RESUMO

Objective:To screen the potential characteristic gene spectrums and signal pathways of osteoarthritis based on gene chips.Methods:We analyzed 2 microarrays of human joint synovial tissue (GSE82107 and GSE55235) derived from the Gene Expression Omnibus (GEO) database and included for this study 20 osteoarthritis (OA) samples and 17 healthy control samples. The differentially expressed genes (DEGs) between OA and HC were screened by GEO2R tool. Analyses of Gene Ontology function and Kyoto Encyclopedia of Genes and Genomes pathway enrichment were performed using the Database for Annotation, Visualization and Integrated Discovery to identify the pathways and functional annotations of DEGs (https://david.ncifcrf.gov/). Protein-protein interaction of these DEGs was analyzed based on the Search Tool for the Retrieval of Interacting Genes database and visualized by Cytoscape software (http://www.string-db.org/).Results:191 up-regulated DEGs and 49 down-regulated DEGs were screened out from the 2 microarray databases. Enrichment of DEGs was mainly found in regulation of such biological functions as "inflammation" , "bone cell differentiation" and "positive apoptotic cell regulation" , HTLV-I infection, silk crack on the original amp-activated protein kinase (MAPK) signaling pathway, swine flu, tumor necrosis factor (TNF) signaling pathway, the nf-kappa B signaling pathway, PI3 kinase/Akt pathway, toll-like receptor pathway, legionella, salmonella and other 14 signaling pathways. In 2 modes of MNC and Degree, the top 10 core genes were screened, of which interleukin-6 (IL6), JUN, chemokine 8 (CXCL8), early reaction growth factor (EGR1) and cyclin (CCND1) were identified as valuable biomarkers of OA.Conclusions:Based on GEO chips, 10 characteristic gene profiles such as IL6, JUN, CXCL8, EGR1, CCND and 14 signal pathways such as tumor necrosis factor (TNF) signal pathway, NF-κB signal pathway, PI3 kinase/Akt pathway and Toll-like receptor pathway were screened, which may provide new clues for understanding of the pathogenesis of osteoarthritis.

RESUMO

Objective To explore the protective effect of emodin on kidney damage in high-fat diet-induced obese mice. Methods Male C57bl/ 6 mice were divided into groups according to diet and treatment. Collagen Ⅳ(Col4), phosphatidylinositide 3-kinase(PI3K), phosphorylated phosphoinositol 3-kinase(p-PI3K), protein kinase (Akt), and phosphorylated protein kinase(p-Akt) were measured by Western blotting method. Results Col4 was increased, while p-PI3K and p-Akt were decreased in kidney tissue in high-fat diet-induced obese mice. However, there was lower level of Col4, but higher levels of p-PI3K and p-Akt in kidneys. Conclusion Kidney damage of high-fat diet-induced obese mice seems to be alleviated by emodin via improving insulin sensitivity.

RESUMO

To analyze the fuzzy language and explore the translation methods of the fuzzy language in traditional Chinese medicine,examples are given and discussed in this essay.Based on the analysis of the problems in the application of the fuzzy theory to TCM translation,a flexible translation method like a combination of multiple translation method has been suggested in addition to appropriate translations such as transliteration,literal translation,free translation and borrowing translation,which can promote understanding the characteristics of the fuzzy language in TCM so as to further boosting the communication of TCM culture.

ObjectiveTo investigate and compare the clinical features of drug-induced liver injury (DILI) and primary biliary cirrhosis (PBC). MethodsA total of 116 PBC patients and 124 DILI patients who were diagnosed in Shengjing Hospital of China Medical University from January 2005 to December 2013 were enrolled, and clinical features, laboratory indices, and the results of liver biopsy were compared between the two groups. The t-test or Nemenyi rank sum test was applied for comparison of continuous data between the two groups, and the chi-square test was applied for comparison of categorical data between the two groups. ResultsThere were significant differences in age of onset and gender composition between the two groups(both P<005). The serological markers (except albumin), immunological indicators (IgG, IgM, and IgA), and positive rate of autoantibodies (except SMA) showed significant differences between the two groups (all P<0.05). The histological appearances of PBC were hepatocyte edema (30 patients), inflammatory cell infiltration around the bile duct (29 patients), and atypical hyperplasia of the small bile duct (28 patients), and those of DILI were hepatocyte steatosis (15 patients) and spotty necrosis and loss of hepatocytes (14 patients). ConclusionDILI and PBC are different in the aspects of laboratory tests and pathological examinations, which has a certain guiding significance in differential diagnosis in clinical practice.

