Introduction: the persistence of microorganisms in root canals is one of the main factors of endodontic failure. Therefore, the importance of knowing the antimicrobial properties of the different types of sealants. Objective: to make a comparison based on the available evidence on the antimicrobial activity of the different endodontics sealers. Material and methods: the information was collected from PubMed and Google Academic databases in English and Spanish, published in the period 2014-2023. Results: a large number of microorganisms are present in the different types of infections of endodontic origin, more than 500 microbiological species have been identified, including bacteria, fungi, archaea and viruses. Sealer cements can be classified according to their chemical composition, into cements based on zinc oxide-eugenol, calcium hydroxide, based on glass ionomer, silicone, resin and bioceramics. Conclusion: sealer cements that showed the highest antimicrobial activity against persistent microorganisms were zinc oxide-eugenol, resin, and bioceramic-based cements. However, it was identified that each author used different methods and times, therefore, it is not possible to accurately define which sealer cement has the best antimicrobial capacity (AU)

RESUMO

Abstract The study of biologically active substances-secondary metabolites of plants that exhibit geroprotective properties is an actual and popular direction in medicine to prevent early aging. This work aims to select the cultivation parameters for obtaining in vitro cell cultures of meadowsweet containing the largest amount of biologically active substances (BAS) for their further extraction as candidate substances for geroprotectors. To specify the effectiveness of the selected cell culture cultivation parameters, biomass growth for callus and root cultures, growth index, specific growth rate, and viability for suspension cultures was carried out. The study results made it possible to select the nutrient media for the cultivation of cell cultures of meadowsweet. It has been found that the greater the antioxidant activity of the extracts, the greater the antimicrobial properties it exhibits. In this study, cell cultures in vitro and alcohol extracts from the plant Filipendula ulmaria were considered as raw materials rich in candidate substances for geroprotectors. According to the data obtained, the plant is rich in hydroxybenzoic and salicylic acids, spireoside, avicularin, and hyperoside.

Abstract In order to ensure the timely and uninterrupted supply of medicinal plant raw materials, the methods of cultivation of plant cell cultures, namely, the production of plant root cultures, are relevant. In this paper, the geroprotective potential of Hedysarum neglectum Ledeb and Panax ginseng C. A. Mey root cultures is studied. They were cultured under in vitro conditions by transforming the rhizome (H. neglectum) and seed seedlings (P. ginseng) with Agrobacterium rhizogenes 15834 Swiss. To identify the geroprotective potential, the antimicrobial disc-diffusion method and the antioxidant activity were analyzed by titration of KMnO4 extracts of plant root cultures. The qualitative and quantitative composition was analyzed using high-performance liquid chromatography, thin-layer chromatography, and gas chromatography with mass spectrometry. In the course of the work, the presence of antimicrobial and antioxidant activity of plant root culture extracts was established. Biologically active substances contained in extracts of Hedysarum neglectum Ledeb root crops and Panax ginseng C. A. Mey are characterized by geroprotective potential, so they can act as a source of natural antioxidants in the functional nutrition of the geroprotective orientation.

RESUMO

Introduction: Intracanal medication with antibiotics is used to ensure the success of treatments. However, no studies evaluating the efficacy of triantibiotic paste after several hours of preparation have been reported. Objective: To evaluate the antimicrobial activity of the triantibiotic paste against Enterococcus faecalis, according to the time of application and storage of the components used for its preparation. Methods: An experimental in vitro study was carried out in the microbiology laboratory of Universidad Nacional Mayor de San Marcos. The sample consisted of three colonies of Enterococcus faecalis, formed in bile-esculin agar. On three specific days (0, 14 and 28), the antimicrobial activity of the conventional (ciprofloxacin/metronidazole/minocycline) and modified (cefaclor/metronidazole/minocycline) paste was evaluated, measuring (mm) the inhibition zones. The freshly obtained components were used to prepare the paste on day 0, and the stored components (powdered antibiotics kept in amber glass bottles at room temperature) were used on days 14 and 28. Two interventions were performed on each specific day (morning and afternoon). Freshly prepared pastes were used in the morning (immediate application), while pastes stored for 6 hours (delayed application) were used in the afternoon. Results: On day 0, it was found that the modified triantibiotic paste of immediate application had higher antimicrobial activity than the one of delayed application (p = 0.046). On day 28, the conventional triantibiotic paste for immediate application showed higher antimicrobial activity than that for delayed application (p = 0.049). Pasta prepared with fresh components (day 0) had higher antimicrobial activity than pasta prepared with components stored for 14 and 28 days. Conclusions: The application time of the triantibiotic paste and the storage times of the components could influence the antimicrobial activity for the eradication of Enterococcus faecalis(AU)

