RESUMO

OBJECTIVES@#The common differentially expressed mRNAs in brain, heart and liver tissues of deceased sudden infant death syndrome (SIDS) and infectious sudden death in infancy (ISDI) confirmed by autopsy was screened by bioinformatics to explore the common molecular markers and pathogenesis of SIDS and ISDI.@*METHODS@#The datasets of GSE70422 and GSE136992 were downloaded, the limma of R software was used to screen differentially expressed mRNA in different tissue samples of SIDS and ISDI decedents for overlapping analysis. The clusterProfiler of R software was used to conduct gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis. The protein-protein interaction (PPI) network was constructed by STRING database, while the hub gene was screened by cytoHubba plug-in.@*RESULTS@#Compared with the control group, there were 19 significant differentially expressed genes in the tissue samples of SIDS and ISDI decedents, among which 16 in the heart tissue and 3 in the liver tissue, and the astrotactin 1 (ASTN1) gene expression difference in the heart tissue was most significant. The PPI network identified Ras homolog family member A (RHOA), integrin subunit alpha 1 (ITGA1), and H2B clustered histone 5 (H2BC5) were hub genes. The analysis of GO and KEGG showed that differentially expressed genes were enriched in the molecular pathways of actin cytoskeleton regulation, focal adhesion and response to mycophenolic acid.@*CONCLUSIONS@#ASTN1, RHOA and ITGA1 may participate in the development of SIDS and ISDI. The enrichment of differentially expressed genes in immune and inflammatory pathways suggests a common molecular regulatory mechanism between SIDS and ISDI. These findings are expected to provide new biomarkers for molecular anatomy and forensic identification of SIDS and ISDI.

The present study aimed to explore the effect of Erxian Decoction on proteomics of osteoblasts stimulated by hydrogen peroxide(H_2O_2) and its protective mechanism with the H_2O_2-induced cell model of oxidative stress. The primary osteoblasts were cultured from the skulls of newborn rats(within 24 hours) and divided into a control group, a model group, a Fosamax group, and an Erxian Decoction group. Blank serum was added in the control group and model group, and the drug-containing serum was added correspondingly to the remaining two groups. After 45 hours, H_2O_(2 )stimulation was conducted for three hours except for the control group, followed by protein extraction. Nano-LC-LTQ-Orbitrap system was used for protein detection, Protein Discovery for protein identification, and SIEVE for quantitative and qualitative analysis. Furthermore, following the blocking of PI3 K signaling pathway by LY294002(10 μmol·L~(-1)), a control group, a model group, an LY294002 group, an Erxian Decoction group, and an Erxian Decoction + LY294002 group were set up to observe the effect of Erxian Decoction on cell proliferation, alkaline phosphatase(ALP) activity, and the relative expression of BMP-2, OPG, p-Akt, p-FoxO1 of osteoblasts stimulated by H_2O_2 under LY294002 intervention. The results revealed that 78 differential proteins were discovered between the Erxian Decoction group and model group, which were involved in the regulation of PI3 K/Akt, glucagon, estrogen, insulin, and other signaling pathways. LY294002 blunted the promoting effect of Erxian Decoction on osteoblast proliferation and significantly down-regulated the expression of OPG and p-FoxO1, whereas its down-regulation on the expression of BMP-2 and p-Akt was not significant. Both LY294002 and Erxian Decoction increased the ALP activity of osteoblasts, which may be related to the cell state and the cell differentiation. The above results suggest that Erxian Decoction can protect osteoblasts stimulated by H_2O_2, with the PI3 K/Akt signaling pathway as one of the internal mechanisms.

RESUMO

Exosome is a goblet-shaped extracellular vesicle with a lipid bilayer structure, and its diameter is about 30-150 nm. Exosomes participate in cell-to-cell communication by transporting proteins, miRNAs, and other biologically active factors, thereby affecting recipient cells. In recent years, a lot of studies have shown that exosomes derived from bone-related cells can not only mediate intercellular communication within the bone marrow microenvironment in a range of physiological conditions, but also contribute to the multiple pathological conditions in skeletal disorders. Here, we summarized the regulation of miR-NA in exosomes derived from bone-related cells on bone formation.

OBJECTIVE: To investigate the clinical characteristics and the feasibility of laparoscopic treatment of ureteral endometriosis.METHODS: Retrospective analysis of the clinical characteristics,treatment methods and prognosis of 16 patients with ureteral endometriosis from Renji Hospital of Shanghai Jiao Tong University School of Medicine from January 2015 to February 2017.RESULTS: Ureteral stricture is the main symptom for all these 16 patients.15 patients merged with other deep infiltrating endometriosis(DIE)(15/16).All patients underwent laparoscopic surgery.Ureterolysis plus ureteral stent placement were performed in 8 patients,while laparoscopic ureteroneocystostomy in 6 and 2 abandoned surgery.Radical resection of DIE lesions were performed in 15 patients,hysterectomy in 4 patients,adenomyotomy in 2 patients,one-sided or double-sided ovarian cyst removal in 5 patients,ovariocystectomy and adnexectomy in 3 patients.More than 2 surgeries were performed in 1 patient.15 patients preserve ovarian function.No serious postoperative complications.CONCLUSION: Ureteral endometriosis is rare and difficult to diagnosis as lacking of specific symptoms.Laparoscopic treatment can achieve satisfactory therapeutic outcome.

