RESUMO

Objective:To investigate eye movement desensitization and reprocessing on ICU memory and post-traumatic stress disorder among patients with ventilator associated pneumonia.Methods:Totally, 66 cases of ICU patients with ventilator associated pneumonia enrolled at Guangzhou Red Cross Hospital from August 2017 to August 2020 were collected as research object, they were randomly divided into intervention group and control group, with 33 cases in each groups. The control group received routine nursing care and cognitive training, in the intervention group, patients implemented EMDR therapy for 8 times. Before and after intervention, the effect was assessed by ICU memory scale and PTSD Checklist-Civilian Version, respectively.Results:After intervention, the scores of factual memories, emotional memories, delusional memories dimension and total ICUMS were (19.48±5.14), (10.45±2.62), (7.06±2.62) and (37.00±6.12) in the intervention group, those scores in the control group were (24.58±7.38), (13.58±4.07), (9.48±3.57) and (47.64±9.99), the difference was statically significant (t values were 3.14-5.22, all P<0.05). The re-experiencing, avoidance, hyperarousal dimensions scores and total PCL-C scores were (8.09±2.13), (10.42±2.62), (7.24±2.69) and (25.76±4.25) in the intervention group, and in the control group those scores were (9.30±2.35), (12.73±2.11), (9.24±2.51) and (31.27±4.97), the difference was statically significant (t values were 2.20-4.85, all P<0.05). Conclusions:Eeye movement desensitization and reprocessing therapy can effectively alleviate ICU memory and improve PTSD symptoms of patients with ventilator associated pneumonia.

Objective To study the expression and clinical significance of ARHGAP4 in colorectal cancer Method Real?time PCR,Western blot and immunocytochemistry were used to detect the expression of ARHGAP4 in colorectal cancer tissues and cell lines ,and the correlation between its expression and clinical features of patients was analyzed Results ARHGAP4 is overexpressed in colorectal cancer tissues and cell lines and its overexpression is correlated with T stage, N stage, clinical stage, and metastasis. Conclusion ARHGAP4 may promote the progression of colorectal cancer ,and have the potential to be a novel prognosis marker.

Objective:To study the effects of down-regulated Smad4 expression on the proliferation and apoptosis of breast carcinoma MCF-7 cells,and to explore their mechanisms.Methods:The human breast carcinoma MCF-7 cells and MDA-MB-231 cells were cultured in vitro.RT-PCR method was used to detect the expression levels of Smad4 mRNA in MCF-7 cells and MDA-MB-231 cells.The Smad4-shRNA plasmid and Scramble-shRNA plasmid were respectively stably transfected into the MCF-7 cells with high expression of Smad4.The experiment was divided into non-transfected MCF-7 cells (normal control) group,Smad4 gene silencing group,Scramble(negative control) group.The proliferation abilities of the cells in various groups were detected by CCK-8 method.The apoptotic rates of the cells in various groups were detected by flow cytometry.Real-time PCR method wasused to detect the mRNA expression levels of the proliferation-related genes CDKN1A,CDK1 and CDK2 and the apoptosisrelated genes Suvivin,bcl-2,caspase 3 and caspase 9.Results:The proliferation abilities of cells had no statistical significance between various groups (P>0.05).The mRNA expression levels of CDKN1A,CDK1 and CDK2 in the cells had no statistical significance between various groups (P>0.05).Compared with normal control group and negative control group,the apoptotic rate of the cells in Smad4 gene silencing group was significantly decreased (P<0.01),the expression levels of Suvivin and bcl-2 mRNA in Smad4 gene silencing group were significantly increased (P<0.01),and the mRNA expression levels of caspase 3 and caspase 9 in Smad4 gene silencing group were significantly decreased (P<0.05).Conclusion:Smad4 could induce the apoptosis of MCF-7 cells by downregulating the expressions of Suvivin and bcl-2 and up-regulating the expressions of caspase 3 and caspase 9.

PURPOSE: The aim of this study is to explore signal transducer and activator of transcription 3 (STAT3) expression in breast cancer and to analyze the detailed mechanism that STAT3 contributes to the progression of breast cancer. METHODS: We retrospectively analyzed the clinicopathologic characteristics and overall survival (OS) of 140 breast cancer patients after curative surgery, and detected STAT3 expression, phosphorylated STAT3 (pSTAT3) expression, Ki-67 expression, vascular endothelial growth factor (VEGF)-C and -D expression in breast cancer tissues, and adjacent nontumor tissues. Survival analysis and relationship analysis were adopted for demonstrated the important mechanism of STAT3 contribution to progression of breast cancer. RESULTS: STAT3 expression, pSTAT3 expression, Ki-67 expression, VEGF-C expression, and VEGF-D expression in breast cancer tissues were significantly higher than those in adjacent nontumor tissues, respectively. With survival analysis, only number of lymph node metastasis (N stage) was identified as the independent predictors of the OS of breast cancer patients. Besides, we demonstrated there was the most prominent correlation between STAT3 expression and lymph node metastasis in breast cancer tissues by using the multinominal regression method. CONCLUSION: STAT3, a poor survival biomarker potential association with lymph node metastasis, was suitable for predication the OS of breast cancer patients after curative resection.

RESUMO

Musashil is an evolutionarily conserved RNA binding protein. It plays an important role in the aspects of maintaining stem cell state, differentiation and tumorigenesis. Notch signal pathway plays an important role in the proliferation of neural stem cells after cerebral ischemia. This article briefly introduces the structure and molecular characteristics of Musashil and Notch, and reviews the interaction in the neurogenesis of Musashil and Notch signaling pathway after cerebral ischemic reperfusion.
