RESUMO

The study aimed to achieve enhanced targeted cytotoxicity and cell-internalization of cisplatin-loaded deoxyribonucleic acid-nanothread (CPT-DNA-NT),mediated by scavenger receptors into HeLa cells.DNA-NT was developed with stiff-topology utilizing circular-scaffold to encapsulate CPT.Atomic force microscopy (AFM) characterization of the DNA-NT showed uniformity in the structure with a diameter of 50-150 nm and length of 300-600 nm.The successful fabrication of the DNA-NT was confirmed through native-polyacrylamide gel electrophoresis analysis,as large the molecular-weight (polymeric) DNA-NT did not split into constituting strands under applied current and voltage. The results of cell viability confirmed that blank DNA-NT had the least cytotoxicity at the highest concentration (512 nM) with a viability of 92% as evidence of its biocompatibility for drug delivery.MTT assay showed superior cyto-toxicity of CPT-DNA-NT than that of the free CPT due to the depot release of CPT after DNA-NT inter-nalization.The DNA-NT exhibited targeted cell internalizations with the controlled intracellular release of CPT (from DNA-NT),as illustrated in confocal images. Therefore,in vitro cytotoxicity assessment through flow cytometry showed enhanced apoptosis (72.7%) with CPT-DNA-NT (compared to free CPT;64.4%).CPT-DNA-NT,being poly-anionic,showed enhanced endocytosis via scavenger receptors.

RESUMO

The pandemic of coronavirus disease 2019 (COVID-19) has become a major public health threat to the whole world. Although the control of COVID-19 has been in the forefront of interventional practice, most interventional radiologists (IRs) are not equipped adequately to cope with such a crisis. In this review, we share our experience from Chinese IRs' perspective, report on the acute measures instituted within interventional radiology (IR) units, and give recommendations to the prevention and control of COVID-19

RESUMO

Objective: To explore relation of pain threshold and effect of labor analgesia with Anxiety in Nulliparous. Methods: The psychosis according to Hamilton Anxiety Scale(HAS) and Hamihon Depression Scale(HDS) were evaluated in 50 primiparae requiring epidural labor analgesia. The cases were divided into two groups the experiment group subdivided into exp I(24 cases with mild anxiety) and exp H(9 cases with moderate to severe anxiety) and the control group(19 cases without anxiety) There was no Nulliparous with depression(HDS, scores<8) The effect of analgesia the labor course the mod e of delivery the plasma concentrations of epinephrine (E), norepinephrine(NE) and β-EP in the two groups were observed Results: The rate of anxiety in 50 nulliparous was 66 In the experimental group the epidural labor analgesia was weak the degree of nulliparous anxiety was less the latent time prolonged the E level elevated and NE levels lowered as compared with the control(P<0 01), the changes of PPT was significantly negatively correlated to the HAS score(P<0 005) Conclusion: The degree of nulliparous anxiety descrease the pain threshold and influences the effect of the epidural labor analgesia Therefore it is necessary to estimate fully the degree of nulliparous anxiety before delivery and to take individual and to improve the obstetrics quality.

RESUMO

OBJECTIVETo study the clinical significance of piggy-back liver transplantation in treating acute liver failure (ALF).

METHODSFifteen ALF patients (13 caused by HBV and 2 with acute Wilson disease) had piggy-back liver transplantations (PBLT) in our hospital from Sept 1999 to Feb 2006. The outcomes of these patients were retrospectively analyzed.

RESULTSOne year survival rate of the 15 patients was 87% (13/15). Excellent outcome was achieved in the 2 acute Wilson disease cases: their corneal Kayser-Fleischer rings disappeared and serum ceruloplasmin levels returned to normal. Among the 15 cases, one died of severe pulmonary infection and another died of multiple organ system failure on the 6th and 11th postoperative days. HBsAg positivity was observed in 13 cases before liver transplantation. Eleven patients survived and later received anti-HBV treatment recommended by the American Association for the Study of Liver Diseases. Their HBsAg became negative.

CONCLUSIONLiver transplantation is an effective therapy for ALF and can improve survival rate significantly.
