BackgroundDepressive disorders are prevalence among adolescents, with a higher incidence in females compared to males, often accompanied by more severe depressive symptoms. Sertraline is commonly prescribed for adolescent depressive disorder, however, its efficacy is only evident in certain patients. Currently, there is limited research on predicting the therapeutic efficacy of sertraline in female adolescents with depressive disorder. ObjectiveTo explore the relationship between personality traits, psychological resilience, coping style and the clinical efficacy of sertraline treatment in female adolescents with first-episode depressive disorder, in order to provide references for the treatment of this population. MethodsA total of 112 female adolescent patients with first-episode depressive disorder, meeting the diagnostic criteria of the International Classification of Diseases (tenth edition) (ICD-10), and undergoing a 4-week treatment regimen with sertraline, were selected from the Second People's Hospital of Guizhou Province from February 2019 to January 2021.General demographic questionnaire, NEO Five-Factor Inventory (NEO-FFI), Resilience Scale for Chinese Adolescents (RSCA), Coping Style Scale for Middle School Students (CSSMSS), Hamilton Anxiety Scale (HAMA) and Hamilton Depression Scale-17 item (HAMD-17) were used for assessment. Logistic regression analysis was used to investigate the factors influencing the clinical efficacy of sertraline in the treatment of female adolescents with first-episode depressive disorder. ResultsNeuroticism (β=-0.115, 95% CI: 0.807~0.984) could negatively predicted the therapeutic efficacy of sertraline in female adolescents with first-episode depressive disorder, whereas agreeableness (β=0.129, 95% CI: 1.025~1.264) and RSCA score (β=0.062, 95% CI: 1.004~1.128) could positively predict its therapeutic efficacy. ConclusionHigh neuroticism, low agreeableness and low psychological resilience may be the risk factors for the poor therapeutic effect of sertraline in the treatment of female adolescents with first-episode depressive disorder. RESUMO

In this study, we synthesized six tetrazine-dipyrromethene boron difluoride (BODIPY) probes and achieved a remarkable up to 14-fold increase in singlet oxygen yield via tetrazine bioorthogonal click-to-release reactions. We systematically investigated the photodynamic activity of these probes, revealing crucial structure-activity relationships. Additionally, we evaluated the stability and release kinetics of these probes and identified P5 and P6 as ideal candidates for photodynamic therapy in live cells. This innovative strategy opens new avenues for fine-tuning the photodynamic properties of BODIPY dyes, thereby expanding their utility in cancer therapy.

