RESUMO

OBJECTIVE@#To investigate the effect of danusertib (Danu), an inhibitor of Aurora kinase, on the proliferation, cell cycle, apoptosis, and autophagy of hepatocellular carcinoma HepG2 cells and explore the underlying mechanisms.@*METHODS@#MTT assay was used to examine the effect of Danu on the viability of HepG2 cells to determine the IC50 of Danu. The effect of Danu on cell cycle distribution, apoptosis and autophagy were determined using flow cytometry. Western blotting was used to detect the expressions of the proteins related to cell cycle, apoptosis and autophagy. Chloroquine was used to suppress Danuinduced autophagy to test the apoptosis-inducing effect of Danu.@*RESULTS@#Danu significantly inhibited the proliferation of HepG2 cells with IC of 39.4 μmol and 14.4 μmol at 24 h and 48 h, respectively. Danu caused cell cycle arrest in G/M phase in HepG2 cells and led to polyploidy accumulation via up-regulating the expressions of p53 and p21 and down-regulating the expressions of cyclin B1 and DC2. Danu also caused apoptosis of HepG2 cells through up-regulating the expressions of Bax, Puma, cleaved caspase-3, cleaved caspase-9, cleaved PARP and cytochrome C and down-regulating the expressions of Bcl-xl and Bcl-2. Danu induced autophagy via activating AMPK signaling and inhibiting PI3K/PTEN/AKT/mTOR axis, and inhibition of Danu-induced autophagy with chloroquine enhanced the pro-apoptotic effect of Danu.@*CONCLUSIONS@#Danu inhibits cell proliferation and induces cell cycle arrest in G/M phase, apoptosis and cytoprotective autophagy in HepG2 cells.

Objective@#To investigate the association between single nucleotide polymorphisms (SNP) in cytokine IL6, IL10 genes and the susceptibility to primary hepatic carcinoma(PHC) of Shunde district in Guangdong Province.@*Methods@#Patients from two hospitals in Shunde District of Foshan City were selected from October 2010 to October 2012. Case group inclusion criteria includedprimary liver cancer; local residents of Shunde or living in Shunde for more than 10 years. The control group inclusion criteria included: patients visited ENT, general surgery and physical examination department in the same hospital during the same period; Local residents or living in Shunde for 10 years and above. The control group was matched 1∶1 by gender, and age (±3 years old) with case group. A total of 306 subjects were collected. Questionnaires were used to investigate the information including demographic characteristics, PHC status survey and so on. The venous blood was collected from each subject to extract DNA, and to detect label SNP site and genotype. Hardy-Weinberg equilibrium was detected in the control group by the goodness-of-fit χ2 test. Multivariate conditional logistic regression was used to estimate the relationship between IL6 (rs1800796), IL10 (rs1800871, rs1800872)genes polymorphisms and susceptibility to PHC.@*Results@#There were 264 males and 42 females both in the case group and the control group, with an average age of (55.84±11.49) and (55.83±11.67) years old respectively (t=0.011, P=0.992). The frequencies of IL6 (rs1800796), IL10 (rs1800871) and IL10 (rs1800872) genotypes in the control group were in accordance with the Hardy-Weinberg equilibrium, which indicated the population was representative (all P values>0.05). Conditional logistic regression analysis showed that compared with the AA genotype and AA+AC genotype of IL10(rs1800872), CC genotype increased the risk of PHC by 2.02 times (OR=3.02, 95%CI:1.21-7.56)and 1.89 times (OR=2.89, 95%CI:1.19-7.04)respectively after the smoking history, eating fish history, drinking history, chronic hepatitis B infection, and family history of liver cancer adjusted. No statistical association was found between SNP in cytokine IL6 (rs1800796) and the susceptibility to PHC (P>0.05).@*Conclusion@#The results indicated that people who carried CC genotype in rs1800872 of IL10 gene have an increasing risk of PHC.

RESUMO

<p><b>OBJECTIVE</b>To investigate the basic incidence of subjective tinnitus in Xingui Community, Daliang, Shunde District, Guangdong Province, conduct preliminary analysis on its clinical feature, provide scientific evidence for subjective tinnitus prevention and cure in community.</p><p><b>METHODS</b>Performed census in the entire population, totally 17253 people in Xingui Community, then gave tinnitus surveys for the people who have subjective tinnitus, and finally conducted analysis and evaluation.</p><p><b>RESULTS</b>the morbidity of tinnitus in the investigated people is 28.7%. With the increases of age, the morbidity goes up gradually, but it is not statistically significant(P>0.05) among different age group, and between different sex. There is an obvious correlation between tinnitus and hearing loss; Matching tone of tinnitus is related to the nature of hearing loss. The morbidity of decompensation tinnitus is 3.2%, it is not statistically significant among different age group(P>0.05).</p><p><b>CONCLUSION</b>subjective tinnitus is common in aged people, so it is very important to strengthen the work of subjective tinnitus prevention and cure in Community.</p>

Objective To study dexamethasone and higher C-reactive protein(CRP) on effects of platelets concentrates(PC) transfusion for bleeding patients with thrombocytopenia.Methods To investigate peripheral blood platelets count,clinical bleeding before(CRP test simultaneously) and 12~16 hours after PC transfusion for 45 non-fever,non-splenomegaly and non-DIC(disseminated intravascular coagulation,DIC) patients with leukemia,aplastic anemia,or solid tumor chemotherapy,which had bleeding and needed PC transfusion.All cases were divided into two groups:CRP normal and CRP higher one according to their CRP results.Each group were also classified as non-dexamethasone and dexamethasone division(10mg dexamethasone intravenous injection before transfusions) respectively,in order to analyze the relationships between effects of PC transfusion and serum levels of CRP or dexamethasone.Results The peripheral blood platelets counts (33.2?9.2)?109/L after PC transfusions in CRP normal group were more than that (25.4?10.4)?109/L,in CRP higher group,with significant differences(t=4.42,P
