RESUMO

Objective:To compare the efficacy and prognostic evaluation of response evaluation criteria in lymphoma (RECIL) and Lugano classification in patients with 18F-FDG-avid Hodgkin′s lymphoma (HL) and non-HL (NHL) during mid-chemotherapy. Methods:From January 2015 to August 2021, 240 patients (149 males and 91 females, age 50.0(32.0, 62.0) years) with pathologically confirmed lymphoma in Shanxi Cancer Hospital were retrospectively analyzed. Pre-treatment and mid-treatment 18F-FDG PET/CT examinations were performed, and the differences of PET/CT imaging results among patients with different types of lymphoma were analyzed ( χ2 test or Kruskal-Wallis rank sum test). Efficacy evaluation was conducted during mid-chemotherapy, and the results were divided into complete remission (CR), partial remission (PR), stable disease (SD), progressive disease (PD) according to Lugano classification, which were divided into CR, PR, mild remission (MiR), SD, PD according to RECIL. For the better comparison with Lugano classification, MiR was included in PR group (recorded as RECIL-1) and SD group (recorded as RECIL-2) in this study. All patients were followed up, and the progression free survival (PFS) was analyzed. Kappa test, χ2 test or Fisher exact probability method were used to analyze the data, and ROC curve was used to compare the prediction efficiency of different standards. Results:Among 240 patients with different types of lymphoma (96 HL, 30 follicular lymphoma (FL), 114 diffuse large B-cell lymphoma (DLBCL)), there were statistically significant differences in baseline SUV max ( H=54.96, P<0.001) and the sum of longest diameters ( H=15.85, P<0.001). Patients were followed up for 12-89 months, and 26 patients (10.8%) were evaluated as MiR according to the RECIL. The consistency ( Kappa) was 0.84 between results of RECIL-1 and Lugano classification, which was 0.74 between RECIL-2 and Lugano classification (both P<0.001). According to Lugano classification, the PFS rates of patients evaluated as CR, PR, SD and PD were 91.4%(148/162), 57.1%(36/63), 1/3 and 3/12 respectively, which were 91.3%(136/149), 62.8%(49/78), 1/2 and 2/11 for RECIL-1, and 91.3%(136/149), 57.7%(30/52), 71.4%(20/28) and 2/11 for RECIL-2 respectively ( χ2 values: 46.64-52.44, all P<0.001). The AUC of Lugano classification for predicting PFS had a trend to be higher than AUCs of RECIL-1 and RECIL-2 (0.774, 0.758 and 0.746; z values: 1.28, 1.61, P values: 0.200, 0.107). Conclusion:RECIL and Lugano classification have similar efficacy and prognostic evaluation effect on patients with 18F-FDG-avid HL and NHL during mid-chemotherapy.

Objective To explore the correlation between the pulmonary function and air pollutant emissions for the woolen and dust factory workers in Dongguan city .Methods Totally 519 yarn factory (wool group) and 503 dust factory (dust group) workers ,and 326 individuals (control group) undergoing the healthy physical examination were selected ,the respirometer was a‐dopted to detect the pulmonary function .The air sampling was carried out in the above factory areas and non‐factory communities , then the concentrations of silicon dioxide (SO2 ) ,nitrogen dioxide (NO2 ) ,silica (SaO2 ) ,cotton and other PM2 .5 mass were meas‐ured ,and their correlation with the lung function was analyzed .Results Compared with the control group ,the forced vital capacity (VC) ,forced vital capacity (FVC) ,maximal voluntary ventilation(MVV) ,first second forced expiratory volume (FEV1 .0) and ra‐tio of first second forced expiratory volume and forced vital capacity (FEV1 .0 % ) in the workers of wool group and dust group were decreased (P0 .60 ,P<0 .05) .Conclusion The air pollution in the partial factories in our city is heavier and the lung function of workers are injured ;the cotton dust content is high in the yarn factories area and the SO2 content is high in the dust factories ,which may be the main factors of the lung function injury in the woolen yarn and dust factory workers .

RESUMO

Objective To investigate the anti-hepatocarcinoma(anti-HCC) function of HLA-A2-restricted point-mutated Survivin peptide induced CTLs.Methods The HLA-A2-restricted Survivin nonapeptides were evaluated using bioinformatics software.The binding affinity of Survivin peptide to HLA-A2 molecular was determined with flow cytometry analysis.After peptide-induced CTLs were generated in vitro,flow cytometry and ELISA were performed to detect the levels of IFN-γ,which were secreted by reactive CTLs.Peptide-induced CTLs were co-cultured with hepatoma cell lines HepG2 and BEL-7402.The rates of tumor cells lysis were assayed using CytoTox 96(R) and the morphological changes of tumor cells were observed with inverted phase contrast microscope.Results Point-mutated Survivin nonapeptide Sur79M2 (KMSSGCAFL) was filtered out,which was shown higher scores compared with the wild-type peptide Sur79.Consistent with the results of software analysis,Sur79M2 showed higher binding ability in T2 binding assays.At the same time,Sur79M2-induced CTLs could release a large number of IFN-γ after incubated with target cells rather than Sur79.When co-cultured with HCC cell lines HepG2 and BEL-7402,Sur79M2-induced CTLs effectively lysis HepG2 on HLA-A2-restricted manner without killing effect on BEL-7402 that do not express HLA-A2 molecules.Conclusion Sur79M2 could elicited specific cytotoxic T lymphocytes in vitro,which were able to specifically kill HCC cell lines on HLA-A2-restricted manner.

Objective To establish the fingerprints of quaternary ammonium hydrate alkaloids in Radix Zanthoxyli nitidii by means of HPLC and to identify and evaluate the quality of different parts and commercial decoction pieces of Radix Zanthoxyli nitidii.Method The column of Zorbax Eclipse XDB-C_8(4.6?150mm,5?m)was selected.The mobile phase consisted of A:3 % glacial acetic acid-diethylamine(1000:7.8),B:methanol,and C:acetonitrile(non-lin- ear gradient elution).The elution speed was 0.8 mL?min~(-1),the detection wavelength was at 250 nm and 270 nm,and the column temperature was 20℃.Results The HPLC fingerprint of Radix Zanthoxyli nitidii consisted of 21 peaks which were chiefly composed by alkaloids such as Chelerythrine,Nitidine chloride,with a consistent peak-to-peak ratio.The constituents' distribution information provided quality information for assessing medicinal materials.Conclusion It showed that the alkaloids distributed mainly in the cortex of the roots,so the commercial decoction pieces of aged roots shed cortexes are inferior.The stems can not be used equivalently with the roots due to low content distribution of alkaloids.
