This study aims to explore the pharmacodynamic differences of Puerariae Lobatae Radix(PLR), Puerariae Thomsonii Radix(PTR) and their different processed products and the influences of these medical materials on the diversity of intestinal flora. The Sennae Folium-induced diarrhea model, streptozotocin(STZ)-induced diabetes model and L-nitro-arginine methyl ester(L-NAME)-induced hypertension model were used to compare the pharmacodynamic differences in anti-diarrhea, blood glucose reduction and blood pressure lowering among raw, roasted and vinegar-processed PLR and PTR. The effects of raw and processed PLR and PTR on intestinal flora diversity of rats were evaluated by 16 S rDNA high-throughput sequencing. The roasted PLR and PTR performed better in anti-diarrhea, especially the former. PLR and its processed products all presented the efficacy of reducing blood glucose, and the vinegar-processed PLR was the most outstanding. The raw PTR was not that effective in reducing blood glucose, whereas its efficacy was improved after roasting and vinegar processing. Both PLR and PTR were capable of lowering blood pressure to a certain extent, and PLR is superior to PTR in this aspect. Further, the vinegar-processed PLR showed the best effect. The diversity of intestinal flora was different among rats to which different products of PLR and PTR were administered. The roasted PLR led to the highest abundance of Lactobacillus, which was closely related to its best antidiarrheal effect. The highest abilities of vinegar-processed PLR to lower blood glucose and blood pressure were associated with the high abundance of Blautia and Prevotella_9. This study lays a foundation for elucidating the processing mechanisms of PLR and PTR and provides a basis for their further development and application.

Objective:To observe the expression of brown adipose tissue (BAT), cells, proteins and corresponding genes in Yang deficiency model mice induced by Rhei Radix et Rhizoma suspension, and to explore the thermogenesis of processed products of Aconiti Lateralis Radix Praeparata with Jianchang faction characteristics. Method:Twenty mice, half male and half female, were randomly selected as the normal female and male groups. And the other 80 mice were administrated with Rhei Radix et Rhizoma suspension (the content of 0.25 g·mL-1) to establish Yang deficiency model, after the model was established, they were randomly divided into the model female and male groups, female and male groups of Shengfupian, female and male groups of Yinfupian, female and male groups of Yangfupian, 10 mice in each group. Mice were intragastric administrated with corresponding medical solution for two weeks (1.54 g·kg-1·d-1) according to groups. Normal group and model group were given equal volume distilled water. After administration, BAT of scapular region of mice was collected and the changes of BAT cells were observed by hematoxylin-eosin (HE) staining. The expression of uncoupling protein 1 (UCP1) and its mRNA were detected by Western blot and real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). Result:Compared with the normal group of the same sex, the proportion of BAT in the model group decreased significantly (P<0.01). Compared with the model group of the same sex, the proportion of BAT in female mice from Shengfupian and Yinfupian groups increased significantly (P<0.01), while there was no significant difference between each administration group and model group in the male mice. Compared with normal mice of the same sex, there were many scattered vacuoles in BAT cells of the model group, and fewer cells could be observed due to larger vacuoles. Compared with the model group of the same sex, BAT cells in mice from the Shengfupian group showed fewer vacuoles, smaller cells and tight arrangement, the density of BAT cells in mice from the Yangfupian group also increased significantly, while the vacuoles in BAT cells of mice from the Yinfupian group decreased relatively and the cells did not increase significantly. Compared with the same sex mice, the expression level of UCP1 in the model group and the normal group was statistically significant (P<0.05, P<0.01). In the female mice, the expression level of UCP1 in Yangfupian group was significantly higher than that in the model group (P<0.05), each administration group of male mice was significantly different from that of the model group of the same sex (P<0.05), of which Yangfupian was the most significant. The relative expression of UCP1 mRNA in the model group was significantly lower than that in the normal group of the same sex (P<0.05, P<0.01). In the female mice, compared with the model group, the relative expression levels of UCP1 mRNA in Yangfupian group, Shengfupian group and Yinfupian group increased significantly (P<0.05, P<0.01), compared with Yangfupian group, the relative expression levels of UCP1 mRNA in Shengfupian and Yinfupian were also significantly different (P<0.05). In the male mice, compared with the model group, the relative expression of UCP1 mRNA in Yangfupian group was significantly increased (P<0.01), but there was no significant difference in Shengfupian group and Yinfupian group, in addition, compared with Yangfupian group, the relative expression of UCP1 mRNA in Shengfupian group and Yinfupian group had significant difference (P<0.05). Conclusion:Shengfupian, Yinfupian and Yangfupian all have obvious improvement on Yang deficiency syndrome induced by Rhei Radix et Rhizoma suspension. The mechanism may be to promote the expression of UCP1 protein and its mRNA and enhance the activity of BAT. And the effect of Yangfupian is the best.