RESUMO

Green nanotechnology, the science that utilizes various plant resources for the synthesis of nanoparticles without posing any chemical hazard has proved to be highly efficient and environment friendly technique. This opens up options for the synthesis of novel nanoparticles with desirable characteristics required for various application viz., biosensors, biomedicine, cosmetics, nanobiotechnology, as antimicrobials, electronics, sensing etc. In this context, here, we have made an attempt for cost effective and eco-friendly synthesis of silver nanoparticles (AgNPs) using the extract of medicinally important plant Justicia neesii Ramamoorthy. The phytochemical analysis of the extract exhibited the presence of glycosides, flavonoids, lignins, phenols, phytosterols, reducing sugars, saponins, etc. The absorbance peak of the biofabricated nanoparticles at 425 nm as indicated by UV-Vis spectrophotometer broadens with increase in time indicating their poly dispersity nature and particle size analyzer revealed the average size to be in the range of 20-45 nm. The antioxidant and antimicrobial activity of the synthesized AgNPs demonstrated promising results. The kinetics of in vitro drug release profile of the drug loaded AgNPs was carried out and the data obtained was correlated with various mathematical models. The drug release from AgNPs at both the pH’s shows good fit to the First order model which is obvious from the high values of coefficient of correlation which logically means that the release of drug from AgNPs is dependent on the concentration present within the nanoparticles.

Staphylococcus aureus and Staphylococcus epidermidis are the primary etiological agents of bacterial conjunctivitis which are empirically treated with antibiotics. This results in an increase in antimicrobial resistance due to repeated exposure. Currently, natural treatment alternatives are being sought for self-limited bacterial infections of the conjunctiva. Objective: To determine the antimicrobial activity of eight extracts from Colombian plants against bacteria isolated from patients with bacterial conjunctivitis. Materials and methods: Samples were taken from 15 patients with bacterial conjunctivitis which were grown on blood and chocolate agar for 24 h at 37 °C. These samples were identified by the vitek automated system and antimicrobial susceptibility tests by the Kirby Bauer method. Each isolate identified with the genus Staphylococcus was evaluated for susceptibility to the following eight plant extracts of seven plant: Ocimum basilicum (basil), Sambucus nigra L. (elderberry), Delphinium elatum(belladonna), Calendula officinalis (marigold), Bixa orellana (annatto) (aerial part and independent fruit), Clinopodium brownei (pennyroyal), and Laurus nobilis (laurel), with traditional use previously reported for treating eye infections. Results: The most frequently isolated bacteria were S. epidermidis, S. hominis, and S. aureus, which exhibited antimicrobial resistance mainly to oxacillin, tetracyclines, and erythromycin. All isolates were inhibited by O. basilicum extracts (mic > 0.9 mg/mL) and L. nobilis (mic < 15 mg/mL). Conclusion: The extracts of C. officinalis y D. elatum showed antimicrobial activity only against isolates with higher antimicrobial sensitivity. Ethanolic extracts of O. basilicum y L. nobilis can be used as an alternative treatment for infections of the anterior segment of the eye.

RESUMO

In this project, we employed ethanolic (EMI) and aqueous (AMI) extracts of mango (Mangifera indica L., Anacardiaceae) fruit seeds as a modulator of antibiotic resistance against multidrug-resistant (MDR) Pseudomonas aeruginosa and Acinetobacter baumannii to evaluate natural compounds isolated from by-products or waste of edible plants. We also investigated the effect of these extracts alone and in combination with standard classes of antibiotics in the desired strains. M. indica seeds were processed and exploited using ethanol and water. The minimum inhibitory concentrations (MICs) of clinical isolates were examined against EMI and AMI extracts, followed by seven antibiotics of ceftazidime, ciprofloxacin, penicillin, amikacin, meropenem, ampicillin, and colistin. The checkerboard method evaluated the synergistic action between mango kernel extract (EMI) and seven antibiotics. EMI extract significantly revealed antimicrobial properties against MDR A. baumannii and P. aeruginosa with synergistic effects with the applied antibiotics. The considerable antibacterial efficacy of ethanolic extract of M. indica seeds can have great curative value as antibacterial drugs against infections caused by MDR P.aeruginosa and A. baumannii.

