RESUMO

Objective To observe the therapeutic effect of the combination of cinobufacini with DC chemo-therapy in the treatment of advanced stomach cancer .Methods 65 patients who were diagnosed advanced gastric cancer on the basis of histocytology and pathology results , were randomly divided into DC chemotherapy group (31 cases) and combined treatment group (34 cases).The clinical curative effect ,adverse reaction,quality of life ( KPS questionnaire ) and the easement rate of pain were analyzed .Results The two groups had no statistically significant differences in adverse reaction (t=-0.0047,P>0.05) and clinical curative effect (χ2 =0.077,P>0.05),and the clinical response rate of the combined treatment group (35%) was similar to the DC chemotherapy group (32%).Besides,the easement rate of pain in the combined treatment group increased to 71%,which in the DC chemotherapy group increased to 45%(χ2 =19.969,P<0.05).And the improvement rate of quality of life (QOL) in the combined treatment group was 82%,which of the DC chemotherapy group was 55%,the difference was statistically significant(χ2 =13.923,P<0.05).Conclusion Cinobufaconi combined with DC chemotherapy in the treatment of advanced stomach cancer has no significant effect on increase of short -term efficacy ,but it can improve quality of life and alleviate pain obviously .

Objective To quantitatively analyze the characteristics of a panel of murine anti-human papillomavirus(HPV)11 L1-derived virus-like particle( VLP ) monoclonal antibodies ( mAbs ) and establish the mAb-based methods for antigen quality analysis.Methods A panel of 22 murine anti-HPV11 mAbs were characterized in details with their isotype, and binding affinity, conformational sensitivity were examined quantitatively in the direct binding ELISA and Western blot.The hemagglutination inhibition activity of mAbs were identified using the hemagglutination inhibition assay and the pseudovirus ( PsV ) neutralization efficiency were examined quantitatively using the PsV-based neutralization assay.The type-specific, highly conformational sensitive and neutralizing mAbs were selected to be used in the sandwich ELISA assay.Results Based on the quantitative and semi-quantitative results, six type-specific, highly conformational sensitive and neutralizing mAbs (2A2, 4A1-3, 16G7, 14A6, 9C1 and 19C7) were identified.These mAbs, along with 10D6 were screened as the capture mAb or as the detection mAb in the sandwich ELISA.Conclusion The binding affinity, conformational sensitivity and neutralization efficiency of anti-HPV11 mAbs were characterized in details.A mAb-based sandwich ELISA assay (14A6:Ag:9C12-HRP) were developed, which could be used in the in vitro potency analysis of HPV11 VLP-based vaccine.

RESUMO

Salmonella is a pathogenic bacteria to human and animals ,which can cause seriously complication and death . Salmonella pathogenicity is from the reactions of SP‐1 and SP‐2 T3SS effector proteins .In this paper ,the functions of different T3ss effector proteins in different infection periods is reviewed to provide a reference for further understanding the pathogenesis mechanisms of Salmonella .

RESUMO

Objective To evaluate the clinical effects of combined therapy of Fuyuan Huoxue decoction and transcatheter hepatic arterial chemoembolization in the treatment of primary hepatic carcinoma. Methods 80 patients with primary hepatic carcinoma were randomly divided into a control group, treated by transcatheter hepatic arterial chemoembolization, and a treatment group, additionally treated by Fuyuan Huoxue decoction on the basis of the control group. By observing the change of gross tumor volume、tumor markers、clinical symptoms、Karnofsky Performance Status(KPS) score、quality of life and so on,compare the clinical effects and quality of life between the two groups. Results The effective rate of solid tumor was 47.50%and 35%in the treatment and the control group respectively, with no significant difference(χ2=-1.229, P>0.05);The total effect rate was 87.50%and 32.50%in the treatment and the control group respectively, with significant difference(χ2=-5.633, P0.05);The accumulated scores change of quality of life(QOL) has asignificant difference(χ2=-3.025, P<0.05) between the two groups after the treatment. Conclusion The combined therapy of Fuyuan Huoxue decoction and transcatheter hepatic arterial chemoembolization can alleviate the clinical symptoms, improve treatment effects and quality of life of patients with primary hepatic carcinoma.

AIM:To study the relationship between the genetic polymorphism of CYP3A5 gene and the blood concentration/dose(C/D) ratio of cyclosporine combined with calcium channel blockers(CCBs) in renal transplant patients.METHODS:Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) analysis was applied to determine the genotype of CYP3A5 in 69 renal transplant patients.The blood concentration/dose(C/D) ratio of cyclosporine was compared in patients coadministrated of CCBs with different genetic polymorphism of CYP3A5.RESULTS:The cases of genotype of CYP3A51/1,CYP3A51/3 and CYP3A53/3 were 8,23 and 38,respectively.The mutation rate of CYP3A53 in renal transplant patients was 71.74%.The CsA C/D ratio of patients of CYP3A51/1 and CYP3A51/3 were 1.73 and 2.43 times higher than that in patients of CYP3A53/3.The CsA C/D ratio of patients(CYP3A51/1,CYP3A51/3) combined with CCBs had been obviously elevated(19.90?7.66 vs 36.01?8.18,34.33?7.87 vs 46.16?9.63 ng?mL-1 per mg?kg-1;P
