RESUMO

OBJECTIVE To study the protective effects of mangiferin against oxidative stress injury of myocardial cells induced by hydrogen peroxide (H2O2), and its effects on the expression of nuclear factor of activated T cell cytoplasmic 4(NFATc4). METHODS H9c2 myocardial cells were cultured in vitro and divided into blank group, H2O2 group, and 50, 100, 150 μmol/L mangiferin groups. Mangiferin groups were treated with different concentrations of mangiferin for 12 h, and then were subjected to H2O2 (200 μmol/L) stimulation for 12 hours together with the H2O2 group; relative survival rate was detected in each group, and the levels of superoxide dismutase (SOD), catalase (CAT) and malondialdehyde (MDA) in cell supernatant and reactive oxygen species (ROS) in H9c2 cells were measured. Meanwhile, the expressions of apoptosis-related proteins [B cell lymphoma 2 (Bcl- 2), Bcl-2 associated X protein (Bax), cleaved caspase-3] and nuclear protein NFATc4 were determined. Furthermore, the NFATc4 interference sequence was transfected, and the effects of NFATc4 on oxidant stress indexes and apoptosis-related proteins in H2O2- induced myocardial cells were investigated. RESULTS Compared with blank group, relative cell viability, the levels of SOD and CAT, relative expression of Bcl-2 were decreased significantly, while the levels of MDA and ROS, relative expressions of Bax, cleaved caspase-3 and nuclear protein NFATc4 were increased significantly (P<0.05 or P<0.01). Compared with the H2O2 group, the above indexes of 100 and 150 μmol/L mangiferin groups were reversed significantly (P<0.05). After the transfection of the NFATc4 interference sequence, the expression of nuclear protein NFATc4 was down-regulated significantly; the levels of MDA and SOD, the protein expressions of Bax and cleaved caspase-3 were all decreased/down-regulated significantly, while the levels of SOD and CAT, and the protein expression of Bcl-2 were all increased/up-regulated significantly, compared with H2O2 group (P< 0.05). CONCLUSIONS Mangiferin can relieve H2O2-induced oxidative stress of H9c2 cells, reduce the apoptosis and inhibit the nuclear translocation of NFATc4, thereby alleviating myocardial cell damage; reducing the nuclear level of NFATc4 protein is related to reducing H2O2-induced oxidative stress and cell apoptosis.

RESUMO

Brain metastases are the most common intracranial malignancies, and radiotherapy is an effective treatment of controlling the localized lesions. However, conventional radiotherapy techniques have their own shortcomings that limit the effectiveness of radiotherapy. Metastatic brain tumors are highly likely to recur or progress after treatment. Clinical studies have shown that arginine can penetrate the blood-brain barrier and subsequently improve the radiosensitization of metastatic brain tumors. In this article, the mechanisms underlying the effect of arginine on the radiosensitization of metastatic brain tumors by inhibition of tumor glycolytic metabolism, reduction of DNA damage repair and alteration of tumor hemodynamic parameters were reviewed, aiming to provide new ideas for clinical research and treatment of brain metastases.

OBJEC TIVE To establish the qu ality standard of Hylocereus undatus ,and to provide reference for its quality control. METHODS The sample of H. undatus medicinal materials was collected for character observation ,powder microscopic identification and thin-layer chromatography (TLC)identification. Moisture content ,total ash ,acid-insoluble ash ,water-soluble extracts and alcohol-soluble extracts were determined according to the corresponding methods in the general provisions of the 2020 edition of Chinese Pharmacopoeia (part Ⅳ). The contents of kaempferol and isorhamnetin in H. undatus were determined by high performance liquid chromatography. RESULTS The medicinal materials of H. undatus were in brown or yellowish brown irregular long bundles ;the calyx tubes were twisted in bundles ;the scales on the outside of the flower were shrunken ,and many stamens were inserted on the inside. The powder was brown-green or brown-yellow ,and pollen grains ,ducts and non-glandular hairs were found.In the TLC diagram of test sample ,fluorescent spots of the same color were displayed on the corresponding position of the chromatogram of substance control (kaempferol, isorhamnetin) and reference material. The moisture content , total ash , acid-insoluble ash ,water-soluble extract and alcohol-soluble extract of the 15 batches of samples ranged from 10.70% to 12.23%, 7.48% to 11.29%,0.25% to 0.70%,30.34% to 49.91%,and 25.27% to 36.92%,respectively. The average values were 11.44%,9.51%,0.46%,40.13%,32.33%,respectively. The contents of kaempferol and isorhamnetin were 1.787-3.785 and 0.597-2.211 mg/g,respectively. CONCLUSIONS This study add microscopic identification ,TLC identification and inspection items such as moisture content ,ash and extract on the basis of the existing quality standards of H. undatus . It is preliminarily proposed that the moisture content in H. undatus shall not exceed 13.0% and total ash content shall not exceed 12.0%,and the water-soluble extract and alcohol-soluble extract shall not be less than 30.0% and 25.0% respectively;the contents of kaempferol and isorhamnetin shall not be less than 1.780 and 0.590 mg/g,respectively. The established quality standard can be used for quality control of H. undatus .

