RESUMO

Objective:To explore new methods to assist the diagnosis of glucose transporter type 1 deficiency syndrome (GLUT1-DS).Methods:Sixteen children with epilepsy and/or movement disorder carrying the SLC2A1 mutation who admitted to Department of Pediatrics, the First Medical Center, Chinese People′s Liberation Army General Hospital and Department of Nutrition, Shanghai Deji Hospital from October 2019 to October 2020 were retrospectively analyzed.GLUT1-DS was diagnosed based on clinical phenotype, glucose level in CSF and/or genetic testing results.Forty-four healthy children who underwent physical examination in the First Medical Center, Chinese People′s Liberation Army General Hospital during the same period were selected as healthy control group.Glucose transporter 1 (GLUT1) level on the membrane surface of peripheral red blood cells and erythrocyte glucose uptake rate were measured by flow cytometry and glucose oxidase method, respectively.Their differences between groups were compared by the rank sum test.The receiver operating cha-racteristic (ROC) curve was plotted to assess their diagnostic value. Results:Sixteen children were diagnosed as GLUT1-DS.GLUT1 levels of 16 children with GLUT1-DS were significantly lower than those of healthy control group [17.96% (13.43%, 22.12%) vs.27.93% (24.76%, 34.30%), Z=5.249, P<0.001]. Area under curve (AUC) was 0.946, and weighted Kappa was 0.791 ( P<0.001). The erythrocyte glucose uptake was measured in 12 children with GLUT1-DS, which was significantly lower than that of healthy control group [23.14% (14.80%, 26.45%) vs.27.40% (24.61%, 32.82%), Z=2.366, P=0.018]. AUC and weighted Kappa were 0.724 and 0.344, respectively ( P<0.001), showing a poor consistency. Conclusions:GLUT1 level on the surface of human erythrocyte membrane measured by flow cytometry may be a new method to assist the diagnosis of GLUT1-DS.The erythrocyte glucose uptake rate test requires stricter experimental conditions and needs further investigation.

RESUMO

Aiming at the current situation of high cost, huge volume, complex operation and difficulty in real application of pulse analyzer, this study designs and implements a portable pulse detection system based on IoT. The design utilizes Raspberry Pi 3B+, STM32 series MCU and cloud server to collect, store, display and recognize pulse signals at CUN, GUAN and CHI. The system is small in size and low in cost, which can be connected with cloud server through network to make full use of resources. The experimental results show that the recognition accuracy of the main feature points of the pulse signal by the portable pulse analyzer is higher than 97%, which has a broad prospect of development and application.

RESUMO

Infantile spasm is the most common epileptic encephalopathy in infancy.Without early diagnosis and effective treatment, serious consequences and economic burden maybe caused.One of the main characteristics of infantile spasm is the hypsarrhythmia on electroencephalography (EEG). It is important to analyze EEG accurately and reliably by scoring EEG.In order to further improve the EEG scoring system and better play the role of EEG in the diagnosis, treatment and prognosis implications of infantile spasm, in this paper, the advantages and disadvantages of various EEG scoring systems were analyzed, so as to provide clues by summarizing current EEG scoring systems and their applications in clinical practices.

