RESUMO

Industrialization has added extremely toxic metalloid arsenic into the environment which at high concentration severely threatens the biota. Naturally, some microbes possess the ability to bio-accumulate metals and also to transform arsenite (As III) a toxic form to a non-toxic arsenate As V. The present study aimed to isolate arsenic resistant bacterias from the arsenic contaminated soil and water. Among eleven bacterial isolates, three FAs 1, 4 and 9 exhibited tolerance against sodium arsenite at 100mM concentration by achieving growth of 7.48×109,1.57×109 and 2.23×109 C.F.U./ml, respectively. Optimization at differentconditions such as temperature, pH and arsenic concentration revealed high arsenic tolerance from isolate FAs 4 (5.33×108) at 37°C and FAs 1 (4.43×108 C.F.U./ml) at pH 7. Arsenic resistance at optimum conditions for the bacterial strains FAs 1, FAs 4 and FAs 9 showed maximum growth at 80mM concentration of arsenite. These bacterial isolates did not show redox ability to oxidize arsenite As III to arsenate As V. However bacterial isolates FAs 1, FAs 4 and FAs 9 were able to accumulate arsenic 39.16, 148 and 125 µ g/L on the 4th, 3rd and 5thday of incubation, respectively. The isolates FAs 1, FAs 4 and FAs 9 were identified as Gram negative non endospore forming rods. In future, these novel isolates possess a great potential in biotechnology field, as bioremediation of arsenic contaminated soil and water can be done by employing arsenic accumulating bacteria which is an eco-friendly and cost effective method.


A industrialização adicionou arsênico metalóide extremamente tóxico ao ambiente que, em alta concentração, ameaça severamente a biota. Naturalmente, alguns micróbios possuem a capacidade de bio-acumular metais e também transformar arsenito (As III) uma forma tóxica a um arsenato não-tóxico Como V. O presente estudo visa o isolamento de bactérias resistentes ao arsênico do solo contaminado com arsênico e água . Entre onze isolados bacterianos, três FAs 1, 4 e 9 exibiram tolerância à concentração de 100 mM de arseatura de sódio, obtendo crescimento de 7,48 × 109,157 × 109 e 2,23 × 109 C.F.U./ml, respectivamente. Otimização em diferentes condições como temperatura, pH e concentração de arsênio revelaram alta tolerância ao arsênico do isolado FAs 4 (5,33 × 108) a 37 ° C e FAs 1 (4,43 × 108 UFC / ml) em pH 7. Resistência ao arsênico em condições ótimas para as cepas bacterianas FAs 1, FAs 4 e FAs 9 apresentaram crescimento máximo na concentração de 80 mM de arsenito. Estes isolados bacterianos não mostraram capacidade redoxpara oxidar o arsenito As III para arseniar como V. No entanto, os isolados bacterianos FAs 1, FAs 4 e FAs 9 foram capazes de acumular arsênico 39,16, 148 e 125 µ g / L no 4º, 3º e 5º dia de incubação, respectivamente. Os isolados FAs 1, FAs 4 e FAs 9 foram identificados como bastonetes gram-negativos não endoscópicos. No futuro, esses novos isolados possuem um grande potencial no campo da biotecnologia, já que a biorremediação de solo e água contaminados com arsênico pode ser feita empregando-se bactérias que acumulam arsênico, o que é um método ecologicamente correto e econômico.

Contamination of groundwater by inorganic arsenic is one of the major public-health problems in Bangladesh. This cross-sectional study was conducted (a) to evaluate the quality of life (QOL) and mental health status of arsenic-affected patients and (b) to identify the factors associated with the QOL. Of 1,456 individuals, 521 (35.78%) were selected as case and control participants, using a systematic random-sampling method. The selection criteria for cases (n=259) included presence of at least one of the following: melanosis, leucomelanosis on at least 10% of the body, or keratosis on the hands or feet. Control (nonpatient) participants (n=262) were selected from the same villages by matching age (±5 years) and gender. The Bangladeshi version of the WHOQOL-BREF was used for assessing the QOL, and the self-reporting questionnaire (SRQ) was used for assessing the general mental health status. Data were analyzed using Student’s t-test and analysis of covariance (ANCOVA), and the WHOQOL-BREF and SRQ scores between the patients and the non-patients were compared. The mean scores of QOL were significantly lower in the patients than those in the non-patients of both the sexes. Moreover, the mental health status of the arsenic-affected patients (mean score for males=8.4 and females=10.3) showed greater disturbances than those of the non-patients (mean score for males=5.2 and females=6.1) of both the sexes. The results of multiple regression analysis revealed that the factors potentially contributing to the lower QOL scores included: being an arsenic-affected patient, having lower age, and having lower annual income. Based on the findings, it is concluded that the QOL and mental health status of the arsenic-affected patients were significantly lower than those of the non-patients in Bangladesh. Appropriate interventions are necessary to improve the well-being of the patients.

