Objective@#To investigate the expression of SMARCA4 (BRG1) and SMARCB1 (INI-1) protein in endometrial dedifferentiated carcinoma (DDC) and undifferentiated carcinoma (UDC), and their correlation with clinicopathologic features.@*Methods@#Clinicopathological information was gathered for 26 cases of DDC and UDC and consulting hospitals from January, 2006 to December, 2018 in Fudan University Shanghai Cancer Center, including 10 cases of DDC and 16 cases of UDC. Morphologic features and diagnosis were reviewed by two pathologists. Immunohistochemistry for expression of BRG1 and INI1 protein was performed. The correlations with clinicopathologic features were analyzed.@*Results@#BRG1 and INI1 loss were present in 14 of 26 cases of DDC/UDC, including 12 BRG1-deficient cases and 2 INI1-deficient cases, respectively. Six cases demonstrated variable amounts of rhabdoid cells in 14 BRG1/INI1-deficient cases, and only 1 case showed rhabdoid cells in the 12 intact expression cases. However, there was no significantly statistical difference (P=0.060). Age, invasive depth, lymph node status and FIGO stage were not associated with the expression of the BRG1 and INI1 (P=0.437, P=0.672, P=0.242, P=0.348). Remarkably, the BGR1/INI1-deficient patients had worse survival than those with intact expression (4.7 vs. 22.9, P=0.033).@*Conclusion@#BRG1/INI1-deficient is observed in approximately half of DDC and UDC. Identification of these tumors is clinically relevant due to their more aggressive behavior and poor prognosis. Hence, BRG1 and INI1 immunohistochemical stains should be performed for DDC and UDC in order to help the pathologists to distinguish these tumors from other carcinomas, and to predict the clinical prognosis.

OBJECTIVE: To analyze the clinical manifestations and characteristics of adverse drug reactions (ADR) induced by osimertinib mesylate, and to provide reference for clinical safe use of drugs. METHODS: Retrieved from PubMed, Wanfang database, CNKI and VIP during database establishment-Aug. 2018, ADR cases of osimertinib mesylate were analyzed retrospectively in respects of patient’s age, gender, nationality, usage and dosage of osimertinib mesylate, occurrence time of ADR, clinical manifestations, treatment measures and outcome, etc. RESULTS: A total of 20 articles were included, involving 21 patients. The age of the patients was 32-82 years old, and the proportion of patients aged 57-79 was the largest (80.9%). The female with ADR (14 cases) were more than the male. The patients were from 5 countries, in which Japan took the most ratio (13 cases, 61.9%). The dose of osimertinib mesylate was 160 mg/d in a patient and commonly recommended dose 80 mg/d for other patients. The most ADR cases (16 cases, 76.2%) occurred within 3 months, and no reports of ADR occurred more than 12 months. Organs/systems involved in ADRs were mainly respiratory system (11 case times, 45.8%) and digestive system (6 cases, 25.0%), in addition, ADR also occurred in cardiovascular system, hematological system, systemic reactions, skin and eyes. Among 21 patients, 3 cases were mild ADR so that they were given same dose with same frequency. A patient suffered from interstitial pneumonia and was given medicine every other day and symptomatic treatment; the symptoms of the patient were relieved. Other 17 cases were relieved after drug withdrawal and symptomatic treatment, but 2 patients died of tumor progression. CONCLUSIONS: In clinical application of osimertinib mesylate, attention should be paid to ADR monitoring, especially short-term ADR, especially ADR of respiratory, digestive system.

Radiotherapy is a very important treatment for early or advanced stage of prostate cancer (CaP).Radioresistance is the most important challenge for current radiotherapy.It is of great importance to study the mechanisms of radioresistance and to develop new treatment strategies that can overcome radioresistance.In CaP patients,microRNAs (miRNAs) are usually abnormally expressed,and radiotherapy can significantly alter the expression levels of miRNAs.There are increasing evidences that miRNAs are closely related to tumor radioresistance.The mechanism of action of miRNAs in regulating radioresistance is very complex.The possible mechanisms include DNA damage response,cell cycle checkpoint activation,apoptosis,autophagy,tumor stem cells,hypoxia,etc.The previous studies showed that miRNAs hold potential as prognostic biomarkers and therapeutic targets for CaP radiotherapy.New technologies targeting miRNAs combined with radiotherapy are expected to overcome CaP radioresistance,which will help to achieve precise radiotherapy,and bring hope to patients.

