RESUMO

1. Lipase enzyme was produced from A. ustus under optimized cultural conditions [C/N ratio was 0.4/1% i. e: the weight of waste material, soy-bean litters, equivalent to 0.4% lipid is 6.48%, and peptone, 1% for 3 days at pH 6.98 in presence of 1 mg/ml FeCl[3] under shake conditions]. 2. The enzyme was extracted and purified by precipitation with [NH[4]][2] SO[4] then by chromatography on DEAE-Sephadex A[50] followed by Gel-filtration on Sephadex G[100]. Recovery was 27.67%. 3. The enzyme was clearly distinguished from lipases previously characterized, On using SDS-PAGE, the final fraction splits into two bands with electrophoretic mobility 0.24 and 0.56, indicating that the molecule consists of two peptide chains with molecular weight 34.670 and 66.70 Daltons with PI 7.1. km and V[max] values were 5.8 g/100ml and 31.2 U/mg protein, respectively. The maximum activity occurred at 37°C and pH 7.5 for 2.5 hrs. The enzyme has thermostability properties. 4. The enzyme protein is rich in essential amino acids as compared with FAO and it seems to be related to the same group of porcine pancreatic lipase

RESUMO

Batch shake cultures of Bacillus cereus grown in tryptic soy broth medium gave the highest beta-lactamase activities, [Intracellular, [ICL] 2.34; Extracellular [ECL], 2.39; Cell-bound [CL], 1.56 m mol/ min /ml] as well as protein content [Intracellular proteins, [ICP], 5.77; Extracellular proteins [ECP], 8.88; Cell-bound proteins [CP], 2.61 mg/ml and biomass [600 mg/ml medium] after an incubation period of 48 hr. at 30 °C and pH 7.3. Sodium salts of different organic acids had varying influences on the fermentation yields. RESUMO

The effect of salt stress on the growth parameters of S.cerevisiae Y-1347 was studied by using Nacl. The increase in Nacl level was accompanied by a parallel increase in the efficiency of the cultures to convert glucose to glycerol. Maximum glycerol and glycerol phosphate dehydrogenase production [700 mg/l00ml and 198 U/ml respectively] were achieved with 5.8 g% Nacl 3% [W/V] NaHSO[3] and 1 g/l beef extract in presence of tween 80 with a[w] = 0.24k[-1]

RESUMO

Variations in the mineral composition of the medium, its hydrocarbon and nitrogen sources, played a significant role in the productivity of lipids and protein by Candida parapsilosis wild type, KSh 21. Among the petroleum fraction tested were gasolene, kerosene and solar as well as El-Alameen crude oil. Solar at a concentration of 3% v/v proved to be the best carbon source utilized by the organism giving the highest frementation yields. Factors such as fermenation time course, solar level, pH relations [initial and biffered], nature and concentration of nitrogen source, phosphate level and addition of magnesium sulphate and soduim chloride were also tested
