Expression of Fas/FasL in PC12 cell apoptosis induced by ropivacaine / 临床麻醉学杂志

Objective To detect the expression of Fas and FasL in ropivacaine-induced rat phechromocytoma (PC12)cells apoptosis and the mechanism of its neurotoxicity.Methods PC12 cells were treated with different concentrations of ropivacaine (0.1,0.5,1,2,and 4 mmol/L)for 24 h to establish a cellular neurotoxicity model,and the cell viability were assessed by CCK-8.The cells were finally divided into 3 groups randomly:0.5 mmol/L group,2 mmol/L group and control group. After the cells were cultured for 24 h,morphological changes of cells were observed under optical mi-croscope (add the 1 mmol/L group),apoptosis was assessed by flow cytometer,the expression of Fas and FasL were assessed by immunofluorescence.Results Compared with the control group,the cell viability of 0.5 mmol/L and 2 mmol/L group decreased significantly (P<0.05),the cells exhibited obvious morphologic abnormalities (including the 1 mmol/L group),the apoptotic rate increased sig-nificantly (P<0.05),the expression of Fas and FasL increased significantly (P<0.05);Compared with 0.5 mmol/L group,the apoptotic rate and expression of Fas,FasL of 2 mmol/L group increased significantly (P<0.05).Conclusion Ropivacaine explosure induces apoptosis in PC12 cells,which might be related with the up-regulation of Fas/FasL.

Clinical therapy of Zisheng decoction recipe for chronic atrophic gastritis with intestinal metaplasia / 中国中药杂志

To explore the therapeutic effect and security of Zisheng decoction recipein treatment of the chronic atrophic gastritis (CAG) with intestinal metaplasia(IM). A total of 147 eligible cases were randomly divided into the traditional Chinese medicine group, Western medicine group and the combined group,47 cases in each group. Zisheng decoction recipe, famotidine, as well as Zisheng decoction recipe + famotidine were respectively given in the above three groups, with a treatment course of 30 d. The symptoms of traditional Chinese medicine, pathological score of gastric mucosa and the negative rate of Helicobacter pylori before and after treatment were observed in each group.The changes in pepsinogen Ⅰ (PGⅠ), pepsinogen Ⅱ (PGⅡ), gastrin-17 (GAS-17) and endothelin-1 (ET-1)were also detected to compare the efficient and safety indexes in the three groups. The combined group was better than the traditional Chinese medicine groupand the Western medicine group in total effective rate (P<0.05), pathological score of gastric mucosa and the negative rate of Helicobacter pylori, and serum indexes improvement (P<0.05). The improvement in TCM symptom score was more obvious in traditional Chinese medicine group and combined group than the Western medicine group (P<0.05). In the comparison ofincidence of complications,heart, liver and renal dysfunction, the traditional Chinese medicine group (2 case,4.8%)< the combined group (7 case,15.2%) <the Western medicine group (20 case,41.3%) (P<0.05). In the comparison of the blood and urine routine: the traditional Chinese medicine group (7 case,16.7%)< the combined group (14 case,30.4%) < the Western medicine group (24 case,52.2%) (P<0.05).The results showed that there was no significant difference in total effective rate between Zisheng decoction recipe and Western medicine in the treatment of CAG with IM, indicating Zisheng decoction recipe was effective; meanwhile, it had higher safety than famotidine. Zisheng decoction recipe combined with famotidine showed synergistic effect in the treatment of CAG with IM, can reverse the pathological changes of gastric mucosa in a certain extent, and the effect was better than that of Zisheng decoction recipe alone and famotidine alone.

Meta-analysis of laparoscopic versus open partial nephrectomy / 中华泌尿外科杂志

Objective To provide Meta-analysis evidence of laparoscopic partial nephrectomy (LPN) vs open partial nephrectomy (OPN) in assisting clinical decision making.Methods By searching CHKD,PUBMED,Wanfang and VIP database self-built library up to June 30,2012,both Chinese and English literatures of LPN and OPN efficacy in controlled study were included with strict exclusion criteria by two independent screenings of the literature.Data extraction and quality assessment were done by using the RevMan 5.1 META analysis software.Results A total of four English and six Chinese literature were included in this Meta-analysis.There were 1636 cases of partial nephrectomies.Of these patients,794 cases were treated with LPN,842 cases were treated with OPN.Meta analysis results showed that:in terms of operative time (SMD =0.10,95％ CI-O.40-0.59,P =0.70),surgical complication (OR =1.03,95％ CI0.73-1.44,P =0.88),positive surgical margin (OR =1.64,95 ％ CI0.83-3.23,P =0.16),warm ischemia time (SMD =1.07,95％ CI-0.02-2.16,P =0.05),postoperative tumor recurrence (OR =0.58,95％ CI 0.26-1.30,P =0.18),there was no significant difference.But in terms of intraoperative blood loss (SMD=-1.08,95％CI-1.57--0.59,P＜0.01),postoperative hospital stay (SMD=-0.81,95％CI-0.97--0.65,P ＜0.01),the differences were significant in favor of LPN.Conclusion Comparing with OPN,LPN has advantages in intraoperative blood loss and post-operative hospital stay,no obvious advantages in operative time,surgical complications,positive surgical margin,warm ischemia time and tumor recurrence.
