Abstract Due to extensive application of antibiotics as growth promoters in animal feed, antimicrobial resistance has been increased. To overcome this challenge, rumen microbiologists search for new probiotics to improve the rate of livestock production. The present study was aimed to isolate and evaluate breed-specific lactic acid bacteria (LAB) as potential animal probiotics. The current study was conducted during 10 months from July 2020 to April 2021, in which a total of n=12 strains were isolated from different samples including milk, rumen, and feces of Nilli Ravi Buffaloes. These isolates were evaluated for their antimicrobial potential against common animal pathogens (Bacillus spp., E. coli, Staphylococcus aureus, Salmonella spp., Listeria spp.). All the isolates were identified using 16S rRNA gene sequencing and the phylogenetic analyses inferred that these strains showed close relations to the species of various genera; Enterococcus lactis, Pediococcus pentosaceus, Bacillus subtilis Weissella cibaria, Weissella soli, Bacillus tequilensis, Weissella bombi, Bacillus licheniformis, Lactococcus lactis, Bacillus megaterium, Lactobacillus ruminis, and Lactococcus lactis. NMCC-Ru2 has exhibited the enormous potential of antimicrobial activity, 28 mm, for Salmonella typhimurium;23 mm for Listeria monocytogenes 21 mm for E.coil. Highest resistance was seen in NMCC-Ru2 agasint test antbiotic, like 25.5 mm for Tetracycline. Overall results revesl that the probiotic profile of isolates was achieved using standard criteria, particularly with animal probiotic properties


Resumo Devido à extensa aplicação de antibióticos como promotores de crescimento na alimentação animal, a resistência aos antimicrobianos aumentou. Para superar esse desafio, os microbiologistas do rúmen buscam novos probióticos para melhorar a produtividade do gado. O presente estudo teve como objetivo isolar e avaliar bactérias lácticas específicas de raças (BAL) como potenciais probióticos animais. 12 cepas foram isoladas de diferentes amostras, incluindo leite, rúmen e fezes de búfalos Nilli Ravi. Esses isolados foram avaliados quanto ao seu potencial antimicrobiano contra patógenos animais comuns (Bacillus spp., E. coli, Staphylococcus aureus, Salmonella spp., Listeria spp.). Todos os isolados foram identificados por meio do sequenciamento do gene 16S rRNA e as análises filogenéticas inferiram que essas cepas apresentaram estreita relação com as espécies de vários gêneros; Enterococcus lactis, Pediococcus pentosaceus, Bacillus subtilis, Weissella cibaria, Weissella soli, Bacillus tequilensis, Weissella bombi, Bacillus licheniformis, Lactococcus lactis, Bacillus megaterium, Lactobacillus ruminis e Lactococcus lactis. O perfil probiótico dos isolados foi obtido usando critérios padrão, particularmente com propriedades probióticas animais.

The supplementation of Selenium-enriched probiotics is effective in reducing oxidative stress and maintaining meat quality stability in broiler chicken especially under heat stress. An experimental study was conducted to perform Comparative analysis of Selenium yeast with inorganic Se in broilers under heat stress. A total of 120 broilers chicks of one day were assigned to 4 groups each consisting 30 chicks fed on same basal diet but different selenium sources. The basal diet of group D1 was not supplemented with Se source (Negative control), group D2 basal diet was supplemented with inorganic selenium (Sodium selenite 0.22mg/Kg starter phase and 0.15mg/Kg finisher phase), group D3 basal diet was supplemented with commercially available organic selenium (Seleno-methionine 0.22mg/Kg starter phase and 0.15mg/Kg finisher phase) and group D4 basal diet was supplemented with self-developed organic selenium (Se-enriched yeast 0.22mg/Kg starter phase and 0.15mg/Kg finisher phase). The performance parameters i.e. feed intake (FI), live body weight (BW) and FCR were not significantly (p>0.05) effected by selenium supplementation in the starter phase but were significantly (p<0.05) effected in the finisher phase. Selenium supplementation significantly (p<0.05) effected serum Se level in different supplemented groups. Higher serum Se value (58.20±0.06) was recorded in D4 group. Similarly significantly lower selenium value was recorded for D4 and higher was recorded for D1 (11.36±0.08). However lower serum Paraoxonase (PON) value was recorded for D4 (13.24±0.01) and higher for D1 (13.33±0.03). Comparatively self-developed Se enriched yeast increased the Se accumulation and improved antioxidant system. Glutathione peroxidase (GPx) was found higher in D4 (12.333±0.03) followed by D3, D2 and D1 respectively. Whereas superoxide dismutase (SOD) was significantly lower (p<0.05) in D4 (0.1437±0.003) followed by D3 (0.1457±0.002). Selenium supplementation increased the [...].


