RESUMO

Objective To observe the effects of different doses of Sijunzi decoction(SJZD) on soleus(SOL) atrophy in tail-suspended rats.Methods Weightlessness was simulated by tail suspension in female Sprague-Dawley rats.Six rats in each group were assigned to a 5-group design randomly: control(CON);hind limb unloading(HLU) plus intragastric water instillation(HLU-W),as well as plus intragastric high dose(HLU-H),middle dose(HLU-M) and low dose(HLU-L) SJZD instillation respectively.Immunohistochemical method was used to observe the effects of them on expression of myosin heavy chain(MHC) isoforms in soleus muscle.Results Compared with HLU-W,muscle wet weight(MWW) of SOL in high and middle dose group increased by 33.11%and 37.57%respectively,as well as no significant difference with CON;three doses of SJZD groups increased the percentage of type Ⅰ myosin by 34.04%~54.90% and its cross sectional area(CSA) by 8.50%~36.86%,moreover SJZD of middle dose group showed no significant difference with that of CON group.Conclusion Three doses of SJZD groups can significantly prevent soleus atrophy induced by simulated weightlessness,and middle dose is the more effective one than high and low dose.

RESUMO

Objective To study the effects of choline chloride on activities of myosin adenosine triphosphatase (mATPase) of muscle fibres and muscle atrophy in tail-suspended rats. Method Twenty-four adult female Sprague-dawley rats were averagely divided into 3 groups matched for body mass:control group(CON),tail suspension group(TS) and tail suspension group associated with choline chloride(TS+Cch).Weightlessness was simulated by 14 d tail suspension of rats;gastric lavage with choline chloride was given to 1/2 of the rats during tail suspension (15 mg/kg body weight.i.g.).Activities of myosin adenosine triphosphatase (mATpase) of intrafusal and extrafusal fibres in soleus (SOL) muscle were determined by Ca 2+-ATPase method.Paraffin sections of the soleus muscle were prepared by routine histochemistry method. RESUMO

Objective To study the influence and the optimal dosages of Sijunzi Decoction (SJZD) on treating digestive and immune function of mice with Pi-deficiency syndrome (PDS). Methods The experimental model of mice with PDS was induced by rhubarb decoction. Then the mice were treated with three dosages of SJZD (12.5, 25, and 37.5 g/kg), respectively. The body weight, body temperature, mean retention time of chyme in digestive tract, gastric emptying velocity, gastrin and motilin levels, and visceral index of mice were measured. Results Being compared with the normal group, the PDS mice showed a slower growth of body weight and a lower body temperature, a shorter mean retention time of chyme in digestive tract and faster gastric emptying velocity, a lower gastrin content, and a higher motilin content. Also splenic and thymus indexes were found to follow a downtrend in the PDS mice. Three dosages of SJZD showed a restoration effect on the above all indexes, and the high dosage was the most effective one, especially. Conclusion The high dosage (37.5 g/kg) of SJZD possibly is the optimal dosage on treating the digestive and immune diseases of PDS mice.