Arsenic contamination of tubewell water is a major public-health problem in Bangladesh. In the recent years, the use of shallow and deep tubewell water for irrigation and the use of excess amount of cheap fertilizers and pesticides containing cadmium pose a serious threat of contamination of arsenic and cadmium in food. In an exploratory study, arsenic and cadmium were measured in foods from Matlab, a rural area in Bangladesh, that is extensively affected by arsenic and the economy is agriculture-based. Raw and cooked food samples were collected from village homes (households, n=13) and analyzed to quantify concentrations of arsenic and cadmium using atomic absorption spectrophotometry. Washing rice with water before cooking reduced the concentration of arsenic in raw rice by 13-15%. Rice, when cooked with excess water discarded, showed a significant decrease in arsenic concentration compared to that cooked without discarding the water (p<0.001). In contrast, concentration of cadmium did not decrease in cooked rice after discarding water. Cooked rice with discarded water had significantly lower concentration of arsenic compared to raw rice (p=0.002). Raw rice had higher concentration of arsenic compared to raw vegetables (p<0.001); however, no such difference was found for cadmium. Compared to raw vegetables (e.g. arum), concentration of arsenic increased significantly (p=0.024) when cooked with arsenic-contaminated water. Thus, the practice of discarding excess water while cooking rice reduces the concentration of arsenic but not of cadmium in cooked rice. However, water generally not discarded when cooking vegetables to avoid loss of micronutrients consequently retains arsenic. The results suggest that arsenic and cadmium have entered the food-chain of Bangladesh, and the cooking practices influence the concentration of arsenic but not of cadmium in cooked food.

Technical and social performances of an arsenic-removal technology—the sono arsenic filter—in rural areas of Bangladesh were investigated. Results of arsenic field-test showed that filtered water met the Bangladesh standard (<50 μg/L) after two years of continuous use. A questionnaire was administrated among 198 sono arsenic filter-user and 230 non-user families. Seventy-two percent of filters (n=198) were working at the time of the survey. Another 28% of the filters were abandoned due to breakage. The abandonment percentage (28%) was lower than other mitigation options currently implemented in Bangladesh. Households were reluctant to repair the broken filters on their own. High cost, problems with maintenance of filters, weak sludge-disposal guidance, and slow flow rate were the other demerits of the filter. These results indicate that the implementation approaches of the sono arsenic filter suffered from lack of ownership and long-term sustainability. Continuous use of arsenic-contaminated tubewells by the non-user households demonstrated the lack of alternative water supply in the survey area. Willingness of households to pay (about 30%) and preference of household filter (50%) suggest the need to develop a low-cost household arsenic filter. Development of community-based organization would be also necessary to implement a long-term, sustainable plan for household-based technology.

RESUMO

Arsenic is known as a toxic metalloid, which mainly exists in inorganic forms such as arsenite and arsenate in the natural environment. A number of microorganisms have evolved different resistant mechanisms for arsenic detoxification to cope with the widespread distribution of the poisonous arsenic. Four distinct microbial arsenic-resistant mechanisms have been described including As(III) oxidation, cytoplasmic As(V) reduction, respiratory As(V) reduction, and As(III) methylation. These mechanisms confer arsenic resistance in microorganisms that play important roles in the transformation and geological cycle of arsenic. This review mainly focuses on the researches on these molecular mechanisms and potential application for environmental arsenic bioremediation using microorganisms.
