RESUMO

Aim To design and synthesizea new pyrazolo[4,3-d] pyrimidine derivative for the purpose of developing novel anti-inflammatory agents, and to revealthe possible anti-inflammatory mechanisms of thenew compound using preliminary studies. Methods The changes of the cell morphology were investigated via the microscope. The influence of title compound on the NO production of the L P S - stimulated RAW264. 7 cells was measured by Griess assay. The gene relative transcription levels of TNF-a, IL - 6 and IL-lß were evaluated by qRT-PCR. COX-2 expression was measured by qRT-PCR and western blot. NF-KB/NLRP3 inflammasome signaling pathway were investigated by Western blot. The changes of lung tissue were observed by HE staining in vivo experiments of mice. Results Preliminary mechanism researches revealed that the ti - tle compound could inhibit nitric oxide (NO) generation as well as the expression of COX-2, TNF-a, IL - 6, and IL - lβ by NF-KB/NLRP3 inflammasome signaling pathway in RAW264. 7 cells. Furthermore, it could simultaneously improve the morphology of the cells and relieve acute lung injury in mice. Conclusions The new pyrazolo [4, 3-α] pyrimidine derivative has anti-inflammatory activity through NF-KB/NLRP3 inflammasome signaling pathway.

RESUMO

AIM: To evaluate the clinical results of capsular tension ring (CTR) implantation in phacoemulsification for eyes with congenital lens subluxation.METHODS: This study comprised 18 patients (31 eyes) with congenital ectopia lentis.All patients received phacoemulsification with CTR and intraocular lens (IOL) implantation.Visual acuity before and after surgery were examined.IOL decentration were measured with Image-Pro Plus image processing software.The complications were also recorded preoperatively and postoperatively.RESULTS: Uncorrected vision of all eyes increased after surgery, and all cases had different degree of IOL decentration after surgery.Two eyes received CTR scleral fixation 3mo after surgery because the IOL decentration aggravated during the follow-up period.CONCLUSION: Phacoemulsification with CTR and IOL implantation is effective and safe for congenital lens subluxation.The implantation of CTR increase the stability of the capsular bag, improve the safety of surgery, and maintain the right position of IOL.CTR scleral fixation is a effective supplement for progressive cases.

RESUMO

NSCL/P is a common congenital defect and gene-environmental factors involve in this disorder. Periconceptional intake of folate may reduce the risk of NSCL/P. The present study investigated three SNPs (rs1801198, rs955516, and rs3733890) in three folate pathway genes, including TCN2, MTR, and BHMT among 481 patients and 558 healthy subjects. Rs955516 showed allelic association with NSCL/P. More patients carry rs955516 AA and rs3733890 AA genotypes. The gene-gene interaction test showed trans-phase combination effects for MTR and BHMT genes. Our study suggests that the interaction of MTR and BHMT genes play a vital role in the pathogenesis of NSCL/P in Chinese population.

<p><b>OBJECTIVE</b>To investigate the clinical effect of spanishneedles leaves on middle and severe xerophthalmia of menopausal females.</p><p><b>METHOD</b>This study was a prospective random controlled trial. Ninty-six menopausal females diagnosed with xerophthalmnia (aged from 40 to 50) were randomly divided into in two groups: group A' the spanishneedles leaves group (n=48) and group B' the control group (n=48). Both groups were treated with Forte eye drops. All patients were detected at 3, 7, 28 h before and after treatment to evaluate subjective symptoms, OSDI and four tear film indicators. Variance analysis and differential analysis on sample average or median were made on both groups before and after treatment.</p><p><b>RESULT</b>There were no significant difference in symptom and diction indicators between both groups before treatment. For 28 d after treatment, among middle and severe xerophthalmia samples of the spanishneedles leaves group, the mean differences showed significant improvement compared with that before treatment , OSDI and four tear film indicators also showed improvement to varying degrees. For 28 d after treatment, among middle and severe xerophthalmia samples of the vitamin C group, the mean differences showed no significant improvement compared with that before treatment , OSDI and four tear film indicators also showed no remarkable improvement. There were significant differences in OSDI, BUT, SIT, height of tear meniscus and FL between both groups.</p><p><b>CONCLUSION</b>Spanishneedles leaves can effectively improve symposiums and signs of middle and severe xerophthalmia among menopausal females and thus showing clinical significance to some extent.</p>

Objective In order to provide a reference for analysis of the pathophysiological changes of human arterial tree, the effects of the arterial parameters on input impedance of arterial tree were studied by proposing a recursive algorithm to calculate the input impedance. Method A distributed electric network model was constructed for the arterial tree which consisted of 55 segments accounting for all the central vessels and major peripheral arteries. By setting up the peripheral resistance of the model and constructing a unilateral data link list of the arterial tree, the recursive algorithm was employed to compute the input impedance of ascending aorta in the arterial tree. Then, comparisons of the effects of arterial compliance, peripheral resistance, length, internal radius and wall thickness on the input impedance were made based on the recursive algorithm. Results The computational results were in accord with experimental data and results of other models, and proved the validity of the model and algorithm. The input impedances of the arterial tree showed significant influence with different characteristics for different parameters. Conclusions The input impedance of arterial tree can reflect the changes of the hemodynamic parameters effectively, which is an important reference for the pathophysiological diagnosis of human arterial tree.

