RESUMO

Objective:To analyze the clinical value of relative expression of peroxisome proliferators-activated receptors mRNA (PPAR mRNA) and protein in placental tissues of pregnant women with gestational diabetes mellitus (GDM) .Methods:52 pregnant women (the study group) with GDM who were admitted to our hospital from Jan. 2019 to Nov. 2020 and 50 normal pregnant women (the control group) who underwent physical examination and gave birth during the same period were selected. Serum samples were collected to detect the islet cell function. Placental tissue samples of the two groups of pregnant women were collected after delivery to measure the content of PPARγmRNA and protein in placental tissues and adipose tissues. The correlations between PPARγ in placental, adipose tissues of GDM pregnant women with islet cell function were analyzed. Results:The HOMA-IR level of the study group was higher than that of the control group [ (3.45±1.06) % vs (1.40±0.43) %], and the HOMA-β level [ (126.59±23.59) % vs (153.12±27.34) %] was lower than that of the control group ( P<0.05). The PPARγ mRNA [ (1.65±0.21) vs (0.93±0.16) ] and PPARγ protein content [ (1.89±0.51) vs (1.02±0.23) ] of placenta tissue in the study group were higher than those in the control group ( P<0.05), the PPARγmRNA [ (0.49±0.12) vs (1.15±0.26) ] and PPARγ protein content [ (0.43±0.11) % vs (0.96±0.22) %] in adipose tissue were lower than those of the control group ( P<0.05). Adipose tissue PPARγ mRNA and PPARγ protein were negatively correlated with HOMA-IR ( r=-0.45, -0.33), and positively correlated with HOMA-β ( r=0.47, 0.43) ( P<0.05) ; placental tissue PPARγ mRNA and PPARγ protein were positively correlated with HOMA-IR ( r=0.40, 0.37), and negatively correlated with HOMA-β ( r=-0.44, -0.35) ( P<0.05) . Conclusion:The levels of PPARγ mRNA and PPARγ protein are low expressed in adipose tissue of GDM patients, and highly expressed in placental tissues, and PPARγ expression is significantly correlated with HOMA-IR and HOMA-β, which can provide new clinical treatment for GDM Target and direction.

RESUMO

Objective To explore the association of genetic polymorphism of fucosyltransferase (FUT) 2 and FUT3 and expression of Lewis antigen with ulcerative colitis (UC) in Chinese Zhejiang Han population. Methods We genotyped FUT2 (rs281377, rs1047781 and rs601338) and FUT3 (rs28362459, rs3745635 and rs3894326) in 485 UC patients and 580 healthy controls using SNaPshot. By immunohistochemistry method, we also evaluated expression of Lewis a and b antigens in the sigmoid colon of 10 UC patients and 10 patients with benign colonic polyps. Results The frequencies of mutant allele (A) and genotype (GA+AA) in FUT3 rs3745635 were higher in UC patients than in controls (P=0.016, 95%CI 1.339-1.699;P=0.038, 95%CI 1.330-1.742, respectively). Stratified analyses revealed that the frequencies of mutant allele (G) and genotype (TG+GG) of FUT3 rs28362459 were significantly lower in patients with extensive colitis than in those with distal colitis (P=0.001, 95%CI 0.567-0.786;P<0.001, 95%CI 0.503-0.742, respectively). Similar conclusions were drawn for the mutant allele (A) and genotype (GA+AA) of FUT3 rs3745635 in patients with extensive colitis compared to those with distal colitis (P=0.011, 95%CI 0.621-0.900;P=0.006, 95%CI 0.553-0.845, respectively). Although expression of Lewis b antigen in the sigmoid colon did not differ between UC patients and controls, Lewis a antigen expression was higher in the crypt epithelium of both inflammatory and non-inflammatory sigmoid colon of UC patients than in controls (P=0.028).Conclusion Polymorphisms of FUT3 and expression level of Lewis a antigen might be associated with UC.

