RESUMO

Objective: To research the effect of Fugan Huaxian Decoction on MAPK signaling pathway in rats with hepatic fibrosis (HF) and explore the mechanism of anti-HF. Methods: A total of 120 SD rats were randomly divided into control group, model group, colchicine group and Fugan Huaxian Decoction group (high, medium and low dose groups), with 20 rats in each group. The rats in the normal group were SD rats, and the rats model in the remaining groups were established into HF rats with syndrome of qi deficiency, poison weakness and blood stasis on the basis of the HF model induced by carbon tetrachloride (CCl4). Moreover, the rats were also received tail clamping, forced swimming, abnormal of starvation and full as well as rhubarb gavage. Liver pathology was performed on all rats after six weeks of modeling. After the validation model was successful, each group was given different doses of gavage, colchicine group (2 mg/kg), high, medium and low dose groups of Fugan Huaxian Decoction were intra-gastrically administered (67.08, 33.54, and 16.77 g/kg), normal group and model group were given pure water 2 mL/d for three weeks continuously. On the second day of last gavage, the serum levels of ALT, AST, ALB, LN, HyP and PIIINP were detected in each group. HE staining and Masson staining were performed on liver tissues. The expression of ERK1/2, JNK1/2 and P38-related protein and their phosphorylated protein in liver MAPK signaling pathway were detected by Western blotting. The results of quantitative analysis were expressed as p-ERK/ERK, p-JNK/JNK, p-p38/p38 ratio. Results: In model group, hepatic tissue cells were severely infiltrated and fibrotic, while, the degree of liver injury and fibrosis were significantly reduced in other groups. Compared with normal group, the serum levels of ALT, AST, Hyp, LN and PIIINP in model group were increased significantly, ALB was decreased significantly (P < 0.01), and the protein expression of p-JNK, p-ERK and p-p38 was increased significantly (P < 0.01). Compared with model group, Fugan Huaxian Decoction decreased serum ALT, AST, Hyp, LN and PIIINP levels of rats, increased ALB content and down-regulated the expression of phosphorylated protein of p38, ERK1/2 and JNK1/2, and it showed in a dose-dependent manner, the high dose group worked the best. Conclusion: The model of qi deficiency, poison weakness and blood stasis combined with HF model rats was successfully established. Fugan Huaxian Decoction not only protects hepatocytes, alleviates liver injury and inhibits HF, but also regulates the protein expressions of p-JNK, p-ERK, p-p38, as well as inhibits MAPK signaling pathway activation, which may be one of the mechanisms of its anti-HF function.

<p><b>OBJECTIVE</b>To detect the expressions of Ras and Sos1 proteins in human epithelial ovarian cancer (EOC) tissues and explore their correlation with the clinicopathological features of the patients.</p><p><b>METHODS</b>The expressions of Ras and Sos1 proteins were detected immunohistochemically in 62 EOC tissues, 5 borderline ovarian cancer tissues, 15 benign epithelial ovarian neoplasm tissues, and 18 normal ovarian tissues.</p><p><b>RESULTS</b>The EOC tissues showed significantly higher expression levels of both Ras and Sos1 than the other tissues tested (P<0.05). In EOC tissues, Ras and Sos1 proteins were expressed mostly on the cell membrane and in the cytoplasm. The expression level of Ras was correlated with pathological types of the tumor (P<0.05) and was the highest in serous cystadenomcarcinoma; Sos1 expression did not show significant correlation with the clinicopathological indexes of the patients. High expressions of both Ras and Sos1 proteins were associated with shorter progression-free survival of the patients, but this association was not statistically significant.</p><p><b>CONCLUSIONS</b>Ras and Sos1 protein may participate in in the occurrence and development of EOC. The tissue-specific variation of Ras expression can lend support to a specific diagnosis of ovarian serous adenocarcinoma. The association of Ras and Sos1 protein expression with the tumor-free survival time of the patients awaits further investigation with a larger sample size.</p>

<p><b>BACKGROUND</b>Diabetes mellitus has become epidemic in recent years in China. We investigated the prevalence of hyperglycaemia and inadequate glycaemic control among type 2 diabetic inpatients from ten university teaching hospitals in Guangdong Province, China.</p><p><b>METHODS</b>Inadequate glycaemic control in diabetic patients was defined as HbA1c = 6.5%. Therapeutic regimens included no-intervention, lifestyle only, oral antiglycemic agents (OA), insulin plus OA (insulin + OA), or insulin only. Antidiabetic managements included monotherapy, double therapy, triple or quadruple therapy.</p><p><b>RESULTS</b>Among 493 diabetic inpatients with known history, 75% had HbA1c = 6.5%. Inadequate glucose control rates were more frequently seen in patients on insulin + OA regimen (97%) than on OA regimen (71%) (P < 0.001), and more frequent in patients on combination therapy (81% - 96%) than monotherapy (75%) (P < 0.05). Patients on insulin differed significantly from patients on OA by mean HbA1c, glycemic control rate, diabetes duration, microvascular complications, and BMI (P < 0.01).</p><p><b>CONCLUSIONS</b>This study showed that glycaemic control of type 2 diabetic patients deteriorated for patients who received insulin and initiation time of insulin was usually delayed. It is up to clinicians to move from the traditional stepwise therapy to a more active and early combination antidiabetic therapy to provide better glucose control.</p>

Objective Abstract:Objective To inquire into the reason that chronic diabetic foot ulcer(CDF) was difficult to be cured by clarifying their interaction between interin-1?(IL-1?) and fibroblast proliferation,the activity and protein expression of matrix metalloproteinase-2(MMP-2),matrix metalloproteinase-9(MMP-9)in fibroblasts from CDF.Method Fibroblast collected from the edge area of the diabetic foot ulcer was cultured for 4-6 generations,and added in with different concentration of IL-1? at the 24 hr,48 hr and 72 hr,then the effect of IL-1? on the activity and expression of MMP-2 and MMP-9 in fibroblasts were observed by methods of zymography and Western blott.Results There was a significant difference of IL-1? between CDF and acute diabetic lesions(ADL);IL-1? in 0.5 ng/ml-5 ng/ml could promote fibroblast proliferation obviously,but weakened gradually with increasing of the IL-1?,but no affection on fibroblast proliferation when IL-1? was in 50 ng/ml,and inhabited fibroblast proliferation over 50 ng/ml of IL-1?;IL-1? in 50 ng/ml increased the activity and protein expression of MMP-2 and MMP-9 in fibroblasts from CDF. Conclusion The reason that CDF was difficult to be cured were related to significant increase of IL-1?.The increase of IL-1? not only inhibits the fibroblast proliferation,but also promotes the activity and protein expression of MMP-2 & MMP-9,and it results in the increase of extracellular matrix degradation.
