RESUMO

Aplastic anemia (AA) is a Tlymphocyte-mediated autoimmune disease. The research on AA was focused on three areas, which were the pathogenesis of immune dysfunction, hematopoietic stem cell damage and abnormal hematopoietic microenvironment. In recent years, more attentions have been paid on abnormal immune mechanisms in the blood. There are complex intracellular cytokine and signal transduction pathway of pathogenesis in AA. And T-bet/IFN-γ signaling pathway plays an important role in development and progression of AA. This article aimed to review T-bet/IFN-γ signaling pathways in AA.

RESUMO

This study was aimed to investigate the effect of Bu-Shen Y i-Sui Sheng-Xue (BSYSSX) method on pro-liferation and differentiation mechanisms of hematopoietic progenitor cells. The rat models were established by 60Co-γrays and cyclophosphamide. Compound Chinese medicine was gavaged to rats of the normal control group, model group, stanozolol group, Yi-Sui Sheng-Xue (YSSX) group, Wen-Shen Sheng-Xue(WSSX) group and Zi-Shen Sheng-Xue (ZSSX) group. Then, serum of rat was prepared. Rat bone marrow cells were incubated with AA rats serum ac-counted for 20% and the number of hematopoietic progenitor cells colony-forming units (CFU) were counted. The level of GATA-1 and PU.1 mRNA in colony cells were detected with RT-PCR. The results showed that compared with the normal control group, the number of bone marrow cells, CFU-E, BFU-E, CFU-GM, as well as the expres-sion of GATA-1 and PU.1 mRNA in the model group decreased significantly (P< 0.01). Compared with the model group, the number of bone marrow cells, CFU-E, BFU-E, CFU-GM of each treatment group were significantly in-creased (P< 0.01). CFU-E and BFU-E of the ZSSX group were better than the YSSX group (P < 0.01). CFU-GM of the ZSSX group was better than the YSSX group and the WSSX group. The expression of GATA-1 and PU.1 mR-NA in each treatment group were significantly higher than the model group (P< 0.01). The expression of GATA-1 mRNA in the ZSSX group was better than the YSSZ group and WSSX group (P< 0.05). The expression of PU.1 mR-NA in the ZSSX group was higher than the YSSX group and WSSX group. It was concluded that BSYSSX method may increase the expression of GATA-1 and PU.1 mRNA in order to promote the proliferation and differentiation of bone marrow hematopoietic progenitor cells. The ZSSX method was better than the YSSX method and WSSX method.

OBJECTIVE: To investigate the clinical efficacy and safety of Yisui Shengxue Granule (YSSXG), a compound traditional Chinese herbal medicine for reinforcing kidney and nourishing blood, in treating hemoglobin H (HbH) disease. METHODS: YSSXG was given orally to 25 patients with HbH disease in Guangxi Zhuang Autonomous Region (high incidence area for HbH disease in China) for 3 months as one therapeutic course, 3 times a day, 10 g YSSXG was given each time (dose of YSSXG for children should be reduced properly), and blood transfusion was not given to HbH patients during the course of treatment. The levels of hemoglobin (Hb), red blood cell (RBC), HbH and reticulocyte (Ret) were observed before and after YSSXG treatment, and side effects were observed during the course of treatment. Meanwhile, the genotype was examined, and the clinical efficacy of YSSXG in treating HbH patients with different genotype was evaluated. RESULTS: The levels of Hb, RBC and Ret were obviously increased after YSSXG treatment from the first month to the end of treatment (P<0.01). After YSSXG treatment, the levels of Hb, RBC, Ret in 12 HbH patients with gene deletion were elevated (P<0.05, P<0.01), and the levels of Hb and Ret in 13 HbH patients with gene non-deletion were increased obviously (P<0.05, P<0.01). The total response rate was 84% after 3-month treatment, and there was no statistical difference in clinical efficacy between gene deletion HbH patients and non-deletion HbH patients. No adverse effect was observed during the course of treatment. CONCLUSION: YSSXG is effective and safe for treatment of HbH disease. YSSXG can improve the levels of Hb, RBC and Ret in HbH patients, especially in gene deletion HbH patients.

OBJECTIVE: To investigate the efficacy and safety of Yisui Shengxue Granule (YSSXG), a compound traditional Chinese herbal medicine, in treating beta-thalassemia. METHODS: A randomized single-blinded trial was designed. Sixty patients with beta-thalassemia were divided into two groups: 30 patients in YSSXG-treated group and 30 in placebo parallel-control group. The patients in the two groups were assigned to receive either YSSXG or placebo for three months. The patients' symptoms and their blood indexes such as hemoglobin (Hb), red blood cell (RBC), reticulocytes (Ret) and fetal hemoglobin (HBF) were examined before and after the treatment. Meanwhile, the liver and spleen were examined with B-mode ultrasound. RESULTS: In the YSSXG-treated group, the blood indexes (Hb, RBC, Ret and HBF) and the symptoms of the patients were improved after three-month treatment, with statistical significance compared to those before treatment (P0.05). CONCLUSION: YSSXG demonstrates obvious clinical efficacy and no adverse reactions in treating beta-thalassemia.

Objective To investigate the clinical usage of Traditional Chinese Medicine Theory "kidney produce marrow,marrow produce blood,marrow and blood originate from kidney" in aging delay.Methods Several animal models(including radiation damaged,hemolytic anemia,Myrelan induced haematopoietic impedient anemia,emasculated rooster,emasculated rat,aged rat,glucose induced senescence rat/mouse,ozone induced aged mouse,aged Golden hamster,aged partridge and the fifth instar larva of silkworm etc)were applied to study the mechanism of the aging delay,and "Bushenshengxue" medicine were also used in treating ?-thalassemia patients.Results Significantly results were observed both in aging delay study guided by Traditional Chinese Medicine Theory "kidney produce marrow,marrow produce blood,marrow and blood originate from kidney" and in ?-thalassemia clinical study.The results indicated the science connotation of this theory.Conclusion The Traditional Chinese Medicine Theory "kidney produce marrow,marrow produce blood,marrow and blood originate from kidney" is the main basic mechanism of aging delay and single gene heredity disease treating with TCM.

Objective To investigate the survival situation of thalassemia cases treated with Bushen Yisui Method. Methods The survival situation of 197 cases were investigated by telephone. Results The total survival rate of 197 cases is 87.3%, among which the thalassemia major is 65.71%, the thalassemia intermedia is 99.2%, the ?-thalassemia is 100%, the ?-thalassemia is 85.0%. 158 of 197 cases have been analyzed genotype, among which 63.16% is the survival rate of homozygote, 89.16% of the heterogenic duplex heterozygote, 100% of the heterozygote. RESUMO

Objective To study the clinical effect of compound Yisui Shengxue Keli(YSSXKL) on treating ?-thalassemia. Methods By using self-control study method,30 cases of major and intermediate ?-thalassemia had been first treated with placebo for three months,then were treated with YSSXKL in the followed three months in Guangxi province,the high incidence area of the disease. The clinical symptoms,traditional Chinese medicine symptoms and dynamic changes of the blood levels of Hb,RBC,Ret and HbF were observed before and after each treatment. The ingredients of compound "Baxianjiangtang" are Radix Rehmanniae, Radix Polygoni Multiflori preparate, Rodix Astragali, Radix Ophiopogonis etc. Pharmacological tests showed that it can decrease LPO and increase the activity of SOD in blood and liver of Alloxan induced diabetic rats and mice.It also increased that phagocytosis of phagocytes of experimental anim als. It can increased tolerance to fatigue at low and high temperature conditions. According tothe above results, it can be considered as safe and effective.
