ObjectiveTo study the correlation between the appearance color of the sample powder and the contents of five non-sugar components of wine-processed Polygonatum kingianum rhizoma during processing, and determine the feasibility of color quantitative value for judging the processing end point of the wine-processed products, and to screen steroidal saponins and flavonoids as markers for the control of the wine-processed products during processing. MethodThe changes of apparent color of the sample powder at different time points of the wine-processed products were measured by colorimeter, and the total color value (E*ab),the total color difference value (ΔE*ab) were calculated. The contents of protodioscin, pseudoprotodioscin, dioscin, diosgenin and narcissoside in the wine-processed products (No. S0-S10) after processing for 0, 5, 10, 14, 16, 18, 20, 22, 24, 26, 28 h were determined simultaneously by ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). Cluster analysis (HCA), principal component analysis (PCA), partial least squares discriminant analysis (PLS-DA) and Pearson correlation analysis were used to analyze the chromaticity value of the sample powder and the content of the five components. ResultDuring processing of wine-processed P. kingianum rhizoma, E*ab of the sample powder showed a decreasing trend and the apparent color changed from light yellow to lacquer black. The contents of the five components showed an obvious dynamic change trend with time, and showed different laws. HCA results showed that the processing process of the wine-processed products could be divided into three stages, namely, the early stage (samples S0-S1), the middle stage (samples S2-S4) and the late stage (samples S5-S10). PCA results showed that there were significant differences in color and contents of five components between the initial sample and the processing samples, and the difference between samples S8 and S9 was the smallest. PLS-DA results showed that the variable importance in the projection (VIP) values of b*, the contents of pseudoprotodioscin, narcissoside, diosgenin and protodioscin were >1. Pearson correlation analysis showed that the contents of protodioscin, diosgenin and narcissoside had a significant positive correlation with E*ab (P<0.01), the content of diosgenin had a significant negative correlation with E*ab (P<0.01), while the content of pseudoprotodioscin had no linear correlation with E*ab. ConclusionIn the process of wine-processed P. kingianum rhizoma, there is a certain linear correlation between color quantitative value and chemical composition, and the processing end point can be determined objectively. It can be considered that protodioscin can be used as a marker for the control of the wine-processed products.

ObjectiveTo compare the differences in resistance and structure of skin between acupoints and non-acupoints, and to study the difference in skin permeability characteristics of Corydalis Rhizoma total alkaloid patches (CTTP) after administration at Shenque acupoint and non-acupoint, so as to provide experimental support for its clinical acupoint application to prevent and treat chronic pain. MethodTaking corydaline (CD), tetrahydropalmatine (THP) and corydalis L (CDL) as evaluation indexes, and the quantitative analysis was carried out by high performance liquid chromatography (HPLC). The mobile phase was methanol-0.04 mol·L-1 phosphoric acid aqueous solution (70∶30, pH 6.0 adjusted with triethylamine), the detection wavelength was 281 nm. In vitro transdermal test in Franz diffusion cell and in vivo transdermal test were used to study the skin permeability characteristics of CTTP through Shenque acupoint and non-acupoint administration. At the same time, the skin resistance between Shenque acupoint and non-acupoint was measured before and after the administration, and the distribution of the drug in each layer of the skin was compared by freezing sectioning, and visual verification was performed with fluorescence inverted microscope. ResultAfter 24 h of administration, the results of in vivo and in vitro experiments showed that the cumulative permeation and retention of CD, THP and CDL at Shenque acupoint skin were higher than those at non-acupoint skin (P<0.05, P<0.01), the skin resistance of Shenque acupoint was lower than that of non-acupoint at all time points. The fluorescence microscopic observation results showed that the drug content of each layer of the skin was all Shenque acupoint>non-acupoint, indicating that the skin of Shenque acupoint had better effect on drug penetration and storage than non-acupoint. ConclusionThe 24 h cumulative permeation and retention of CTTP in Shenque acupoint skin are higher than those in non-acupoint skin, and the mechanism may be related to the thin skin, low electrical resistance and large number of hair follicle bodies at Shenque acupoint.

