RESUMO

Objective:To investigate the effect of paeoniflorin on toll-like receptor 4(TLR4)/nuclear transcription factor(NF-κB) signaling pathway of streptozotocin combined with ovariectomized mice, and to explore whether it can improve the cognitive impairment of ovariectomized diabetic mice.Methods:Ninety female C57BL/6J mice were divided into SHAM group, ovariectomy group, diabetes group(intraperitoneal injection of STZ 50 mg·kg -1·d -1 for 5 consecutive days), dual model group(DM modeling and OVX operation), paeoniflorin low-dose intervention group(OVX+ STZ+ L-PF 50 mg·kg -1·d -1), paeoniflorin high-dose intervention group(OVX+ STZ+ H-PF 100 mg·kg -1·d -1; all groups n=15). After 8 weeks of paeoniflorin intervention, their cognitive function was tested by behavioral experiments(Morris water maze and Y maze). The pathological changes of hippocampal tissue were observed by HE and Nissl staining. The mRNA expressions of TLR4, tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β) and interleukin-6(IL-6) in hippocampal tissues were detected by real-time fluorescence quantitative PCR. The expression of TLR4, NF-κB P65, TNF-α, IL-6, IL-1β, β-amyloid protein(Aβ), tau proteins, and p-tau proteins were detected by Western blot. Results:Compared with SHAM group, the learning and memory ability of ovariectomy group, diabetes group and dual model group decreased, hippocampal cells were damaged, and the expression of related gene mRNA and protein were increased, especially in dual model group; Compared with dual model group, paeoniflorin intervention could delayed the learning and memory impairment, improve cognitive function, reduce the degree of hippocampal injury, and decrease the expression levels of related gene mRNA and protein, The above changes were the most pronounced at paeoniflorin high-dose intervention group.Conclusion:Paeoniflorin improves cognitive dysfunction in ovariectomized diabetic mice by inhibiting TLR4/NF-κB signaling pathway.

Objective:To explore the associations of stress, coping strategies and quality of life in globus patients.Methods:A total of 180 patients diagnosed with globus were retrospectively analyzed between September 2018 to July 2020 in the First Affiliated Hospital of Zhengzhou University.The questionnaire included baseline characteristics and assessment scales. Quality of life was measured by short-form health survey-36 (SF-36), which included physical composite score (PCS) and mental composite score (MCS). Perceived stress was measured by perceived stress scale 10 (PSS-10). The coping strategy was evaluated by medical coping modes questionnaire (MCMQ). We analyzed the relationship between baseline characteristics, stress, coping strategies, and quality of life, and the influential factors of quality of life.Results:PCS was affected by the number of previous chronic illness, age, stress, confrontation, and avoidance ( F=3.647, r=-0.263, -0.634, 0.249, -0.329, all P<0.05). MCS was related to monthly income, marital status, stress, confrontation, and resignation ( F=1.963, 5.764, r=-0.312, 0.384, -0.360, all P<0.05). Based on the data of multiple linear regression analysis, stress was negatively correlated with both PCS and MCS ( t=-3.883, -9.708, all P<0.01), confrontation was positively correlated with both PCS and MCS ( t=2.030, 2.798, P=0.044, 0.006), and resignation was negatively correlated with MCS ( t=-1.585, P=0.025). Besides, age was negatively correlated with PCS ( t=-2.736, P=0.007), and monthly income was positively correlated with MCS ( t=2.497, P=0.013). Conclusion:Aging, low income, over stress, resignation rather than confrontation as a coping style impair the quality of life in globus patients.

RESUMO

Different microorganisms can cause intraperitoneal infection. This study was to distinguish different microbial infections by urine analysis. Rats were intraperitoneally injected with Escherichia coli, Staphylococcus aureus, and Candida albicans, separately. Urine samples were collected from rats at 0, 12, 36 and 72 h after infection. Urinary proteins were profiled using liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). Compared with the control (without infection), a total of 69 differential proteins were identified in rats injected with E. coli. A total of 31 differences proteins were identified in rats injected with S. aureus. A total of 38 differential proteins were identified in rats injected with C. albicans. Urine proteome was different when rats were infected by different microorganisms, suggesting that urine may have the potential for differential diagnosis of different intraperitoneal infections.

@#The metabolic reprogramming in cancer cells has recently attracted more and more attention from researchers. Lipid metabolism is involved in many cell processes such as cell growth, apoptosis, exercise, membrane homeostasis, chemotherapy response and drug resistance. This article summerizes the advances in research on fatty acids, cholesterol and phospholipid metabolism in non-small cell lung cancer, which may provide new ideas for the prevention, early diagnosis and treatment of non-small cell lung cancer.

Objective To investigate the abnormal functional connectivity (FC) between the substantia nigra (SN) and the brain motor area in rapid eye movement sleep behavior disorder (RBD) by Unified Parkinson′s Disease Rating Scale (UPDRS), Hoehn-Yahr Scale and resting state functional magnetic resonance imaging (rfMRI).Methods A total of 34 subjects (14 with RBD (RBD group), 12 with Parkinson′s disease (PD group), and 8 age, sex-matched normal controls (control group)) recruited in the Department of Neurology, Henan Provincial People′s Hospital from 2014 to 2015 were enrolled in this study according to international diagnosis criteria.All subjects were examined by UPDRS, Hoehn-Yahr Scale and rfMRI.Results UPDRS scores and Hoehn-Yahr staging were 0.00 (0.00, 3.75) and 0.00 (0.00, 0.50) respectively in the RBD group, which were significantly different from that in the PD group (30.5 (18.75, 33.00) and 1.75 (1.50, 2.50), respectively;Z=-3.782, P0.05 and Z=1.10, P>0.05).The FC analysis showed alterations from the right SN to bilateral cerebellum respectively among the three groups (Fright cerebellum=12.975, Fleft cerebellum=6.144;P0.05).Conclusions There was abnormal FC from the SN to motor areas in RBD patients, and partial alterations were similar as PD patients.rfMRI provided an evidence that RBD might be presymptom of PD.
