RESUMO

Objective:To investigate the psychological feelings of parents of children with fracture of ulna and radius during radiological examination, and to provide the basis for taking scientific and effective intervention program.Methods:Using the purposive sampling methods, a total of 30 parents of children who were treated in the pediatric orthopedics department of the emergency department of Beijing Jishuitan Hospital affiliated to Capital Medical University from January 2022 to June 2023 were selected as the research objects. A descriptive research method was adopted to conduct semi-structured interviews with parents of children, and Colaizzi′s phenomenological analysis was applied to analyze, refine, and summarize the interview information.Results:Four themes were summarized: negative emotions, information needs, seeking multi-party support and post-traumatic growth.Conclusions:The medical staff should pay close attention to the psychological status of the parents, meet their needs for information during the radiological examination, and provide targeted support measures to promote the prognosis of the children, improve the psychological adjustment and quality of life of the children and their parents.

@#Objective To explore the role and mechanism of LRG1 in the cell model of Aβ1-42-induced Alzheimer’s disease (AD).Methods Aβ1-42 stimulated SH-SY5Y cells was used to build AD cell model in vitro.The cell counting kit-8 (CCK-8) was used to detect cell aviability.LRG1 transcriptional level was measured through real-time fluorescence quantitative PCR (RT-qPCR).The phosphorylation level of p38 and the protein levels of LRG1,Bcl-2 and Bax were detected by Western blot.Apoptosis rate was checked by flow CytoMetry.Results The results showed that treatment of Aβ1-42 in SH-SY5Y cells significantly reduced cell aviability and increased LRG1 protein level.LRG1 silencing promoted the cell aviability and inhibited apoptosis that induced by Aβ1-42.Silencing LRG1 reversed the changes of Bcl-2 and Bax protein levels which induced by Aβ1-42.Silencing LRG1 inhibited the phosphorylation of p38 that induced by Aβ1-42.In addition,U-46619 (p38 specific activator) reversed the protective effect of LRG1 silencing on Aβ1-42-treated SH-SY5Y cell damage.Conclusion These results suggest that LRG1 silencing alleviates cell injury that induced by Aβ1-42 through deactivating of the p38/MAPK signaling pathway.

RESUMO

This study examined the promoter methylation of APO-1/CD95 (Fas) gene in bladder urothelial carcinoma and analyzed the relationship between the Fas promoter methylation and the biological behavior of bladder cancer. Promoter methylation of Fas gene was detected by methylation-specific PCR (MSP) in 4 bladder cancer cell lines, 50 human bladder urothelial carcinoma samples and l0 normal bladder tissue samples. Correlation of the aberrant methylation of Fas promoter with the clinicopathological parameters was statistically analyzed. The results showed that Fas was down-regulated at both mRNA and protein level in bladder cancer cell lines and tissue samples of bladder urothelial carcinoma. The positive rate of Fas protein expression in bladder urothelial carcinoma was 34.0% (17/50), significantly lower than that in normal bladder tissues (70.0%, 7/10) (P<0.01). Fas promoter methylation was detected, and the positive rate of Fas promoter methylation in bladder urothelial carcinoma was 42.0% (21/50), which was obviously higher than that in normal bladder tissues (0.0%, 0/10) (P<0.01). The aberrant methylation of Fas promoter was reversely correlated with Fas protein expression (P<0.05). Furthermore, the positive rates of Fas promoter methylation in high-grade and low-grade bladder urothelial carcinoma were 73.3% (11/15) and 34.2% (12/35), respectively, with significant difference shown (P<0.05). No statistical significance was found in the Fas promoter methylation among different clinical stages of bladder cancer. It was concluded that Fas promoter hypermethylation plays an important role in the pathogenesis of bladder urothelial carcinoma and may serve as a prognostic indicator of bladder urothelial carcinoma.
