RESUMO

Objective @#To explore the effect of ATP binding cassette subfamily A member 1 (ABCA1) on the inva- sion,migration and epithelial-mesenchymal transition(EMT) of human gastric cancer and investigate the underlying mechanism.@*Methods @#The expression of ABCA1 in human gastric cancer and its relationship with the prognosis of gastric cancer patients were analyzed by Bioinformatics analysisbased on network databases. RT-qPCR was per- formed to detect the expression of ABCA1 in 25 pairs of clinical gastric cancer tissue samples ; RT-qPCR and West- ern blot were used to analyze the expression of ABCA1 in different human gastric cancer cell lines ; Gene silencing technology was used to construct stable low-expressing ABCA1 gastric cancer cell line.Cell migration and invasion were determined by transwell assay ; Western blot was used to detect the expression of related marker proteins in EMT and epidermal growth factor receptor (EGFR) / signal transducer and activator of transcription(STAT3) signa- ling pathway. @*Results @#The results of bioinformatics analysis indicated that ABCA1 was highly expressed in gastric cancer,which was positively correlated with advanced grade and stage as well as the poor prognosis of gastric canc- er patients. RT-qPCR results showed that ABCA1 was highly expressed in gastric cancer tissues and gastric cancer cells HGC-27 ; The results of Transwell indicated that knockdown of ABCA1 could inhibit the invasion and migra- tion of HGC-27 cells.Western blot results showed that the expression of mesenchymal cell marker proteins (Vimen- tin,N-Cadherin) and EMT-related transcription factors (Twist,Zeb1 and Snail) in the ABCA1 knockdown group was down-regulated,while the expression of epithelial cell marker protein E-Cadherin was up-regulated.Western blot results showed that compared with the control group,knockdown of ABCA1 inhibited the expression of EGFR downstream molecule STAT3,and reduced the phosphorylation level of EGFR and its downstream molecule STAT3 . @*Conclusion @#ABCA1 is up-regulated in gastric cancer and is correlated with cell invasion,migration,EMT and in- dicate poor prognosis of gastric cancer patients.ABCA1 may affect the expression and activation of STAT3 by regu- lating EGFR , thereby inhibiting the invasion,migration and EMT of gastric cancer cells.

Aim To study the effects of total saponins of Panax japonicus(TSPJ) on liver inflammation of natural aging rats.Methods The experimental rats were allocated into seven groups (twelve rats in each group): three months group, nine months group, fifteen months group, twenty-four months group, and TSPJ low-, mid-and high-dose groups(10, 30, 60 mg·kg-1).When the rats were eighteen months old, the TSPJ low-, mid-and high-dose groups of rats were given lavage treatments with TSPJ 10,30, 60 mg·kg-1 respectively until twenty-four months.During lavage, we stopped a day every week for six consecutive months.HE staining was used to observe the pathological morphological changes.Western blot was utilized to test IL-1β and TNF-α protein expressions.RT-PCR method was adopted to test IL-1β, IL-6, IL-12, IL-17α, TNF-α, IFN-γ mRNA expressions.Results HE staining observation showed that as the rats grew older the hepatic cord and sinusoid were arranged in more severe disorder, and the fat vacuole and inflammatory cells were increased significantly.While every dose group of TSPJ could improve these pathological changes distinctly.The IL-1β, TNF-α protein and IL-1β, IL-6, IL-12, IL-17α, TNF-α, IFN-γ mRNA expressions were increased gradually as the rats grew older, and every dose group of TSPJ could reduce their expressions to some extent.Conclusion TSPJ could protect the aging rat liver to some extent by inhibiting the liver inflammation.