Objective:To analyze the changes in biological characteristics including infectivity, growth and pathogenicity of Chlamydia muridarum ( Cm) after serial passage in vitro in special conditions in order to provide reference for screening attenuated live vaccines and virulence-related genes. Methods:Wild-type Cm strain (G0) was cultured for several passages using conventional cell culture method under alternate unassisted and assisted culture conditions. Then, the 28th generation (G28) of Cm was selected and compared with the parental G0 strain in terms of centrifugation dependence, attaching ability, intracellular growth curve, plaque size and fallopian tube lesions after genital tract infection in a mouse model. Results:Compared with the parental G0 strain, the G28 strain showed significantly decreased dependence on centrifugation during cell infection ( P<0.05) and increased attachment capacity to cells ( P<0.05). No significant differences were observed in the growth curves 32 h after cell infection or in the plaque sizes between the parental G0 and G28 strains. In the in vivo virulence test, fallopian tube lesions were observed in 87.5% of G0-infected mice and 37.5% of G28-infected mice ( P<0.05). Conclusions:Compared with the parental G0 strain, the G28 strain showed significantly enhanced in vitro infection ability, but decreased in vivo pathogenicity, which brought hope for further identification of virulence genes, isolation of attenuated strains with single genotype and development of live attenuated Chlamydia vaccines.

OBJECTIVE:To study the effects of different amounts of Euphorbiae Semen fatty oil in self-assembly micelles on intestinal absorption of 4 kinds of euphorbia (euphorbia L 1,L2,L3,L8)in rats. METHODS :The self-assembled micelle solution containing 4 kinds of euphorbia was prepared by adding 4 kinds of euphorbia (40 mg/L)in excess ,using the fatty oil of Euphorbia Semen(0.2,0.4,1,4 g/L)and sodium deoxycholate as carriers. Totally 60 rats were collected to establish in-situ one-way intestinal perfusion model. Different intestinal segments (duodenum,jejunum,ileum,colon)were perfused with drug-containing intestinal perfusion fluid according to different dosage of Euphorbiae Semen fatty oil. HPLC method was adopted to determine the contents of 4 kinds of euphorbia in the intestinal perfusate before and after perfusion. The absorption rate constant (Ka)and apparent absorption coefficient (Peff)of 4 kinds of euphorbia in different intestinal segments were calculated. The ileum segment with better absorption was selected as the object to investigate and calculate the ac cumulative absorption of 4 kinds of euphorbia. RESULTS:The self-assembled micelles formed by different concentrations of fatty oil of Euphorbiae Semen could significantly increase the absorption of 4 kinds of euphorbia in different intestinal segments to different extents. When the dosage of Euphorbiae Semen fatty oil was 0.4 g/L,the intestinal absorption effect of 4 kinds of euphorbia were all the best ;the Peff was significantly increased,compared with no fat oil group (P<0.05 or P< . According to the order of Ka and Peff of each intestinal : segment in different fatty oil dosage groups ,the absorption 0531-89628590。E-mail:1310394709@qq.com effect of 4 kinds of euphorbia in each intestinal segment was the best in jejunum and the worst in colon. Compared with no fatty oil group ,when the amount of Euphorbiae Semen fatty oil w as 0.2-4 g/L,accumulative amount of 4 kinds of euphorbia in the ileum of rats increased significantly (P<0.05 or P<0.01), and the highest in 0.4 g/L Euphorbiae Semen fatty oil group. CONCLUSIONS :The self-assembly micelle s composed of Euphorbiae Semen fatty oil and deoxycholate can increase the absorption of euphorbia L 1,L2,L3,L8 in each intestinal segment to different extent,and the jejunum is the main absorption segment.