A suplementação de probióticos enriquecidos com selênio é eficaz na redução do estresse oxidativo e na manutenção da estabilidade da qualidade da carne em frangos de corte, especialmente sob estresse por calor. Um estudo experimental foi conduzido para realizar uma análise comparativa da levedura selênio com o Se inorgânico em frangos de corte sob estresse térmico. Um total de 120 pintos de um dia foi dividido em 4 grupos, cada um consistindo de 30 pintos alimentados com a mesma dieta basal, mas com diferentes fontes de selênio. RESUMO

Tuberculosis is an universal health problem worldwide. In Iraq the problem is aggravated by drug resistance. In Silico studies usually pave the way for more investigations of the real problem .On the other hand Mycobacterium tuberculosis does not lend itself for deep wet lab studies, therefore, In Silico studies must precede many aspects of experimental work. In Silico studies were carried out using most virulent strain and a model of studies M. tuberculosis H37Rv to investigate some of the hypothetical proteins which compromised about 39% of the annotated proteins. The studied Rv hypothetical proteins were distributed among cellular compartment fractions with high existence in the cytoplasmic fraction (about 67%). Major function prediction of these proteins were found in cellular process section using different approaches of predictions .However , some of these proteins were still await to be included in the important databases such as COG and GO which concerned mainly with function annotation.

The Helicobacter pylori CagA gene is a major virulence factor that plays an important role in gastric pathologies. The size variation of CagA gene, which is dependent on the 3' repeat region, contains one or more Glu-Pro-Ile-Tyr-Ala [EPIYA] motifs and CagA multimerization [CM] motifs. Four segments flanking the EPIYA motifs, EPIYA A, B, C, or D, were reported to play a crucial role in the pathogenesis of H. pylori infection. The aim was to determine the roles of EPIYA segments and CM motifs in gastroduodenal pathogenesis in an Iraqi population. Gastric biopsies were collected from 210 patients with gastritis, duodenal ulcer [DU], gastric ulcer [GU], and gastric cancer [GC]. The EPIYA motif genotyping was determined by polymerase chain reaction and sequencing. The differences in age, gender, and CagA EPIYA motifs of H. pylori between GC, DU, GU and gastritis patients were analyzed using a 2-test. A total of 22 [45.8%] strains had three copies of EPIYA [ABC type], 2 [4.16%] had four copies [ABCC type], 6 [12.7%] had five copies [ABCCC type], 13 [27.08%] had two copies [AB type], 3 [6.25%] had the BC, and 2 [4.17%] had AC motif. The alignment of the deduced protein sequences confirmed that there were no East Asian type EPIYA-D sequences in Iraqi strains. A significant association was found between increase in number of EPIYA-C motifs and GU [P 0.01] compared with gastritis. The structure of the 3' region of the CagA gene in Iraqi strains was Western type with a variable number of EPIYA-C and CM motifs. A significant association was found between increase in number of EPIYA-C motifs and GU compared with gastritis indicating predictive association with the severity of the disease. The GenBank accession numbers for the partial CagA nucleotide sequences determined in this study are JX164093-JX164112

