RESUMO

Objective@#This study aimed to evaluate the epidemiological, clinical and mycological characteristics of invasive candidiasis (IC) in China.@*Methods@#A ten-year retrospective study including 183 IC episodes was conducted in a tertiary hospital in Beijing, China.@*Results@#The overall incidence of IC from 2010-2019 was 0.261 episodes per 1,000 discharges. Candidemia (71.0%) was the major infective pattern; 70.3% of the patients tested positive for @*Conclusion@#The incidence of IC has declined in the recent five years.

RESUMO

OBJECTIVE: To investigate the clinicopathological features, prognosis and genes mutations of pancreatic gastrointestinal stromal tumors. METHODS: Four cases of pancreatic gastrointestinal stromal tumors were analyzed retrospectively. The cases were diagnosed in Affiliated Hospital of Southwest Medical University from May 2010 to October 2018. PubMed and main domestic databases were searched to retrieve the domestic and foreign literatures for literature review. RESULTS: Including our 4 cases, 59 cases were reported, with 30 males and 29 females. The most common initial symptoms were abdominal pain or discomfort(38 cases, 64.4%), abdominal mass(10 cases, 16.9%) or health examination finds(11 cases, 18.7%). A total of 53 cases were composed of complete imaging and histological type data. CT examination showed that the tumors were more uneven and low-density shadow, with solid-cystic(21 cases,39.7%), solid(20 cases, 37.7%) or cystic(12 cases, 22.6%). The most common histological types were spindle cell type(44 cases, 83.0%), mixed type(7 cases, 13.2%) or epithelioid cell type(2 cases, 3.8%). The rates of 5-year overall survival(OS) and disease-free survival(DFS) were 79.7% and 39.3% respectively. By univariate and multivariate analysis, the main indicator for adverse outcome was mitoses >5/50 HPF(P5/50 HPF is an independent influencing factor of prognosis.Usually, the size of tumor is large and risk is high. C-kit gene mutations are common.

<p><b>Background</b>The hedgehog signaling system (HHS) plays an important role in the regulation of cell proliferation and differentiation during the embryonic phases. However, little is known about the involvement of HHS in the malignant transformation of cells. This study aimed to detect the role of HHS in the malignant transformation of human bronchial epithelial (16HBE) cells.</p><p><b>Methods</b>In this study, two microfluidic chips were designed to investigate cigarette smoke extract (CSE)-induced malignant transformation of cells. Chip A contained a concentration gradient generator, while chip B had four cell chambers with a central channel. The 16HBE cells cultured in chip A were used to determine the optimal concentration of CSE for inducing malignant transformation. The 16HBE cells in chip B were cultured with 12.25% CSE (Group A), 12.25% CSE + 5 μmol/L cyclopamine (Group B), or normal complete medium as control for 8 months (Group C), to establish the in vitro lung inflammatory-cancer transformation model. The transformed cells were inoculated into 20 nude mice as cells alone (Group 1) or cells with cyclopamine (Group 2) for tumorigenesis testing. Expression of HHS proteins was detected by Western blot. Data were expressed as mean ± standard deviation. The t-test was used for paired samples, and the difference among groups was analyzed using a one-way analysis of variance.</p><p><b>Results</b>The optimal concentration of CSE was 12.25%. Expression of HHS proteins increased during the process of malignant transformation (Group B vs. Group A, F = 7.65, P < 0.05). After CSE exposure for 8 months, there were significant changes in cellular morphology, which allowed the transformed cells to grow into tumors in 40 days after being inoculated into nude mice. Cyclopamine could effectively depress the expression of HHS proteins (Group C vs. Group B, F = 6.47, P < 0.05) and prevent tumor growth in nude mice (Group 2 vs. Group 1, t = 31.59, P < 0.01).</p><p><b>Conclusions</b>The activity of HHS is upregulated during the CSE-induced malignant transformation of 16HBE cells. Cyclopamine can effectively depress expression of HHS proteins in vitro and prevent tumor growth of the transformed cells in vivo.</p>

