RESUMO

Bacteremia induced by periodontal infection is an important factor for periodontitis to threaten general health. P. gingivalis DNA/virulence factors have been found in the brain tissues from patients with Alzheimer's disease (AD). The blood-brain barrier (BBB) is essential for keeping toxic substances from entering brain tissues. However, the effect of P. gingivalis bacteremia on BBB permeability and its underlying mechanism remains unclear. In the present study, rats were injected by tail vein with P. gingivalis three times a week for eight weeks to induce bacteremia. An in vitro BBB model infected with P. gingivalis was also established. We found that the infiltration of Evans blue dye and Albumin protein deposition in the rat brain tissues were increased in the rat brain tissues with P. gingivalis bacteremia and P. gingivalis could pass through the in vitro BBB model. Caveolae were detected after P. gingivalis infection in BMECs both in vivo and in vitro. Caveolin-1 (Cav-1) expression was enhanced after P. gingivalis infection. Downregulation of Cav-1 rescued P. gingivalis-enhanced BMECs permeability. We further found P. gingivalis-gingipain could be colocalized with Cav-1 and the strong hydrogen bonding between Cav-1 and arg-specific-gingipain (RgpA) were detected. Moreover, P. gingivalis significantly inhibited the major facilitator superfamily domain containing 2a (Mfsd2a) expression. Mfsd2a overexpression reversed P. gingivalis-increased BMECs permeability and Cav-1 expression. These results revealed that Mfsd2a/Cav-1 mediated transcytosis is a key pathway governing BBB BMECs permeability induced by P. gingivalis, which may contribute to P. gingivalis/virulence factors entrance and the subsequent neurological impairments.

RESUMO

The spleen is the most vulnerable abdominal parenchymal organ. Accurate detection, diagnosis and evaluation of the severity of splenic parenchyma and vascular injury by radiologists is crucial for selection of clinical treatment strategies. Multidetector computed tomography (MDCT) is the first choice for hemodynamically stable patients. The American Association for Surgery of Trauma-Organ Injury Scale (AAST-OIS) scale is the most widely used tool to guide clinical treatment based on spleen parenchymal injury. The authors conduct a review from aspects of MDCT diagnosis, AAST-OIS classification and treatment, prognosis evaluation and imaging follow-up of patients with blunt splenic injury, in order to provide more help for radiologists and trauma surgeons.

Objective:To study the application and effect of hybrid teaching based on the rain classroom in the teaching of periodontology for undergraduates.Methods:Sixty-four stomatological undergraduates in Phase 101 were selected as experimental group, and 62 in Phase 100 as control group. The hybrid teaching based on the rain classroom were conducted in experimental group, and mind mapping were used before and after class to help students understand and expand the content of class. The traditional teaching was adopted in the control group. SPSS 17.0 was applied to compare the differences between the written test scores and teaching satisfaction evaluation of the two groups. The learning behavior data of the students in experimental group before, during and after class were collected and analyzed.Results:The score of the written test in the experimental group were significantly better than that in the control group ( t=2.97, P<0.05). The result of questionnaire showed that the experimental group was significantly more satisfied with the improvement of lecture efficiency ( t=2.71, P<0.05) and self-learning ability ( t=4.35, P<0.05). The majority (82.55%) of students in the experimental group are in favor of the rain classroom; 89.06% of students finished the preview before class, and 67.19% of them completed the review after class. Conclusion:The hybrid teaching based on the rain classroom can be applied to the teaching of periodontology, which can effectively improve the learning quality and initiative of the students.

Objective To evaluate the rehabilitation efficacy of music training in pediatric cochlear implants (CIs).Methods The 14 subjects were divided into the training and control groups.The age of the training group was 5 to 8 years old,with an average of 6.12±1.46.The age of the control group was 5 to 7 years old,with the average of 5.86±0.90.The subjects in the training group received speech rehabilitation training together with music training,while those in the control group only received speech rehabilitation training.We performed an analysis to compare the music perception,speech perception,mental state,and the quality of life between the two groups before and after training.Results For training group,the score of social anxiety scale for children(SASC) was 11± 3.27 before training,and 15.71±1.89 after training;the score of note discriminate test was 30.86±13.61 before training,and 42.29± 13.24 after training.For control group,the scores of SASC was 12.71±2.93 before training,and 14.57±3.51 after training;the pre-training score of note discriminate test was 33.14±11.48,and post-training score was 36.57± 10.44.After training,the scores of the two test items in training group were significantly higher than those in control group (P<0.05).Conclusion Not only does music training promote music and speech perception in pediatric CIs,it also has proved to be an effective psychological construct for the hearing-impaired children that helps develop self-confidence and improve social status.Moreover,children's improvement in hearing and emotional well-being helps alleviate the psychological stress and anxiety experienced by the children's parents.

