RESUMO

@#Excellent lead compounds have a profound influence on drug development and can improve the success rate of product launch. It is expensive and time-consuming to discover lead compounds by traditional methods, yet artificial intelligence (AI) can discover good lead compounds efficiently.This article systematically summarizes the research progress of obtaining lead compounds through the screening and generation models of AI, classifies different models according to the type of information input, focuses on drug repurposing by screening model and multi-objective drug design by generation model, and discusses the development prospect of AI in the research field of lead compounds, aiming to provide new research ideas for the application of AI in lead compounds.

RESUMO

Exosomes are nanometer-sized vesicles that contain various types of biologically active components, including proteins, nucleic acids, carbohydrates, and lipids, which vary with the type and physiological state of the cell. In recent years, several studies have showed that exosomes can provide new non-invasive diagnostic and prognostic biomarkers in patients affected by cancers, including bladder cancer (BC), and the lipid bilayer membrane structure makes exosomes as promising delivery vehicles for therapeutic applications. Exosomes have the characteristics of high abundance, high stability, tissue specificity, and wide distribution in body fluids, and are secreted as various types by cells in different states, thereby possessing great potential as biomarkers for BC. Herein, we briefly summarize the functions and roles of exosomes in the occurrence and development of BC and the current progress of research on exosomes in BC, while focusing on potential clinical applications of the diagnosis, treatment, and prognosis of BC.

OBJECTIVE@#To investigate the effect of Piwil2-induced cancer stem-like cell (Piwil2-iCSC)-derived exosomes on the proliferation,migration and invasion of human umbilical cord blood-derived mesenchymal stem cells (hucMSCs).@*METHODS@#Piwil2-iCSC-derived exosomes were isolated by ultracentrifugation and identified using transmission electron microscopy,nanoparticle tracking analysis and Western blotting.Exosome uptake assay was used to identify the pathway that Piwil2-iCSCderived exosomes utilized.HucMSCs were divided into control group,PBS intervention group and exosome intervention group,and CCK-8 assay,wound healing assay,Transwell assay,Western blotting and cell karyotype analysis were used to observe the proliferation,migration,invasion,expression levels of MMP2 and MMP9 proteins,and chromosome structure of hucMSCs.@*RESULTS@#The diameter of Piwil2-iCSC-derived exosomes ranged from 50 nm to 100 nm,and most of them were oval or spherical capsules rich in CD9,CD63 and Piwil2 proteins.Exosomal uptake assay showed that the exosomes executed theirs functions after entering the cells.Compared with the control cells and PBS-treated cells,hucMSCs treated with the exosomes showed significantly increased number of proliferating cells (<0.05) with accelerated healing rate (<0.05 at 24 h;<0.01 at 48 h),increased invasive cells (<0.01),enhanced protein expressions of MMP2(<0.05 PBS group;<0.01 control group) and MMP9(<0.05),but their karyotype still remained 46XY without any abnormalities.@*CONCLUSIONS@#Piwil2-iCSC-derived exosomes can promote the proliferation,migration and invasion but does not cause cancer-like heterogeneity changes in hucMSCs.

RESUMO

Calcifying epithelial odontogenic tumor (CEOT) is a rare benign epithelial tumor of odontogenic origin. CEOT is a benign but a locally infiltrative tumor. CEOT has two clinical variants: intraosseous (central) CEOT and extraosseous (peripheral) CEOT. The peripheral type is rare. In this paper, we report two cases of CEOT. The diagnoses of the cases were verified by histopathology. This study aims to explore the clinical and imaging appearances of CEOT and improve the understanding of the disease.

The keratocystic odontogenic tumor (KCOT) is a common cystic lesions of jaw, which has a high recurrence rate but rarely undergo malignant transformation. This study reported a patient with primary intraosseous squamous cell carcinoma developed from KCOT.

<p><b>OBJECTIVE</b>To assess the clinical and imaging features of cementoblastoma for diagnosis and differential diagnosis.</p><p><b>METHODS</b>Clinical and imaging data of 14 cases were selected for this retrospective study.</p><p><b>RESULTS</b>A total of 14 cases of cementoblastoma patients with ages at diagnosis ranging from 11 years to 58 years (mean age, 27.6 years; median age, 21.0 years). The tumor affected 7 patients from each gender. The mandible was the main site of occurrence. The most common complaints were swelling and pain. Radiographic results showed that cementoblastoma typically presented as a well-circumscribed and radiopaque mass confluent with the involved tooth root and surrounded by a thin and uniform radio-lucent border. Tumor parenchyma was generally mixed-density, which could be divided into two types, namely, homogeneous and heterogeneous patterns. Other significant imaging features included the periphery bone of the lesion sclerosis and cortical thickening of the lower edge of the mandible.</p><p><b>CONCLUSION</b>Obvious characteristics are found in the clinical and imaging findings of cementoblastoma. Accurate recognition of this lesion is significant in clinic to avoid misdiagnoses.</p>

Objective To study the curative effects of fixation with absorbable screw and fixative stick to treat ankle joint fracture accompanied by ankle joint dislocation.Methods 27 Patients with ankle joint fracture accompanied with ankle joint dislocation were fixed with absorbable screw and fixative stick since 1995 in our hospital.Internal,outer,backankle with SR-PLLA fixation and the distal tibiofibular syndesmosis separation by SR-PGA with absortive screw and fixative stick.Results All 27 Patients were followed up.The fracture healed up in about 10 weeks.All fracture had no displacement during the course of the treatment and no influencing function after operation.They were evaluated with Sarkision criterion.The successful rate was 92 6%.Conclusion SR-PLLA fixation,PGA absorbable screw and fixative stick are ideal fixation in treating ankle joint fracture accompanied by dislocation.
