RESUMO

OBJECTIVE To investigate the improvement effects of 3,5,6,7,8,3′,4′-heptamethoxyflavone (HMF) of Fructus Aurantii on rats with damp blockage of the middle energizer. METHODS The rats were randomly divided into normal group, model group, positive control group (Raceanisodamine tablet, 1 mg/kg), HMF low-dose, medium-dose and high-dose groups (0.3, 0.6, 0.9 mg/kg), with 7 rats in each group. Except for the normal group, the other groups were modeled by internal and external composite factors. After successful modeling, the rats in each group were given the corresponding drug or normal saline, once a day, for 14 days. The general behavioral states such as dietary intake, water intake and mental state of the rats were observed, and the fecal water content rate and saliva flow rate were measured. Hematoxylin-eosin (HE) staining was used to observe the pathological and morphology in gastric and small intestinal tissues of rats. The plasma content of aldosterone was detected, and the expression of aquaporins (AQP3) in the gastric tissue of rats was determined. RESULTS Compared with the normal group, the dietary intake and water intake of the model group rats were significantly decreased (P<0.01), the fecal water content rate, salivary flow rate, plasma content of aldosterone and the expression of AQP3 in gastric tissue were increased significantly (P<0.01). Gastric tissue injury invaded the mucosal muscle layer, resulting in mucosal muscle layer rupture; pathological and morphological changes such as small intestinal villous erosion and glandular structure destruction were observed in the small intestine. Compared with the model group, the dietary intake and water intake of rats were increased in HMF groups; fecal water content rate, salivary flow rate, plasma content of aldosterone, the expression of AQP3 in gastric tissue were decreased, most of the above differences were statistically significant (P<0.05 or P<0.01). The pathological and morphological changes in the gastric and small intestine tissues of rats had been improved to varying degrees. CONCLUSIONS HMF of Fructus Aurantii with dry property HMF could improve the symptoms of rats with damp blockage of middle energizer, the mechanism of which may be associated with reducing the content of plasma aldosterone and down-regulating the expression of gastric AQP3.

Objective@#To explore the role of PinXl in the migration and invasion of melanoma cells, and study the mechanism from the perspective of extracellular matrix .@*Methods@#PinX1-siRNA was transfected into melanoma cells A375 and MV3 respectively. Detect the effect of PinXl on proliferation abilities of melanoma cells A375 and MV3 by CCK-8 assay. The migration and invasion abilities of the two cell lines were detected by Transwell migration and invasion assay experiments.The protein expression levels of PinXl, TIMP-1 and MMP-9 were detected by Western Blot analysis.@*Results@#The proliferation, migration and invasion abilities of melanoma cells A375 and MV3 were significantly increased after PinX1 gene knockdown. The expression level of MMP-9 was significantly increased (P<0.01) and the expression level of TIMP-l was reduced significantly(P<0.01).@*Conclusions@#Down-regulation of PinX1 may activate the MMP-9 pathway by down-regulating the TIMP-1 protein, which improve the migration and invasion ability of melanoma cells.

Objective To observe the effects of Yuanzhi San (YZS) on the ethology and cerebral acetylcholinesterase(AchE) activity of mouse model of memory disorder induced by scopolamine. Methods Sixty mice were randomly divided into six groups, namely blank control group, model group, positive medicine group, and low-dose, middle-dose and high-dose YZS groups. Except for the blank control group and model group were given the normal saline, the mice in other groups were administered with the corresponding drugs for 10 days. And then, mice in the medication groups were given subcutaneous injection of scopolamine in the dose of 3mg/kg to induce memory disorder model. Morris water maze test and step-down test were adopted for the observation of the learning-memory ability of the mice, and at the end of the tests, the activity of AchE in mouse cerebral cortex was measured by a biochemical method. Results Compared with the model group, escape latency was decreased, and retention time and swimming distance in the effective area in Morris water maze test were prolonged in YZS groups (P0.05). Conclusion YZS exerts certain effect on improving learning-memory ability of memory disorder mice induced by scopolamine , and the mechanism might be related with the inhibition of AchE activity in the cerebral cortex of model mice.