RESUMO

OBJECTIVE: To figure out the regulatory models and concepts of FDA in drug review process that promotes surrogate endpoints development. METHODS: This paper made a thorough research to the FDA regulations, guidance documents and guidelines and literatures, and through historical retrospective of surrogate endpoints used in clinical drug trials and regulatory acceptance, interpretation of FDA's established development model in scientific use of surrogates, combined with the status quo surrogate endpoints in the field of serious diseases as cancer, AIDS, diabetes. RESULTS AND CONCLUSION: Then conclusion of FDA's experience in promoting regulation and utility of surrogate endpoints is summed up, including the development of guidelines to promote common understanding of specific use of surrogates; combining a variety of programs promote the development of surrogate endpoints; and using a risk-based classification in favor of the allocation of resources.

<p><b>OBJECTIVE</b>To investigate the clinical and pathological features, diagnosis and treatment of Castleman disease (CD).</p><p><b>METHODS</b>Clinical features and related information on diagnosis and treatment of eight cases of CD were retrospectively analyzed. The size of involved lymph nodes ranged from (2 cm x 2 cm x 3 cm) - (4 cm x 3 cm x 2 cm). The lymph nodes were found in level I (1 case), level II (3 cases), level III (3 cases) and level IV (1 case). CT examination in eight patients showed the lesions manifested as ellipse soft masses. Dynamic contrast CT scan in four patients showed ring-enhanced area around the masses. Blood routine examination in eight patients were generally normal, with six patients had mild anemia.</p><p><b>RESULTS</b>Based on the clinical classification, all lesions in this group were localized CD. Histopathology indicated that all lesions were of hyaline-vascular type. After surgery, there was no recurrence during the follow-up period.</p><p><b>CONCLUSIONS</b>Patients with localized CD mainly have lymphadenectasis in a single location. The CT scan can give some evidence. Surgery should be given first priority.</p>

RESUMO

Iliac artery rupture is a rare complication of post-stenting angioplasty and can lead to massive life-threatening haemorrhage. Conventional surgery can not repair the damaged vessel easily and may cause substantial blood loss and high operative morbidity and mortality. We report our experience with a self-expanding covered endoprosthesis for endovascular repair of the rupture of an iliac artery caused by stenting angioplasty.

<p><b>OBJECTIVE</b>To study effects of hypericin associated with light irradiation on human laryngeal squamous cell carcinoma strain Hep-2.</p><p><b>METHODS</b>Using techniques of tumor cells culture in vitro, Hep-2 cells were exposed to different concentration hypericin as 0.5, 1.0, 2.0, 3.0, 5.0 microg/ml, then 10 minutes 7.5 J/cm2 light irradiation was given after an hour. Other groups Hep-2 culture cells were also exposed to different concentration hypericin as 5.0, 10.0, 20.0, 25.0 microg/ml, without light irradiation. In all groups, contrast groups were set up. And 48 hours later, growth characteristics of Hep-2 cells were studied by morphological observation, fluorescence microscope, MTT assay and flow cytometry.</p><p><b>RESULTS</b>In normal contrast group, Hep-2 cells grew intensively and contacted with each other. However, cells which were treated with hypericin, combination with light irradiation were declined greatly. In higher dose hypericin group, necrosis could be found. MTT assay showed that hypericin associated with light irradiation inhibited growth of laryngeal cell with dose dependence manner. Flow cytometry showed that hypericin with light irradiation could block cell growth at G0/G1 phase, inducing apoptosis of laryngeal cell. Under fluorescence microscope, some sings of cell apoptosis including coagulation of chromatin, fragmentation of nuclei and apoptotic body could be found.</p><p><b>CONCLUSION</b>Human laryngeal squamous cell carcinoma strain Hep-2 can be inhibited and induced into apoptosis by treated with hypericin combination with light irradiation.</p>

<p><b>OBJECTIVE</b>To observe the clinical effect of Xiao Gu San on cirrhosis ascites.</p><p><b>METHOD</b>Groups were randomly divided into treating group and group of western medicine comparision.</p><p><b>RESULT</b>30 cases of the treating group were observed with the following result: first class curative effect 7; second class 12; the third class 9; effectiveless 2; total effective rate 93.33%. 30 cases of the Group of Comparison: first class curative effect 2; second class 6; the third class 15; effectiveless 7; total effective rate is 76.67%.</p><p><b>CONCLUSION</b>The curative effect of Xiaogu San combined with Western medicine on cirrhosis ascites is better than that of Western medicine alone.</p>