RESUMO

Objective:To observe the clinical efficacy modified Chaihu Shugansan combined with Xuanfu Daizhetang on anxiety and depression of patients with gastroesophageal reflux disease (GERD) and Qi stagnation and phlegm obstruction syndromes in clinic and the effect on neuropeptide factor and pro-inflammatory factor. Method:Patients 200 cases were divided into control group and observation group. Patients in control group got omeprazole enteric-coated tablets, 20 mg/time, 1 time/day, flupentixol and melitracen tablets, 1 tablet/time, 2 times/day, and Dalitong granules, 1 bag/time, 3 times/day. In addition to omeprazole enteric-coated tablets in control group, patients in observation group were also added with syndrome differentiation-based treatment of modified Chaihu Shugansan combined with Xuanfu Daizhetang, 1 dose/day. The treatment lasted for 8 weeks. Before and after treatment, scores of acid regurgitation, heartburn, poststernal pain, syndrome of Qi stagnation and phlegm obstruction, gastroesophageal reflux disease questionnaire (GerdQ), Hamilton anxiety Scale-14 (HAMA-14) and Hamilton depression scale-17 (HAMD-17) were scored; And upper gastrointestinal endoscopy, levels of peripheral vasoactive intestinal peptide (VIP), 5-hydroxytryptamine (5-HT), tumor necrosis factor-<italic>α </italic>(TNF-<italic>α</italic>), interleukin-1 (IL-1), IL-6 were detected. All the patients (GerdQ<8) got follow-up for 16 weeks, and the recurrence and safety were recorded. Result:After treatment, scores of GerdQ, endoscope, main symptoms and syndrome of Qi stagnation and phlegm obstruction on observation group were lower than those in control group (<italic>P</italic><0.01), and scores of HAMA-14 and HAMD-17 decreased in both groups (<italic>P</italic><0.01), but with no statistically significant difference in both groups. The comprehensive efficacy of main symptoms in observation group was better than that in control group (<italic>Z</italic>=2.076, <italic>P</italic><0.05). The curative effect of traditional Chinese medicine (TCM) syndrome in observation group was superior to that in control group (<italic>Z</italic>=2.151, <italic>P</italic><0.01). The effect of endoscope was better than that in control group (<italic>Z</italic>=2.103, <italic>P</italic><0.05). And levels of VIP, 5-HT, TNF-<italic>α</italic>, IL-1 and IL-6 in observation group were lower than those in control group. During the 16-week follow-up, the recurrence rate in observation group was 19.23% (15/78), which was lower than 35.38% (23/65) in control group (<inline-formula><alternatives><mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML" id="M2"><mml:msup><mml:mrow><mml:mi>χ</mml:mi></mml:mrow><mml:mrow><mml:mn mathvariant="normal">2</mml:mn></mml:mrow></mml:msup></mml:math><graphic specific-use="big" xmlns:xlink="http://www.w3.org/1999/xlink" xlink:href="alternativeImage/EF9122E2-D647-4d34-AB25-83CBA259DE55-M002.jpg"><?fx-imagestate width="3.30199981" height="3.64066648"?></graphic><graphic specific-use="small" xmlns:xlink="http://www.w3.org/1999/xlink" xlink:href="alternativeImage/EF9122E2-D647-4d34-AB25-83CBA259DE55-M002c.jpg"><?fx-imagestate width="3.30199981" height="3.64066648"?></graphic></alternatives></inline-formula>=4.741, <italic>P</italic><0.05). The average recurrence time of the observation group was lower than that of the control group(<italic>P</italic><0.01). Conclusion:Modified Chaihu Shugansan combined with Xuanfu Daizhetang can significantly improve the main symptoms and TCM syndromes, relieve depression, anxiety and other adverse emotions, promote the healing of gastroesophageal mucosa, reduce the recurrence rate and delay the recurrence time among patients with GERD and Qi stagnation and phlegm obstruction syndromes. The mechanism of action may be related to the expression of neuropeptide factor and the inhibition of pro-inflammatory factor.

Objective:To observe the effect of acupuncture on the expression of mitochondrial proteome in hippocampus of senescence-accelerated mouse prone g (SAMPg) mice models with Alzheimer disease (AD),and to explore the possible protective mechanism of acupuncture on mitochondria.Methods:Sixty 6-month-old male SAMP8 mice were randomly divided into an acupuncture at acupoint group,an acupuncture at non-acupoint group and a model group,20 mice in each group.The 20 male senescence-accelerated mouse/resistance 1 (SAMR1) mice of the same age were used as a normal control group.Shenshu (BL 23),Baihui (GV 20),Xuehai (SP 10) and Geshu (BL 17) were selected for acupuncture intervention in acupuncture at acupoint group.After an 8-week intervention,mitochondrial tissues were extracted from the hippocampus.Differentially expressed proteins were identified by subcellular organelle proteomics.Western blot was used to verify the expressions of some related proteins in hippocampal mitochondria.Results:Compared with the model group,there were 13 differentially expressed protein spots in the acupuncture at acupoint group,of which,9 were up-regulated,including neurofilament light polypeptide (NFL),actin (cytoplasmic 1,database ID:ACTB),tubulin beta-2A chain (TBB2A),tropomodulin-2 (TMOD2),pyruvate dehydrogenase E1 component subunit beta (PDHE1-β),NADH-ubiquinone oxidoreductase 75 kDa subunit (database ID:NDUS1),heat shock cognate 71 kDa protein (HSC71),pyruvate dehydrogenase E1 component subunit alpha (PDHE1-α) and ATP synthase beta subunit (ATP-β);4 were down-regulated,including glial fibrillary acidic protein (GFAP),pyruvate dehydrogenase phosphatase 1 (PDP1),mitochondrial-processing peptidase subunit alpha (MMP-α) and adenosine kinase (ADK).According to the information provided in the protein database,most of the differentially expressed proteins involve the regulation of mitochondrial function and structure.The expression levels of NFL and TBB2A in the normal control group and the acupuncture at acupoint group were significantly higher than those in the acupuncture at non-acupoint group (P<0.05).ATP-β and NDUS1 expression levels were significantly higher in the acupuncture at acupoint group than those in the acupuncture at non-acupoint group (P<0.05);there was no significant difference between the acupuncture at non-acupoint group and the model group (P>0.05).Conclusion:Acupuncture may achieve the potential therapeutic effect on AD by regulating the structure and functional proteins of hippocampal mitochondria.

