateperoxidase(APX), malondialdehyde(MDA)] and yield and growth related indexes of soil without A. senticosus (group 1), soil with A. senticosus (group 2) for three consecutive years and soil with A. senticosus (group 3) for many years were measured respectively. Result::There were significant differences in plant height, SPAD, protein, SOD, CAT, POD, APX and MDA between seedlings of A. senticosus planted for three consecutive years (group 1), two successive years (group 2) and three successive years (group 3). The biomass, MDA, CAT, POD and SOD of A. senticosus seedlings in the soil without A. senticosus (group 1) were higher than those in the soil with A. senticosus (group 2 and 3), while the protein and APX were lower than those in the soil with A. senticosus (group 2 and 3). Conclusion::Plant and soil shows negative feedback regulation during the growth of A. senticosus seedlings, which reduced the activity of antioxidant enzymes. Therefore, the soil without planting A. senticosus has more advantages for the growth of A. senticosus seedlings. The results provide a basis for explaining the effect of plant-soil feedback on the growth of A. senticosus, and a theoretical basis and technical support for the technical standards of A. senticosus cultivation in farmland.

RESUMO

Objective::To study the effect of different N application rates on the growth and development of Acanthopanax senticosus and the changes of antioxidant enzyme activity, and screen out the suitable amount of nitrogen fertilizer for its growth and development, in order to provide scientific evidence for rational fertilization of Acanthopanax senticosus in artificial cultivation. Method::By single-point, single-factor field experiment, the study samples were one-year-old seedlings of growing evenly A. senticosus.Five nitrogen application treatment groups were set up in the fields. They were N1 (30 g·m-2), N2 (60 g·m-2), N3 (90 g·m-2), N4 (120 g·m-2), N5 (150 g·m-2) and CK (0 g·m-2) in the control group.Three months later, the raw weight of plant, root, leaf and stem were measured at harvest time. After drying to constant weight, plant dry weight, stem dry weight, leaf dry weight and root dry weight were measured.Fresh leaves of plants were collected to measure malondialdehyde(MDA) content and activities of catalase(CAT), superoxide dismutase(SOD), peroxidase(POD), acid phosphatase(ACP) and aseorbateperoxidase(APX) after harvesting seedlings. Result::The biomass of A. senticosus in group N4 (120 g·m-2) was the highest, the protein content in group N3 (90 g·m-2) was the highest, and the activity of all antioxidant enzymes in group N3 (90 g·m-2) was the lowest. Conclusion::There is a dose-effect relationship between seedlings and the nitrogen application rate. That is to say, low nitrogen application rate and high nitrogen application rate will cause stress on Acanthopanax senticosus, and the activity of antioxidant enzymes under low nitrogen application rate is higher than that under high nitrogen application rate.

RESUMO

Objective::To study the effect of different nitrogen application rates on the growth and carbon metabolism of Acanthopanax senticosus seedlings, and screen out the rational fertilization conditions, in order to provide basis and guidance for scientific fertilization of artificially cultivated A. senticosus. Method::A single-point, single-factor field experiment was conducted to study the seedlings of growing evenly A. senticosus.Five different nitrogen application treatment groups were set up to treat the seedlings, namely N1 group (30 g·m-2), N2 group (60 g·m-2), N3 group (90 g·m-2), N4 group (120 g·m-2), N5 group (150 g·m-2) and CK group (0 g·m-2), respectively.Three months later, plant height, root circumference, stem circumference, root-shoot ratio and SPAD were measured at harvest time. The contents of protein, sucrose, starch, soluble sugar and reducing sugar in fresh leaves were measured. Result::N3 treatment was the best treatment method for the growth and development of A. senticosus seedlings, and the growth of A. senticosus seedlings was the best under this treatment. The protein content of A. senticosus seedlings in N3 treatment was the highest.Starch and sucrose were best accumulated in N5 treatment group and CK treatment group.N5 treatment had the highest soluble sugar content and reducing sugar content. Conclusion::There is a dose-effect relationship between the growth and development of A. senticosus seedlings; that is to say, low and high nitrogen application treatments will cause stress on A. senticosus seedlings. In conclusion, the suitable nitrogen application rate for A. senticosus growth is 90-120 g·m-2.

RESUMO

Cerebral infarction is a clinical disease with corresponding neurological symptoms caused by cerebral ischemia and hypoxia caused by cerebral blood supply disorder. It is one of the most common cerebrovascular diseases and a serious threat to human health. The prevention and treatment of cerebral infarction has an important social significance. Angiogenesis is the key starting point for medical treatment of cerebral infarction, and signal transduction and transcriptional activators (STAT)/hypoxia inducing factor-1(HIF-1)/vascular endothelial growth factor (VEGF) pathway are important pathways to mediate angiogenesis after cerebral infarction. This paper took the angiogenesis as the starting point and the upstream molecules of STAT/HIF-1/VEGF signal pathway STAT3 and miRNA as the main study objects, and comprehensively discussed the results of chip sequencing, experimental research, traditional Chinese medicine (TCM) pathogenesis and TCM treatment. Based on the regulatory mode of "TF-miRNA" and the idea of "micro-whole", it is suggested that under the guidance of the basic theory of TCM, cubic compound prescriptions of TCM and its active components might activate the STAT/HIF-1/VEGF signal pathway through STAT3/miRNA feedback loop to promote angiogenesis after cerebral infarction, which puts forward a deep molecular mechanism and new direction for the treatment of cerebral infarction with TCM.
