RESUMO

Exosomes are one of the most important ways of cell-to-cell communication in living lives. They are involved in major physiological and pathological processes, including drug resistance, infection propagation, cancer development and cardiovascular diseases. The biological functions of exosomes made it possess characteristics of low immunogenicity, high delivery efficiency, ability to cross multiple biological barriers and targeting capacity, which also encourage people to try to use it as a drug carrier to overcome the disadvantages of poor stability, low solubility, low bioavailability and high toxicity of some drugs. In this paper, the latest progress of exosomes in the delivery of antitumor drugs, including small chemotherapeutic drugs, biological macromolecules and nucleic acid drugs, is reviewed. In addition, the isolation, drug loading, and modification method and the application prospect of exosomes are also discussed.

RESUMO

<p><b>OBJECTIVE</b>Disturbances of the synthesis and breakdown of the extracellular matrix of arterial walls have emerged as key features of the atherosclerotic process. We observed the changes of circulating procollagen marker for type III collagen turnover rate, the N-terminal propeptide P III NP and vascular resistance in hypertensive patients treated with various antihypertensive regimens.</p><p><b>METHOD</b>A total of 130 light to moderate hypertensive patients were randomly assigned to receive enalapril (group B, n = 43), enalapril + spirolactone (20 mg/d, group A, n = 44) and anti-hypertensive drugs not directly affecting RAAS (calcium antagonist, beta-blocker, group C, n = 43) for 1 year. Target blood pressure is < 130/80 mm Hg.</p><p><b>RESULTS</b>Target blood pressure was reached in all treated patients and was similar among various groups. Under the same blood pressure controlling precondition, serum P III NP were similar at baseline among various groups and remained unchanged in group B [(3.4 +/- 0.3) microg/L vs. (3.7 +/- 0.3) microg/L, P > 0.05] and significantly decreased in group A [(2.3 +/- 0.2) microg/L vs. (3.8 +/- 0.2) microg/L, P < 0.05] while significantly increased in group C [(3.9 +/- 2.0) microg/L vs. (3.2 +/- 1.5) microg/L, P < 0.05]. Vascular resistance was similar among groups before therapy and all significantly decreased after 1 year antihypertensive therapy and the decrease was more significant in group A [(1064.3 +/- 158.6) dyn.s(-1).cm(-5)] than that in group B [(1200.8 +/- 298.7) dyn.s(-1).cm(-5)] and group C [(1205.1 +/- 206.4) dyn.s(-1).cm(-5)].</p><p><b>CONCLUSION</b>Spironolactone in conjunction with enalapril is a more favorable antihypertensive regimen in decreasing P III NP and improving vascular resistance than enalapril alone or antihypertensive drug regimens not directly affecting RAAS.</p>

Objective To observe the effects of low-intensity(650 nm,5 mW)laser irradiation on the pathological changes associated with aortic atherosclerosis using a rabbit model.Methods Thirty-six male Zelanian rabbits were randomly divided into 4 equal groups:a control group,a laser irradiation group,a simvastatin treatment group and a laser plus simvastatin group,and were treated accordingly.After being fed the basic diet for a week,all the animals were fed a high fat diet during the experiment.Blood samples were taken for lipid assay at the 60th day. The animals were sacrificed at the 61st day and their aortas removed for gross and microscopic examination.Any pathological changes were graded as mild,intermediate or severe according to the indicators of atherosclerosis observed.Results There were no significant differences in blood fat levels among the four groups before the experi- ment,but significant differences in serum triglyceride prevailed after the treatments.Low-density lipoprotein(LDL)- C level in the control group was significantly higher than those in the laser irradiation,simvastatin treatment and laser plus simvastatin groups.There was also a very significant difference in high-density lipoprotein(HDL)-C levels among the laser irradiation group,the laser plus simvastatin group and the control group.The pathological changes observed were correlated with blood fat levels.Mild atherosclerosis was found in the treated groups,but severe or in- termediate atherosclerosis was more prevalent in the control group.Conclusion Low-intensity laser irradiation a- lone or combined with simvastatin can significantly decrease blood lipid levels and the severity of pathological changes associated with aortic atherosclerosis in this animal model.
