RESUMO

Related substances in pharmaceutical formulations are associated with their safety, efficacy and stability. However, there is no overall study already published on the assessment of related substances in the Compound Ketoconazole and Clobetasol Propionate Cream. In this work, a reliable HPLC-TOF-MS qua-litative method was developed for the analysis of related substances in this preparation with a quick and easy extraction procedure. Besides the active pharmaceutical ingredients, two compounds named ke-toconazole impurity B′ optical isomer and ketoconazole impurity E were identified. Furthermore, a new HPLC method for qualitative and quantitative assessment on related substances and degradation pro-ducts, which were found in the stability test, was established and validated. The single standard to determine multi-components method was applied in the quantitative analysis, which was an effective way for reducing cost and improving accuracy. This study can provide a creative idea for routine analysis of quality control of the Compound Ketoconazole and Clobetasol Propionate Cream.

RESUMO

To investigate the effect of ethanol on the zero-stress state of rabbit artery, a rabbit animal model of blood ethanol concentration gradient was established by intragastric administration of ethanol (IGAE). The blood ethanol concentration of each group was measured. The zero-stress state characterized by opening angle was observed by an image processing system. The results revealed that the blood ethanol concentration of each group increased after IGAE. The magnitude of the blood ethanol concentration came to the peak at 1.5-2 h after IGAE. The opening angle decreased with the increase of the magnitude of blood ethanol concentration and the opening angle in each group. These findings suggest that the zero-stress state of the rabbit artery has close relationship with ethanol concentration.

RESUMO

The changes in biomechanics of rat thoracic aorta at different post mortem interval (PMI) (0-7 d) were observed when it was stretched in one direction. Some significant changes were disclosed by statistical analyses. Maximum stretch, maximum strain and maximum energy showed a significant relationship with PMI, the coefficients of determination being 0.822, 0.716 and 0.669 respectively, P < 0.001. The stretch experiment of thoracic aorta may be a novel method to determine PMI.

OBJECTIVE@#To study the correlation between quantity of the connexin43 express and ischemia time in rabbit.@*METHODS@#Left coronary arteries were ligated to create early myocardial ischemia during distinct time, HBFP staining was used in the ischemia spot and SP method was used to show the expression of connexin43 and the ischemia are were measured by the computer image analysis.@*RESULTS@#Negative correlation was observed between the connexin43 quantity and the ischemia time. The significant was difference.@*CONCLUSION@#With ischemia time raised, expression of connexin43 was descented.

AIM: To study the expression of c-fos mRNA in brain following moderate lateral fluid percussion brain injury in rats, and to observe the temporal patterns of its expression following percussion. METHODS: Male Sprague-Dawley rats were divided into normal control, sham operation control and injury group. The rats of injury group subjected to moderate lateral fluid percussion injury (0.2 mPa). The injury groups were then subdivided into 5 min, 15 min, 30 min, 1h, 2h groups according to the time elapsed after injury. The expression of c-fos mRNA was studied with reverse transcription polymerase chain reaction (RT-PCR) semi-quantitatively.RESULTS: At 5 min after percussion, the induction of c-fos mRNA was increased, and remained elevated up to 2 h after brain injury.CONCLUSION: The induction and expression of the c-fos mRNA in cortex and brain stem after fluid percussion brain injury were increased rapidly.

AIM: To study the expression of c-jun in brain stem following moderate lateral fluid percussion brain injury in rats, and to observe the temporal patterns of its expressions following percussion.METHODS: Male Sprague-Dawley rats were divided into normal control, sham operation control and injury groups. The rats of injury group subjected to moderate lateral fluid percussion injury (0.2 mPa), and then were subdivided into 5 min, 15 min, 30 min, 1 h, 2 h, 4 h, 8 h and 12 h groups according to the time elapsed after injury. The expression of c-jun was studied by immunohistochemistry and in situ hybridization. RESULTS: After percussion for 15 min, Jun positive neurons increased in brain stem progressively, and peaked at 12h. At 5min after percussion, the induction of c-jun mRNA was increased, and remained elevated up to 1h-2h after brain injury. CONCLUSION: The induction and expression of the c-jun in brain stem after fluid percussion brain injury were increased rapidly and lasted for a long time.

Enzyme-histochemical investigations on experimental incised injury of thirty rabbits fortiming of wound were reported.In peripheral areas of wounds,strong reaction of six kinds ofdehydratase(ACP.AKP,ATP.ANAE.?—GA,?—Gr).were demonstrated 1~2 hrs followingincision while no reaction of four kinds of dehydrogenase(LDH.SDH.NADH.?—GPDH)werefound.The activity of dehydratase are explained by the enhanced metabolism of fibrocytes and theaggregation of acute inflammatory cells.The author suggests that the positive enzyme histochemicalreactions indicate the vital reaction in the first two hours,and PTAH staining is helpful for thediagnosis of micro—exudation of fibrin in the early stage of injury