<p><b>OBJECTIVE</b>To compare the effects and mechanism of blood enriching on mouse model of blood deficiency syndrome induced by cyclophosphamide of albiflorin and paeoniflorin.</p><p><b>METHOD</b>Albiflorin and paeoniflorin were determined by using animal models of blood deficiency syndrome induced by cyclophosphamide. The amount of WBC, RBC, HGB, index of thymus gland and spleen, and the changes of GM-CSF, IL-3 and TNF-α in serum were detected after the treatment.</p><p><b>RESULT</b>Compared with the model group, the amount of WBC in the group of 30 mg x kg(-1) albiflorin and 30 mg x kg(-1) paeoniflorin were increased obviously (P < 0.01). The amount of RBC in the group of 30 mg x kg(-1) albiflorin and 30 mg x kg(-1) paeoniflorin were increased obviously (P < 0.01, P < 0.001), which did not had a significant difference compared with the same dose. The index of thymus gland in the group of 30 mg x kg(-1) albiflorin was superior to the model group (P < 0.01), the difference was significant compared with the same dose of paeoniflorin (P < 0.05). The GM-CSF in serum in all groups of 30 mg x kg(-1) albiflorin, 15 mg x kg(-1) albiflorin, 30 mg x kg(-1) paeoniflorin and 15 mg x kg(-1) paeoniflorin increased obviously (P < 0.01, P < 0.05, P < 0.01, P < 0.05); The IL-3 in serum in both group of 30 mg x kg(-1) albiflorin and 30 mg x kg(-1) paeoniflorin also increased (P < 0.001). The content of TNF-α in group of 30 mg x kg(-1) albiflorin and 30 mg x kg(-1) paeoniflorin were reduced (P < 0.01), which showed the obvious difference compared with the same dose group (P < 0.01).</p><p><b>CONCLUSION</b>Albiflorin had the effect of blood enriching by regulating the immune function, same with the paeoniflorin. The probable mechanism of nourishing blood and liver of Paeoniae Radix Alba was not only the better effect of adjusting the content of TNF-α, but also might act synergistically with paeoniflorin.</p>

<p><b>OBJECTIVE</b>To discuss the clinical manifestations and CT features of acute moderate or severe hydrogen sulphide poisoning fishermen.</p><p><b>METHODS</b>The clinic and CT datas of 8 acute moderate or severe hydrogen sulphide poisoning cases were retrospectively analysed.</p><p><b>RESULTS</b>The lesions located at two pulmonary each leaf in 3 cases, located at both lower lungs in 3 cases and asymmetry leaves in 2 cases. Lesions form: little patchy shadow in 8 cases (8/8), fibrous band shadow in 4 cases (4/8), ground glass shadow in 3 cases (3/8), peripheral fuzzy interlobular nodule in 4 cases (4/8); 5 cases complicated with pleural effusion; After treatment 6 ∼ 10 days, the pulmonary shadows were absorbed obviously; Fibrous band shadow was residual in a case after a follow-up of 2 months.</p><p><b>CONCLUSION</b>With definite history of hydrogen sulphide inhalation and the corresponding clinical manifestations, and if fishermen contacted for a long time, who would be made chemical pulmonary damage; Combined with the imaging manifestations, the diagnosis as well as the severity of pulmonary damage could be made.</p>