Objective To explore the clinical effect of mosapride combined with rabeprazole in the treatment of elderly patients with reflux esophagitis .Methods Clinical data of 126 elderly patients with reflux esophagitis were retrospectively studied.64 cases in the study group were treated with mosapride combined with rabeprazole ,62 cases in the control group were treated with mosapride combined with omeprazole .The clinical effects between the two groups were compared.Results Before treatment,there were no statistically significant differences in the scores of pain of patients with heartburn,acid reflux and chest between the two groups (t =0.512,0.181,0.228,all P >0.05). After treatment,the symptom scores of the study group were significantly lower than those of the control group (t =3.689,4.892,5.571,all P <0.01).The clinical symptoms and esophageal mucosal total effective rate in the study group were significantly higher than those in the control group (96.88% vs.87.10%,93.75% vs.80.65%,χ2 =4.121,4.479,all P <0.05).The scores of quality of life scale of the study group were significantly higher than those of the control group in cognitive function ,mental function,physical function and social function (t =4.920,6.853, 4.153,6.163,all P <0.01).Conclusion Mosapride combined with rabeprazole in the treatment of elderly patients with reflux esophagitis can effectively relieve symptoms of heartburn ,acid reflux and chest pain,help patients with mucosa recovery as soon as possible ,improve the quality of life of patients,and it is worthy of clinical application .

Objective: To investigate the effects of progranulin on migration and invasion of human colon cancer cells


Study Design: An experimental study


Place and Duration of Study: Department of Histology and Embryology, College of Basic Medical Sciences, Jilin University, Changchun, China, from October 2015 to July 2017


Methodology: Cell transfection was used to obtain progranulin knocked down/overexpressing cells. MTT and soft agar assay were applied to investigate the cell growth. Migration and invasion were detected using transwell assay


Results: Tumor growth, migration and invasion decreased in progranulin knocked down cells and increased in progranulin overexpressing cells. The decrease was recovered when exogenous progranulin was added. Progranulin-induced migration and invasion were inhibited by neamine


Conclusion: Progranulin has a direct effect on tumor growth, migration, and invasion of human colon cancer cells. It may serve as a potential therapeutic target for human colon cancer

Objective To analyze the clinical effect of anti-angina pectoris combined with anxiolytic-depressive drugs in patients with angina pectoris and anxiety and depression and its influence on psychological fluctuation .Methods 100 patients with coronary heart disease and angina pectoris who were treated with anxiety and depression were randomly divided into the observation group and the control group according to the digital table method.The control group were treated with conventional angina pectoris, and the control group were treated with anti-anxiety and depression medication.The left ventricular end-systolic volume ( LVESV ) , left ventricular ejection fraction ( LVEF ) , left ventricular end-diastolic volume (LVEDV), left ventricular end-diastolic volume (LVEDV) and left ventricular end-diastolic volume (LVEDV) and anxiety and depression scores were compared between the two groups before and after treatment.Results The total effective rate of the treatment group was significantly higher than that of the control group (P<0.05).There was no serious adverse reaction in the two groups.The treatment compliance rate in the observation group was significantly better than that in the control group (P<0.05), and the level of cardiac function after treatment in the observation group was significantly better than the control group (P<0.05); observation group before and after treatment anxiety and depression scores were statistically significant (P<0.05), while the control group before and after treatment anxiety and depression score was not statistically significant.Results The combination of anti-angina pectoris and anxiety-depression drugs had better clinical curative effect on patients with angina pectoris and anxiety and depressive patients, and could effectively control the onset of angina pectoris.Meanwhile, they had higher safety and could improve the compliance of patients with clinical treatment.Anxiety and depression, so as to improve the quality of life of patients.

Objective To investigate the factors affecting non-neoplasma touch technology operation by operation room nurses for malignant tumor patients.Methods The qualitative methods was used in the study to interview 15 operation room nurses phenomenologically using semi-structured interviews.Colaizzi principles were used for data analysis and summarization of themes.Result The themes brought out from the data included lack of knowledge of no-neoplasma touch technology operation,huge workload,less importance attached to the management,over-consideration of medical costs.Conclusions It is a need to improve the level of knowledge of operating room nurses on non-neoplasma technology operations.The importance should be attached to management of the technology operation.The training on the operation should be strengthened so as to improve their operation of the technology.