Objective To observe the effect of omeprazole combined with Kangfuxin liquid on immune function,gastrin family and NO levels in patients with gastric ulcer.Methods 120 cases of patients with gastric ulcer from October 2014 to September 2016 in our hospital were selected and randomly divided into the observation group and the control group with 60 cases in each group.The control group was given omeprazole based on conventional treatment, and the observation group was given Kangfuxin liquid based on the control group.For four weeks, to observe the differences of immune function (CD3 +,CD4 +,CD8 +,CD4 +/CD8 + and CD4 +CD25 +FoxP3 +Treg), gastric gastrin family (GAS, CCK, MTL) and NO levels between the two groups.Results After treatment, the CD3 +, CD4 +, CD8 +, CD4 +/CD8 +and CD4 +CD25 +FoxP3 +Treg levels of the two groups were compared, the difference was statistically significant (P<0.05), CD3 +, CD4 +, CD4 +/CD8 + of the groups showed a gradually increasing, the difference was statistically significant (P<0.05) and CD8 +, CD4 +CD25 +FoxP3 +Treg showed a gradually decreasing, the difference was statistically significant (P<0.05), and the change range of the observation group was more than that of the control group, the difference was statistically significant (P<0.05), the GAS,CCK,MTL levels of the two groups were compared, the difference was statistically significant (P<0.05), the GAS, CCK of the two groups showed a gradually decreasing, the difference was statistically significant (P<0.05), and MTL showed a gradually increasing, the difference was statistically significant ( P<0.05 ) , and the change range of the observation group was more than that of the control group , the difference was statistically significant (P<0.05), the NO levels of the two groups were compared, the difference was statistically significant (P<0.05), NO of the groups showed a gradually increasing, the difference was statistically significant (P<0.05), and the change range of the observation group was more than that of the control group, the difference was statistically significant (P<0.05).Conclusion Omeprazole combined with Kangfuxin liquid can get better regulating results of immune function and gastrin family, NO in patients with GU, and have more effective than omeprazole alone.

Objective To investigate clinical effect of esomeprazole, mosapride, magnesium plus aluminum in patients with reflux esophagitis. Methods 98 patients with reflux esophagitis who were treated in our hospital from October 2014 to October 2016 were selected and randomly divided into control group and observation group, with 49 cases in each group.In the control group, patients were treated with esomeprazole and mosapride.In the observation group, patients were given esomeprazole, mosapride and magnesium plus aluminum.The levels of serum gastrin, serum motilin and 24-hour pH in the esophagus were compared before and after treatment .Results After treatment,The levels of serum gastrin and serum motilin in the observation group were significantly higher than those in the control group, the difference was statistically significant (P<0.05), while the total time percentage of pH<4 at standing position, total time percentage of pH <4 at supine position, total reflux time percentage of pH <4, and the duration of continuous reflux≥five mins in the observation group were significantly lower than those in the control group, the difference was statistically significant (P<0.05). Conclusion Esomeprazole, mosapride and magnesium plus aluminum have significant clinical effect in patients with reflux esophagitis, and can effectively reduce 24-hour pH in the esophagus, decrease the number of reflux, increase serum gastrin and serum motilin levels.

Aim To investigate SIRT1 activity and an-ti-inflammatory effects of Chikusetsu oleanane saponin ( COS) on the RAW264. 7 macrophage cells induced by lipopolysaccharide ( LPS ) . Methods The nitric oxide ( NO) release was detected with Griess method. Western blot was used to detect the expression of tumor necrosis factor-α( TNF-α) , interleukin 1β( IL-1β) . Nuclear factor-κB ( NF-κB) and silent information ad-justment factor 1 ( SIRT1 ) were tested by immunofluo-rescence. Results 1 mg · L-1 LPS co-cultured with COS at 25~300 mg·L-1 had no significant effect on the growth of RAW264. 7 cells. Compared with the LPS group, COS effectively inhibited the NO release and suppressed the expression of TNF-α and IL-1β, and also inhibited the translocation of NF-κB and up-regulation of SIRT1 . Conclusion COS has protective effects on RAW264. 7 cells stimulated by LPS, which may be related to up-regulating the expression of SIRT1 and promoting the deacetylation of NF-κB, thereby in-hibiting the translocation of NF-κB and reducing the expression of TNF-α and IL-1β.

RESUMO

A method to determine phytolith-occluded carbon ( PhytOC) was explored. Sodium hydroxide solution was used to dissolve the silicon compound(SiO2 ·nH2O) which occluding on the carbon, and then potassium dichromate-sulphuric acid solution was used to oxidize the released organic carbon. The concentration of Gr3+produced from the reaction was measured by spectrophotometry with its absorbance at 590 nm wavelength. The organic carbon concentration was calculated based on the amounts of potassium dichromate consumed. This analytical method was verified well accuracy and repeatability by running standard addition and recovery experiments. It has the advantages of low carbon loss, easy to operate, low cost;and hardly any influence to body health, as compared with acid dissolution-Elementar Vario MAX CN method. The results obtained from this method have a good correlation with these determined by Elementar Vario MAX CN ( Germany) .