<p><b>OBJECTIVE</b>To investigate the association between a polymorphism of methylenetetrahydrofolate reductase with Non-syndromic cleft lip with or without cleft palate (NSCL/P) in Chinese population.</p><p><b>METHODS</b>The polymerase chain reaction (PCR)-based restriction fragment length polymorphism (RFLP) technique was used to detect a single nucleotide polymorphism (SNP), rs1801131, at the methylenetetrahydrofolate reductase (MTHFR) gene in both 158 patients with NSCL/P and 192 healthy individuals. The Hardy-Weinberg equilibrium for genotypic distributions was estimated by the goodness-of-fit test. The UNPHASED program was applied to perform the association analysis.</p><p><b>RESULTS</b>The genotypic distribution of A1298C was not deviated from the Hardy-Weinberg equilibrium in both controls and patients. No association was found between cleft lip with or without palate (CL/P) and controls. There was significant difference of cleft palate only (CPO) and the healthy individuals (χ(2) = 4.256, P = 0.039). The frequency of AC + CC genotype was higher in control group than that in CPO group (OR = 0.8, 95%CI = 0.381 - 1.683), 26 among 100 healthy individuals carried AC + CC genetypes, which were carried by 22% of CPO patients.</p><p><b>CONCLUSIONS</b>The polymorphism of MTHFR A1298C may be involved in the occurrence of non-syndromic cleft palate only in Chinese population.</p>

<p><b>OBJECTIVE</b>To analyze the feasibility and reliability of free anterolateral thigh flap transfers in head and neck reconstruction.</p><p><b>METHODS</b>Twenty-two consecutive free anterolateral thigh flap transfers from January 2002 to January 2006 were reviewed. Data concerning the operation included date of surgery, defect description and site, stage and histology of tumor, design of anterolateral free flap, type of perforators, recipient vessel and type of anastomosis used and complications.</p><p><b>RESULTS</b>There were 13 males and 9 females in this group, with age range of 35 to 70. Among 26 perforators, there were 6 septocutaneous perforators and 20 musculocutaneous perforators. One flap developed venous thrombosis 48 hours after operation, and the flap was removed after failed salvage. No vessel thrombosis occurred during and after operation with other flaps, which survived completely. All the donor sites were closed directly without skin graft. Apart from one case, all the donor sites healed uneventfully.</p><p><b>CONCLUSIONS</b>Free anterolateral thigh flap is a safe and reliable donor site in the reconstruction of head and neck defects. It overcomes the shortcomings of traditional free radial forearm flap and free rectus abdominis myocutaneous flap.</p>

RESUMO

Herba leonuri (HL) is a famous Chinese traditional medicine with long history. Recent studies have shown that the effects of HL are versatile. In addition to its activating effect on blood flow which has been widely used in gynecological and obstetrical disorders, HL has been demonstrated to have beneficial effects on ischemic diseases including myocardial infarction (MI). Experimental studies have shown that HL ameliorated myocardial ischemia, increased coronary blood flow, and improved heart functions. The underlying mechanism is proved to be its antioxidant effects including scavenging free radicals and inhibiting the formation of reactive oxygen species in ischemic myocardium. The antioxidant effects of HL are exerted only under the condition of oxidative stress. A significant angiogenesis promoting effect is also counted for its cardioprotective effects. Clinic trials also indicated that HL could inhibit blood platelet congregating, resist coagulation, restrain thrombosis, and improve hemorheology in patients with coronary heart diseases. This paper gives a brief review on HLos cardioprotective effects based on recent research progress.

RESUMO

<p><b>OBJECTIVE</b>A new injury severity score system was proposed in this study to evaluate maxillofacial injuries.</p><p><b>METHODS</b>Based on AIS-90, all clinical samples were given diagnosis. Their month opening limitation, malocclusion and facial deformity were recorded as indices in scoring system. Severity injury scores for different groups and types were statistically. analyzed.</p><p><b>RESULTS</b>It was demonstrated that there was a significant difference in scores between injuries of soft tissue and bone tissue and between different fracture groups.</p><p><b>CONCLUSIONS</b>The proposed scoring was system useful to predict the cost of medical care and the days of in-patient, reliable, sensitive and specific in evaluation the maxillofacial injury severity.</p>