Objective To establish a method for determination of phenylethanoid glycoside and acteoside in Plantago Herba. Methods UV-visible spectrophotometric method was used for the determination of the content of phenylethanoid glycosides compounds in Plantago Herba. HPLC method was used for the determination of acteoside in Plantago Herba. Chromatographic column with C18 ODS2 (4.6 mm × 150 mm, 5 μm) was used. Acetonitrile-0.1%formic acid (13:87) was as mobile phase; the flow rate was 1 mL/min; the detection wavelength was 332 nm; the column temperature was 30 ℃; the sample volume was 10 μL. Results The contents of phenylethanoid glycoside in Plantago Herba from different producing areas were among 1.03%–3.47%. Acteoside with peak area over the 0.0062–1.55 mg range showed a good linear relationship; the sample recovery rate was 98.9%, and the RSD was 1.6%. The contents of acteoside in Plantago Herba from different producing areas was among 0.18%–0.56%. Conclusion The method is simple, stable and reproducible, which can be used for the determination of phenylethanoid glycoside and acteoside in Plantago Herba from different producing areas and provide experimental basis for quality control of Plantago Herba.

Objective To explore the natural courses of 11 patients with cesarean scar pregnancy (CSP) with expectant management. Methods Eleven patients with CSP who were diagnosed in the first trimester in the Third Affiliated Hospital of Guangzhou Medical University from January 2015 to March 2017 were recruited. All of them received expectant management. Nine pregnancies continued to the third trimester (the third trimester group), and 2 patients were expected to the second trimester (the second trimester group). The gestational age at diagnosis, CSP type, gravidity, parity, miscarriage and previous cesarean section history, gestational weeks at termination, amount of postpartum hemorrhage, prenatal and postnatal hemoglobin levels, pregnancy outcomes and obstetric complications were compared between the two groups. Results The third trimester group terminated pregnancies between 33 + 2 and 36 + 5 weeks. The second trimester group terminated in the second trimester because of rupture of uterus (at 17+2,17+3 weeks). There was no statistical difference between the two groups regarding the number of gravidity, parity and previous cesarean section(all P>0.05). The number of miscarriage in the second trimester group was 4.0 ± 2.8,and in the third trimester group was 1.3 ± 1.1(P<0.05). In the third trimester group, 7 cases were CSP typeⅠand 2 cases were CSPⅡ. In the second trimester group, 2 cases were both CSP typeⅢ. Eleven cases were diagnosed placenta accreta pathologically. There was no maternal death,and 6 cases received hysterectomy (6/11). The amount of postpartum hemorrhage increased remarkably and all neonates survived (pregnancy terminated between 33+2 and 36+5 weeks). Conclusions For those diagnosed as CSP typeⅠandⅡwho urge to continue pregnancies, it′s plausible to have expectant management with fully consent of obstetric hemorrhage, rupture of uterus and hysterectomy and close monitoring in tertiary hospital. The detailed expectant management of CSP is needed further exploration.

Objective: To determine total phenylethanoid glycoside and acteoside in Plantago Herba to provide reference for evaluating the quality of medicinal materials.Methods: With acteoside as the control sample, a UV visible spectrophotometric method was used to determine total phenylethanoid glycosides in Plantago Herba.An HPLC method was applied to determine acteoside in Plantago Herba , and the conditions were as follows: an ODS2 C 18 (150 mm× 4.6 mm ,5 μm) chromatographic column was used with acetonitrile-0.1% formic acid (13∶87) as the mobile phase at a flow rate of 1.0 ml·min-1 , the detection wavelength was 332nm, the column temperature was 30℃, and the sample volume was 10 μl.Results: The reference solution and the sample solution had the maximum absorption at 332 nm, and the linear relationship was good within the range of 0.003 1-0.155 0 mg·ml-1 (r=0.999 5).The content of total benzene alcohol glycosides in 3 batches of samples was 2.73% , 2.61% and 2.84% , respectively;acteoside over the range of 0.000 6-0.155 0 mg·ml-1 (r=0.999 1) showed a good linear relationship with peak area,the sample recovery was 98.5% and the RSD was 1.6% (n =6), and the acteoside content in 3 batches of samples respectively was 0.54% , 0.51% and 0.56%.Conclusion: The method is simple, accurate and reproducible, and can be used for the determination of total phenylethanoid glycosides and acteoside in Plantago Herba.