Objective:Increased D-dimer level has been recently correlated with the progression and high mortality rate of some cancers. This study aimed to determine whether the plasma D-dimer level was a predictor of non-Hodgkin lymphoma (NHL). Methods:Clinical data of 425 with NHL were retrospectively reviewed. Variables such as clinicopathological factors, plasma D-dimer levels, and the international prognostic index (IPI) were evaluated. Results: The plasma D-dimer level was closely correlated with the IPI. The overall 1-, 3-, and 5-year survival rates of the low plasma D-dimer levels were 80%, 75%, and 50%;those for the high plasma D-dimer levels were 41%, 33%, and 9%, respectively (P<0.001). Compared with an IPI of zero or one, IPI values≥4 were predictive of poor prognosis (P<0.001). The Ann Arbor stage, pathological classification, plasma D-dimer level, and IPI were found to be significantly as-sociated with mortality. However, similar to IPI, the plasma D-dimer level was the independent risk factor affecting multivariate analy-sis. Conclusion:High D-dimer levels are a potential prognostic marker for NHL.

Objective To elucidate the effects of thalidomide on proliferation of human renal cell line 786-0 and the expression of basic fibroblast growth factor (bFGF) in this cell line. Methods Cell was treated with different doses of thalidomide(6.25 μg/ml, 25 μg/ml, 100 μg/ml) respectively or normal saline as control; cell survival rate was analyzed by MTT assay. The mRNA level of bFGF was detected by reverse transcriptase-polymerase chain reaction (RT-PCR). bFGF protein expression in 786-0 cells was detected by flow cytometry (FCM). Results Thalidomide, ranging from 6.25 μg/ml to 100 μg/ml, suppressed the proliferation of 786-0 cell line in vitro significantly. After application of thalidomide for 48 and 72 hours, the IC50 was 46.42 μg/ml and 19.56 μg/ml respectively. Apoptosis rate increased from 12.43 % to 30.30 %, accompanying with reducing expression of bFGF. Application of thalidomide (25 μg/ml) induced the most significant inhibition to the bFGF in the cell line. Conclusion Thalidomide down-regulates bFGF expression, inhibit the proliferation, and induce apoptosis in 786-0 cell line.

BACKGROUND: It has been proved that the rare earth compounds can obviously restrain the growth of tumor in vitro and in vivo.OBJECTIVE: To check the effect of chlorinated lanthanum (LaCl3) on the expressions of cyclin D1 and cyclin dependent kinase 4 (CDK4) of hepatocellular carcinoma cells.DESIGN: Controlled observation.SETTING: Department of Nephropathy, First Clinical Hospital, Jilin University. MATERIALS: The experiment was carried out in the cell culture laboratory (province-level) of Department of Histology and Embryology, Jilin University from July 2002 to June 2003. Wells of the microwell plate seeded with CBRH-7919 cells were randomly divided into 4 groups according to the dose of LaCl3: control, 0.01, 0.1, 1.0 mmol/L LaCl3. And each group was further divided into three subgroups according to the culture time: 1, 3, 5 days, with 6 wells for each.METHODS: The growth of CBRH-7919 cells was monitored following treated with 0.01, 0.1, 1.0 mmol/L LaCl3 for 1, 3, 5 days in vitro, respectively. ① Absorbance(A) of each well was measured by MTT assay.② The changes of distribution of cells into different phases of cell cycle were detected by flow cytometry. ③ Cyclin D1 and CDK4 expression was detected by immunocytochemical analysis.MAIN OUTCOME MEASURES: The expression of cyclin D1 and CDK4 in CBRH-7919 cells of different doses of LaCl3 at different time.RESULTS: Effect of LaCl3 on the growth of CBRH-7919: On days 3 and 5, the A value of each well in the 0.1 and 1.0 mmol/L LaCl3 groups was respectively significantly lower than that in the control group (day 3: 1.140±0:070, 0.706±0.092,1.461±0.087; day 5: 1.888±0.020,0.625±0.037,2.544±0.032, respectively) (P < 0.05,0.001 ). ②Effect of LaCl3 on the percentage of CBRH-7919 cells at G0/G1 stage: On days 1, 3, and 5, the percentage of CBRH-7919 cells at G0/G1 stage in the 1.0 mol/L LaCl3 group was respectively significantly higher than that in the blank control group [(60.70±0.20)%, (39.49±0.67)%; (61.66±0.97)%, (45.56±1.00)%; (69.92±0.18)%, (49.24±0.27)%, (P < 0.01,0.001 )]; On day 5, the percentage of CBRH-7919 cells at G0/G1 stage in the 0.1 mol/L LaCl3 group was respectively significantly higher than that in the blank control group[(58.88±0.73)%, (49.24±0.27)%, P < 0.05]. ③ Effect of LaCl3 on the expressions of cyclin D1 and CDK4 of CBRH-7919 cells: On day 1, the expressions of cyclin D1 and CDK4 of CBRH-7919 cells in the 0.1 and 1.0 mmol/L LaCl3 groups were respectively lower than those in the blank control group (expression of cyclin D1: 562±35,453±22,860±82;expression of CDK4: 705±84,680±28,762±16, P < 0.05, 0.01 and 0.001).CONCLUSION: LaCl3 inhibited the growth of CBRH-7919 cells correlating down-regulating cyclin D1 and CDK4 as reflected on the cell cycle arrest of CBRH-7919 cells from G1 stage to S stage.

