@#ObjectiveTo determine the effect of measles and rubella combined attenuated live vaccine(MR)supplementary immunization activities(SIA)of enterprise employees in Jinshan District,Shanghai from 2019 to 2021.Methods A total of 437 enterprise employees immunized with MR were collected for the serum samples of 2 ml before and one month after immunization in Jinshan District from 2019 to 2021. The IgG level against measles was determined by ELISA,and the geometric mean concentration(GMC)was calculated. The antibody against measles was judged as positive when the GMC of IgG antibody was more than 200 mIU/mL,while the protective antibody was positive when the GMC was more than800 mIU/mL;After MR vaccination,the immunity was judged to be effective when measles IgG antibody changed from negative to positive or IgG level against measles increased more than 4 times.ResultsThe antibody positive rate,positive rate of protective antibody and IgG antibody GMC of measles before MR vaccination were 90. 85%,38. 44% and795. 29 mIU/mL,while 99. 31%,76. 20% and 1 481. 51 mIU/mL after vaccination,respectively. The vaccination success rate was 26. 54%,which decreased with the increase of antibody level before vaccination(χ2= 119. 022,P < 0. 001).ConclusionThe effect of MR SIA in the population with low measles antibody level was good,which suggested that SIA of measles-containing vaccine should be carried out in enterprise employees with low level of measles antibody.

@#Abstract:Objective To analyse the level of measles antibody and serological susceptibility of the enterprise employees in Jin⁃ shan District,Shanghai in 2019,to provide references for measles prevention and control of enterprise employees. Methods A total of 1 796 employees were selected from 6 enterprises in Jinshan District by using a stratified random sampling method,of which serum samples from intravenous blood were collected and determined for IgG antibody level of measles by ELISA. Geo⁃ metric mean concentration(GMC)of measles IgG antibody,antibody positive rate and antibody protection rate were calculated and the susceptibility factors of measles in enterprise employees were analyzed by using the unconditional multivariate Logistic regression model. Results The GMC of measles antibody in 1 796 monitoring objects was 639. 71 mIU/mL,the positive rate was 87. 58% and the protection rate was 29. 06%. Multivariate Logistic regression analysis showed that the odds ratio(OR)of measles susceptibility was 1. 55(95% CI:1. 14 ~ 2. 12)in native subjects compared with non⁃resident subjects,while 2. 35 (95% CI:1. 35 ~ 4. 08)in subjects aged 40 ~ 49 years compared with those aged ≥ 50 years. Conclusion In 2019,the measles antibody level of enterprise employees in Jinshan District of Shanghai was low,and people aged 40 ~ 49,especially the nativeadults,wereathighriskofsusceptibilitytomeasles. Supplementaryimmunizationwithmeaslesvaccineshouldbecarried out for high⁃risk enterprise employees to reduce the risk of measles infection and control the epidemic situation of measles.

Objective@#To understand the impact of screen time on duration and quality of sleep, so as to provide scientific basis for the development of interventions.@*Methods@#A total of 3 584 preschool children in Haikou City were selected by stratified sampling method from March to June in 2022, and the Children s Sleep Habits Questionnaire (CSHQ) and self designed questionnaire were used.@*Results@#The average sleep duration of preschool children was (10.41±0.98)h/d, the rate of sleep deprivation was 28.71 %, and the report rate of sleep problems was 50.47%. The average sleep duration in general as well as on weekdays and weekends decreased by age ( F=21.00, 29.53, 3.26, P <0.05), and insufficient sleep duration rate significantly varied by age groups ( χ 2=29.85, P < 0.01 ). The average screen time was (1.15±0.37)h/d, and the screen exposure rate was 55.39%. The total sleep problems ( 52.14 %), poor bedtime habits (43.02%), daytime sleepiness (67.10%), irregular sleep duration (53.05%), and abnormal sleep latency ( 24.99 %) were all higher than those in the non exposed group (48.41%, 39.59%, 63.29%, 48.91%, 19.57 %) ( χ 2= 4.94 , 4.31, 5.69 , 6.08, 14.85, P <0.05). Multiple linear regression analysis showed that age and weekend average screen time were negatively associated with sleep duration ( β =-0.01, -0.06), weekday and weekend average outdoor activity duration were positively correlated with sleep duration ( β =0.08, 0.08) ( P <0.05).@*Conclusion@#Screen exposure to preschool children s sleep time,sleep quality was closely related. Parents should cultivate good sleep habits of preschool children, and limit children s screen time to maximize outdoor activities, so as to ensure healthy development of children.