Objective:To investigate the value of event-related potential (ERP) P3 in the assessment of visual attention impairment in patients with cerebral small vessel disease (CSVD).Methods:Twenty-five patients with CSVD diagnosed in the Shandong Provincial Hospital Affiliated to Shandong First Medical University from July 2019 to December 2020 were selected as the CSVD group, while 25 healthy subjects who underwent physical examination in the same period were selected as the control group.The neuropsychological evaluation of CSVD group and control group was carried out by Montreal cognitive assessment (MoCA), 7-items generalized anxiety disorder (GAD-7) and patient health questionnaire-9 (PHQ-9). Magnetic resonance imaging brain white matter high signal of CSVD group and control group was carried out by Fazekas score.The amplitude and latency of ERP component P3 were measured by visual tristimulation Oddball experimental paradigm.SPSS 23.0 software was used for statistical analysis.The differences of amplitude and latency between the two groups were compared by repeated measurement analysis of variance.Pearson or Spearman correlation analysis were used to explore the correlation between P3 amplitude, latency and related scale scores.Results:(1)In the amplitude of P3, the interaction effect between group and stimulation was significant( F(2, 96)=3.922, P=0.023). The main effect between groups was significant( F(1, 46)=15.976, P<0.01). The main effect of stimulation was significant( F(2, 96)=86.212, P<0.01). Further simple effect analysis showed that compared with the control group((9.82±5.14)μV, (11.12±4.72)μV) the P3 amplitude induced by target stimulation ((6.59±4.22)μV, F(1, 48)=7.363, P=0.009) and novel stimulation ((7.08±3.91)μV, F(1, 48)=13.907, P=0.001) in CSVD group decreased significantly.(2)In the latency of P3, the main effect between groups was significant( F(1, 48)=4.870, P=0.032). The main effect of stimulation was significant( F(2, 96)=86.212, P<0.01). The interaction effect between group and stimulation was significant( F(2, 96)=4.561, P=0.013). The main effect of stimulation was significant( F(2, 96)=16.299, P<0.01). Further simple effect analysis showed that the P3 latency induced by novel stimulation in CSVD group was longer than that in control group( F(1, 48)=17.124, P<0.01). (3)P3 amplitude induced by target stimulation was positively correlated with MoCA score ( r=0.255, P=0.027). The P3 amplitude ( r=-0.502, P<0.01) and P3 latency ( r=-0.265, P=0.022) induced by novel stimulation were negatively correlated with Fazekas score. Conclusion:The speed and ability of patients with CSVD to process visual spatial information are impaired, especially for rare stimulation.ERP examination may be a rapid, objective and sensitive method for the diagnosis of visual attention impairment in patients with CSVD.

Exosomes are nanoscal-scale vesicles containing a variety of proteins(HSP70, actin, Alix, etc.), lipids, mRNAs and miRNAs.They are commonly found in various tissues and cells of the organism.They are involved in processes such as intercellular communication, immune regulation and cell signaling pathway regulation, and then play an important role.Recent studies have shown that exosome-mediated intercellular signaling plays an important role in the progression of Alzheimer's disease(AD). This paper reviewed the general characteristics of exosomes, clarifies the possible role of exosomes in the pathogenesis of AD, and also discussed the application prospect and the related challenges of exosomes as a new potential option in the future treatment of AD.