Objective?To investigate the effect of lung protection ventilation on postoperative pulmonary complications in patients with esophageal cancer.?Methods?98 patients underwent thoracoscopic laparoscopic esophagectomy for radical resection from January 2014 to January 2017 were enrolled in this study. Patients were divided into observation group (n = 49) and control group (n = 49) randomly. Two groups of patients underwent intraoperative single lumen tracheal catheter supplemented with CO2 pneumothorax. The control group received conventional ventilation. The protective group of patients in the observation group were given protective ventilation. The time of tracheal intubation after 10 min (T1); single lung ventilation 1 h (T2); at the end of surgery (T3); 24 h (T4) after pulmonary function parameters, inflammatory response, blood gas were analyzed. The patients were divided into recurrence group (n = 24) and non-recurrence group (n = 74), pulmonary complications and its related factors were analyzed according to whether the patient had recurrent or not pulmonary complications after recurrent (n = 24) or non-recurrent (n = 74).?Results?Pplat, airway peak pressure (Ppeak), airway resistance (Raw), interleukin-6 (IL-6), and interleukin-8 (in the two groups) at time T1. Interleukin-8, (IL-8), soluble intercellular adhesion molecule-1 (Sicam-1), partial pressure of carbon dioxide in artery (PaCO2), partial pressure of oxygen There was no significant difference in the index of oxygen, PaO2, and oxygenation index (PaO2/FiO2) (P > 0.05); the changes in Pplat, Ppeak, and Raw in the control group between T1 and T3 were significantly greater than those in the observation group. The changes of IL-6, IL-8, and Sicam-1 in the control group from T1 to T4 were significantly greater than those in the observation group (P < 0.05). T1 to T4 The change of PaO2 in the control group was significantly greater than that in the observation group, and the difference was statistically significant (P < 0.05). There were multiple pulmonary complications in some patients in both groups. There were 18.36% in the observation group and 30.61% in the control group combined with various types of pulmonary complications. The difference was statistically significant (P < 0.05). Preoperative smoking, operation time, degree of radicalization, clinical pathological stage, lymphatic infiltration, microvascular infiltration, postoperative adjuvant chemotherapy, and pulmonary complications in patients undergoing radical surgery for esophageal cancer were closely related (P < 0.05).?Conclusion?Lung protection ventilation can relieve the thoracoscopic laryngeal esophageal cancer surgery in the airway resistance, pressure and inflammation, increased oxygen saturation, should strengthen the high risk of pulmonary complications in patients with monitoring, targeted prevention, reduce postoperative the incidence of pulmonary complications.

Objective To investigate the effects of inflation pressures on mechanical environment of pathological carotid after stent implantation. Methods The carotid artery models with lipid and calcified plaques were constructed based on high-resolution MRI images. The artery-stent interaction model was developed in the finite element software. Based on the models, the von Mises stress distributions on the vascular wall and plague under 3 different inflation pressures (909, 1212 and 1515 kPa) were simulated. Results High inflation pressure would induce large Von Mises stress on the artery-stent interface. Moreover, the stress on the lipid plaque increased significantly with the increase of inflation pressure. However, the differences in stress distributions on the calcified plaque were very small under different inflation pressure. Conclusions Higher inflation pressure may severely damage the lipid plaque and artery-stent interface after stenting, which will contribute to the failure of stent. This research finding may provide clinical guidance for the selection of inflation pressure for arterial stent deployment and the assessment of plague stability after stent implantation.