Objective To investigate the effects of Bushen Huoxue acupuncture method on the behavioral changes and the proliferation of endogenous neural stem cells (NSCs) in hippocampus of Alzheimer disease (AD) model mice. Methods 18 male SAMP8 mice, seven months old, were randomly divided into acupuncture group, non-acupoint control group, and model control group. And another age-matched 6 male SAMR1 mice were prepared as normal control group. Mice in acupuncture group were intervened by acupuncture method in the acupoints of "Shenshu", "Baihui", "Xuehai", and "Geshu". Mice in non-acupoint control group were treated by stimulating the fixed non-point under the bilateral rib, while mice in model control group and normal control group were raised without special treatment but administered the stimulation of catching with the same time and the same stimulus intensity. All treatrment lasted for 8 weeks. After the intervention, the learning and memory abilitities and brain hippocampus Brd U positive cells of all groups were detected. Results Compared with the nomal control group, mice in the model control group had longer escape latency and less time spent in former platform quadrant (P<0.05); the number of Brd U positive cells decreased significantly (P<0.05). Compared with the model control group and the non-acupoint control group, mice in the acupuncture group had shorter escape latency and more time spent in former platform quadrant (P<0.05); the number of Brd U positive cells increased significantly (P<0.05). Conclusion Bushen Huoxue acupuncture method can improve the learning and memory abilities of SAMP8 mice AD model by inducing the proliferation of hippocampal endogenous NSCs.

<p><b>OBJECTIVE</b>To detect the JAK2, CALR and MPL gene mutations in patients with BCR/ABL1 negative chronic myeloproliferative diseases(BCR/ABL1-CMPD)and to evaluate their diagnostic value.</p><p><b>METHODS</b>Two hundred and eight cases of BCR/ABL1-CMPD comprising of 146 cases of essential thrombocythemia(ET), 37 cases of polycythemia vera(PV)and 25 cases of primary myelofibrosis(PMF)from March 2012 to December 2015 were enrolled in the BCR/ABL1-CMPD, while 124 cases of secondary thrombocythemia and 73 cases of secondary polycythemia were enrolled in the control group. The genomic DNA and total RNA Were isolated from bone marrow or peripheral blood, then the exons 12 to 20 of JAK2 gene, exon 10 of MPL gene and exons 3 to 9 of CALR gene were analyzed by using DNA sequencing.</p><p><b>RESULTS</b>among 146 ET patients, the JAK2, CALR or MPL mutations were found in: 138 cases(94.5%)including 86 cases with JAK2V617F mutation(58.9%)and 2 cases(1.4%)with exon 12 of JAK2 mutations. CALR mutations were detected in 41 cases(28.1%), among them type 1(c.1092_1143del)in 22 cases, type 2(c.1154_1155insTTGTC)in 11 cases, and type 5(c. 1091_1142del), type 8(c.1104_1137del), type 41(c.1107_1137del), type 42(c.1125_1125del)in one case respectively. In addition, 4 cases were detected withother mutations of the CALR gene(c.1107_1115del, c.1111_1144 del, c.1101 A>C, c.1112_1117del). Moreover, 9 cases harbored MPL mutations(6.2%). Secondly, 31 patients were detected with JAK2V617F mutation(83.8%)in 37 cases of PV, and JAK2 exon 12 mutations were found in 2 cases(5.4%). Besides, CALR mutations were detected in 2 cases(5.4%), including 1 case of type I, the other of novel mutation of CALR. Thirdly, 19 in 25 cases of PMF were detected with JAK2V617F mutation(76%), 2 cases with CALR mutations(8%). 4 patients(16%), JAK2, CALR or MPL mutations were not detected, but among them 3 cases were found harboring other genetic abnormalities. Fourthly, no mutations of JAK2, MPL and CALR genes were detected in 124 patients with secondary thrombocytosis and 73 cases with secondary polycythemia.</p><p><b>CONCLUSION</b>Combined detection of JAK2, CALR and MPL gene mutations can cover the vast majority of patients with BCR/ABL1-negative myeloproliferative neoplasms. For higher frequencies of the mutations of CALR in ET patients, CALR mutation can be used as a new diagnostic marker in ET patients with JAK2 and MPL wild type.</p>

