RESUMO

A bacterial community has a central role in nutrient cycle in aquatic habitats. Therefore, it is important to analyze how this community is distributed throughout different locations. Thirty-six different sites in the upper Paraná River floodplain were surveyed to determine the influence of environmental variable in bacterial community composition. The sites are classified as rivers, channels, and floodplain lakes connected or unconnected to the main river channel. The bacterial community structure was analyzed by fluorescent in situ hybridization (FISH) technique, based on frequency of the main domains Bacteria and Archaea, and subdivisions of the phylum Proteobacteria (Alpha-proteobacteria, Beta-proteobacteria, Gamma-proteobacteria) and the Cytophaga-Flavobacterium cluster. It has been demonstrated that the bacterial community differed in density and frequency of the studied groups. And these differences responded to distinct characteristics of the three main rivers of the floodplain as well as to the classification of the environments found in this floodplain. We conclude that dissimilarities in the bacterial community structure are related to environmental heterogeneity, and the limnological variables that most predicted bacterial communities in the upper Paraná River floodplain was total and ammoniacal nitrogen, orthophosphate and chlorophyll-a.

Arraial do Cabo is where upwelling occurs more intensively on the Brazilian coast. Although it is a protection area it suffers anthropogenic pressure such as harbor activities and sporadic sewage emissions. Short-time studies showed a high variability of bacterial production (BP) in this region but none of them evaluated BP during long periods in a large spatial scale including stations under different natural (upwelling and cold fronts) and anthropogenic pressures. During 2006, we sampled surface waters 10 times (5 in upwelling and 5 in subsidence periods) in 8 stations and we measured BP, temperature as well as the concentrations of inorganic nutrients, pigments and particulate organic matter (POM). BP was up to 400 times higher when sewage emissions were observed visually and it had a positive correlation with ammonia concentrations. Therefore, in 2007, we did two samples (each during upwelling and subsidence periods) during sewage emissions in five stations under different anthropogenic pressure and we also measured particles abundance by flow cytometry. The 12 samples in the most impacted area confirmed that BP was highest when ammonia was higher than 2 μM, also reporting the highest concentrations of chlorophyll a and suspended particles. However, considering all measured variables, upwelling was the main disturbing factor but the pressure of fronts should not be neglected since it had consequences in the auto-heterotrophic coupling, increasing the concentrations of non fluorescent particles and POM. Stations clustered in function of natural and anthropogenic pressures degrees and both determined the temporal-spatial variability.

Dissolved organic carbon (DOC) is nowadays recognized as the main substrate and source of energy for aquatic microbial community. The great part of available organic carbon for bacterioplankton might be formed after photolytic degradation of humic material, which constitutes the major part of DOC in almost all natural waters. The effects of DOC photo-degradation were evaluated, as was its utilization by bacterioplankton, through a two-step experiment, one involving photo-degradation of DOC and the other bacterial growth on the photo-degraded substrate. Photo-degradation was responsible for the consumption of 19% of DOC, reduced SUVA254, an increase in the E2/E3 and E3/E4 ratios, in addition to modifications in the fluorescence spectra that indicated a rise in the labile fraction of DOC. However, these alterations on DOC were not reflected in differences in bacterioplankton growth, as shown by the fact that there were no significant differences in density, biomass, bacterial production, bacterial respiration and bacterial growth efficiency between treatment and control.


O carbono orgânico dissolvido (COD) é reconhecido atualmente como o principal substrato e fonte de energia para a comunidade microbiana aquática. Grande parte do COD biodisponível para o bacterioplâncton pode ser gerada após degradação fotolítica do material húmico dissolvido, que constitui a maior parte do carbono orgânico total. Neste trabalho foram avaliados os efeitos da fotodegração no COD de um ambiente húmico e a sua posterior utilização pelo bacterioplâncton, por meio de um experimento em duas etapas, uma de fotodegradação do COD e outra de crescimento microbiano sobre o substrato fotodegradado. A fotodegradação causou consumo de 19% do COD, diminuição na SUVA254, aumento nas razões E2/E3 e E3/E4 e modificações nos espectros de fluorescência apontando aumento da fração lábil. No entanto, as alterações no COD não se refletiram em diferenças no crescimento do bacterioplâncton, não havendo diferenças significativas com relação à densidade, biomassa, produção, respiração e eficiência de crescimento bacteriano.