@#[摘 要] 目的:探讨甲基转移酶样蛋白7B(METTL7B)在胶质瘤组织中的表达及其与患者临床病理特征和预后的相关性。方法:基于CGGA数据库胶质瘤数据和GTEx数据库正常脑组织数据,分析METTL7B基因在胶质瘤与正常脑组织中的表达差异,并用GEPIA数据库数据和免疫组织化学染色法进行验证。用Kaplan-Meier生存分析、单因素Cox分析、多因素Cox分析及ROC曲线分析等评估METTL7B在胶质瘤患者预后中的价值,用CGGA数据库数据分析METTL7B表达与胶质瘤患者临床病理特征的相关性,用CIBERSORT及TIMER数据库进行肿瘤免疫细胞浸润分析,进行KEGG通路富集分析及GO功能富集分析,通过基因共表达分析确定与METTL7B相关的基因。结果:METTL7B在胶质瘤组织中明显上调(均P<0.05),METTL7B表达是胶质瘤患者独立的不良预后因素。METTL7B高表达与高龄(>41岁)、肿瘤分级增加、肿瘤复发或继发性肿瘤、IDH野生型、1p19q非共缺失以及肿瘤的恶性病理学有关联(均P<0.01);METTL7B表达与B细胞、CD4+ T细胞、CD8+ T细胞、单核细胞、中性粒细胞、巨噬细胞、活化肥大细胞等免疫细胞有关联(均P<0.05)。KEGG通路富集及GO功能分析结果显示,肿瘤相关信号通路及多种免疫反应在METTL7B高表达表型中显著富集。基因共表达分析结果表明,METTL7B与TNFRSF12A、CHI3L1、EMP3表达呈正相关(r=0.807、0.804、0.783,均P<0.01),与ELFN2、REPS2、SHANK2表达呈负相关(r=-0.642、-0.627、-0.602,均P<0.01)。结论:METTL7B在胶质瘤组织中的表达上调是预后不良的指标,且与肿瘤免疫浸润相关。

OBJECTIVES@#To study the effect of intrapartum antibiotic prophylaxis (IAP) of group B streptococcus (GBS) infection on the incidence and bacteriological profile of early-onset neonatal sepsis (EONS).@*METHODS@#A retrospective analysis was performed on the medical data of 494 pregnant women with positive GBS screening results and 526 neonates born by these women. According to whether the pregnant woman received IAP, the neonates were divided into two groups: IAP (n=304) and control (n=222). The two groups were compared in terms of clinical indices, incidence rate of EONS, and distribution of pathogenic bacteria in blood culture.@*RESULTS@#Compared with the control group, the IAP group had a significantly lower proportion of children with abnormal clinical manifestations (P<0.001) and a significantly lower incidence rate of EONS (P=0.022). In the IAP group, Escherichia coli (2.3%) was the most common type of pathogenic bacteria in blood culture of the neonates with EONS, while GBS (3.2%) was the most common type of pathogenic bacteria in the control group. The IAP group had a significantly higher detection rate of ampicillin-resistant Escherichia coli than the control group (P=0.029).@*CONCLUSIONS@#Although IAP can significantly reduce the incidence rate of EONS in neonates born to pregnant women with positive GBS screening results, the infection rate of ampicillin-resistant Escherichia coli may increase after IAP treatment. Therefore, it is needed to enhance the monitoring of blood culture results of neonates with EONS and timely adjust treatment plan according to drug susceptibility test results.

ABSTRACT Purpose: To explore the role and mechanisms of octreotide in neurofunctional recovery in the traumatic brain injury (TBI) model. Methods: Rats were subjected to midline incision followed by TBI in the prefrontal cortex region. After 72 hours, the behavioural and neurological deficits tests were performed, which included memory testing on Morris water maze for 5 days. Octreotide (15 and 30 mg/kg i.p.) was administered 30 minutes before subjecting to TBI, and its administration was continued for three days. Results: In TBI-subjected rats, administration of octreotide restored on day 4 escape latency time (ELT) and increased the time spent in the target quadrant (TSTQ) on day 5, suggesting the improvement in learning and memory. It also increased the expression of H2S, Nrf2, and cystathionine-γ-lyase (CSE) in the prefrontal cortex, without any significant effect on cystathionine-β-synthase. Octreotide also decreased the TNF-α levels and neurological severity score. However, co-administration of CSE inhibitor (D,L-propargylglycine) abolished octreotide-mediated neurofunctional recovery, decreased the levels of H2S and Nrf2 and increased the levels of TNF-α. Conclusions: Octreotide improved the neurological functions in TBI-subjected rats, which may be due to up-regulation of H2S biosynthetic enzyme (CSE), levels of H2S and Nrf2 and down-regulation of neuroinflammation.