Objective To observe the effects of acupuncture on the behaviors and the expressions of Fis1 and OPA1, as well as mitochondrial ultrastructure in the hippocampus of mice with Alzheimer disease (AD); To explore the mechanism of action of acupuncture for AD.Methods Forty male SAMP8 mice were randomly divided into acupuncture group and model group, with 20 mice in each group. Another 20 male natural aging mice with the same age (SAMR1 mice) were set as the normal group. Acupuncture group chose Shenshu, Baihui, Xuehai and Geshu for intervention. 8 weeks later, Morris water maze was used to test the mice behaviors, and then hippocampus organization was taken. Western blot was used to detect the expressions of Fis1 and OPA1 and mitochondrial ultrastructure in hippocampal neurons of mice was observed by transmission electron microscopy.Results Compared with the model group, the escape latency of acupuncture group was significantly shortened (P<0.05), and the stay time in the former platform quadrant and former platform crossing times were significantly increased (P<0.05). The expression of Fis1 in the hippocampus of acupuncture group decreased significantly (P<0.05), while the expression of OPA1 increased significantly (P<0.05). The mitochondrial ultrastructure in hippocampus in the acupuncture group was effectively improved, and the mitochondrial surface density and body density were both increased in the acupuncture group compared with the model group (P<0.05).Conclusion Acupuncture may play a potential therapeutic role in AD by decreasing the expression of Fis1, increasing the expression of OPA1, recovering the injury of mitochondrial ultrastructure.

Objective To investigate the effect of acupuncture on behaviors and its mechanism of action in a SAMP8 mouse model of Alzheimer disease.Methods Sixty 6-month-old male SAMP8 mice were randomized to acupuncture,model and non--acupoint groups.Twenty male SAMP 1 mice of the same age constituted a normal control group.The acupuncture group received intervention by acupuncture at Shenshu,Baihui,Xuehai and Geshu.After eight weeks,a behavioral test was performed using the Morris water maze in every group of mice.The expressions of Flotillin-1,NEP and Aβ42 in mouse hippocampus were determined by western blot.Results The Morris water maze test showed that as compared with the model group,escape latency shortened significantly (P<0.05) and the time spent in the former platform quadrant and the number of former platform position crossings increased (P<0.05) in the acupuncture group of mice;escape latency,the time spent in the former platform quadrant and the number of former platform position crossings had no significant differences in the non-meridian-acupoint group of mice (all P>0.05).As compared with the model group,Flotillin-1 expression decreased significantly in the acupuncture group (P<0.05) but had no significant difference in the non-meridian-acupoint group (P>0.05);NEP expression increased significantly in the acupuncture group (P<0.05) but had no significant difference in the non-meridian-acupoint group (P>0.05);Aβ42 expression decreased significantly in the acupuncture group (P< 0.05) but had no significant difference in the non-meridian-acupoint group (P>0.05).Conclusions Acupuncture can markedly improve learning and memory abilities in a SAMP8 mouse model of AD and reduce Flotillin-1 content and upregulate NEP expression in SAMP8 mouse hippocampus to decrease Aβ42 expression,relieve neurotoxicity and produce a neuroprotective effect.

<p><b>OBJECTIVE</b>To investigate the hemostatic effect of spleen-invigorating, qi-replenishing and blood-containing formula on simvastatin-induced zebrafish hemorrhage model, and to compare with the effect of clearing heat and cooling blood formula.</p><p><b>METHODS</b>Zebrafishes from breed A B line were treated with 0.5 µmol/L simvastatin for 24 hours to establish zebrafishes hemorrhage model. Under strict blinded experimental conditions, the above mentioned zebrafishes were then treated with experimental drug of different concentrations at the maximum non-lethal dose. The intervention effect of spleen-invigorating, qi-replenishing and blood-containing formula was comprehensively assessed by examining the main observational parameters, such as bleeding reduction rate and hemostasis rate while referring to additional parameters, such as blood flow, improvement rate of blood flow, velocity of movement, improvement rate of motion, which are characteristics of spleen qi deficiency.</p><p><b>RESULTS</b>When the hemostatic effect of experimental drug B1 at the concentrations of 500 and 1 000 µg/ml, zebrafish bleeding rates were 30% and 15%, the hemostatic rate was 60% and 80%, respectively; when the experimental drug B2 at concentration of 500 and 1 000 µg/ml, Zebrafish bleeding rates were 45% and 40%, the hemostatic rate was 40% and 47%, respectively, showing that experimental drug B1 was superior to B2 in terms of decreasing bleeding rate and improving hemostatic effect in zebrafish. In the equal concentration, the experimental drug B1 was superior to B2 in terms of increasing and improving the blood flow of hemorrhagic zebrafish. Promotion and improvement of motion: in equal concentration, experimental drug B1 was superior to B2 in terms of promoting the motion velocity and increasing the improving rate of motion in zebrafish.</p><p><b>CONCLUSION</b>The spleen-invigorating, qi-replenishing and blood-containing formula displays a good hemostatic effect on simvastatin-induced hemorrhage of zebrafish. It also boosts the blood flow and motion velocity in hemorrhagic zebrafish, therefore, providing an experimental basis for the treatment of syndrome of spleen failing to control blood by spleen-invigorating, qi-replenishing and blood-containing formula.</p>