Con el propósito de evaluar la participación de bacterias heterotróficas en el flujo de carbono en la cadena trófica de un lago de inundación amazónico, recolectas mensuales de estos microorganismos fueron realizadas durante el año hidrológico de diciembre de 2007 hasta noviembre de 2008, en las regiones litoral, pelágica y de macrófitas acuáticas del lago Catalão, en la Amazonia central brasilera. Las bacterias fueron multiplicadas in vitro, usando como sustrato carbono orgánico disuelto (COD) del lago en cada una de las regiones estudiadas, para posterior análisis de la abundancia de isotopos estables de carbono y nitrógeno. Estos datos fueron confrontados con los valores de COD de los cuatro periodos limnológicos del lago (seca, inundación, llena y bajada de aguas). En general se encontró que la fuente principal de carbono de las bacterias heterotróficas fue aquella de origen C4, que presentó un aporte mínimo de 75%, en la medida en que el d13C de las bacterias presentó valor medio de -17,72‰ ± 2,25. Confrontando ese valor, con el d13C del zooplancton en el mismo período (- 33,04‰ ± 3,81) se concluye que el aporte de las bacterias heterotróficas al flujo de carbono hacia niveles tróficos superiores en el lago estudiado es mínimo.


In order to evaluate the role of heterotrophic bacteria on carbon flow in food chains of an Amazonian floodplain lake, monthly collections of these organisms were made during the hydrological year from December 2007 to November 2008. Littoral, pelagic, and aquatic macrophyte regions of the Catalão Lake in Central Amazonia were sampled and bacteria were multiplied in vitro, using dissolved organic carbon (COD) of each one of the regions studied as a substrate. The bacterial biomass obtained was used for stable isotope analyses of carbon and nitrogen. These data were confronted with COD values of the four hydrological periods of the lake (dry, rising, flood and fall). In general, it was found that the main source of carbon for heterotrophic bacteria was that of C4 origin, which presented a minimum contribution of 75% of bacterial biomass, to the extent that the bacteria d13C average value was -17.72‰ ± 2.25. Comparing this value with the d13C of zooplankton in the same period (-33.04‰ ± 3.81) permit concludes that the contribution of heterotrophic bacteria in the carbon flow to higher trophic levels in the Catalão lake is minimal.

In this work, the density of bacterioplankton, bacterial biomass and environmental variables were monitored in two seasons (summer and winter), two times each month (spring tide and neap tide sampling), over a 12 h period, comprising a tidal cycle (semidiurnal), from subsurface and bottom waters, in a fixed station in the Estuarine Turbidity Maximum Zone (ETMZ) of Paranaguá Bay, Brazil. The data were treated with multivariate analyses methods in order to indentify the key controlling factors of the bacterial community dynamics. The microbial community seemed to be structured by a close relationship with the nutrients concentration, mainly by total phosphorous and nitrate. Regardless of variations throughout the tidal cycles, free-living bacteria had a dominant role on the Paranaguá's Bay ETMZ.

Since the introduction of the Microbial Loop concept, many studies aimed to explain the role of bacterioplankton and dissolved organic carbon (DOC) in aquatic ecosystems. Paraná River floodplain system is a very complex environment where these subjects were little explored. The aim of this work was to characterize bacterial community in terms of density, biomass and biovolume in some water bodies of this floodplain and to verify its temporal variation and its relation with some limnological variables, including some indicators of DOC quality, obtained through Ultraviolet-visible (UV-VIS) and fluorescence spectroscopic analysis. Bacterial density, biomass and biovolume are similar to those from other freshwater environments and both density and biomass were higher in the period with less rain. The limnological and spectroscopic features that showed any relation with bacterioplankton were the concentrations of N-NH4 and P-PO4, water transparency, and some indicators of DOC quality and origin. The analysis of these relations showed a possible competition between bacterioplankton and phytoplankton for inorganic nutrients and that the DOC used by bacterioplankton is labile and probably from aquatic macrophytes.