Periodontal disease and systemic diseases are inter-related, each influencing one another. Adequate knowledge on periodontal disease among medical doctors will enable them to refer their patients to dentists for timely management. This study aimed to assess knowledge, awareness and attitudes of medical doctors towards periodontal disease in Dar-es-Salaam, Tanzania. Cross sectional study on medical doctors working at Muhimbili National Hospital in Dar-es-Salaam, Tanzania. The study involved 151 medical doctors drawn randomly and who are practicing at Muhimbili National Hospital in Dar-es-Salaam, Tanzania. Data were obtained via a structured questionnaire and included demographic information's, knowledge regarding periodontal disease, their awareness and their attitude towards periodontal disease. They were entered into the SPSS statistical software for analysis. Frequencies and percentages were calculated and association between variables was done using the chi-square test. Of the 151 questionnaires administered, 124 were returned [response rate of 82.12%]. One hundred and eleven [89.5%] medical doctors believed poor oral hygiene was the main cause of periodontal disease, only 49[39.5%] told their patients to brush properly, 47[37.9%] believed that there was a bidirectional relationship between periodontal disease and systemic disease and only 35[28.2%] referred all patients with systemic diseases to dentists. 34[27.4%] believed that medical doctors have adequate knowledge regarding periodontal disease and 18[14.5%] believed that the medical curriculum provides enough knowledge regarding periodontal disease. Medical doctors had inadequate information about periodontal disease and this should be addressed in the medical curriculum so as to enable proper patient management

RESUMO

Perioperative blood transfusion is usually given to sickle cell disease patients to reduce or prevent perioperative morbidity. Assessment of such a practice was the subject of our study. A retrospective one year survey of sickle cell disease patients undergoing surgery at Salmaniya Medical Complex, Bahrain was conducted. The medical records were reviewed to characterize the surgical procedure, transfusion management and perioperative complications. 85 sickle cell disease patients who underwent surgery were studied. Preoperatively, 21.2% had exchange transfusion [ETX], 24.7% had simple transfusions [STX] and 54.1% had no transfusion [NTX]. 14.1% of all patients had postoperative complications, and 50% of those, had complications from the laparoscopic cholecystectomy group. The incidence of sickle cell crisis postoperatively was 22.2% in ETX group, 9.5% in STX group and 4.34% in the NTX group. The incidence of acute chest syndrome postoperatively was found to be 5.55% in the ETX group, 4.76% in the STX group and 4.34% in the NTX group. No intraoperative complications were recorded in all groups. All patients who had postoperative complications had a preoperative HBSS > 40%.Exchange transfusion does not prevent perioperative complications of sickle cell disease patients. HBSS > 40% carries a higher risk of postoperative complications

RESUMO

The effect of frequent subculturing, freezing and lyophilization in presence of cryoprotectives on antagonistic activity and viability of saccharomyces boulardii was studied. Results indicated that early subculturings led to increasing the antagonistic activity but this activity fell down gradually and diminished after 10 -11 of subculturing which indicated the cytoplasmic responsibility i.e., could be coded by plasmids or virus-like particles. Freezing at -20°C had no effect on the viability in the presence of cryoprotectives like skim milk powder, Mg-stearate or without them, since the cultures recovered at 99-99.2% after [4] days. Lyophilization had very overt effect on viability .Skim milk provided the best protection [8% reduction] followed by using distilled water and Mg-stearate .Saline had the greatest effect and caused [15% reduction]. Storage results of lyophilized samples at room temperature and refrigerator showed that skim milk was the best at both temperatures followed by distilled water, saline and then Mg-stearate which showed the highest reduction in viable count of yeast

RESUMO

Serum and synovial fluid levels of transforming growth factor beta [TGF-beta], interleukin-1 beta [IL-1 beta] and hyaluronan in osteoarthritis and rheumatoid arthritis were estimated. Twenty- three and twenty-two serum and synovial fluid [SF] samples were obtained from patients with osteoarthritis [OA] and rheumatoid arthritis [RA] and thirteen serum samples from age and sex matched controls. Serum TGF-beta, IL-1 beta and HA levels were significantly elevated in OA and RA patients compared with control group. The significant increase of these previous serum markers was observed in RA patients compared with OA patients, whereas no significant differences were obtained in SF TGF-beta levels between the two groups. On the other hand, SFHA levels were reduced significantly in OA and RA patients with high index radiological progression, whereas serum HA and SF IL-1 beta levels were elevated significantly in RA patients with high index radiological progression. In RA patients, SF IL-1 beta levels were also correlated significantly with the clinical variables. Concerning the biochemical markers interrelationship, SFHA levels were negatively correlated with SF IL-1 beta level and RA, respectively. Also, SFHA levels were correlated with SF TGF-beta-levels in OA and RA, respectively. Moreover, negative correlation was observed between serum HA and SFHA in RA patients
