RESUMO

ObjectiveTo analyze the monitoring data of schistosomiasis from 2004 to 2021 in Suzhou New District, Jiangsu Province, and to provide evidence for improving schistosomiasis elimination strategies. MethodsFollowing the Opinions on Prevention and Control of Schistosomiasis, Parasitic Diseases and Endemic Diseases in Suzhou and the Technical Plan for Prevention and Control of Schistosomiasis, Parasitic Diseases and Endemic Diseases in Suzhou, the monitoring of schistosomiasis in the population and snail habitats from 2004 to 2021 was conducted. The Mann-Kendall method and Joinpoint regression method were employed to analyze the trend of epidemic indicators (such as seropositive rate, prevalence of snail frames, etc.). Time series analysis (exponential smoothing model) was conducted to predict snail occurrence. ResultsFrom 2004 to 2021, a total of 73 680 people were serologically tested for schistosomiasis, with a positive rate of 0.084%. The seropositivity rate showed statistically significant differences between different years (χ2=70.73, P<0.05), but there was no significant trend over time. In addition, 3 053 fecal tests were conducted and no positive result was found. The snail habitats covered an area of 70.11 hm2 and showed a decreasing trend (Z=-1.97, P<0.05). A total of 30 093 frames were surveyed, of which 19.038% contained snails. The difference in the prevalence of snail frames between different years was statistically significant (χ2=7 203.09, P<0.05), with a decreasing trend in the prevalence of snail frames (Z=-2.05, P<0.05). A total of 26 296 live snails were seized and density of live snails was 0.874 snails per frame, showing a decreasing trend in the density of live snails (Z=-2.35, P<0.05). A total of 12 391 snails were dissected and no infected snail was found. The areas treated with molluscicides remained stable at 264.60 hm2. An area of 27.77 hm2 achieved the goal of snail eradication through environmental modification, with a decreasing trend (Z=-2.44, P<0.05). It is estimated that the prevalence of snail frames and snail density will remain relatively stable from 2022 to 2026, but the snail habitat area will fluctuate significantly, showing an increasing trend. ConclusionNo indigenous cases of schistosomiasis and no infected snails are reported, indicating the successful consolidation of schistosomiasis prevention and control measures. However, the snail habitat area fluctuates greatly with an increasing trend, suggesting the need for long-term Oncomelania snail monitoring in local areas.

Objective:To explore the mechanism of Ginseng Radix et Rhizoma- Notoginseng Radix et Rhizoma- Chuanxiong Rhizoma medicinal pair in delaying heart aging based on animal experiments, network pharmacology and molecular docking. Methods:Mice were divided into control group, aging group, metformin group and TCM group according to random number table method. All the groups were injected subcutaneously by D-galactose except the control group to build the subacute aging model. Two weeks later, the metformin group was given metformin suspension (150 mg/kg), the TCM group was given Ginseng Radix et Rhizoma- Notoginseng Radix et Rhizoma- Chuanxiong Rhizoma lyophilized powder solution (650 mg/kg), and the control group and aging group were given an equivalent volume of ultrapure water by gastric gavage, once a day, six times a week, for 10 weeks. The level of heart TERT mRNA was detected by PCR; the expression of heart p53 was observed by immunohistochemical staining; the morphology of heart tissue was observed by HE staining. TCMSP and SwissTargetPrediciton databases were used to retrieve the active components and targets of Ginseng Radix et Rhizoma- Notoginseng Radix et Rhizoma- Chuanxiong Rhizoma medicinal pair; TTD, OMIM, Gene, HAGR, DisGeNET and other data platforms were used to screen the targets of heart aging; after the drug and disease targets were intersected, the active components of them were collected; STRING database, Cytoscape 3.8.0 software, etc. were used to make PPI of the intersection targets, and screen out the key targets; FunRich was used to perform enrichment analysis of cellular components, molecular functions, biological processes, and biological signal pathways for key targets; Schr?dinger Maestro software was used to do the molecular docking of the screened active components and key targets, and docking results were visualized via PyMOL 2.1 software. Results:Experiment results showed that Ginseng Radix et Rhizoma- Notoginseng Radix et Rhizoma- Chuanxiong Rhizoma could significantly ameliorate the damage of aging heart tissues, elevate TERT mRNA level, while significantly reducing the positive expression of p53. A total of 32 active components from the medicinal pair were screened, corresponding to 637 target genes. There were 263 targets for heart aging, and 67 intersection targets of drug active component targets and heart aging targets. 31 key targets were obtained after screening. Enrichment analysis showed that molecular functions were related to transcription factor activity and protein-tyrosine kinase activity. Biological processes involved signal transduction and cell communication. Signaling pathways mainly involved PDGFR-beta, PI3K-Akt, S1P1, Glypican, TRAIL, and Glypican 1. The molecular docking results showed that kaempferol, suchilactone, and ginsenoside Rg5_qt in the medicinal pair had a strong binding ability to p53. Conclusion:Ginseng Radix et Rhizoma- Notoginseng Radix et Rhizoma- Chuanxiong Rhizoma may achieve the effect of delaying heart aging by inhibiting p53 expression, providing a foundation for further research on mechanism of invigorating qi and activating blood circulation drugs to delay heart aging.