RESUMO

One new dicyclopeptide cyclo-(L-N-methyl Glu-L-N-methyl Glu) (1), together with one new natural dicyclopeptide cyclo-(L-methyl Glu ester-L-methyl Glu ester) (2), and two known dicyclopeptides cyclo-(L-methyl Glu ester-L-Glu) (3), and cyclo-(L-Glu-L-Glu) (4), were isolated from the aerial parts of Dianthus chinensis L. Their structures were determined by spectroscopic analyses and chemical methods.

RESUMO

<p><b>OBJECTIVE</b>To study the chemical constituents from the fruits of Leonurus japonicus.</p><p><b>METHOD</b>The compounds were extracted by 70% methanol, then isolated and purified by column chromatography on silica gel, RP-18, Sephedax LH-20 and HPLC. Their structures were determined on the basis of spectroscopic data and physicochemical property.</p><p><b>RESULT</b>Two 28-noroleanane-derived spirocyclic triterpenoids including a new acylated nortriterpenoid were isolated and identified as (17R)-19(18 --> 17)-abeo-2alpha,18beta,23-trihydroxy-3beta-E-feruloyoxy-28-norolean-12-ene (1) and phlomistetraol B (2).</p><p><b>CONCLUSION</b>Compound 1 is a new compound,named as leonujaponin A. Both compounds 1-2 are isolated for the first time from Leonurus plants.</p>

RESUMO

<p><b>OBJECTIVE</b>To study the effects of total alkaloids of Rubus alceaefolius (RAP) on non-alcoholic fatty liver disease (NAFLD) rats and explore its possible mechanisms.</p><p><b>METHOD</b>Sixty SD rats were randomly divided into six groups: control group, model group, compound methionine and choline bitartrate tablets (CMCB)group and three RAP groups treated respectively with low, middle and high dose of RAP. The NAFLD model was induced by feeding fat-rich food. NAFLD rats were administrated with 0.35 g x kg(-1) CMCB and 0.36, 0.72, 1.44 g x kg(-1) RAP for 4 weeks respectively. The weight index of liver was measured. Hepatic histolog ical changes were observed. The concentration in serum of aspartate amino transferase (AST), alanine amino tranferase (ALT), tumor necrosis factor-alpha (TNF-alpha) and interleukin-6 (IL-6) were determined. The mRNA expressions of SOD, MDA, TNF-alpha and IL-6 in hepatic tissue were detected.</p><p><b>RESULT</b>Compared with the model group, degree of steatosis of hepatic lobule was improved, the weight index of liver was decreased, serum levels of ALT, AST, TNF-alpha and IL-6 were significantly lower in the high and middle dose RAP group (P < 0.05 or P < 0.01). The levels of SOD and MDA in hepatic tissue were lower in the high dose RAP group (P < 0.05). The mRNA expressions of TNF-alpha and IL-6 in hepatic tissue were decreased (P < 0.05 or P < 0.01).</p><p><b>CONCLUSION</b>RAP can protect liver in experimental NAFLD, and its possible mechanisms may be concerned with clearing the oxygen free radical, reducing the product of lipid peroxidation, inhibiting the release of inflammatory cytokines and reducing nflammatory response.</p>

Objective To investigate the effects of keratinocyte growth factor(KGF) and keratinocyte growth factor receptor(KGFR) on the malignant transformation of gestational trophoblastic disease(GTD).Methods Immunolocalization of KGF/KGFR was performed on sections prepared with the samples from 26 hydatidiform mole,18 invasive mole and 12 choriocarcinoma.The in situ hybridization was used to detect the mRNA of KGF/KGFR in the tissues of hydatidiform mole and GTD.Analysis was performed according to intensity of staining and number of positive cells.Results It was revealed that specific staining for mRNA and protein of KGF/KGFR existed in hydatidiform mole and gestational trophoblastic tumor(GTT).The mRNA and protein of KGF/KGFR were allocated in cytoplasm of syncytiotrophoblasts and cytotrophoblasts of malignant hydatidiform mole,and the KGF/KGFR protein was also expressed in benign tissue,while the expression of KGFR in malignant hydatidiform mole was significantly higher than that in benign tissue(?2=12.775,P