RESUMO

OBJECTIVE:To observe clinical efficacy of clomiphene combined with metformin in the treatment of polycys-tic ovary syndrome. METHODS:112 patients with polycystic ovary syndrome admitted in gynaecology and obstetrics depart-ment of our hospital were selected,and randomly divided into control group and observation group,with 56 cases in each group. Control group was treated with clomiphene orally,since fifth day of menstruation,once a day,50 mg/time;observa-tion group was additionally treated with metformin orally,30 min before meal,3 times a day,500 mg/d. Treatment course last-ed for 2 months. Biochemical indicators,control time of disease,actual treatment time,HOMA-IR,HOMA-β and clinical effi-cacy of patients were observed in two groups. RESULTS:After treatment,LH,FSH,T and FINS of patients in observation group were lower than before treatment and control group,with statistical significance(P<0.05);control time of disease and actual treatment time of observation group were significantly shorter control group (P<0.05),with statistical significance;HOMA-IR and HOMA-β of observation group were significantly lower than those of control group,with statistical significance (P<0.05);clinical effective rate of patients in observation group was significantly higher than control group,with statistical significance (P<0.05). CONCLUSIONS:Clomiphene combined with metformin in the treatment of polycystic ovarian syn-drome can improve biochemical indicators,shorten control time of disease and actual treatment time and enhance clinical effi-cacy.

BACKGROUND:Pueraria is a member of isoflavone characterizing by decreasing the toxic effects of alcohol,relieving the effect of alcohol withdrawal,anti-oxidative effect,removing oxygen free radicals,and preventing cell injury induced by lipid peroxidation.OBJECTIVE:To investigate the inhibitory effect of puerarin on adipogenic differentiation of alcohol-induced human bone marrow mesenchymal stem cells (hBMSCs).DESIGN,TIME AND SETTING:An in vitro observation based on cytology was performed at Laboratory of Biochemistry and Molecular Biology,Basic Medical College of Zhengzhou University in October 2008.MATERIALS:The bone marrow was taken from the adult healthy volunteers from the Department of Orthopedics of the First Affiliated Hospital of Zhengzhou University.Puerarin standard was provided by the National Institute for the Control of Pharmaceutical and Biological Products.METHODS:The primary hBMSCs were purified from the adult marrow by gradient centrifugation.The second passaged cells were divided into 3 groups.Blank control group:Cells were cultured with DMEM culture media containing 10% fetal bovine serum;Model group:Cells were cultured with 0.09 mol/L alcohol;Experimental group:Cells were cultured with 0.09 mol/L alcohol and 10-6 mol/L puerarin;while,the culture liquid was changed every two or three days.Cells were cultured for 7 days in each group.MAIN OUTCOME MEASURES:PPARγ 2 gene expression with RT-PCR;amount of adipocyte with oil red "O" staining;alkaline phosphatase activity.RESULTS:PPAR-γ2 mRNA expression in the model group was significantly greater than experimental group and blank control group (P <0.05),while the expression in the experimental group was slightly greater than blank control group,but there was no significant difference (P >0.05).The number of adipocytes was the most in the model group,and the lipid droplet was large and abundant.The number of adipocytes in the experimental group was significantly less than model group (P < 0.05),and there were few adipocytes in the blank control group.Compared with blank control group,alkaline phosphatase activity was decreased in the model group (P<0.05);but the activity in the experimental group was increased compared to model group (P < 0.05).CONCLUSION:Puerarin inhibits differentiation of hBMSCs into adipocytes induced by alcohol.

RESUMO

Objective To analyze CT characteristics of spherical pneumonia in order to differentiate it from other lesions especially lung cancer. Methods The CT and X-ray film of 25 cases diagnosed clinically as spherical pneumonia were analysed retrospectively.Results CT features of 25 patients were summarized as follow : ⑴Lesions were usually located in the posterior lung fields.(2)The central part of the lesions showed high density, while periphery part of lesion were low in density and blurred. (3)The bilateral margin of lesions were straight, and the lesions appeared square or triangular in shape. ⑷Margins of the lesions may be irregular and blurred. ⑸There were marked pleural reaction around the lesions and some times pleura diffusely thickening occurred. ⑹Vascular marking around the lesions were exaggerated increase and thickened, nevertheless they were not stiff. Conclusion CT is valuable in diagnosing and differential diagnosing spherical pneumonia.