RESUMO

The current study aimed to detect the antimicrobial effect of cell free supernatant (CFS) of Lactobacillus plantarum strain against Gram +ve and Gram ve bacterial strains. The strain of Lactobacillus plantarum was isolated using selective media MRS agar. The strain was characterized on the basis of the gram staining, colony morphology, the biochemical tests and the DNA sequencing based method of 16S ribotyping. A total of four test strains (The three already isolated and reported strains (E.coli, S. aureus and B. subtilis) and the one recently identified novel strain (B. cereus) were used for the analysis of antagonistic activity of bacteriocin produced by L. plantarum strain. The CFS of L. plantarum showed zone of inhibition against all the test strains (Gram +ve and Gram ve bacteria). The conditions favoring the growth of bacteria were associated with the antimicrobial efficacy of CFS. Bacteriocin activity of CFS remained effective after exposure to temperature stress. Wide range of antagonistic potential of CFS of L. plantarum provides an alternative for antibiotics in pharmaceutical industry. Heat resistant feature of bacteriocin suggests its application in food industry.

Secondary metabolites produced by endophytes are an excellent source of biologically active compounds. The newly isolated natural products terezine E and 14-hydroxyterezine D are endophytic metabolites exhibiting anticancer activity recently identified by our team (https://doi.org/10.1080/14786419.2018.1489393). In our current study, we evaluated their affinity for binding to the active site of histone deacetylase (PDB ID: 4CBT) and matrix metalloproteinase 9 (PDB ID: 4H3X) by molecular docking using AutoDock Vina software after having tested their cytotoxic activities on three cell lines (human ductal breast epithelial tumor cells (T47D)-HCC1937), human hepatocarcinoma cell line (HepG2)-HB8065), and human colorectal carcinoma cells (HCT-116)-TCP1006, purchased from ATCC, USA)). Additionally, their antimicrobial activities were investigated, and their minimum inhibitory concentration (MIC) values were determined against P. notatum and S. aureus by the broth microdilution method. Higher cytotoxicity was observed for terezine E against all tested cell lines compared to 14-hydroxyterezine D. Molecular docking results supported the high cytotoxicity of terezine E and showed higher binding affinity with 4CBT with an energy score of 9 kcal/mol. Terezine E showed higher antibacterial and antifungal activities than 14-hydroxyrerezine D: MIC values were 15.45 and 21.73 µg/mL against S. aureus and 8.61 and 11.54 µg/mL against P. notatum, respectively.

RESUMO

Abstract Present study analysed the therapeutic potential of traditionally acclaimed medicinal herb Nanorrhinum ramosissimum, using plant parts extracted with different solvents (10 mg/mL). Shoot extracts exhibited comparatively better antimicrobial properties, in comparison to root extracts. Total phenolic content was estimated, to ascertain its dependency on antioxidant properties of plant extracts. Antioxidant assay revealed promising results in comparison to IC50 value of standard ascorbic acid (52.2±0.07 µg/mL), for methanolic extracts of shoot (61.07±0.53 µg/mL and 64.33±0.33 µg/mL) and root (76.705±0.12 µg/mL and 89.73±0.28 µg/ mL) for in vivo and in vitro regenerants respectively. Correlation coefficient R2 values ranged between 0.90-0.95, indicating a positive correlation between phenolic contents and antioxidant activity. Plant extracts were also able to inhibit DNA oxidative damage again indicating their antioxidative potential. Antidiabetic potential was confirmed by alpha amylase inhibition assay where shoot methanolic extracts (invivo, in vitro) exhibited the best IC50 values (54.42±0.16 µg/mL, 66.09±0.12 µg/mL) in comparison to standard metformin (41.92±0.08 µg/mL). Ethanolic extracts of roots (in vitro, invivo) exhibited the relative IC50 values (88.97±0.32µg/mL,96.63±0.44 µg/mL) indicating that shoot parts had a better alpha amylase inhibition property; thus proving the herb's bioactive potential and its prospective therapeutic source for curing various ailments.