Objective To sum up and analyze the clinical and pathological characteristics in patients with both IgA nephropathy (IgAN) and diabetes mellitus. Methods A total of 500 patients were recruited, including 25 patients with both IgAN and diabetes mellitus, and 475 patients with IgAN only, who were diagnosed by renal-biopsy during Jan 2015 to Jan 2017 at the First Affiliated Hospital of Zhengzhou University. The clinical and pathological data were collected and analyzed using SPSS 22.0. Propensity Score Matching was used to match and select the patients in the both groups, and thereafter the depth of the basement membrane from the matched patients were compared using electron microscopy. The data of the patients whose follow - up time was ≥3 months were retrospectively collected, and Kaplan-Meier analysis was used to compare the difference of the prognosis. Results Compared to the patients with IgAN only, patients with both IgAN and diabetes mellitus were older [(46.36±13.49) years vs (34.00±13.80) years, P<0.001], had higher level of serum triglyceride [2.06(1.52, 3.11) mmol/L vs 1.51(1.01, 2.25) mmol/L, P=0.012] and thicker basement membrane [(384.33 ± 61.20) nm vs (346.72 ± 52.65) nm, P=0.044]. The patients with both IgAN and diabetes mellitus were more prone to reach the composite endpoint [4/7(57.14%) vs 25/265(9.33%), P<0.001] and had worse prognosis (Log-Rank test, P=0.004). Conclusions IgAN patients with diabetes mellitus have different clinical, pathological characteristics and prognosis from patients with IgAN alone. These patients need to be closely monitored and actively treated.

Objective To observe the efficacy difference between nerve-trunk stimulation plus electroacupuncture at antagonistic points and the control group. Method Patients were randomized into group A (nerve-trunk stimulation plus electroacupuncture at antagonistic points), group B (electroacupuncture at antagonistic points), and group C (basic treatment), 45 cases in each group. The interventions were given once a day, 5 times a week, totally for 4 weeks. The changes of spasm degree and motor function of upper limb and activities of daily life were observed. Result After intervention, there were significant differences between group B and group C, and between group A and group B (P<0.05); there was a significant difference between group A and group C (P<0.01). Conclusion Compared to the control group, nerve-trunk stimulation plus electroacupuncture at antagonistic points can produce more significant efficacies in improving the spasm degree and motor function of upper limb and activities of daily life.

Objective To explore the relationship of therapeutic compliance and complications in children with severeβ-thalassemia.Method One hundred and eight children with severeβ-thalassemia involved in the survey using convenient sampling to investigate the relationships between compliances with blood transfusion and iron removal treatment and complications.Results Of all the patients,66 complied with blood transfusion and the rest 42 did not;73 complied with the therapy and the rest 35 did not. Those complying with the blood transfusion had significant lower rates of developmental retardation,unusual faces,hepatosplenomegaly(P<0.01). Those complying with the therapy had significant lower rates of injuries in liver,heart and endocrinal system(P<0.01).Conclusion Therapeutic compliance in children with severeβ-thalassemia is at lower level so that the incidence of complications in them is higher.

This study was aimed to optimize the alcohol precipitation techniques for honeysuckle extract in order to standardize the production of honeysuckle extract of Qingkailing Injection and reduce the differences among batches. The orthogonal test was applied in this study. The content of chlorogenic acid and galuteolin were taken as compre-hensive indicators. Multi-index comprehensive scoring method was used in the data analysis. Three influencing fac-tors, which were the fluid temperature, the stirring speed and the speed of adding alcohol, were optimized in the al-cohol precipitation techniques. The results showed that the optimal alcohol precipitation techniques were when the fluid temperature was 20℃, the stirring speed was 240 rpm and the speed of adding alcohol was one time of the ma-terial per minute. It was concluded that the optimized alcohol precipitation process was stable and feasible.

Objective To analyze clinical characteristics of migraine complicated with cerebral edema.Methods We first time reported a case of familiar hemiplegic migraine complicated with diffuse cerebral edema in China,another 19 cases of migraine complicated with cerebral edema were reported in literature.The clinical characteristics of these 20 cases were analyzed.Results In 20 cases,the male:female ratio was 1.2∶ 1,the mean age of onset was (16.0 ± 10.2) years.All 20 cases were complicated with cerebral edema,among whom 8 (40%) were familiar hemiplegic migraine,8 (40%) were sporadic hemiplegic migraine,4 (20%) were migraine with aura.Common neurological symptoms and signs included paralysis (75%),aphasia (65%),disturbance of consciousness (60%),disordered vision (40%),Paresthesia (35%).Eleven cases (55%) had permanent neurological deficit.Conclusions The clinical manifestations of migraine with cerebral edema are complicated,the neurological disturbance is severe and usually has a long duration.The early diagnosis and treatment is beneficial,and preventive treatment in intermittent period is recommended.