Objective@#To explore current status of sleep disturbances and its influencing factors in preschool children in Haikou city, and to provide a reference for improving chidlren sleeping quality by taking effective measures.@*Methods@#Layered random cluster sampling method was used to select 1 200 preschool children from kindergartens in Haikou city in 2018. Sleep status questionnaire was used to investigate children’s sleep disorders and analyze the influencing factors.@*Results@#The detection rate of sleep disturbances was 50.00%, the rates in each group were 50.90% for boys, 48.87% for girls(χ2=0.49, P>0.05), and 48.45% for 3 years old group, 50.33% for 4 years old group, 50.55% for 5 years old group, and 49.55% for 6 years old group(χ2=0.24, P=0.97). The total daily sleep time of children was(10.53±1.05)h, and the duration of continuous sleep at night was(8.69±0.78)h, the length of sleep during the day was(1.84±0.67)h, sleep time decreased with the increase of age(F=7.52, 3.04, 7.88, P<0.05). The univariateand multivariate Logistic regression showed that, asthma(OR=2.65), frequent cold(OR=1.67), carpet at home(OR=1.40), poor appetite(OR=1.18), changing caregivers(OR=1.25), inconsistent rearing ideas(OR=1.42), parents falling asleep late (OR=1.39) and mothers with sleep problems (OR=1.73) were the risk factors for sleep problems, children and parents sleeping separately(OR=0.75), children’s sleeping rule (OR=0.32) were the protection factors(P<0.05).@*Conclusion@#It is necessary to do a good job in the sleep management of preschool children and reduce the detection rate of sleep problems from many aspects, such as family rearing environment, health knowledge of caregivers, parents undefined sleep habits, children undefined sleeping style and so on.

OBJECTIVE To investigate effects of Wudang cherry on urate excretion and renal function and examined whether renal organic ion transporters were involved in potassium oxonate-induced hyperuricemic mice. METHODS The model of hyperuricemic mice was induced by intraperitoneal injection of potassium oxonate (250 mg·kg- 1) for 7 d. Water extracts of Wudang cherry at 500 mg·kg- 1 were orally administered to hyperuricemic mice for 7 d, benzbromarone (20 mg·kg- 1) and allopurinol (20 mg · kg- 1) were given as positive controls, vehicle control group was given equal normal saline.Serum and urine levels of uric acid were measured in hyperuricemic and normal mice. Simultaneously, the mRNA and protein levels of mouse urate transporter 1 (mURAT1), glucose transporter 9 (mGLUT9), organic anion transporters (mOAT1 and mOAT3), ATP- binding cassette, subfamily G, membrane 2 (mABCG2) and organic cation/carnitine transporters (mOCT1, mOCT2, mOCTN1 and mOCTN2) in the kidney were analyzed by Western blot, RT-PCR, immunohistochemical and immuno?fluorescent assay, respectively. RESULTS Wudang cherry significantly reduced serum uric acid levels and increased urine uric acid levels in hyperuricemic mice. And it effectively reversed potassium oxonate-induced alterations in renal mURAT1, mGLUT9, mOAT1, mOAT3 and mABCG2 mRNA and protein levels, resulting in the enhancement of renal urate excretion in mice. Moreover, Wudang cherry increased renal mOCT1, mOCT2, mOCTN1 and mOCTN2 mRNA and protein levels, and improved renal impairment in this model. CONCLUSION Wudang cherry processes uricosuric and nephroprotective actions by regulating renal organic ion transporters in hyperuricemic mice.

RESUMO

A gene encoding NAD+-dependent sorbitol dehydrogenase (SDH) in peach fruit was cloned and expressed in Escherichia coli. Recombinant SDH protein with 6×His-tagged was localized exclusively in the cytoplasmic soluble fraction of E. coli when the strains were grown for 4-5 h at 37 ºC. Highly pure protein was isolated by Ni2+-resin chromatography with 150 mM imidazole in 50 mM Tris, pH 8.0, by elution. In order to ensure that the recombinant SDH could be used for further study, the fluorescence and ultraviolet spectrum of the recombinant SDH were detected. Recombinant SDH was confirmed to be capable of oxidizing sorbitol by enzymatic activity assay. The activity of the recombinant SDH was 2.73 U mg-1min-1, which was similar with that directly extracted from peach fruits. The activities of SDH extracted from the fruits in different periods (30, 60, 90 days after flowing) were 7.75, 5.95, 3.26 U mg-1min-1, respectively.