Objective:To study the different region (DFR) genotyping of 15 strains of Yersinia pestis from Inner Mongolia. Methods:Fifteen strains of Yersinia pestis isolated from Otog Banner and Darhan Muminggan Joint Banner in Meriones unguiculatus plague foci in Inner Mongolia Plateau, Zhengxiangbai Banner in Spermophilus dauricus plague foci in the Songliao Plain in 2019 were amplified by PCR, and identified DFR genotyping. Results:Two strains of Yersinia pestis in Otog Banner jointly deleted DFR loci 1, 6, 7, 13, 15, 16, 17, 18, 23, and 13 strains of Yersinia pestis in Darhan Muminggan Joint Banner and Zhengxiangbai Banner jointly deleted DFR loci 1, 6, 7, 12, 13, 15, 16, 17, 18 and 23. G17 and G20 were classified as genotyping by comparison with DFR locus distribution and genotyping results. Conclusion:The genotyping of Yersinia pestis in the natural foci of Meriones unguiculatus plague foci in Inner Mongolia Plateau and Spermophilus dauricus plague foci in the Songliao Plain is the same as that of the historical strain.

Objective To investigate the effects of Tianxiang capsule on Neurotransmitters and Hormone Level of rats with motion sickness. Methods Male SD rats were randomly divided into six groups, including blank control group, model control group, positive drug control group, low-dose, mid-dose and high-dose Tianxiang capsule groups with the method of random digital table, and every group had 10 mice. Except the normal group, the rats in the other groups were intragastrically pre-administered for 1 hour, and the low, medium and high doses of Tianxiang capsule were 0.91, 1.82, 3.64 g/kg, the positive drug control group was given scopolamine 1 mg/kg, and then the rat motion sickness model was induced by a rotary stimulation device. After the modeling, the feces, urine, standing hair, trembling were immediately observed and recorded, and the halo response index of the rats was calculated. The blood from the heart puncture was taken and the vestibular nucleus were put on the ice. Then, the content of histamine (HIS) in the vestibular nucleus and plasma was detected by ELISA. The expression of plasma cortisol (Cort) and arginine vasopressin (AVP) were measured by radioimmunoassay. Results Compared with the model control group, the motion sickness index of rats with low, medium and high doses of Tianxiang capsule (6.56 ± 2.16, 6.10 ± 1.35, 4.46 ± 2.50 vs. 8.90 ± 2.61) significantly decreased (P<0.05 or P<0.01). The HIS content in the vestibular nucleus (12.70 ± 3.86 μg/L, 11.45 ± 1.57 μg/L, 10.02 ± 1.30 μg/L vs. 17.50 ± 4.82 μg/L) significantly decreased (P<0.05 or P<0.01). The plasma content of HIS (4.24 ± 1.75 μg/L vs. 7.69 ± 3.06 μg/L), Cort (286.90 ± 8.72 ng/ml vs. 329.26 ± 29.04 ng/ml) and AVP (16.54 ± 2.48 pg/ml vs. 22.35 ± 3.08 pg/ml) in the high doses of Tianxiang capsule significantly decreased (P<0.05 or P<0.01). Conclusions The Tianxiang capsule could effectively reduce the motion sickness index of rats with motion sicknes, which might be related to the down-regulation of HIS in Vestibule Nucleus and HIS, Cort and AVP in plasma.