Objective To study the effects of LaCl3 on P16 and P21 expressions of hepatocellular carcinoma cells.Methods The experiment was divided into 4 groups.In experiment groups,CBRH-7919 cells were cultivated with 0.01,0.10 and 1.00 mmol?L-1 LaCl3 in DMEM containing 10% calf serum.In control group,CBRH-7919 was cultivated in DMEM containing 10% calf serum without LaCl3.The growth of CBRH-7919 cells was observed following treated with 0.01,0.10,1.00 mmol?L-1 LaCl3 for 1,3,5 d in vitro, respectively,the changes of cell cycle were detected by flow cytometry.At the same time,P16 and P21 were detected by immunocytochemical analysis.Results Compared with control group,the inhibitory rate of growth of CBRH-7919 cells was increased evidently when treated with 0.1 and 1.00 mmol?L-1 LaCl3 for 3 and 5 d(P

Objective:To discuss the effects of Rhodosin,the extractant of medical plants na med Rhodiola Sachinonsis A.Bor,on aging rats and the model rats of Alzheier′s d isease (AD).Methods:Step down and water maze tests were used to determine the content of ace tylcholine (Ach) and the activity of acetylcholine transferase (ChAT) of hippoca mpus,lipid peroxide (LPO) and activity of superoxide dismute (SOD) of the cerebr al cortex,and cerebellar cortex and spinal cord were observed respectively.At th e same time,the general light microscope and eletron microscope were used to obs erve the changes in morphology.Results:The Rhodosin as an antioxidant could enhance the content of Ach and the activity of ChAT,reduce the formation of LPO,and increase activity of SOD.The Rh odosin could facilitate metabolism,enhance activity,and inhibit degeneration of aging rat cells.Conclusion:Rhodosin had obvious anti-aging effects on aing rats and also had p reventive,protective,anti-dementia effects on experimental dementia.

ObjectiveTostudytheincidenceandclinicalcharacteristicsoftheco-infectedsexuallytransmittedinfections(STI)inpatientswithgenitalherpes.MethodsTheclinicaldataof287caseswithherpeticlesionsorpatientswithsuspectedherpeslesionswerecollected,andthepathogensofsexuallytransmittedinfectionsweredetected.ResultsGenitalherpeswasconfirmedin64.8%(186/287)oftherecruitedcases.HIVantibodiesweredetectedin68cases,andnoHIVantibodywasdetected.Theco-infectionssuchascondylomaacuminatum,activeorlatentsyphilis,genitalcandidiasisandotherSTIswerediscoveredin23.1%(43/186)ofpatientswithgenitalherpes.Allgenitalherpescasesco-infectedwithotherSTIswerecausedbyHSV-2.ConclusionTheco-infectionsarecommoninpatientswithgenitalherpes,andthefeaturesofthelesionsmaybechangedbytheseco-infections.

Objective To detect and type herpes simplex virus (HSV) in genital lesions of the patients attending STD clinic. Methods Clinical data were collected and analyzed from patients with anogenital non-herpetic lesions including induration or furuncle, fissure, folliculitis, single ulcer and so on. HSV was detected and typed by culture and PCR with specimens taken from these lesions. Results One hundred and five cases were recruited in this study. Among them, 18 cases presented induration (furuncle), 15 fissure, 16 folliculitis, 7 abrasion, 12 single ulcer, 25 nonspecific erythema and 12 balanoposthitis with edema and exudation. HSV was found in 33.3%(6/18), 20%(3/15), 37.5%(6/16), 28.6%(2/7), 33.3%(4/12), 20%(5/25) and 50%(6/12) of these lesions, repectively, by PCR, while in 22.2%(4/18), 13.3%(2/15), 25%(4/16), 14.3%(1/7), 33.3%(4/12), 8%(2/25) and 41.7%(5/12), repectively, by viral culture. The positive rates of HSV from all these non-herpetic lesions were 30.5% (32/105) and 21% (22/105), respectively (? = 0.095, P = 0.114), by PCR and viral culture. The results of HSV typing were consistent between PCR and immunofluorescence with type-specific monoclonal antibodies. Among those with HSV infections, HSV-1 infection acounted for 9.4% (3/32), and HSV-2 90.6% (29/32). Conclusions The clinical manifestations of genital HSV infections vary, and HSV could be isolated from lesions of induration (furuncle), fissure, folliculitis, abrasion, single ulcer, nonspecific erythema and balanoposthitis with edema and exudation. HSV-2 is the predominant type.