<p><b>OBJECTIVE</b>To evaluate the feasibility of electroporation-mediated transfection of recombinant plasmid to mandibular distraction area of rabbit in vivo.</p><p><b>METHODS</b>New-Zeland rabbit were employed. The mandible was distracted 3 days after operation at a rate of 0.8 mm per day for 7 days. The rabbits were randomly divided into 3 groups as group A (recombinant plasmid pIRES-VEGF165-EGFP), group B (recombinant plasmid plRES-VEGF165-EGFP) and group C (normal saline). The rabbits were sacrified at 3 hours, 1, 3, 7 and 14 d after injection respectively. The tissue at the distraction area was taken out for frozen section. The gene expression was assessed by the detection of expression of green fluorescence protein (GFP) using fluorescence microscope. The liver and kidney function test (ALT, AST, BUN, Scr) and the histological examination of heart, liver and kidney were also performed.</p><p><b>RESULTS</b>GFP was seen in the distraction area in group A and group B 3 hours after injection, which increased at the 1st day, reached peak value at the 3rd day, decreased at the 7th day and was very lower at 14th day. The GFP expression was much stronger in group A than in group B. GFP was not expressed in group C. There was no statistical difference in the concentration of ALT, AST, BUN and Scr in serum of rabbits among the three groups.</p><p><b>CONCLUSIONS</b>Electroporation-mediated transfection of recombinant plasmid can be expressed in the distraction area of rabbits, and there was no toxicity to the liver and kidney of rabbits. Electroporation could obviously improve transfection efficiency in vivo. It indicates that electroporation-mediated transfection of recombinant plasmid to distraction area tissue of rabbits is feasible.</p>

<p><b>OBJECTIVE</b>To compare therapeutic effects of Tongdu Tiaojin needling method and routine acupuncture method on blepharoptosis.</p><p><b>METHODS</b>Fifty-eight cases of blepharoptosis were randomly divided into an observation group and a control group, 29 cases in each group. The observation group was treated by Tongdu Tiaojin needling method with Baihui (GV 20), Fengchi (GB 20), Houxi (SI 3), Yintang (EX-HN 3) selected, and the control group by routine acupuncture method with Cuanzhu (BL 2), Yangbai (GB 14), Sizhukong (TE 23) selected. They were treated for 3 courses and one course was constituted by 10 sessions.</p><p><b>RESULTS</b>The cured rate of 41.4% and the effective rate of 82.8% in the observation group were better than 24.1% and 48.2% in the control group (P<0.05); after treatment, the palpebral fissure widened and the level of the palpebra superior shading the cornea was alleviated in the two groups, but the observation group was superior to the control group (P<0.05).</p><p><b>CONCLUSION</b>Tongdu Tiaojin needling method has a definite therapeutic effect on blepharoptosis.</p>

OBJECTIVE@#To investigate the expression of hypoxia-inducible factor 1-alpha (HIF1-alpha) in the heart, lung, liver and kidney in rats died of two typical models of asphyxia.@*METHODS@#Two asphyxia models were made and tissue samples of the dead rats were collected from different groups at various postmortem duration. The expression and the changes of HIF1-alpha in various tissues were examined by immunohistochemistry and image analysis techniques. Results Significant expression of HIF1-alpha was observed in the myocardial fibers, kidney cells, liver cells and lung cells in both asphyxia models, but not in the control group. The expression of HIF1-alpha in various tissues in the rat died of nitrogen gas breathing was found in the nuclei at 0 hour and the expression level decreased gradually thereafter. The HIF1-alpha expression level and duration in various tissues of the rat died of hanging were higher and longer than that of the former group, with a peak of the expression level observed 6 hours after death, and then started to decline in all tissues except the heart where the expression still showed an increase 24 hours after death. The control groups showed a steady expression in the cytoplasm but not in the nuclei.@*CONCLUSION@#HIF1-alpha appears to be a valuable biomarker in the diagnosis of asphyxia within 24 hours after death.

Objective: To detect the expression of fragile histidine triad (FHIT) protein and aberrant methylation of its gene in lung carcinoma tissue and discuss their significance in tumorigenesis of lung cancer. Methods: The expression of FHIT protein was determined by immunohistochemical SP staining and its aberrant methylation was detected by methylation-specific PCR (MSP) in lung cancer and adjacent lung tissues (n = 60). The sequencing of the amplified products of MSP was performed. The 60 patients were followed up. Results: The expression of FHIT protein in adjacent lung tissues was obviously higher than that in lung cancer tissues. There was significant difference between them (76.7% vs 50%, P 0.05). The survival time was longer in FHIT-positive patients than FHIT-negative patients (P <0.05). The FHIT protein was the risk factor associated with the disease-free survival time of patients (P <0.01). Conclusion: The aberrant methylation of FHIT gene occurrs frequently and the expression of FHIT protein is significantly down-regulated in lung cancer. FHIT may play an important role in the tumorigenesis and development of lung cancer. FHIT protein could be used as an important factor for predicting the prognosis of patients.