Objective To explore the effect of dexamethasone(Dex)treatment on the expression of interleukin(IL)?6 and IL?8 in human peri?odontal ligament cells(hPDLCs). Methods hPDLCs were subjected to one of the following treatments for 24 h or 48 h:10-9 mol/L Dex;10-6 mol/L Dex;10μg/mL Porphyromonas gingivalis(P. g)?lipopolysaccharide(LPS);10-9 mol/L Dex+10μg/mL P. g?LPS;10-6 mol/L Dex+10μg/mL P. g?LPS;or 0.1%absolute ethyl alcohol(control). Protein and mRNA expression was detected using ELISA and real?time PCR ,respectively. Results At 24 h and at 48 h,IL?6 and IL?8 protein expression in the 10-9 mol/L Dex?and 10-6 mol/L Dex?treated groups was significantly lower than that in the control group(P<0.05). At 48 h,IL?6 mRNA expression in the 10-9 mol/L Dex?and 10-6 mol/L Dex?treated groups was signifi?cantly lower than that in the control group(P<0.05),while IL?8 mRNA expression in the 10-6 mol/L Dex?treated group was significantly higher than that in the control group(P<0.05). At 24 h and at 48 h,IL?6 protein and mRNA expression in the 10-9 mol/L Dex+10μg/mL P. g?LPS?treated group and the 10-6 mol/L Dex+10μg/mL P. g?LPS?treated group was significantly lower than that in the 10μg/mL P. g?LPS?treated group (P<0.05). At 24 h,IL?8 protein expression in the 10-9 mol/L Dex+10μg/mL P. g?LPS?treated group and the 10-6 mol/L Dex+10μg/mL P. g?LPS?treated group was significantly lower than that in the 10μg/mL P. g?LPS?treated group(P<0.05),while no such significant difference exist?ed at 48 h. At 48 h,IL?8 mRNA expression in the 10-6 mol/L Dex+10μg/mL P. g?LPS?treated group was significantly higher than that in the 10μg/mL P. g?LPS?treated group(P<0.05). Conclusion Dex inhibits the innate and P. g?LPS?induced expression of IL?6 in hPDLCs. However, Dex exerts profound effects on IL?8 expression,and treatment with high doses of Dex may promote IL?8 expression over an extended period.

<p><b>OBJECTIVE</b>This study was intended to determine the salivary protein factors related to adult periodonititis.</p><p><b>METHODS</b>Twenty-five patients with adult periodontitis (AP group) and twenty-five normal controls (NC group, 25) were adopted in the study. Salivary protein composition in unstimulated whole saliva samples obtained before and after basic treatment was analyzed by SDS-PAGE.</p><p><b>RESULTS</b>The volume of proteins with 66 kD, 18 kD, 15 kD, 13 kD in AP group before treatment were all remarkably higher than those in NC group. In addition, 51 kD, 34 kD, 19 kD, 17 kD protein bands were more frequently observed in AP group by comparison with NC group. However, after initial therapy, all the variables mentioned above decreased remarkably.</p><p><b>CONCLUSION</b>The results demonstrated that protein bands with 18 kD, 15 kD, 13 kD might be closely related to the occurrence and recovery of periodontitis, and serum-derived proteins in saliva increased in patients with adult periodontitis.</p>

RESUMO

Objective: Our aim was to analyze the relationship between the concentration of some minerals in saliva and periodontitis.Methods:Twenty-five patients with adult periodontitis (AP group), 25 age and sex matched healthy people(N group) were selected. Unstimulated whole saliva samples were collected and qualified for the concentration of TCa,P,Mg2+, Na+,K+, and Cl-. Clinical parameters on periodontitis were also assessed for the patients. All the aforementioned indices were measured repeatedly at the end of the fourth week after initial therapy. Results:The clinical condition of the patients improved significantly after treatment. The level of saliva Ca elevated before and after treatment in AP group than that in N group. Before treatment, the concentration of Na+ also increased significantly, but it decreased to normal level after initial therapy. The findings also indicated the positive relationship between the concentration of salivary Na+ and pocket probing depth as well as gingival index before treatment.Conclusion:The elevated level of saliva Ca is a sign of periodontitis and may help the occurrence and progress of periodontitis. The level of saliva Na+ may indicate the pocket depth and degree of gingivitis.The saliva Na+ is closely associated with the clinical condition of periodontitis.

Objective:To investigate the effects of Candida albicans(C. albicans) on the cell cycle distribution of oral epithelial KB cells in vitro. Methods:KB cells at 3?106/ml were seeded and cultured for 48 h, then the bacterial filament and the spore of C. albicans at 102 CFU/ml were added to the cultures respectively. KB cell culture without C. albicans was used as the control. After 48 h culture cell suspensions were prepared and subjected to FACS flow cytometry for detecting the cell cycle distribution. RESUMO

The pharmacokinetics of domestic or imported nimesulide tablet was determined following a single oral dose of 200 mg given to 10 volunteers in randomized crossover study. The plasma concentration of nimesulide was assayed by HPLC method. The concentration -time curve of nimesulide conformed to a one-compartment model and the main parameters of domestic nimesulide were as follows: T1/2Ke 3. 61?1.43 h; Tpeak 2. 07?0. 63 h; Cmax=9. 46?2. 06 mg?L-1 ; AUC = 76. 39?17. 62 mg?L-1?h-1 respectively. The relative bioavailability of domestic tablet was 92.2%. The results of three factors analysis of variance and Bayesian method showed that two formulation were bioequiva-lent.