Objective To establish a monkey model of muscle atrophy by fixing the right lower limb and set up e-valuation criteria for that model. Methods Twelve healthy,6-7 year old male cynomolgus monkeys(body weight 6-7 kg)were used in this study. All animals were normal and had no malformed lower limb bones. Right lower limbs of the an-imals were fixed with fiberglass bandage(from knee joint to anklebone)for 15 weeks. Body weight,crus perimeter and crus volume were recorded every two weeks. MRI examinations of the gastrocnemius muscle were conducted at weeks 11,13 and 15. Muscle biopsy was taken for pathological examination at week 15. Results There were no obvious changes of body weight during the experiment. At 15 weeks after modeling,the right crus perimeter and crus volume of the experimental an-imals were significantly decreased(P <0.05),and some biochemical indexes such as serum creatinine,gloucose and al-kaline phosphatase were significantly decreased(P <0.05), while cholesterol was significantly increased(P <0.05).MRI showed atrophy of the right gastrocnemius muscle. Muscle biopsy also showed muscle atrophy and muscle fibers be-came thinner in the right gastrocnemius,and the cross-section area of the muscle fibers was significantly decreased. Con?clusions It is easy to operate and establish a muscle atrophy model in Cynomolgus monkeys by fixing the right lower limb. This model is suitable for objective and easy evaluation of muscle atrophy by measurement of crus perimeter and crus volume of the limb,blood biochemical parameters,MRI examination,and histopathological examination in combination.

Objective To explore the effect of nursing support on negative emotion and quality of life in patients with brain tumor. Methods A total of 139 cases of brain tumor in Shandong Provincial Cancer Hospital of Shandong University from September 2014 to February 2016 by convenient sampling , according to the random number table, they were divided into the control group and the intervention group. The patients in the control group were given routine discharge follow-up, while the patients in the intervention group were given emote team nursing support on the basis of the control group. We used anxiety self rating scale (SAS), depression self rating scale (SDS) and quality of life assessment scale (QLICP-BN) evaluation two groups of patients with anxiety, depression and quality of life level in the patients when they were discharged from hospital and discharged after 1 month, 3 months. Results Comparing the two groups of patients with anxiety and depression level and quality of life in the hospital, there was no statistically significant difference (t=0.187,0.174, P > 0.05);1 month after discharge, the scores of SAS and SDS respectively (62.97 ± 485), (63.83 ± 5.24) points in the intervention group, the scores of SAS and SDS respectively (64.58 ± 5.15), (65.17 ± 5.11) points in the control group, the difference is statistically significant (t=2.753, 2.321, P<0.05);3 month after discharge, the scores of SAS and SDS respectively(61.04±4.13),(62.25±3.95)points in the intervention group, the scores of SAS and SDS respectively(63.91 ± 4.73),(64.83 ± 4.29)points in the control group, the difference is statistically significant (t=4.621,5.196, P < 0.01); 1month, 3 months after discharge, the total quality of life scores respectively (65.28 ± 12.53), (68.71 ± 12.78) points in the intervention group, the total quality of life scores respectively (62.07 ± 11.27), (63.86 ± 12.13) points in the control group, the difference is statistically significant (t=2.439, 3.803, P < 0.01). The intervention effects and time effects of anxiety, depression and quality of life were statistically significant (W Mauchly's were 0.823, 0.782, 0.757, P <0.05). Conclusions Remote team nursing support can effectively reduce the negative emotions of patients with brain tumor, improve the quality of life, can be used in the department to promote the quality of clinical care.

Objective:To investigate the effect of persimmon leaf extract (PLE) on HEK293-APPswe transgenic cells (20E2).Methods:To determine whether the 20E2 cells model was successfully established,the level of Aβ1-40 in SH-SY5Y was detected and 20E2 cells(HEK293 cells stably expressing Swedish mutant APP)cultured in vivo by ELISA kit,and the expression of APP protein level was detected by Western blot.Cell viability was assayed by CCK-8 method and then selected the best concentration.Set groups:SH-SY5Y as normal control group (NC group),20E2 as model group (20E2 group),treating with PLE as treating group (20E2+PLE group).Reactive oxygen species (ROS) levels of each group were detected with DCFH-DA fluorescent probe.The extracellular level of Aβ1-42 were detected by ELISA kit.Cytoplasmic Nrf2,Nuclear Nri2,Whole-cell HO-1 were detected by Western blot.Results:Compared with model group,the expressions of ROS,Aβ1-42 were down-regulated and the Nuclear Nrf2 and Whole-cell HO-1 were up-regulated in 20E2+PLE group.Conclusion:PLE can reduce the level of oxidative stress of model group effectively,it possibly reduce the aggregation of Aβ1-42 and prevent oxidizing via activating Nrf2/HO-1 pathway.