Objective To explore the effect of preoperative platelet (PLT) count on the prognosis of patients with laryngeal squamous cell carcinoma.Methods The clinical data of 286 patients with laryngeal squamous cell carcinoma were retrospectively analyzed to determine the optimal critical value of PLT count for end point of recurrence and death.The effects of preoperative PLT count on the recurrence and 5-year survival rates of patients with laryngeal squamous cell carcinoma after surgery were analyzed.Results The optimal critical value of PLT count for end point of recurrence was 242.5 × 109/L.The patients were divided into PLT ≥242.5 × 109/L group (n =115) and PLT <242.5 × 109/L group (n =171).Single factor analysis indicated that the recurrence was not related to age (x2 =0.005,P =0.942),gender (x2 =0.309,P =0.579) and pathological differentiation (Z =2.858,P =0.240),and was related to T staging (x2 =10.509,P =0.001),lymph node metastasis (x2 =7.297,P =0.007),primary tumor site (x2 =16.797,P < 0.001) and preoperative PLT count (x2=12.081,P =0.001).Multivariate analysis indicated that T staging (OR =0.518,95 % CI:0.281-0.954,P =0.035),primary tumor site (OR =2.371,95 % CI:1.283-4.382,P =0.006),and PLT count (OR =2.885,95% CI:1.607-5.179,P < 0.001) were the independent factors affecting the recurrence of laryngeal squamous cell carcinoma.The optimal critical value of PLT count for end point of death was 251.5 × 109/L.The patients were divided into PLT≥251.5 × 109/L group (n =94) and PLT < 251.5 ×109/L group (n =192).Single factor analysis indicated that the 5-year survival rate was not related to age (x2 =0.030,P =0.863),gender (x2 =0.000,P =0.945) and pathological differentiation (x2 =4.050,P=0.133),and was related to T staging (x2 =41.630,P < 0.001),lymph node metastasis (x2 =58.110,P <0.001),primary tumor site (x2 =36.250,P < 0.001) and preoperative PLT count (x2 =4.790,P =0.029).Multivariate analysis indicated that T staging (HR =0.353,95% CI:0.193-0.645,P =0.001),primary tumor site (HR =2.151,95 % CI:1.312-3.526,P =0.002),lymph node metastasis (HR =2.819,95 % CI:1.633-4.867,P<0.001),and PLT count (HR=1.853,95%CI:1.160-2.960,P=0.010) were the independent factors affecting 5-year survival rates of laryngeal squamous cell carcinoma.Kaplan-Meier survival analysis indicated that the 5-year survival rate of PLT≥251.5 × 109/L group and PLT < 251.5 × 109/L group were 58.23%,67.87%,with significant difference (x2 =4.79,P =0.029).Conclusion Preoperative PLT count is the influence factor of recurrence and 5-year survival rate of laryngeal squamous cell carcinoma patients,which has important significance to the prognosis of laryngeal squamous cell carcinoma patients.

Objective To illustrate more accurately the feasibility of DNA barcode technology in species identification in different geographical areas.Methods Four plague host animals were selected from three plague natural foci in Inner Mongolia for DNA barcode comparison,GC base content,codon preference and genetic distance calculation,and a phylogenetic tree was constructed.Results Compared between different foci of A llactaga sibirica,the difference of base content was statistically significant (F =5.34,P < 0.05);the variation of Meriones unguiculatus focus Allactaga sibirica base content was larger,and compared with other foci Allactaga sibirica,the difference was statistically significant (all P < 0.05).Different foci between one species rarely exist in codon sites.The genetic distance of intraspecific was less than 1%,and the genetic distance of interspecific was more than 18%.Conclusions DNA barcode of each species is specific,with no interspecific species sharing,and does not change with geographical position.Molecular identification of plague host animal is effective by DNA barcode.