RESUMO

Human RNase A (human RNase A) superfamily contains 13 members (RNase 1~RNase 13) with different biological activities. In addition to the conserved catalytic sequence, its protein structure also has a significant diversity of sequences, which determines that human ribonuclease A can exert biological functions other than ribonuclease activity. Human RNase A superfamily members are expressed in various immune cells such as eosinophils, neutrophils, monocytes, and macrophages and can be secreted to exert various biological functions, including anti-microbial activity, promote host defense, participate in angiogenesis and sperm maturation, etc. Some members of the human ribonuclease A superfamily can exert antimicrobial and antiparasitic activities through direct actions such as hydrolysis of viral RNA, inhibition of viral replication, destruction of bacterial cell walls, promotion of microbial aggregation, damage to parasitic cell membranes and mitochondrial membranes, and indirect actions mediated by host innate immune cells, thereby participating in host defense. This article reviews the antimicrobial (including viruses, bacteria, and fungi) and antiparasitic activities of human ribonuclease A and their mechanisms of action. It also prospects the prospects of human ribonuclease A as an active antimicrobial substance and innate immune molecule for the treatment of severe and drug-resistant microbial infections.

RESUMO

@#The present study was conducted to investigate the antimicrobial potential of essential oils of Curcuma longa and Syzygium aromaticum against multidrug-resistant pathogenic bacteria. Four identified bacterial isolates including Methicillin-resistant Staphylococcus aureus (MRSA), Escherichia coli, Klebsiella pneumoniae, and Acinetobacter baumannii were selected and their antibiotic sensitivity was checked by disc diffusion assay. C. longa and S. aromaticum were subjected to steam distillation to obtain their essential oils. The crude essential oils were fractioned by employing column chromatography. Crude essential oils and their fractions were evaluated for their antibacterial activity by agar well diffusion assay and minimum inhibitory concentrations were calculated. All the selected bacterial isolates showed resistance to three or more than three antibiotic groups and were declared as multidrugresistant (MDRs). Crude essential oils of C. longa and S. aromaticum exhibited antimicrobial activity against all selected isolates but S. aromaticum activity was better than the C. longa with a maximum 19.3±1.50 mm zone of inhibition against A. baumannii at 1.04 µL/mL MIC. GC/MS analysis revealed the abundance of components including eugenol, eugenyl acetate, b- caryophyllene, and a- Humulene in both crude oil and fractions of S. aromaticum. While the main components of C. longa essential oil were Ar-tumerone, a–tumerone, b- Tumerone, I-Phellandrene, a-zingibirene, b- sesquiphellandrene, and p- Cymene. This study highlights that plant-based essential oils could be a promising alternative to antibiotics for which pathogens have developed resistance. C. longa and S. aromaticum carry compounds that have antimicrobial potential against multiple drug-resistant bacteria including MRSA. E. coli, K. pneumoniae and A. baumannii.

RESUMO

This study aimed to evaluate methods for studying the in vitro antimicrobial activity of lactic acid bacteria (LAB) against Brucella abortus and to evaluate the antagonistic effect of LAB on the viability of this pathogen. A total of 18 LAB strains (Lactobacillus plantarum, n = 11; Pediococcus acidilactici, n = 1; Lactobacillus rhamnosus, n = 4; and Lactobacillus brevis,n = 2), isolated from Minas artisanal cheeses produced in three regions (Canastra, Campos das Vertentes, and Araxá) of Minas Gerais State, Brazil, were tested for their antimicrobial activity against B. abortus using three methods: spot-on-lawn, agar well diffusion assay, and antagonistic activity of the culture supernatants. None of the tested LAB strains could inhibit B. abortus in the spot-on-lawn and agar-well diffusion assays. The supernatants produced by LAB had an acidic pH, with intensity depending on bacterial growth and strain, and could inhibit the growth of B. abortus. In contrast, pH-neutralized (pH 7.0) LAB supernatants did not suppress the growth of B. abortus. The results showed that the best technique to study the in vitro antagonism of LAB against B. abortus was the antagonistic activity of culture supernatants. The growth of B. abortus may have been inhibited by acid production.(AU)