<p><b>OBJECTIVE</b>To investigate the role of cytogenetic analysis in the detection of bone marrow (BM) involvement in patients with non-Hodgkin's lymphoma (NHL).</p><p><b>METHODS</b>The bone marrow samples of 74 patients with NHL were detection by using morphology, cytogenetic test, flow cytometry and molecular biological assay. The detected results of morphology, cytogenetic test, flow cytometry and molecular biological assay alone and thier combined detection were compared, the detective rate and consistencies of the 4 methods were analyzed.</p><p><b>RESULTS</b>The detection rates of BM involvement by using morphology, cytogenetic, flow cytometry, and molecular biological assays were 21.6%, 17.6%, 23.0% and 33.8% respectively. The detective rate was enhanced to 44.6% by combining the 4 methods. Cytogenetic test showed the result consistent with the other methods.</p><p><b>CONCLUSION</b>Although cytogenetic test shows a lower detective rate than the other methods, but in some patients the cytogenetic test can detect the abnormality of bone marrow which can not be detected by other methods alone, the combination test of 4 detection methods can enhance the detectable rate of BM involvement.</p>

<p><b>OBJECTIVE</b>To observe the effects of Compound Zhebei Granule (, CZBG) combined with chemotherapy on surface markers of leukemia stem cell (LSC) in the bone marrow of patients with acute myeloid leukemia (AML).</p><p><b>METHODS</b>Seventy-eight patients with AML received bone marrow aspiration and the percentages of CD34(+) CD123(+) and CD33(+) CD123(+) cells were tested using flow cytometry method. A total of 24 refractory or relapsed AML patients were enrolled and treated with one cycle of standard chemotherapy combined with CZBG. Bone marrow samples were obtained before and after treatment, and the percentages of CD34(+) CD123(+) and CD33(+) CD123(+) cells were examined by flflow cytometry.</p><p><b>RESULTS</b>Compared with refractory or relapsed AML patients, patients achieved remission had a significant lower percentage of CD34(+) CD123(+) cells(P<0.01) and CD33(+) CD123(+) cells (P<0.01), indicating that controlling the LSC percentage may be important for patients with AML to achieve sustainable remission. Compared with those before treatment, the expression levels of CD34(+) CD123(+) were significantly decreased after CZBG combined with chemotherapy treatment (P<0.01). The percentages of CD34(+) CD123(+) cells and CD33(+) CD123(+) in patients achieving complete remission after CZBG combined with chemotherapy treatment were both significantly lower than those in patients with nonremission (P<0.01).</p><p><b>CONCLUSION</b>CZBG combining chemotherapy could reduce the percentages of CD34(+) CD123(+) and CD33(+) CD123(+) LSC, which might improve the clinical efficacy of refractory or relapsed AML.</p>

In order to establish the quality classification criteria of Paeonia suffruticosa seeds, thirty-one batches of P. suffruticosa seeds from different provenances were selected. The seed rooting rate, seed germination rate, seed purity, seed viability, 1,000-seed weight and moisture content were determined and analyzed through SPSS 20.0 software. Seed rooting rate, seed germination rate and seed purity were selected as the main index for classification, while 1,000-seed weight, seed viability and moisture content could be used as important references. The seed quality grading of P. suffruticosa was set as three grades. The seed quality of each grade should meet following requirements: For the first grade seeds, seed rooting rate ≥ 80%, seed germination rate ≥ 80%, seed purity ≥ 90%, seed viability ≥ 80%, 1,000-seed weight ≥ 250 g, moisture content, ≤ 10. For the second grade seeds, seed rooting rate ≥ 50%, seed germination rate ≥ 60%, seed purity ≥ 70%, seed viability ≥ 75%, 1,000-seed weight ≥ 225 g, moisture content ≤ 10. For the third grade seeds, seed rooting rate ≥ 20%, seed germination rate ≥ 45%, seed purity ≥ 60%, seed viability ≥ 45%, 1,000-seed weight ≥ 205 g, moisture content ≤ 10. The quality classification criteria of P. suffruticosa seeds have been initially established.

