RESUMO

The dexamethasone suppressed and normal wound healing effect of ethanolic extract of leaves of Hyptis suaveolens (L) poit. (EEHS) at 400mg/kg and 800mg/kg was studied in Wistar rats. Two wound models viz. incision and excision wounds were used in this study. The parameters studied were breaking strength in case of incision wounds, period of epithelization and wound contraction in case of excision wound. The dexamethasone treated group showed a significant (P<0.05) reduction in the wound breaking strength in incision type of wound model and significant increase in epithelization period and reduction in percentage of wound contraction in excision type of wound model as compared to control group. Extract treated groups showed significant (P<0.05) improvement in all the wound healing parameters of incision and excision wound models as compared to control. In incision wound model, Co administration of EEHS at both the dosage levels with dexamethasone had significantly (P<0.05) increased the wound breaking strength as compared to dexamethasone only treated group and was seen more at dosage level of 400mg/kg. Similarly, in excision wound model, there was significant improvement (P<0.05) in both the parameters when EEHS was co administered with dexamethasone as compared to dexamethasone only treated group but improvement was more at dosage level of 800mg/kg.

Ethanolic extract of leaves of O. sanctum was investigated for normal wound healing and dexamethasone depressed healing using incision, excision and dead space wound models in albino rats. The extract of O. sanctum significantly increased the wound breaking strength in incision wound model. The extract treated wounds were found to epithelialize faster and the rate of wound contraction was significantly increased as compared to control wounds. Significant increase in wet and dry granulation tissue weight, granulation tissue breaking strength and hydroxyproline content in dead space wound model was observed. The extract significantly decreased the antihealing activities of dexamethasone in all the wound models. The results indicated that the leaf extract promotes wound healing significantly and able to overcome the wound healing suppressing action of dexamethasone. Histological examination of granulation tissue to determine the pattern of lay-down for collagen confirmed the results.

Effect of two calcium channel blockers (CCBs) nifedipine and amlodipine, was studied on normal and steroid depressed wound healing in albino rats, using the dead space wound model. The drugs enhanced normal healing as evidenced by increase in tensile strength of 10 days old granulation tissue. There was neither a significant change in the hydroxyproline level (or collagen) nor a change in the glycosaminoglycan content in granulation tissue. However, lysyloxidase level was increased significantly. The increase in tensile strength could thus be attributed to better cross-linking and maturation of collagen rather than collagen synthesis per se. The drugs were also able to overcome steroid depressed wound healing. It is likely that the prohealing effects may be related to the improved antioxidant status too, since superoxide dismutase levels were observed to be higher in the CCB- treated animals.

The wound healing effect of two calcium channel blockers, nifedipine and amlodipine was studied in rats using incision and excision wound models. In incision wound, two straight paravertebral skin thickness incision were made and on tenth day skin tensile strength was measured by using continuous water flow technique. In excision wound, circular piece of skin excised in its full thickness and wound contraction monitored by alternate day wound tracing and epithelisation period was monitored by noting the number of days required for escher to fall. Drugs enhanced the skin tensile strength in incision wound model. In excision wound model, wound contraction is increased on 4th and 16th day but epithlisation period was not significantly altered. In conclusion, calcium channel blockers can be used to enhance wound healing, especially if wound healing was suppressed by steroids.

Ethanolic extract of leaves of Hyptis suaveolens was evaluated for its wound healing activity in ether-anaesthetized Wistar rats at two different doses (400 and 800 mg/kg) using incision, excision, and dead space wound model. Significant increase in skin breaking strength, granuloma breaking strength, wound contraction, hydroxyproline content and dry granuloma weight and decrease in epithelization period was observed. A supportive study made on granuloma tissue to estimate the levels of catalase and superoxide dismutase recorded a significant increase in the level of these antioxidant enzymes. Granuloma tissue was subjected to histopathological examination to determine the pattern of lay-down for collagen using Van Gieson and Masson Trichrome stains. Enhanced wound healing activity may be due to free radical scavenging action of the plant and enhanced level of antioxidant enzymes in granuloma tissue. Better collagenation may be because of improved antioxidant studies.

RESUMO

Septilin, a proprietary preparation claimed to be useful in inflammatory conditions was tested for anti-inflammatory and wound healing effects in albino rats. It significantly enhanced gain in tensile strength in incision wounds and wound contraction and epithelization in excision wounds. It also suppressed acute inflammation (rat paw edema) significantly without affecting chronic inflammation (cotton pellet granuloma).

RESUMO

Human thyroid monoamine oxidase from hyperfunctioning thyroids was isolated and purified by separating the mitochondria by differential centrifugation followed by ultrasonication. The suspension containing the active enzyme from the DEAE-column, on passing through a Sephadex G-200 column gave three peaks corresponding to molecular weights of approximately 220,000, 23,000 and 4000 of which the component with molecular weight 4000 showed enzyme activity. The blood of the patients was analyzed for plasma MAO, RBC cholinesterase, plasma histaminase and plasma catecholamines. Histology and histochemistry of the thyroid tissues were also done. The data are examined from the point of view of the prevalent idea of the possible existence of MAO in multiple forms.