Objective To detect the impact of ultrasound elastography in diagnosis of prostatic cancer,and to evaluate its capability in differentiating benign from malignant lesions.Methods From June 2013 to February 2015, we selected 50 patients with different prostatic lesions,and suspicious for malignancy were included.All patients had a conventional B-mode ultrasound examination and color Doppler imaging,and then real time ultrasound elastography was performed in the same session.Finally,the results were compared to the histopathological results of those lesions. Results The addition of Strain ratio parameter for evaluating the elastography images showed the highest sensitivity of 74.2%,specificity of 73.7% and accuracy of 74.0% at a best cut off point of 5.5 between benign and malignant lesions.Conclusion Prostate US combined with elastography can be a helpful tool for guiding malignant lesions.Also it can help in targeting the biopsy site.

OBJECTIVE:To establish an HPLC-ELSD method for the determination of the content of astragaloside Ⅳ in Danqi piantan capsule.METHODS:The determination was performed on Intertex C18 column(250 mm?4.6 mm,5 ?m) with column temperature kept at room temperature. The mobile phase consisted of acetonitrile-water(32∶68) at a flow rate of 1 mL?min-1.RESULTS:Astragaloside Ⅳ showed good linear relationship in the range of 0.091 5~0.915 mg?mL-1(r=0.999 5),and the average recovery of astragaloside Ⅳ was 101.12%(RSD=1.75%,n=6).CONCLUSION:The method is simple,accurate and reproducible,and it is applicable for the quality control of Danqi piantan capsule.

Objective To investigate the effects of local mild hypothermia treatment on plasma vaso-active substance such as neuropeptide Y(NPY),neurotensin(NT),calcitonin gene-related peptide(CGRP) and endotheline(ET) in patients with acute cerebral hemorrhage. Methods 70 patients were randomly divided into two groups: local mild hypothermia group (35 patients) and routine treatment group (35 patients).Nineteen healthy persons served as control group. Routine treatment was used in the both groups, and the local mild hypothermia therapy was only used in local mild hypothermia group. Objective To investigate the effect of local mild hypothermia on stress hormones in patients with acute cerebral hemorrhage.Methods 78 patients were randomly divided into two groups, mild hypothermia group (38 cases) and routine group (40 cases). Conventional treatment was used in both groups, and local mild hypothermia therapy was applied in mild hypothermia group. The concentrations of corticotropin-releasing hormone (CRH), adrenocorticotropic hormone (ACTH), corticosteroid (CS) and aldosterone (ALD) in plasma pretherapy and at 3rd, 7th day after treatment were measured. Objective To evaluate the reliability of transcranial color duplex sonography(TCCD) in the detection of stenosis of the middle cerebral artery(MCA).Method 168 patients with cerebrovascular di sease were underwent both cerebral angiography and TCCD examination.Result 18 stenosis of the MCA were diagnosed by angiography, 15 of the 18 stenosis were detected by TCCD. But 2 MCA stenosis diagnosed by TCCD were not proved by angiography. TCCD diagnosed MCA stenosis with sensitivity(83%) and specificity(88%) respectively.Conclusion MCA stenosis diagnosed by TCCD was better than by transcranial Doppler(TCD).

Objective To observe the effects of mild hypothermia on heat shock protein 70 (HSP70) and glial fibrillary acidic protein (GFAP) expression in cerebral tissue after cerebral ischemia and reperfusion. Methods 30 male Wistar rats were randomly divided into sham-operated group, normal temperature group and mild hypothermia group. The model of focal cerebral ischemia was made by middle cerebral artery occlusion (MCAO) method. After 2 h of MCAO following 48 h of reperfusion, HSP70 and GFAP positive neurons were detected by immunohistochemistry technique in ischemic regions of rats. Results Many necrotic neurons were appeared in normal temperature group and no necrotic neurons in sham-operated group. Fewer necrotic neurons were fond in mild hypothermia group than in normal temperature group. The positive cells of HSP70 and GFAP in sham-operated group and mild hypothermia group were less than normal temperature group. Image analysis revealed that the average optical density of HSP70 and GFAP in sham-operated group and mild hypothermia group was significantly lower than in normal temperature group (all P