The temporal and spatial fluctuations of Bacterioplankton in a fluvial-lagunar system of a tropical region (Pitimbu River and Jiqui Lake, RN) were studied during the dry and the rainy periods. The bacterial abundance varied from 2.67 to 5.1 Cells10(7)mL-1 and did not show a typical temporal variation, presenting only small oscillations between the rainy and the dry periods. The bacterial biomass varied from 123 µgC L-1 to 269 µgC L-1 in the sampling sites and the average cellular volume varied from 0.12 to 0.54µm³, showing a predominance of the rods. The temperature showed a positive correlation with the cellular volume of the rods (R=0.55; p=0.02) and vibrio (R=0.53; p=0.03). Significant spatial differences of biomass (Mann Whitney: p=0.01) and cellular volume of the morphotypes (Mann Whitney: p=0.003) were found between the sampling sites. The strong positive correlations of the water temperature and oxygen with bacterioplankton showed a probable high bacterial activity in this system.


A variação temporal e espacial do bacterioplâncton em um sistema fluvial-lagunar de região tropical foi estudada em períodos seco e chuvoso. As médias da abundância bacteriana variaram de 2,67 a 5,1 x 10(7) e não exibiram uma variação temporal marcante, tendo apresentado apenas pequenas oscilações entre os períodos chuvoso e seco. A biomassa bacteriana variou de 123 µg C L-1 a 269 µg C L-1 entre os locais de coleta e o volume celular médio de 0,12µm³ a 0,54µm³, ocorrendo predominância de bacilos. A temperatura mostrou correlação positiva com o volume celular de bacilos (R=0,55; p=0,02) e de vibriões (R=0,53; p=0,03). Foram encontradas diferenças espaciais significativas de biomassa (Mann Whitney: p=0,01) e volume celular dos morfotipos (Mann Whitney: p= 0,003), entre os locais de coleta. As fortes correlações positivas da temperatura da água e do oxigênio, com o bacterioplâncton, são sugestivas de uma provavelmente elevada atividade bacteriana nesse sistema.

We measured the vertical and seasonal distribution of picoplankton (0.2-2 µm) and nanoplankton (2-20 µm) in the photic layer of Cuban southern oceanic and coastal waters. The concentration of the different fractions was estimated by epifluorescence microscopy. Heterotrophic components from the different fractions showed higher vertical stratification in the oceanic station in comparison to the coastal one. The autotrophic components showed an irregular vertical distribution pattern, both in coastal and oceanic stations. In all the analyzed stations, the heterotrophic bacteria showed an inverse correlation with the autotrophic (r= -0.98), and the heterotrophic nanoplankton (r= -0.96). Auto and heterotrophic nanoplankton probably regulate bacteria abundance by predation, although autotrophic nanoplankton may represent a source of organic matter for microorganisms. Rev. Biol. Determinamos la distribución vertical y estacional del picoplancton (0.2-2 µm) y el nanoplancton (2-20 µm) en la capa fótica de las aguas oceánicas y costeras al sur de Cuba. La concentración de las diferentes fracciones fue estimada mediante microscopía de epifluorescencia. Los componentes heterotróficos de las diferentes fracciones mostraron una elevada estratificación vertical en la estación oceánica en comparación con la estación costera. Por otro lado, los componentes autotróficos presentaron un patrón de distribución vertical irregular tanto en la estación costera como en la oceánica. En los análisis realizados las bacterias heterótrofas mostraron una correlación inversa con el nanoplancton autótrofo (r= -0.98), y con el nanoplancton heterótrofo (r= -0.96). Estos resultados sugieren que el nanoplancton (autótrofo y heterótrofo) probablemente regula la abundancia de bacterias mediante la depredación, además que el nanoplancton autótrofo pudiera representar una fuente de materia orgánica para los microorganismos.