OBJECTIVE@#To analyze the correlated factors of cervical lymphatic metastasis of T3 and T4 glottic carcinoma.@*METHOD@#We did a retrospective analysis of 91 glottic carcinoma patients' clinical data to analyze cervical lymph node metastasis on different T stage, pathologic degree and the tumor sites.@*RESULT@#The cervical lymph node metastasis rate of 91 cases of T3 and T4 glottic carcinoma was 21.98%. T3 group's metastasis rate was 18.06% (13/72), T4 group's metastasis rate was 36.84% (7/19), P > 0.05. Grouped according to the degree of pathological differentiation, well-differentiated squamous cell carcinoma metastasis rate is 13.89% (5/36), middle-differentiated squamous cell carcinoma metastasis rate is 26.00% (13/50), and poorly-differentiated squamous cell carcinoma metastasis rate is 40.00% (2/5), P > 0.05. Cervical lymph node metastasis rate was 16.22%, when the tumor invading supraglottic region. Cervical lymph node metastasis rate was 15.38%, when the tumor invading subraglottic region. Cervical lymph node metastasis rate was 46.15%, when the tumor invading supraglottic and subraglottic region (P < 0.01).@*CONCLUSION@#Cervical lymph node metastasis in cN0 patient with supraglottic carcinoma is effected by T classification, cervical lymphatic metastasis of T3 and T4 glottic carcinoma is not entirely effected by T stage and pathologic degree. When the tumor invades supraglottic and subraglottic region, cervical lymph node metastasis is significantly higher. Therefore, the area of tumor invasion is an important factor for lymph node metastasis.

OBJECTIVE@#To analyze the related factors of difficult laryngeal exposure under retaining laryngoscope.@*METHOD@#We did a retrospective analysis of 287 retaining laryngoscope surgery patients' clinical datas to observe the relationship between difficult glottis exposure and patients' gender, degree of mouth opening, BMI, neck circumference, head and neck flexion, TMD, HMD and SMD.@*RESULT@#By ROC curve analysis, we determine the optimal threshold for TMD was 7.35 cm, HMD was 6.33 cm, SMD was 14.75 cm. Univariate analysis showed that gender, and glottis exposure had no significant correlation with difficult laryngeal exposure under retaining laryngoscope. Degree of mouth opening, BMI, neck circumference, head and neck flexion, TMD, HMD and SMD had correlation with difficult laryngeal exposure. Multivariate analysis showed that neck circumference, head and neck flexion, TMD, SMD were independent factors of difficult laryngeal exposure under retaining laryngoscope.@*CONCLUSION@#Measurement of neck circumference, head and neck flexion, TMD, SMD before the operation is important for the prediction of difficult laryngeal exposure under retaining laryngoscope.

Objective To determine a method for amplification of cytochrome C oxidase subunit Ⅰ (CO Ⅰ) gene of Brandt's vole.Methods 发he Brandt's Voles were captured in Abagaqi Xilingol League Inner Mongolia,and DNA was extracted from liver tissue.CO Ⅰ gene was amplified by nested PCR and sequenced afterwards.Results A band of 657 bp and 1 132 bp was amplified by internal and external PCR primers,respectively,which were consistent with expected sizes.A total of 12 segments of Brandt's Vole CO Ⅰ gene sequences were amplified by PCR and verified by sequencing.一he sequence number was KF182196-KF182207 in GenBank.After gene sequence alignment of the 12 CO Ⅰ gene sequences,it was found that the similarity was 100%,and no base mutation.Conclusion CO Ⅰ gene of Brandt's Vole could be amplified by nested PCR without pseudo gene.

