RESUMO

<p><b>AIM</b>To search for and application of new method for data processing in metabonomic studies.</p><p><b>METHODS</b>The paper proposed that in the processing of metabonomic data, robust PCA method can be used to diagnose outliers; and unstable variables judged by comparison between difference within class and difference among classes should be excluded before data analysis; moreover, the data should be properly scaled before further processing. The proposed methods were used to preprocess metabolomic data of four genotypes of the Arabidopsis thaliana plants.</p><p><b>RESULTS AND CONCLUSION</b>The outcome demonstrated that the application of these methods can obviously improve clustering and biomarker identifying results.</p>

RESUMO

<p><b>OBJECTIVE</b>To investigate the effect of plant growth substances on induction and culture of callus from Rhodiola quadrifida and also to analyze salidroside contents in the callus.</p><p><b>METHOD</b>The optimum combination of plant growth substances in MS solid medium for induction and culture of callus was established using orthogonal design. The contents of salidroside was analyzed by HPLC.</p><p><b>RESULT</b>MS medium containing 2,4-D 1 mg x L(-1), NAA 2 mg x L(-1), 6-BA 0.5 mg x L(-1) and KT 0.1 mg x L(-1) could induce the callus from R. quadrifida most effectively;the induction rate was 83.3%. The optimized combination of plant growth substances for callus subculture was 2,4-D 1 mg x L(-1), 6-BA 0.1 mg x L(-1) and KT 0.5 mg x L(-1). The dry weight could reach 11.77 g x L(-1) when the callus was cultured in the optimum medium for 30 d and salidroside content was 0.28%.</p><p><b>CONCLUSION</b>The quantities of plant growth substances required for induction and culture of callus are different in R. quadrifida. The callus could produce salidroside.</p>

RESUMO

<p><b>AIM</b>To investigate the paclitaxel-loaded nanoparticles of poly(ethylene glycol)-b-poly(D,L-lactic acid) amphiphilic diblock copolymer (PMT).</p><p><b>METHODS</b>PMT was prepared by solid dispersion technique. The average size and size distribution were determined by dynamic light scattering (DLS). The morphology was characterized by transmission electron microscopy (TEM) and 1HNMR. The influences of the copolymer molecular weight and the paclitaxel-fed amount on PMT were studied. Therapeutic effect of PMT was studied on Kunming mice liver cancer H22.</p><p><b>RESULTS</b>PMT showed nanometer size and spherical morphology with core and shell. The sizes of PMT increased with increasing the molecular weight of the hydrophobic segment in PEDLLA or increasing the drug-loaded amount. The tumour inhibiting effect of PMT was similar with that of Taxol.</p><p><b>CONCLUSION</b>It will provide an experiment basis for the development of new kind of intravenous administration of paclitaxel.</p>

RESUMO

<p><b>OBJECTIVE</b>To study the adsorption and enrichment of effective compounds of danshensu, and protocatechualdehyde from Danshen water extracts by macroporous resin D301.</p><p><b>METHOD</b>The change of active compounds was observed by detecting the amount of danshensu and protocatechualdehyde with HPLC.</p><p><b>RESULT AND CONCLUSION</b>Macroporous resins D301 can be used in the process of adsorbing and purifying active compounds of danshensu and protocatechualdehyde, and increase the amount of active compounds in water extracts.</p>

RESUMO

The permeability of cell membrane was enhanced by exogenous SA in the culture of Taxus cuspidata. Nuclei condense and fragments were observed in some cells by using fluorescent microscope, and the degraded chromosomal DNA was observed by using agarose gel electrophoresis. The changes in soluble proteins of the suspension cultures of Taxus cuspidata induced by salicylic acids were analyzed by using two-dimensional polyacrylamide gel electrophoresis. Comparing with the control, seven new protein spots were found and six protein spots were not found in the cultures grown with SA at 48h. The results obtained showed that SA could induce the expression of some special proteins that might be related with the action of SA in the suspension cultures of Taxus cuspidata.

RESUMO

Apoptosis was induced by taxol treatment in suspension cultures of Taxus cuspidata cells. Differential display technique was used to investigate the induced-gene expression between the taxol-induced T. cuspidata cells and normal control. Eight different expressed cDNA fragments were cloned and sequenced. These differential expressed fragments were further confirmed by Northern blotting hybridization with their original total RNAs. The result showed that three of the cDNA fragments were from control RNA and five of those were from taxol-induced T. cuspidata cells. The homology of the sequences revealed that one of the clones had 86% homology with ABA-responsive protein gene sequence in Arabidopsis thaliana, two of the clones had 50% homology with endochitinase precursor in tomato and the other 5 clones, which might be new gene fragments, had no significant homology with the known gene sequences in GenBank/EMBL/DDBJ.

RESUMO

A high Streptolydigin-producing strain Streptomyces lydicus AS 4.2501-L8 with genetic stability was obtained from the original strain S. lydicus AS 4.2501-P28 by mutagenesis with UV and acridine orange, followed by resistance selection. Its yield of Streptolydigin was 177.0 ?g/mL, 96.2% higher than the original strain.

RESUMO

The antifungal antibiotic produced by Streptomyces luteogriseus H-103 was purified by means of macroporous adsorbent resin, and the crystal of the antibiotic with high purity was got. In this paper, the methods of purification by adsorbing of microporous adsorbent resin and detection by reversed high performance liquid chromatography with evaporative light scattering detection (HPLC-ELSD) were established. The result appeared that resin X-5 is the best adsorbent, the eluant is 50% ethanol. The antibiotic was successfully separated on Agilent~(TM)20RBA?310SB C_(18 )column (150mm?4.6mm i.d,5?m) , using a mixture of acetonitrile (A)-H_(2)O (B) as a mobile phase under gradient elution at a flow of 0.8mL/min at 30℃.0~4.0 min, V(A)∶V(B)=20∶80, 4.0~9.5min, V(A)∶V(B)=45∶55, then V(A)∶V(B)=80∶20. The drift tube temperature and the air carrier gas flow rate of the ELSD were set at 115℃ and 2.3L/min.
