RESUMO

Objective To investigate the chemical constituents of the fruits of Ficus carica. Methods The chemical constituents were isolated and purified with silica gel chromatography, Sephdex LH-20 and HPLC, and their structures were elucidated by spectral analysis including MS, NMR, IR spectroscopic analysis, elment analyzer, and chemical evidence. Results One new quinoline and two new anhydride compounds were isolated from ethyl acetate extract of the fruits of F. carica. The structures of the three new compounds were respectively defined as 4-hydroxyl-6-formyl-8α-methoxyl-quinoline-2-one (1), 5’β,6’α-[(di-ethyl)-5β,6β-cyclohexyl]- epoxyhexyl, cyclo-pentanhydride-[2,2,1]-2α,3β-cycloheptane (2), and 5’β,6’β-[di-(11-methylbutyl,11’-methylbutyl)-9,9’]-cyclo-8’- hexadiene-5α,6α-epoxyhexyl, cyclo-pentanhydride-[2,2,2,1]-2β,3α-cycloheptane (3). Conclusion These compounds were named figine K-1, figine G-1, and figine G-2, respectively.

RESUMO

Objective: To investigate the chemical constituents from Gleditsiae Spina (the thorns of Gleditsia sinensis). Methods: The compounds were isolated and purified by silica gel, Sephadex LH-20 column chromatographic techniques, and their chemical structures were confirmed on the basis of spectroscopic analysis by the physicochemical properties. Cytotoxic activity using the MTT colorimetry method was performed to measure the inhibitory effect of the compounds on cell proliferation of HepG2, A-549, and EC109. Results: Three coumarins were obtained from the ethyl acetate soluble fraction of the 90% ethanolic extract and their structures were identified as scoparone (1), isoscopoletin (2), and 6-amino-7-methoxycoumarin (3). Conclusion: Compound 3 is a novel coumarin named gledisinmarin A. Compounds 1 and 2 are isolated from this plant for the first time. Compound 2 displays the stronger cytotoxicity against A549 cell with an IC50 value of 34.47 μg/mL, while cisplatin with an IC50 value of 11.50 μg/mL as a positive control.

RESUMO

OBJECTIVETo study the effect of Rhynchosia volubilis Lour ethyl acetate extract (RVLEAE) on male mouse procreation and analyse their chemical composition.

METHODWith the method of solvent extraction, RVLEAE was extracted and concentrated. In the experiment of mice, 80 male mice were randomly and equally divided into four groups: Normal Saline control, positive control with 0.1% triperygium wilfordii glycoside, 100 mg x kg(-1) x d(-1) RVLEAE and 400 mg x kg(-1) x d(-1) RVLEAE. Every mouse was fed with drug 0.01 mL x g(-1), once a day, ig, for eleven consecutive weeks. After two and 10 weeks, male and female mouse naturally mated for one week. The pregnancy rate, number of fetus and nonviable fetus, the viability of spermatozoon in the epididymis cauda, pathological change of testis and epididymis were observed in this experiment. In the analysis of chemical composition, RVLEAE were separated with column chromatography, and chemical compositions were identified with thinlayer chromatography, infrared chromatography and nuclear magnetic resonance.

RESULTThe pregnancy rate of mice was markedly decreased. The number and viability of spermatozoon were slightly reduced in I and II after two and 10 weeks, but the pathological changes of testis and epididymis were markedly occurred. Main chemical compositions were identified as saccharide, glycosides especially analog of fucose, alcohols, and phenols.

CONCLUSIONRVLEAE can inhibit the procreation of male mice, and inhibitory target tissue may be the epididymis. Active mechanism of RVLEAE may be that glycosides interfere the maturation of spermatozoon in the epididymis cauda.