<p><b>OBJECTIVE</b>To investigate the effects of the solid-state fermentation technology in extracting the ingredients of traditional Chinese medicine (TCM).</p><p><b>METHOD</b>Using Danggui Kushen pills as an example, four index ingredients of ferulic acid, matrine, matrine alkaloids and dry extract were compared comprehensively, and the difference between the two water extractions of fermented pills and unfermented pills was evalued.</p><p><b>RESULT</b>The comprehensive evaluated result was that extraction of fermented pills was better than that unfermented.</p><p><b>CONCLUSION</b>The solid-state fermentation is conducive to the extraction of prescription of Danggui Kushen pills.</p>

<p><b>OBJECTIVE</b>To optimize the extraction condtions in the modification of Rhizoma Cyperi, which had been extracted by the technique of supercritical CO2 extraction.</p><p><b>METHOD</b>The semi-bionic extraction (SBE) conditions were optimized through homogeneous design while the extracts (< or = 1 000), the total area of HPLC and dry extracts were adopted as markers.</p><p><b>RESULT</b>The optimized SBE extraction conditions are the following: pH in first extraction was adjusted to 2.005 3. And then pH in second and third extraction adjusted to 6.508 2 and 8.945 6, time of the whole extracted process was 3.912 7 h.</p><p><b>CONCLUSION</b>Combine the faction of production,we make sure the conclusion is pH in first extraction was 2.00, pH in second and third extraction adjusted to 6.50 and 9.00, duration of run was 2.0 h, 1.0 h and 1.0 h, respectively.</p>

RESUMO

Traditional Chinese medicine(TCM) injection,which plays a unique role in the clinical treatment,is Chinese original dosage form.But in recent years due to its quality problems or unscientifically clinical use,some adverse reactions have reported,even several injections had serious adverse reactions.So how to develop TCM injection becomes one of the topics the public and scholars most concerned about.Therefore,the three "bottleneck" problems of TCM injection on the current development,which are the material basis and TCM theory,clinical efficacy and safety,and also quality control,were analyzed and discussed in the article.Then the countermeasures were proposed and provided ideas for future research and development of TCM injection.

Objective:Optimising the technological parameters for the extraction of Maxingshigan Decoction (Herba Ephedrae, Semen Pruni Armeniacae, Gypsum Fibrosum, Radix Glycyrrhizae). Methods:Using the even design and TCL, ephedrine, glycyrrhetinic acid, hydrocyanic acid, calciumion and extract weight were taken as the indexes. Results:Optimal parameters: the pH values of the water for the 1st, 2nd and 3rd decoctions are 2.0 ,6.5 and 9.0,respectively; and the time of 3 decoctions are 2.0,1.0and 0.5h, respectively. Conclusion:Optimal parameters for Maximpshipan Decoction are feasible in extraction.

Objective: The extract technologies for the components of Fuzhuang Capsules were optimized. Methods: Four methods: the semi bionic extraction (SBE), the water extraction (WE), the semi bionic extraction by precipitation with alcohol (SBAE) and the water extraction by precipitation with alcohol (WAE) were used to extract Fuzhuang Capsules, with panaxadiol, panaxatriol, ? sitosterol, icariine, betaine, total sugar and dried extractive taken as the marker, under the same conditions of drug granularity, solvent amount, decocting temperature, filtration, concentration, etc., and then four methods were compared and studied. Results: Through the comprehensive evaluation of seven markers, their comprehensive values Y decreased in the order of SBE, WE, SBAE and WAE. Conclusion: SBE is better than the other three methods in the extraction of the components of Fuzhuang Capsules, which is showed by the fact that the pH values of the water for the 1st, 2nd and 3rd decoctions are 3.4, 7.5 and 8.4, and the extraction times are 2.0h, 1.5h, and 1.5h respectively.