The relationships between cholinergic neurons and SP terminals were examined in the rat sacral ventral horn at the light and electron microscopic levels by means of double immunostaining methods. Cholinergic neurons were labeled by a monoclonal antibody to choline acetyltransferase (CHAT) with the avidin-biotin technique and stained bluish-green by indolyl-?-galactoside reaction products with ?-galactosidase as a marker. On the same sections, SP fibers were labeled by polyclonal antisera to SP after application of the peroxidase-antiperoxidase (PAP) method and stained brown by diaminobenzidine (DAB)reaction. At the light microscopic level CHAT-I neurons stained bluish-green and SP- I fibers Stained brown were found in the ventral horn. At the electron microscopic level, many asymmetrical axodendritic synapses(type I of Gray)were observed between CHAT-I dendrites and SP-I terminals in the ventral horn, but axosomatic synapses and symmetrical synapses (type I of Gray) were hardly detected. These results indicate that SP-I terminals make direct synapses with CHAT-I motoneurons of sacral ventral horn. These synapses may be predominantly excitatory and have importance in the control of muscular constriction.

Specimens of skin, skeletal muscle, adipose tissue, brain, peripheral nerve and catilage were taken from 6 cadavers (divided into 2 groups) perfused and embalmed with formalin. 90 pieces of the above-mentioned tissues were taken in different times and the paraffin sections were stained by routine HE. Electron microscopic observation was made from some skin and skeletal muscle specimens, and histochemical methods were used to demonstrate DNA, RNA, keratin(-SH, -SS), fatty acids and calcium in some skin materials. Loyez stain method for myelin sheath was applied to some brain and nerve sections.1. Skin tissue: In both group A and B, 83.3% and 78.9% of skin materials were appraised as grade I and II (excellent and good) respectively. The morphological changes appeared mainly as karyopyknosis of the epidermal cells and shrinkage of the secretory part of the sweat glands. Most of the skin tissues of the corpses perfused 3~4 hrs after death were well preserved, only 8.3% being in grade Ⅲ (moderate). On the other hand, 43% of skin materials taken from those perfused 30 hrs after death were in grade Ⅲ. Ultrastructurally, the shape of the epidermal cells and their adhensions still remained intact, but karyopyknosis or chromolysis and autolysis of most of the cytoplasmic membraneous structures were found. In those cadavers preserved for less than one year, there was no evidence of progressive changes in microscopic and submicroscopic structures of the skin. The tonofillaments and desmosomes were still kept well in those specimens preserved for many years, and their presence may be the cause of maintaining cell integrity. Histochemically, mercapto-reaction of keratin was kept stable and found to be well shown even in old samples of skin, but the disulfide reaction usually showed marked fading with time, probably due to exofolition of the stratum corneum. However, this reaction was well shown in the hairs and not weakened for many years.2. Fat tissue: Hydrolysis of lipid substances of subcutancous fat and sebaceous gland, as shown by the presence of fatty acid, was found in the corpses preserved even as short as half a year.3. Skeletal muscle: The preservation state of the skeletal muscle was also dependent on the complete perfusion in time. The structure of the muscle fibers was generally worse preserved than that of the skin as shown in severe myolysis. Since stagnation of the small blood vessels frequently accompanied myolysis, obstruction of microcirculation which prevented the permeation of fomalin into the muscle tissues may be the cause of rapid myolysis. The ultrastructural changes of the muscle fibers were the same as those of the epidermal cells. In all specimens even as old as 30 years, two types of myofillaments were well preserved, and bands could be seen distinctly.4. Nervous tissue: The brain tissues were well preserved in those corpses that were perfused shortly after death, but the peripheral nerves usually showed severe autolysis, obviously it is due to the same reasons as those for myolysis.5. Cartilage: The structural changes of cartilage after death were slow and all specimens of cartilage were well preserved.