Background Our previous study showed that curcumin suppresses the proliferation of human lens epithelial cells (LECs) in vitro and has an influence on the signal transduction of cyclic adenosine monophosphate (cAMP) and cyclic guanosinc monophosphate (cGMP).Actually,the regulation for biological behavior of LECs in vivo is complex.Objective This study was to investigate the changes of signal transduction of protein kinase (PK) in inhibition of curcumin on human LECs proliferation.Methods HLE-B3 was cultured and then divided into the blank control group,recombinant human basic fibroblast growth factor (rhbFGF) group and rhbFGF+ curcumin group.rhbFGF of 10 ng/ml was added in the medium in the rhbFGF group,and 20 mg/L curcumin was added into rhbFGF-induced cell medium for 24 hours in the rhbFGF+ curcumin group.The expression rates of PKA,PKC,PKG and calmodulin (CaM) in the cells were assayed using flow cytometry.Results The expression rates of PKA protein were (46.847± 1.673) %,(33.250 ± 2.242) % and (71.645 ±2.432) % in the blank control group,rhbFGF group and the rhbFGF+ curcumin group,respectively,and the expression rate of PKA protein was significantly reduced in the rhbFGF group compared with the blank control group (t =11.904,P<0.01),but the expression rate of PKA protein in the rhbFGF+ curcumin group was significnatly higher than that in the rhbFGF group (t=28.430,P<0.01).In the blank control group,rhbFGF group and the rhbFGF+ curcumin group,the expression rates of PKC protein in the cells were (35.575± 1.937) %,(50.652±2.068) % and (27.662t4.481) %,those of PKG protein were (63.838±0.486) %,(86.562 ± 1.325) % and (40.733 ± 2.175) %,while those of CaM protein were (67.408± 1.391)%,(83.887±3.499)% and (53.785 ± 1.942)%,the expression rates of PKC,PKG and CaM were significantly lower in the rhbFGF group in comparison with the blank control group (all at P<0.01),and those in the rhbFGF+ curcumin group were significantly declined in comparison with the rhbFGF group (all at P<0.01).Conclusions Suppression of curcumin on HLE-B3 proliferation probably is associated with the up-regulation of PKA expression and down-regulation of PKC,PKG and CaM expression in the cells.

OBJECTIVE@#To analyze the recurrencerelative factors of early glottic carcinoma after carbon dioxide laser treatment.@*METHOD@#A retrospective analysis of 134 early glottic carcinoma patients' clinical data was taken to analyze the relationships between recurrence and gender, age, pathologic degree, T stage, involvement of anterior commissure and involvement of thyroarytenoid muscle.@*RESULT@#Recurrent ratio of well-differentiated, middle-differentiated and poorly-differentiated squamous cell carcinoma were 11.83%, 18.42% and 33.33% (P > 0.05) respectively. Recurrent ratio of T1a, T1b, T2 were 13.24%, 18.92%, 23.08% (P > 0.05) respectively. The recurrent rate was 33.33% of tumors offended the anterior commissure, versus 5.43% with no involvement of anterior commissure (P < 0.01). The recurrent rate was 59.09% of tumors offended the thyroarytenoid muscle, versus 5.36% with no involvement of thyroarytenoid muscle (P < 0.01). Unvaried and multivariate analysis indicated that the involvement of anterior commissure and the involvement of thyroarytenoid muscle were the factors of recurrence.@*CONCLUSION@#Recurrence of early glottic carcinoma after carbon dioxide laser treatment is effected by the involvement of anterior commissure and the involvement of thyroarytenoid muscle. Resection can be performed conservatively according to evaluation of macroscopic tumour extension.

ObjectiveTo establish a method for the determination of Nuezhenide and Apiganin 7-O-neohesperidoside in Shanxiangyuan tablets.Methods The Luna C1s column was used,the mobile phase consisted of HCN-0.5%H3PO4(41 ∶ 59),the flow rate was 1.0 ml/min,the column temperature was 30℃,the UV detector was set at 336nm.Results The linear response range was 0.016~0.32 mg/ml (r=0.9997) and 0.006~0.11 mg/ml respectively.The average recovery rate was 99.0% and 97.5% respectively,RSD was 1.83% and 1.57% respectively,ConclusionThe method was simple,accurate and repeatable.It can be applied in quantitative determination of Nuezhenide and Apigenin 7-O-neohesperidoside in Shanxiangyuan tablets.