Objective To observe dynamic changes of levels of monocyte chemotactic protein-1 (MCP-1), tumor necrosis factor-α (TNF-α) and interleukin-8 (IL-8) in patients with acute pancreatitis and to investigate its evaluation value on the severity of acute pancreatitis. Methods A total of 109 patients with acute pancreatitis admitted were divided into mild acute pancreatitis group (MAP group, 42 cases), moderately severe acute pancreatitis (MSAP group, 35 cases) and severe acute pancreatitis (SAP group, 32 cases). ELISA was used to detect the serum levels of MCP-1, TNF-α and IL-8 of patients at day 1, day 4 and day 7 of admission to hospital. Results The serum levels of MCP-1, TNF-α and IL-8 from MAP group, MSAP group and SAP group at day 1 of admission to hospital all significantly increased. There was a significant difference between MAP group and control group, MSAP group and MAP group, SAP group and MSAP group (P 0.05); The serum concentrations of MCP-1, TNF-α and IL-8 from MAP group all reached the highest level at day 4, which were significantly higher than the detection levels at day 1. In MSAP group and SAP group, the serum concentrations of MCP-1, TNF-α and IL-8 were the highest at day 1, which were significantly higher than the detection levels at day 4 and 7. At each detecting timing, the serum concentrations of MCP-1, TNF-α and IL-8 from MSAP group and SAP group were all higher than those of MAP group and MSAP group, respectively. Conclusions The dynamic changes of serum levels of MCP-1, TNF-α and IL-8 in patients with acute pancreatitis have their rules, and the change rule of MAP group was different with that of MSAP and SAP group, which showed the reference value for the diagnosis and illness severity evaluation of acute pancreatitis.

Clinically, abdominal aortic aneurysm (AAA) may continue to expand or even rupture after endovascular aneurysm repair (EVAR) due to endoleak or endotension. The existence of endoleak and endotension can significantly affect the mechanical/hemodynamic environment in AAAs, thus changing the strain and stresses on aneurysm wall, further influencing the transportation of low density lipoprotein (LDL), oxygen and nitric oxide (NO) in AAAs and aneurysm wall, which might eventually alter the biochemical environment and physiological property of aneurysm wall. This review focused on biomechanical mechanism of AAA enlargement after EVAR and its recent research progress, which indicated that reduction of the aneurysm wall strength due to deterioration of biomechanical environment (such as increase of tensile stress on aneurysm wall) and abnormity of biochemical environment (such as increase of LDL deposition, change of oxygen concentration) might be the source leading to AAA enlargement after EVAR. Based on previous studies, the authors propose that comprehensive investigations on AAA enlargement after EVAR from the perspective of biomechanics and mechanobiology have great research values and clinical significance, which will help clarify the mechanism of AAA enlargement after EVAR, as well as optimize the strategies of aneurysm repair surgery and designs of interventional medical devices.

Endothelial cells (ECs) and smooth muscle cells (SMCs) are the most crucial components of the vascular wall, and the interactions between them are essential for the maintenance of normal vascular physiology, as well as the initiation and development of cardiovascular diseases. Various typical co-culture models have been developed to simulate the location and growth situation of ECs and SMCs in vivo. In this review, the co-culture systems combined with the device applying fluid shear stress were introduced with discussion on the strengths and limitations of each system. The influences of ECs-SMCs interaction on the phenotype and alignment of ECs and SMCs, growth and migration of SMCs, and adhesion molecules expression of ECs under shear stress were briefly reviewed. The established evidence indicates that nitric oxide (NO), cytokines and microRNA are the most important signal molecules mediating the interactions between ECs and SMCs.