Over-expression of P-glycoprotein(P-gp),an ATP-dependent drug efflux pump,represents one of the major mechanisms that contribute to multidrug resistance(MDR)in cancer cells.This study examined the effects of troglitazone,a ligand of peroxisome proliferator-activated receptor gamma (PPARγ),on P-gp-mediated MDR in SGC7901/VCR cells(a vincristine-resistant human gastric cancer cell line).The expression of P-gp was detected by RT-PCR and Westem blotting,respectively.The SGC7901/VCR cells were treated with 0.1 mg/L vincristine(VCR)alone or in combination with 1,5,10 μmol/L troglitazone for 24 h.PPARγ was measured by electrophoretic mobility shift assay(EMSA).The intracellular concentration of Rhodamine123(Rh123,a fluorescent P-gp substrate)was assayed to evaluate the activity of P-gp.The cell cycle and apoptosis were measured by flow cytometry.The results showed that the P-gp was increasingly expressed in SGC7901,BGC823 and SGC7901/VCR cells in turn,suggesting that MDR in the SGC7901/VCR cells was mediated by the increased expression of P-gp.In the SGC7901/VCR cells,the expression level of total PPARγ was increased,however,the protein level and activity of PPARγ,in the nuclei of cells decreased significantly.Troglitazone elevated the PPARγ,activity in SGC7901/VCR cells in a dose-dependent manner.Troglitazone decreased the P-gp expression and markedly enhanced the accumulation of Rh123 in SGC7901/VCR cells in a dose-dependent manner.We also found that troglitazone significantly increased the percentage of SGC7901/VCR cells in the G2/M phase and decreased the cell percentage in G1 and S phase in a dose-dependent manner.Troglitazone significantly increased the apoptotic rate of SGC7901/VCR cells treated by VCR or ADR in a dose-dependent manner.It was concluded that P-gp-overexpressed SGC7901/VCR cells have minor endogenous PPARγ activity.Elevation of the PPARγ activity by troglitazone can reverse P-gp-mediated MDR via down-regulating the expression and activity of P-gp in SGC7901/VCR cells.It was suggested that troglitazone can dramatically enhance the sensitivity of P-gp-mediated MDR cancer cells to chemotherapeutic agents.

<p><b>OBJECTIVE</b>To search for the best program increasing clinical therapeutic effect on vertebrobasilar insufficiency (VBI) and to study the mechanism.</p><p><b>METHODS</b>The patients were randomly divided into an acupuncture plus massage group, an acupuncture group and a massage group. Their therapeutic effects were observed and compared. And transcranial Doppler (TCD) was used for detection of systolic flow velocity (Vs), enddiastolic peak flow velocity (Vd), mean peak flow (Vm) and pulsatility index (PI) before and after treatment.</p><p><b>RESULTS</b>The cured rate and the total effective rate in the acupuncture plus massage group were significantly higher than the acupuncture group and the massage group (P < 0.05). And there were significant differences in Vm, PI and DI in the left vertebral artery (LVA), the right vertebral artery (RVA) and the basilar artery (BA) before and after treatment (P < 0.01).</p><p><b>CONCLUSION</b>Acupuncture combined with massage can improve Vm, PI, DI of VA-BA and the clinical therapeutic effect on VBI as well. It is superior to the simple acupuncture therapy and simple massage therapy. At present, it is one of the best therapy for VBI.</p>

<p><b>OBJECTIVE</b>To study the optimum acupuncture treatment program and the mechanism for treatment of cerebral infarction.</p><p><b>METHODS</b>Sixty-three cases were randomly divided into 9 groups. The four factors, times of manipulation, the retaining time of the needle, acupuncture instrument and acupoints,and their corresponding three levels were adopted respectively in treatment of each group. Then nerve function defect score and insulin resistance were observed before and after treatment.</p><p><b>RESULTS</b>The acupoints, the times of manipulation and the retaining time of the needle have significant effects on nerve function defect and insulin resistance (P < 0.01 or P < 0.05), and the acupuncture instrument has a significant effect on insulin resistance (P < 0.01). The choice of acupoints was the most important factor for acupuncture treatment of cerebral infarction.</p><p><b>CONCLUSION</b>Acupuncture for regulating The Governor Vessel with twice manipulations and retaining the needle for 60 min is optimum treatment program for cerebral infarction. The good regulating effect of acupuncture on insulin resistance is one of the mechanisms of achieving the therapeutic results.</p>