RESUMO

Objective To investigate the effect of small bone window hematoma ventriculoscope for clinical treatment of hypertensive cerebral hemorrhage.Methods From January 2014 to June 2017,80 patients with hypertensive cerebral hemorrhage in Zhejiang Rongjun Hospital were selected and randomly divided into two groups according to the digital table,with 40 cases in each group.The control group underwent conventional craniotomy hematoma removal surgery,the observation group received small bone window hematoma ventriculoscope treatment.The operation situation,hospitalization time,clinical efficacy and nerve function defect score,ADL score,the incidence rate of complications were compared between the two groups.Results There was no statisticall significant difference in operation time between the two groups (t =0.284,P > 0.05).The amount of intraoperative bleeding of the observation group was (35.47 ± 10.69) mL,which was significantly less than (96.85 ± 27.54) mL of the control group (t =13.141,P < 0.05).The hospitalization time of the observation group was significantly shorter than the control group[(8.52 ±2.31) d vs.(11.79 ± 3.05) d] (t =5.405,P < O.05).The total effective rate of observation group was 92.5 % (37/40),which was higher than 75.0% (30/40) in the control group (x2 =4.501,P < 0.05).After treatment,the neural function defect score and daily life ability score of the observation group were (17.59 ± 2.98) points,(82.45 ±6.03) points,respectively,which were significantly higher than those of the control group[(22.17 ± 3.06) points,(73.21 ± 5.12) points] (t =6.782,7.388,all P < 0.05).The incidence rate of complications of the observation group was 5.0% (2/40),which was significantly lower than 20.0% (8/40) in the control group (x2 =4.114,P < 0.05).Conclusion Small bone window craniotomy evacuation of hematoma in the treatment of hypertensive cerebral hemorrhage,not only has significant clinical efficacy,but also has the advantages of less intraoperative blood loss,rapid postoperative recovery,less complications and so on.

Objective:To analyze the stress distribution in the fixed denture of edentulous maxilla supported by 6 implants at different implant position.Methods:The skull of a volunteer with total anodontia and moderate alveolar bone absorption was scanned by CBCT.Computer software,Abaqus 6.9,Geomagic Studio 12,and Mimics10.01,were applied for data processing and reconstruction of a 3D finite element model of implant-supported fixed denture with 6 implants in different location;and then,the stress distribution was analisized.Results:3D finite element model of edentulous maxillary implant-supported fixed bridge with good geometric similarity was established.The implant neck bone interface stress value of the distal of the implant fixed bridge at the molar area,the stress concentration area,was the maximum.The main stresses were compressive in the cortical bone adjacent to the mesial or the distal implants.The shift of the denture decreased from anterior to posterior.In model Ⅰ (implants at 13,15,17,23,25 and 27) distal cantilever was not used and the stress distribution was the evenest.Conclusion:The implant position at 13,15,17,23,25 and 27 of edentulous maxillary 6-implants-supported fixed bridge was the best for the stress distribution among the 8 models.

<p><b>OBJECTIVE</b>To explore the detection efficiency of circulating tumor cells (CTCs) in patients with hepatocellular carcinoma (HCC).</p><p><b>METHODS</b>Immunomagnetic negative enrichment by nanometer magnetic beads and label-free capture with Captor(TM) system were used to isolate and enrich CTCs from peripheral blood of HCC patients, and epithelial and HCC markers were applied to identify CTCs by immunofluorescence staining. CTCs were detected in 50 HCC patients before and after hepatectomy to test the method for isolation, enrichment and identification. The dynamic changes of pre- and post-operative CTCs' numbers were compared. The clinical data were analyzed using SPSS 19.0 software.</p><p><b>RESULTS</b>Negative enrichment methods by nanometer magnetic beads and label-free capture using Captor(TM) system were both suitable for CTCs isolation and enrichment in HCC patients. The positive detection rate of CTCs in HCC patients via negative enrichment was 96.0% (48/50), the preoperative median number of CTCs was 16 per 7.5 ml blood, and the postoperative median number was 17 per 7.5 ml blood.</p><p><b>CONCLUSIONS</b>Both negative enrichment and Captor(TM) system are suitable for isolation and enrichment of CTCs in HCC patients. There is a significant difference in the numbers of CTCs before and after operation, and dynamic detection of CTCs will provide helpful prognostic information for HCC patients in clinics.</p>

In proteomic research, to improve protein solubility of membrane proteins and nuclear proteins, buffers containing high concentration of detergent, such as 4% SDS, were widely used. However, high concentration of detergent might severely interfere with the downstream proteomic analysis, including protein quantitation and trypsin digestion. To improve the proteomic compatibility of buffers with high concentration of detergent, we used short gel method to pretreat buffers containing detergent. Protein samples were first separated by a short (2-2.5 mm) SDS-PAGE electrophoresis, and proteins were quantitated by comparing with bovine serum albumin standards via optical density analysis. The gel was then cut and peptides were recovered using in-gel digestion. The quantitative linearity range of this method was 1 to 8 μg. The quantitation was accurate and reproducible. After short gel analysis, recovered peptides generated high mass spectrometry signals. In conclusion, short gel method eliminated the interference of high concentration detergent in the proteomics analysis, and it was suitable for protein samples' pretreatment, and was worth to apply in proteomic research.