Objective To investigate the influence of graft-host diameter ratio on the flow field of fully-occluded artery bypass grafts and provide a theoretical guidance for reducing restenosis in artery bypass graft surgery. Methods Five models were employed to numerically investigate the influence of graft-host diameter ratio on the flow field of fully-occluded artery bypass grafts. The distributions of hemodynamic parameters such as velocity, second flow, wall shear stress (WSS) and wall shear stress gradient (WSSG) and their change in line with the increase of diameter ratio were analyzed. In addition, comparison of hemodynamic differences in a fully-occluded complete model (Model A), a fully-occluded partial model (Model B) and a 75% stenosis complete model (Model C) with graft-host diameter ratio of 1.0 was conducted to validate the fully-occluded complete model established in this study. Results The hemodynamic performance of Model A was totally different from that of Model C, and the velocity distribution at the graft top had an obvious influence on the WSS distribution at host artery bed in the downstream anastomosis, with maximum WSS differences reaching 79%. A large graft-host diameter ratio resulted in a large size of low WSS region at the host artery bed, but with uniformly distributed WSS and small WSSG. A small graft-host diameter ratio resulted in a small size of low WSS region at the host artery bed, but with large WSSG. Conclusions It is necessary to adopt a complete model to study the influence of graft-host diameter ratio on the flow field of fully-occluded artery bypass grafts. The diameter ratio had a significant impact on the flow field of fully-occluded artery bypass graft, thus a large ratio could be helpful to reduce the occlusion resulted from the restenosis at the downstream anastomosis in artery bypass graft.

Objective To investigate effects of pressure and tail suspension on the growth of rat skeletal muscles by establishing the tail-suspended rat model and pressure-induced rat model, respectively. Methods Thirty-six male Sprague-Dawley rats were randomly divided into three groups: the control-group (CON), the tail-suspended group (SUS), the pressure group (PRE), and each group was undergoing two stages (7, 14 d) for observation. At the end of the experiment, the muscle wet weight/body weight (Mww/Bw), muscle fiber cross-sectional area (MFCSA), muscle fiber diameter (MFD) of soleus and extensor digitorum longus (EDL) and the IGF-1 concentration were measured, respectively. Results For both the SUS group and PRE group with 7 d, their Mww/Bw, MFCSA and MFD of soleus were significantly reduced (P<0.05) as compared to the CON group, which were reduced by 23.52%, 14.26%, 13.47% in PRE group, respectively, while these indexes in SUS group were reduced by 23.52%, 33.07%, 25.09%, respectively. Meanwhile, the decrease of Mww/Bw, MFCSA and MFD in PRE group with 14 d was reduced by 20.51%, -10.49%, -5.73%, respectively, which was less than that in PRE group with 7 d. However, the decrease of Mww/Bw in SUS group with 14 d reached 46.15%, showing significantly higher than that of the SUS group with 7 d. For the IGF-1 concentration and EDL changes, no remarkable differences were found among the CON, PRE and SUS groups. Conclusions The impact of pressure on the process of soleus growth is different from that of the suspension (disuse). During the early stage of pressure application, the deep tissue injury mainly with inflammatory responses can be found in the muscle. When the muscle cells adapt to the pressure environment, they may produce a functional adaptation to growth. Therefore, such effect of socket pressure on skeletal muscle should be considered in clinic for prosthetic socket design or rehabilitation training, which will help protect the muscle tissues.

The aorta is the major blood vessel with spiral shaped geometry where the flow pattern is swirling in late systole. The swirling blood flow may have a positive physiological role in preventing cardiovascular diseases by reducing turbulent flow and enhancing mass transport. Several factors have shown to contribute to the formation of the swirling flow in the aorta such as the ventricular twisting, the 3D spiral shaped aorta, the pulsatile blood flow in the aorta and the motion of ascending aorta. Inspired by the above basic researches, the swirling flow mechanism has been applied to the vascular interventional therapies and the design of cardiovascular interventional devices. In the present review, the observation of swirling patterns in the aorta, its physiological significance, the factors contributing to its formation and its potential clinical application are summarized.