<p><b>OBJECTIVE</b>To observe the changing contents of leukotriene B4 ( LTB4 ), leukotriene C4 ( LTC4 ), and leukotriene D4 (LTD4 ) of lung tissue in asthmatic rats, and explore the effect of Shuanglong Capsule (SLC) on it.</p><p><b>METHODS</b>SD rats were randomly divided into the nomal group, asthmatic model group, Dexamethasone group and the high, middle and low dose SLC groups. All rats except those in the normal group were sensitized by ovalbumin and challenged with the antigen, and the contents of LTB4, LTC4 and LTD4 in lung tissue of all the groups were measured by reverse phase-high performance liquid chromatography (RP-HPLC) and compared.</p><p><b>RESULTS</b>The levels of LTB4, LTC4, and LTD4 of asthmatic rats were significantly higher than those of rats in the normal group. Dexamethasone and SLC at the dose of 8. 27 g/kg or 4. 13 g/kg could significantly inhibit the production of leukotrienes of lung tissue in asthmatic rats (P <0.05).</p><p><b>CONCLUSION</b>SLC can significantly inhibit the formation of inflammatory medium LTs of lung tissue in asthmatic rats, it may be one of the key mechanisms of SLC in anti-asthma and anti-inflammatory action.</p>

Objective: To investigate the expression of fragile histidine triad (FHIT) protein in lung carcinoma tissues and its relationship with multidrug resistance protein 1 (MDR1), and to discuss the clinical significance of the changes of FHIT expression. Methods: The expression of FHIT protein was detected in the lung cancerous tissues and the adjacent tissues (n = 60) using S-P staining method. The expression of MDR1 in lung carcinoma tissues was also detected. The expression of FHIT was analyzed in lung cancerous tissues with different clinicopathological parameters and its relationship with MDR1 was analyzed at the same time. Results: The expression of FHIT protein in the adjacent tissues was obviously higher than that in the lung cancerous tissues(76.7% vs 50%, P<0.01). No correlation was found between FHIT expression and the age, sex, smoking, general type, TNM staging, histological type, and lymph node metastasis in the patients with lung cancer. The expression of MDR1 protein in lung cancerous tissues positive for FHIT was obviously higher than those negative for FHIT(83.3% vs 60%, P<0.01). Conclusion: The expression of FHIT protein in lung cancer is evidently down regulated and is correlated with the expression of MDR1. Potential multiple drug resistance should be considered in FHIT gene therapy.

OBJECTIVE@#To investigate the expression of HIF1-alpha in heart and lung tissue died from asphyxia.@*METHODS@#The rats model of asphyxia death was constructed by hanging, different asphyxia groups and control group sets were made according the postmortem time (0,2,6,24 h), immunohistochemistry and half-quantitative RT-PCR methods were used to investigate expression of HIF1-alpha and mRNA changes on heart and lung tissue.@*RESULTS@#The positive staining of HIF1-alpha could be observed in the myocardium and lung tissue. Significant differences were found between the groups of asphyxia and their corresponding control group. HIF1-alpha expression was found in all the asphyxia groups while it was only expressed in the control groups of 2 h, 6 h and 24 h. Nucleic positive staining could be detected in all the asphyxia groups but none was found in the control groups. RT-PCR showed that the expression of mRNA between 0 h asphyxia group and 0 h control group were equal in both cardic muscle and lung, but elevated expression in groups of 2,6,24h compared to their control groups.@*CONCLUSION@#The nuclear positive staining of HIF1-alpha in heart and lung can be a special character of suffocation death.