RESUMO

<p><b>OBJEVTIVE</b>To study the effect of spvB/spvC gene on Salmonella virulence and the Host immune.</p><p><b>METHODS</b>STM.211, STM.211-Delta;spvB, STM.211-Delta;spvC, STM.211-Delta;spvB.spvC and PBS were infected with 0.2 mL 10(5) CFU corresponding strain respectively by intraperitoneal. We observed the mental status, movement, diarrhea, weight, pelage changed hair of the infected mouse. Then the level of IL-10, IL-12, IFN-γ were detected by ELISA. Finally, we observe the pathological changes of liver and spleen with the general view and the microscope.</p><p><b>RESULTS</b>Infection symptoms of STM.211, STM.211-Delta;spvB and STM.211-Delta;spvC were significantly worse than PBS group, but there was no significant difference between STM.211-spvB.spvC group and PBS group. The secretion of IFN-γ and IL-12 of STM.211, STM.211-Delta;spvB, STM.211-Delta;spvC group were significantly lower than those in the STM.211-Delta;spvB.spvC group (P<0.05), but IL-10 secretion was significantly higher than STM.211-Delta;spvB.spvC group (P<0.05). There were no statistical significance among the STM.211, STM.211-Delta;SpvB, STM.211-Delta;spvC groups (P>0.05).</p><p><b>CONCLUSIONS</b>Salmonella virulence can be affected obviously by spvB combined with spvC gene, but not by spvB or spvC. spvB/spvC gene can inhibit the TH1 cytokines (IFN-γ and IL-12) secretion but promote the TH2 cytokines (IL-10) expression, leading immune response trend to TH2 shift. It shows that spvB/spvC gene can help the bacteria evade the host immune defenses, leading to aggravation of infection.</p>

RESUMO

In order to detect endoleaks after endovascular aneurysm repair (EVAR), we developed an implantable micro-device based on wireless power transmission to measure aortic aneurysm sac pressure. The implantable micro-device is composed of a miniature wireless pressure sensor, an energy transmitting coil, a data recorder and a data processing platform. Power transmission without interconnecting wires is performed by a transmitting coil and a receiving coil. The coupling efficiency of wireless power transmission depends on the coupling coefficient between the transmitting coil and the receiving coil. With theoretical analysis and experimental study, we optimized the geometry of the receiving coil to increase the coupling coefficient. In order to keep efficiency balance and satisfy the maximizing conditions, we designed a closed loop power transmission circuit, including a receiving voltage feedback module based on wireless communication. The closed loop improved the stability and reliability of transmission energy. The prototype of the micro-device has been developed and the experiment has been performed. The experiments showed that the micro-device was feasible and valid. For normal operation, the distance between the transmitting coil and the receiving coil is smaller than 8cm. Besides, the distance between the micro-device and the data recorder is within 50cm.

RESUMO

In order to deliver power to the capsule endoscope, whose position and orientation are always changing when traveling along the alimentary tract, a wireless power transmission system based on electromagnetic coupling was proposed. The system is composed of Helmholtz transmitting coil and three-dimensional receiving coil. Helmholtz coil outside the body generates a uniform magnetic field covering the whole alimentary tract; three-dimensional coil inside retrieves stable power regardless of its position and orientation. The transmitter and receiver were designed and implemented, and the experiments validated the feasibility of the system. The results show that at least 320 mW of usable power can be transmitted to capsule endoscope when its position and orientation are changing at random and the transmitting power is 25W.

RESUMO

Objective To evaluate the application of endoscopic needle- knife precut sphincterotomy (PST) in treating acute suppurative cholangitis. Methods After failure of routine papillary intubation during encoscopic retrograde cholangio-pancreatography (ERCP) in papillary incarcerated stone or inflammatory stenosis cases, needle-knife PST was applied to find the lower opening of the common bile duct. After that, routine papillary sphincterotomy or balloon dilation followed. Then net basket for stone extraction and nasobiliary catheter for drainage were made. Results Eleven of the 12 cases′ stones were extracted successfully, the success rate was 91.7%. In the 11 cases, 5 cases′ incarcerated stones dropped into duodenum automatically after sphincterotomy; 9 cases′ stones were extracted successfully in one treatment while 2 cases′ stones were extracted secondarily after stents were implanted; 1 case′s stone could not be extracted and need surgical treatment after nasobiliary catheter drainage because of stenosis of the lower part of the common bile duct. There was no dead case in all the cases. Conclusions Acute suppurative cholangitis patients, who have papillary incarcerated stones or inflammatory stenosis, can receive more efficacious diagnosis and treatment by applying PST when routine endoscopic papillary intubation fails. PST is an important endoscopic treatment for acute calculous suppurative cholangitis