<p><b>OBJECTIVE</b>To investigate the effect of tyrosine kinase inhibitor ZD6474 (Vandetanib) on the proliferative inhibition of K562 cells and its derived imatinib-resistant K562/G cells and its mechanism.</p><p><b>METHODS</b>Imatinib-resistant K562/G cells were obtained by culturing cells in gradually increasing concentrations of imatinib. The changed factors related to drug-resistance were tested by Western blot. ZD6474 and imatinib affected K562/G and parental K562 cells proliferation were analyzed by WST assay. Flow cytometry was used to analyze cell cycle. Direct inhibition of Src activity by ZD6474 was measured by a colorimetric ELISA assay with recombinant human Src kinase.</p><p><b>RESULTS</b>10 µmol/L imatinib failed to inhibit K562/G cells proliferation or induce cell cycle arrest. Compared with that in parental K562 cells, there were marked high levels of p-Src and Src protein in K562/G cells. The expression of Bcl-2 and p-STAT3 also increased in K562/G cells. After 48 hours incubation, the IC(50) values of ZD6474 in K562 and K562/G cells were 1.61 µmol/L and 3.18 µmol/L, respectively. ZD6474 treatment caused accumulation of cells in the G(0)/G(1) fraction and cell apoptosis in K562 and K562/G cells. ZD6474 decreased the expression of p-Src and Src at post-transcriptional level. Moreover, ZD6474 increased the ratio of Bax/Bcl-2 and decreased the expression of p-STAT3 at the same concentration for inducing apoptosis.</p><p><b>CONCLUSIONS</b>ZD6474 is effective in inhibiting the proliferation of imatinib-resistant K562/G cells and parental K562 cells, and induces their apoptasis by significant inhibition of Src kinase activity. Our study provides a reliable experimental basis for chronic myeloid leukemia treatment with ZD6474.</p>

Objective To apply the swirling flow mechanism to the design of arterial prostheses with small diameters and the arterial bypass surgery to resolve the acute thrombus in small diameter prostheses after implantation, and lessen the restenosis of bypassed arteries due to the formation of internal hyperplasia. Method The computational fluid dynamics (CFD) method was used to investigate the flow field and wall shear stress distribution of a new graft, an S type bypass and an axis deviated arterial bypass, which all had the swirling flow. In addition, the platelet adhesion under swirling flow and internal hyperplasia in S-type bypass were measured. Results The swirling flow can apparently enhance the wall shear stress (WSS) and suppress the platelet adhesion and internal hyperplasia. Conclusions The swirling flow can significantly improve the flow field in arterial graft and bypass to inhibit the acute thrombus in small diameter prostheses and internal hyperplasia after bypass surgery.

<p><b>OBJECTIVE</b>To establish a set of suitable and reliable methods for HBV genotyping and to study the distribution of HBV genotypes.</p><p><b>METHODS</b>Type-specific nucleotides were searched through alignment of S genes (more than 1000 sequences) listed in GenBank. Then, type-specific primers were designed and type-specific primer PCR was used to genotype the 238 HBV strains. S genes of the untyped strains were further amplified and sequenced to find out their genotypes with type-specific nucleotide analysis.</p><p><b>RESULTS</b>All the 238 HBV strains were genotyped. 159 (66.8%) cases were genotype B, 69 (28.9%) were genotype C, 6 (2.5%) were mixtures of genotypes B and C and 4 (1.6%) were mixtures of genotypes B and D. No genotypes of A, E, F, G, and H were found.</p><p><b>CONCLUSION</b>Genotypes B and C are the most common types for HBV strains. Mixtures of genotypes B and C or genotypes B and D coinfection rarely existed. There is no relationship between the gender of the patients and HBV genotypes (X2 = 0.794, P more than 0.05).</p>

<p><b>AIM</b>To observe the effect of rhubarb ethanol-extract on hyperlipidemia and liver fatty in rabbits.</p><p><b>METHODS</b>Thirty healthy male white rabbits were divided randomly into five groups, six rabbits in each group. The rabbits in control group were fed with common forage. The rabbits in model group were fed with high lipid forage. The rabbits in three different rhubarb groups were fed with high lipid forage and treated with different level rhubarb ethanol-extract (REE). In the process of experiment, periodically measured serology index of the rabbits and observed common physiology index. The rabbits were killed at the end of tenth week, liver fatty degeneration degree and liver coefficient were measured and compared.</p><p><b>RESULTS</b>REE could decrease serum total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C), and increase serum high density lipoprotein cholesterol (HDL-C), and reduce liver fatty de generation and protect liver cell function. And the dose-effect relation was showed among different dose REE groups.</p><p><b>CONCLUSION</b>REE can significantly reduce blood lipid, prevent and treat hyperlipidemia and liver fatty.</p>