RESUMO

Nowadays, proteomics focuses on quantitative analysis rather than qualitative. In the field of quantitative proteomics, Isobaric tags for relative and absolute quantitation (iTRAQ) is one of the most widely used techniques. The advantage of iTRAQ is high throughput, high stability and free of the restriction of sample property. iTRAQ is suitable for almost all kinds of samples, and up to 8 samples can be analyzed simultaneously by commercially available kit. Along with the development of techniques, more and more mass spectrometry (MS) platforms are used in iTRAQ experiments and the accuracy of iTRAQ has been improved. iTRAQ has been applied to studies of microorganism, animal, plant, medical and protein post-translational modification. Here we review the recent progress in the development of iTRAQ and its applications in quantitative proteomics.

OBJECTIVE@#To observe the CT imaging features of anatomic structure related to endoscopic axilla approach for surgery of the frontal recess and frontal sinus.@*METHOD@#Thirty patients without a history of frontal sinus disease were undergone 16 line high speed spiral computed tomography. The computed tomographic images were analyzed to measure the related structures.@*RESULT@#The vertical distance from the front attachment point of the middle turbinate to the skull base was 13.88 +/- 2.59 mm. The horizontal distance from the top point of the axilla of the middle turbinate to the anterior wall of the frontal sinus outflow tract was 5.77 +/- 12.32 mm, to the anterior wall of the nasal cavity was 13.67 +/- 12.54 mm, to the lamina papyracea or lacrimal sac was 5.89 +/- 1.69 mm.@*CONCLUSION@#Sixteen line high speed spiral computed tomography is helpful to endoscopic axilla approach for surgery of the frontal recess and frontal sinus.

Mass spectrometry-based quantitative proteomics has become an indispensable tool for tumor marker discovery.Stable isotope labeling with amino acid in cell culture(SILAC),as a representative of in vivo metabolic labeling strategy,is a simple and relatively quantitative assay for comparative proteomics.Two kinds of cultured cells are grown in the medium containing either a light or heavy isotope labeled essential amino acids.After several cell doublings,the heavy amino acids are introduced into the nascent polypeptides in a sequence-specific fashion and the natural amino acid is completely replaced by its isotopically labeled analog.Accuracy quantification would be feasible by analyzing the peak intensities of every same peptide pairs labeled with light or heavy isotopes by the mass spectrometry.Compared with chemical labeling,SILAC requires much less amount of labeled proteins,and it is an efficient,straightforward,reproducible and accurate approach in cell-based systems.This method can not only investigate the dynamics of protein abundance,but also monitor the variation of posttranslational modifications and protein-protein interactions qualitatively and quantitatively.It is a powerful and important quantitative proteomics tool for tumor marker discovery.

RESUMO

Copy number variations (CNVs) refer as a DNA segment that is 1 kb or larger and is presented at a variable copy number in comparison with a reference genome. Classes of CNVs include insertions, deletions, duplications and their complex combinations. Because they widely distributed in the genome with some important characteristics, such as heritable, relative stable and heterogeneity, CNVs are considered as novel genomic polymorphism markers. And the alteration of gene dosage which resulted from CNVs could change phenotype, so a novel CNV genome-wide association analysis (CNV-GWAS) strategy appeared recently and began to used for identifying susceptible genes of complex diseases. It was approved that it could complement the tranditional genome-wide association studies based on single nucleotide polymorphisms. Therefore, genomic structure variances are favorable for revealing the molecular mechanisms and genetic foundation of complex diseases.

RESUMO

Effect of Gynostemma pentaphyllum Makino extract on the vitalit y and life-span of thorny nude belly water flea was studied. Resilis showed that all 0. 0125%, 0.0250%, 0.0500% and 0.1000% extract of G. pentaphyllum signifienntly effected the growth, development,reproduction and life span of the water flea.
