RESUMO

Objective To observe the efficacy and toxicity of thermotherapy plus chemotherapy in treatment of retroperitoneal lymph node metastasis after radical gastrectomy and sequential radiochemotherapy. Methods Sixty patients with retroperitoneal lymph node metastasis after radical gastrectomy and sequential radiochemotherapy were randomly divided into of microwave hyperthermia combined with chemotherapy group (experimental group, 30 cases) and chemotherapy group (control group, 30 cases) by using random number table method. The control group: oxaliplatin 130 mg/m2, intravenous drip (2 h), d1; S-1: 80 mg·m-2·d-1, P.O 2 times/d (after breakfast and after dinner), d1-14. 3 weeks was 1 cycle, a total of 4 cycles. The experimental group: chemotherapy on the basis of control group combined with microwave hyperthermia, d1, 8. the efficacy and toxicity of two groups were evaluated. Results The efficacy rate of the experimental group was higher than that of the control group, and the difference was statistically significant [66.7 % (20/30) vs. 33.3 %(10/30), P0.05]. There was no significant difference in bone marrow suppression of two groups [33.3%(10/30) vs. 30.0 % (9/30), P> 0.05]. Conclusion Microwave thermotherapy plus chemotherapy has a good efficacy for retroperitoneal lymph node metastasis after radical gastrectomy and sequential radiochemotherapy, and the patients can well tolerated, it is worthy of clinical promotion.

RESUMO

Objective To explore the mechanism by which liraglutide modulated the differentiation of monocyte subsets in high glucose (HG) conditions.Methods Primary mouse splenocyte suspensions were cultured in HG conditions induced by IFN-γ in the presence or absence of liraglutide.The cells were harvested,co-incubated with antibodies,and analyzed on a BD FACS Calibur.Mononuclear cells (MNCs) were gated according to lower granularity and larger size by SSC and FSC.Monocytes (MCs) were defined as CD11b+MNC and divided into three subsets based on Ly6C expression:Ly6Clow,Ly6Cmid,and Ly6Chi.ROS production in Ly6C+ and Ly6C-MCs was detected by 2',7'-dichlorofluorescein-diacetate (DCFH-DA) staining and ROS-containing MCs were identified as DCF+ cells in both Ly6C+ and Ly6C-MCs.Results HG (15 mmol/L,25 mmol/L,35 mmol/L) dose-dependently increased Ly6Chi and Ly6Cmid MC differentiation and also enhanced the production of ROS in Ly6C+MCs.Liraglutide (100 U,200 U) dose-dependently inhibited HG-induced Ly6Chi and Ly6Cmid MC differentiation and also promoted the differentiation of Ly6Clow MCs.Moreover,liraglutide significantly inhibited HG-induced ROS production in Ly6C+ MCs.Conclusion Liraglutide treatment significantly inhibited inflammatory MC diferentiation induced by HG and also reduced ROS production in inflammatory MCs.

RESUMO

Objective To investigate the possible mechanism of hyperhomocysteinemia (HHcy) in renal dysfunction in patients with chronic stable coronary artery disease (SCAD).Methods We consequentially enrolled patients diagnosed as having SCAD and,according to homocysteine (Hcy) levels,divided them into the HHcy group (Hcy≥15 μmol/L,n =53) and control group (Hcy<15 μmol/L,n =47).We further tested the relationship among plasma Hcy level,renal function,and serum levels of interleukin (IL)-6 and adiponectin (APN).Results IL-6 was significantly higher in the HHcy group than in the control group (P < 0.05).However,APN level was obviously lower in the HHcy group than in the control group (P < 0.05).Correlation analysis revealed that Hcy level positively correlated with creatinine (r =0.379,P < 0.001),uric acid (r =0.238,P =0.019),cystatin C (r =0513,P < 0.001),and IL-6 levels (r =0.561,P < 0.001) but negatively correlated with estimated glomerular filtration rate (eGFR;r =-0.288,P =0.023) and APN level (r =-0.428,P < 0.001).IL-6 level showed positive correlations with creatinine (r =0.406,P =0.002),uric acid (r =0.359,P =0.007),and cystatin C levels (r =0.387,P =0.007) but a negative correlation with eGFR (r =-0.370,P =0.005).Meanwhile,APN level showed negative correlations with creatinine (r =-0.694,P < 0.001),uric acid (r =-0.420,P <0.001),and cystatin C levels (r =-0.553,P < 0.001),but a positive correlation with eGFR (r =0.251,P =0.034).Conclusion Plasma Hcy level may have a predictive value for renal dysfunction in patients with SCAD.Moreover,HHcy probably promoted renal dysfunction by inducing the imbalance of anti-inflammation and pro-inflammation in patients with SCAD.