Objective To suppress the proliferation of lens epithelial cells (LECs) is a primary goal in prevention of after cataract. Recent study demonstrated an effective inhibition of elemene(Ele) on tumor cells. Present study was to investigate the effects of Ele on proliferation and cell cycle of human LECs B3 (HLE-B3). Methods Recombinant human basic fibroblast growth factor(rhbFGF) was utilized to induce proliferation of HLE-B3. Proliferative HLE-B3 was incubated with 80 mg/L Ele in CO_2 incubator for 24 hours. Then the inhibitory effects of Ele on proliferation of HLE-B3 was evaluated by methyl thiazolyl tetrazolium(MTT). The effect of Ele on HLE-B3 morphology was observed under the optical microscope. The effect of Ele on HLE-B3 cell cycle was analyzed by flow cytometer(FCM). Results MTT test showed that the optical density (OD) value of rhbFGF group was remarkably higher than that of control group(0. 599 0 ± 0. 053 1 versus 0. 409 1 ± 0. 042 2) (P < 0. 01), and that of Ele group(0. 450 0 ±0. 061 4) was obviously declined in comparison to rhbFGF group(P <0. 01) . The inhibitory rate of Ele was 24.90 %. In proliferation group, the number of HLE-B3 was increased with the normal cell structure and abundant cytoplasm under the optical microscope. However, in Ele group, the number of HLE-B3 was evidently decreased with less cytoplasm, undistinguished cell structure, condensed and aggregated nucleuses. The result of flow cytometer showed that the percentage of HLE-B3 in G_1 phase in rhbFGF group was 42. 062% ± 1. 270% and that in control group was 46. 422% ±3. 765% with a significant difference(P < 0. 05). HLE-B3 in G_1 phase in Ele group (60. 665% ±2.069%) was evidently increased in comparison with rhbFGF group (P < 0. 01). Conclusion Ele could alter the cell cycle of HLE-B3 and effectively inhibit the HLE-B3 proliferation induced by rhbFGF. Ele may be a reliable and effective drug for prevention and treatment of after cataract.

RESUMO

OBJECTIVE: To investigate the signal transduction mechanism of curcumin in inhibiting the proliferation of bovine lens epithelial cell (LEC) induced by recombinant human epidermal growth factor (rhEGF). METHODS: There were three groups in this experiment, which were normal control group, untreated group and curcumin-treated group. Proliferation of LEC was induced by rhEGF (50 microg/L). The concentration of intracellular Ca(2+) ([Ca(2+)](i)) in LEC was measured with Fura-2/AM by spectrofluorimetry. The contents of intracellular cAMP and cGMP were assayed by radioimmunoassay. RESULTS: The [Ca(2+)]i in LEC was obviously increased in the untrated group as compared with that in the normal control group (P<0.01), and the [Ca(2+)](i) in LEC in the curcumin-treated group was highest among three groups (P<0.01). The content of intracellular cAMP in LEC was decreased while the content of intracellular cGMP was obviously increased in the untreated group as compared with those in the normal control group (P<0.01). The content of intracellular cAMP in LEC was higher in the curcumin-treated group than that in the untreated group, while the content of intracellular cGMP was lower than that in the untreated group (P<0.01). CONCLUSION: The antiproliferation effects of curcumin on LEC may relate to the regulations of multiple processes of signal transduction.

RESUMO

Objective:To investigate the occurrence of antenatal anxiety and possible influencing factors,and analyze its relationship with spontaneous delivery.Methods: 590 healthy puerperal women who delivered in Taihe hospital during Dec 2007 and Dec 2008 were randomly selected and investigated for their psychological status,with T-A1 anxiety scale and SDS depression scale employed.Results: 569 effective questionnaires were gathered,with 114 cases divided into the anxiety group and 159 cases into the depression group.Influencing factors of antenatal anxiety include fear of delivery,worry about the safety of babies,understanding of delivery knowledge,puerperal age,and the family financial status.Conclusion: Factors including a relatively older age of primipara,poor financial status,insufficient support from family,worry about the safety of oneself and baby,and fear for delivery pain all have close connections with antenatal mental disorders.Meanwhile,antenatal mental disorders have a direct influence on delivery quality and the choice of delivery modes,and increase the failure rate of spontaneous delivery.