We analyzed relations among phytoplankton and total bacterioplankton fractions in three lentic ecosystems (Neusa and Prado dams, and Fúquene lagoon) with different physicochemical characteristics, in the Andes of Colombia. Samplings were made in three sites of each water body during three surveys. Neusa dam (meso to oligotrophic) had the lowest bacterial concentration; Prado dam (eutrophic) had a high bacterial and algal abundance, and the Fúquene lagoon (mesotrophic) had lower concentrations of phytoplankton but a high relative concentration of bacteria, probably because of its particular conditions: high organic matter and low nutrient levels in the water. There was a negative correlation of total bacterioplankton with the phytoplankton (Pearson = -0.4479, p = 0.019, n=27) and a positive correlation between phytoplankton and heterotrophic bacteria (Pearson = 0.3866, p = 0.062, n=24) and between total bacterioplankton and DBOs (Pearson = 0.4088, p = 0.034, n=27). Apparently, total bacterioplankton and phytoplankton were not coupling, but cultivable bacteria and the phytoplankton had some degree of relationship.

The aims of this research were to determine the main limiting nutrient to bacterial growth in Imboassica lagoon, southeastern Brazil, to estimate the percentage of dissolved organic carbon (DOC) available for bacterial growth, and to determine the bacterial growth efficiency (BGE) of natural assemblages. Bacterial growth and DOC consumption were determined in batch culture experiments, in which water samples were supplemented with nitrogen and phosphorus together or separately, or incubated without nutrient additions. When added together, N and P stimulated higher bacterial growth rates and production, as well as higher DOC consumption. The BGEs and DOC consumption rates were strongly dependent on the method used to determine bacterial production. The BGE ranged from 11 to 72%. However, only a minor fraction of bulk DOC was consumed by the planktonic bacteria (from 0.7 to 3.4%). The results suggest that low availability of phosphorus and nitrogen coupled with excess organic carbon was the main factor responsible for the relatively low bacterial utilization of DOC in Imboassica lagoon.


Os objetivos desta pesquisa foram: determinar o principal nutriente limitante ao crescimento bacteriano na lagoa Imboassica, estimar a porcentagem de Carbono Orgânico Dissolvido (COD) disponível para o crescimento bacteriano e determinar a Eficiência de Crescimento Bacteriano (ECB) da comunidade bacteriana. O crescimento bacteriano e o consumo de COD foram avaliados em experimentos de culturas de diluição, nos quais nitrogênio e fósforo foram adicionados às amostras de água, juntos ou separadamente, e um controle foi preparado sem adições de nutrientes. Quando adicionados juntos, N e P estimularam um maior crescimento e produção bacterianos, assim como maiores taxas de consumo de COD. ECB e taxas de consumo de COD foram fortemente dependentes do método utilizado para determinar a produção bacteriana. ECB variou de 11% a 72%, porém apenas uma pequena fração do COD total foi consumida pelas bactérias planctônicas (de 0,7% a 3,4%). Sugere-se que as baixas disponibilidades de fósforo e nitrogênio são os principais fatores responsáveis pela baixa utilização de COD pelas bactérias na lagoa Imboassica.

Rodrigo de Freitas lagoon is an eutrophic aquatic environment. The waters from the lagoon are released to the sea at Ipanema and Leblon beaches, through Jardim de Alah channel. In this work, the influence of these waters on the bacterial communities of these beaches was investigated. Eleven sampling stations were set between the lagoon and the beaches, and the samples were analyzed by molecular and microbiological parameters. PCR-DGGE of the DNA extracted from the samples was performed using rpoB primers. Preliminary results indicate that all used approaches could reveal the influence of the lagoon on the beaches bacterial communities.


A lagoa Rodrigo de Freitas é um ambiente aquático eutrofizado, cujas águas são lançadas ao mar nas praias de Ipanema e Leblon através do canal do Jardim de Alah. Nesse trabalho, foi estudada a influência desse aporte na comunidade bacteriana dessas praias. Para isso coletou-se água de onze estações distribuídas entre a lagoa e as praias. Essas amostras foram analisadas quanto a parâmetros moleculares e microbiológicos. Foi realizado também PCR-DGGE utilizando-se iniciadores para o gene rpoB, a partir de DNA extraído das amostras de água coletadas. Resultados preliminares mostram que a influência da lagoa na comunidade bacteriana das praias pode ser verificada por todas as abordagens.