In order to optimize the testing methods for Paeonia suffruticosa seed quality, and provide basis for establishing seed testing rules and seed quality standard of P. suffruticosa. The seed quality of P. suffruticosa from different producing areas was measured based on the related seed testing regulations. The seed testing methods for quality items of P. suffruticosa was established preliminarily. The samples weight of P. suffruticosa was at least 7 000 g for purity analysis and was at least 700 g for test. The phenotypic observation and size measurement were used for authenticity testing. The 1 000-seed weight was determined by 100-seed method, and the water content was carried out by low temperature drying method (10 hours). After soaking in distilled water for 24 h, the seeds was treated with different temperature stratifications of day and night (25 degrees C/20 degrees C, day/night) in the dark for 60 d. After soaking in the liquor of GA3 300 mg x L(-1) for 24 h, the P. suffruticos seeds were cultured in wet sand at 15 degrees C for 12-60 days for germination testing. Seed viability was tested by TlC method.

Current study was carried out to optimize the priming condition of Oldenlandia diffusa seeds, and improve germination rate and seed vigor of 0. diffusa seeds under drought conditions. Uniform design was used to optimize the concentration and priming time of three priming materials (PEG, KNO3, GA3). Different concentrations of polyethylene glycol (PEG) was used to simulate drought stress. The seedling was cultured in 1/4 Hoagland medium for 30 d. The results showed that seed priming treatment with 366 mg x kg(-1) GA3 for 1h resulted in significant increase in germination rate, germination index, vigor, root length, plant height and biomass of O. diffusa seeds under drought stress (15% PEG), while seed priming with 3.0% KNO3 for 1 h showed little effect on germination and growth of O. diffusa seeds under drought stress. Seed priming treatment with appropriate GA3 concentration and priming time could enhance seed germination and drought resistance of O. diffusa in seedling stage.

<p><b>OBJECTIVE</b>To analyze the karyotype stability in hematological malignancies patients before and after allogeneic hematopoietic stem cell transplantation (allo-HSCT) and its prognostic significance of monitoring.</p><p><b>METHODS</b>The karyotypes and clinical data of 21 patients with hematological malignancies at the initial diagnosis and at relapse after allo-HSCT were retrospectively reviewed. Chromosome analysis was performed by standard 24 h-cultured method and R banding.</p><p><b>RESULTS</b>Karyotypes at the initial diagnosis and at relapse after allo-HSCT were different in 11 patients (52.38%), including chromosome 1, 3, 6, 12, 17, 21. Numberical abnormalities and structural chromosomal abnormalities always occurs together. The median survival time of relapse of the patients with karyotype changes was significantly shorter than that of patients without a karyotype change (79 d vs 522 d, P = 0.027), and that of the patients with trisomy 6 was also significantly shorter than that of the patients without trisomy 6 (9 d vs 275 d, P = 0.005).</p><p><b>CONCLUSION</b>Karyotype changes after relapse are associated with the prognosis of allo-HSCT.</p>

To investigate the inhibitory effect of RNA interference (RNAi) on dengue virus I (DENV-1) replication. Small interfering RNA (siRNA) against the PreM gene of dengue virus was synthesized and transfected into C6/36 cells with liposome, which was then attacked by DENV-1 virus. The antiviral effect of siRNA was evaluated by cytopathic effect (CPE), the cell survival rate measured by MTT, and virus RNA quantified by real-time RT-PCR. The results showed that after 7 days post infection of dengue virus, the transfected C6/36 cells showed less CPE. The cell survival rate of the transfected C6/36 cells increased by 2.26 fold, and the amount of virus RNA in the transfected cells was reduced by about 97.54% as well. These findings indicated that the siRNA could effectively inhibit dengue virus RNA replication, and protect C6/36 cells from viral attack, indicating its potential role in prevention and treatment of dengue fever.
